Effects of Chrysanthemum indicum L. Extract on the Function of Osteoblastic MC3T3-E1 Cells under Oxidative Stress Induced by Hydrogen PeroxideJee (감국(Chrysanthemum indicum L.) 추출물이 H2O2로 유도한 산화적 스트레스에서 MC3T3-E1 조골세포 기능에 미치는 영향)

  • Chrysanthemum indicum L. (Asteraceae) is a traditional herbal medicine that has been used for the treatment of inflammation, hypertension, and respiratory diseases due to its strong antagonistic activity against inflammatory cytokines. The effects of Chrysanthemum indicum L. Extract (CIE) for increasing cell growth, alkaline phosphatase (ALP) activity, and collagen content were totally inhibited, suggesting that the effect of CIE might be partly involved with estrogen activity. Furthermore, the protective effects of CIE on the response of osteoblasts to oxidative stress were evaluated. Osteoblastic MC3T3-E1 cells were incubated with hydrogen peroxide and/or CIE, and markers of osteoblast function and oxidative damage were examined. CIE significantly increased cell survival, ALP activity, and calcium deposition, and decreased the production of Reactive Oxygen Species (ROS) and Tumor Necrosis Factor-${\alpha}$ (TNF-${\alpha}$) in osteoblasts. Taken together, these results indicate that the enhancement of osteoblast function by CIE may prevent osteoporosis and inflammatory bone diseases.

Effect of Plant Crowth Regulators on Production of Good Quality Plant for Korean Gerbera Lines (국내 육성 거베라의 우량묘 생산에 미치는 식물생장조절제의 영향)

  • This study was carried out to obtain information about the valuable combination and concentration to produce good plants of cut gerbera flowers of 19 domestically - bred gerbera cultivars. BA and kinetin combination is more effective than NAA and BA combination to produce good plants and mass propagation of domestically - bredgerbera cultivars. Normally almost of 19 cultivars, mass propagation was more effective on the medium containing 1.0 mg/L BA + 0.5, 1.0 mg/L kinetin. But some cultivars, 'Sunnyeo', 'Oksaem','Piny', and 'Pink Light', vitrified plants were induced on MS medium high level of BA concentration(BA 1.0 mg/L), in comparison with those on the medium with low level of BA(0.1 mg/L).Fresh and dry weight, more effective on the medium containing BA 0.5 mg/L . Kinetin 0.1, 0.5 mg/L. Anatomical investigation of vitrified leaf, stomata of vitrified leaves were circular and inflated, where-as those of normal leaves.

Heavy Metal Contents and Antioxidant Activity and Cytotoxic Effect of Red Sea Bream (Pagrus major): Comparative Studies in Domestic and Imported Red Sea Bream (Pagrus major) (국내산 및 수입산 참돔의 중금속 함량 및 항산화 활성과 세포독성 효과 비교)

  • This study compared the heavy metal contents and the effects of extracts from domestic and imported red sea bream on the antioxidant activity and cytotoxicity of human cancer cell lines. The antioxidant activity was measured using the fluorescently sensitive dye, 2’-7’ dichlorofluorescein-diacetate (DCFH-DA), and antiproliferative activity against AGS human gastric adenocarcinoma and HT-29 human colon cancer cell lines, which was determined by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Domestic red sea bream had a higher mercury content when compared to imported red sea bream, but there was no significant difference in the lead content. Treatments with acetone/methylene chloride (A+M) and methanol (MeOH) extracts from domestic and imported red sea bream dose-dependently decreased the H2O2 induced ROS production, compared to the control. The cell viability showed that treatments with the A+M and MeOH extracts had cytotoxicity in the growth of AGS and HT-29 cancer cells. In the case of AGS, the extracts from the domestic red sea bream were higher in inhibiting cancer cell growth, compared to imported red sea bream. Our results demonstrate that the heavy metal contents of domestic and imported red sea bream were below the limit of the Food Code of Korea. The results of the biological activities indicate that the antioxidant activity of extracts from imported red sea bream was more effective, while the extracts from the domestic red sea bream were stronger in cytotoxic activity.

Fabrication of Electrospun Juniperus Chinensis Extracts loaded PVA Nanofibers (향나무 추출물을 함유하는 PVA 나노섬유 제조)

  • Electrospinning is a simple and effective process for producing nanofiber with diameter range from nanometers to micrometers which have high specific surface area. Hence, medicated nanofibers can be readily fabricated using a solution containing a mixture of a plant-extracts and a polymer. It has proved that Juniperus Chinensis can be effectively used for the prevention of UV and SLS-induced advers skin reaction such as radical production, inflammation and skin cell damage. It also found that Juniperus Chinensis has efficient ingredient of antifungal activity and house dust mite repellent effect. The fabrication of PVA nanofibers containing Juniperus Chinensis extracts by electrospinning has been studied. PVA/Juniperus Chinensis extracts composite nanofibers were produced at different Juniperus Chinensis concentrations (0.25, 0.5, 1.5 wt. %). The parameters of electrospinning including polymer contents, voltage and tip-to-collector distance (TCD) were optimized for fabrication process. The study show that 12 wt. % PVA, 10kV applied voltage and TCD 10~20 cm are the best condition to obtain uniform PVA/Juniperus Chinensis extracts composite nanofibers. Morphologies of the electrospun composite nanofiber were observed by using a field emission scanning electron microscope. It has been found that the average diameters of fibers increased by the adding of Juniperus Chinensis extracts. As the results, PVA/Juniperus Chinensis extracts composite nanofibers having a diameter in the range from 310~360 nm were successfully prepared via an electrospinning.

  • We are currently conducting studies on culturing and biocompatibility assessment of various cells such as neural stem cells and induced pluripotent stem cells(IPS cells) on carbon nanotube (CNT), on nerve regeneration electrodes, and on silicon wafers with a focus on developing nerve integrated CNT based bio devices for interfacing with living organisms, in order to develop brain-machine interfaces (BMI). In addition, we are carried out the chemical modification of carbon nanotube (mainly SWCNTs)-based bio-nanosensors by the plasma ion irradiation (plasma activation) method, and provide a characteristic evaluation of a bio-nanosensor using bovine serum albumin (BSA)/anti-BSA binding and oligonucleotide hybridization. On the other hand, the researches in the case of "novel plasma" have been widely conducted in the fields of chemistry, solid physics, and nanomaterial science. From the above-mentioned background, we are conducting basic experiments on direct irradiation of body tissues and cells using a micro-spot atmospheric pressure plasma source. The device is a coaxial structure having a tungsten wire installed inside a glass capillary, and a grounded ring electrode wrapped on the outside. The conditions of plasma generation are as follows: applied voltage: 5-9 kV, frequency: 1-3 kHz, helium (He) gas flow: 1-1.5 L/min, and plasma irradiation time: 1-300 sec. The experiment was conducted by preparing a culture medium containing mouse fibroblasts (NIH3T3) on a culture dish. A culture dish irradiated with plasma was introduced into a $CO_2$-incubator. The small animals used in the experiment involving plasma irradiation into living tissue were rat, rabbit, and pick and are deeply anesthetized with the gas anesthesia. According to the dependency of cell numbers against the plasma irradiation time, when only He gas was flowed, the growth of cells was inhibited as the floatation of cells caused by gas agitation inside the culture was promoted. On the other hand, there was no floatation of cells and healthy growth was observed when plasma was irradiated. Furthermore, in an experiment testing the effects of plasma irradiation on rats that were artificially given burn wounds, no evidence of electric shock injuries was found in the irradiated areas. In fact, the observed evidence of healing and improvements of the burn wounds suggested the presence of healing effects due to the growth factors in the tissues. Therefore, it appears that the interaction due to ion/radicalcollisions causes a substantial effect on the proliferation of growth factors such as epidermal growth factor (EGF), nerve growth factor (NGF), and transforming growth factor (TGF) that are present in the cells.

Characterization of anti-inflammatory effect of soybean septapeptide and its molecular mechanism (대두 septapeptide의 항염 효과 및 분자 기작 규명)

  • Activation of nuclear factor kappa B ($NF{\kappa}B$) leads to the inflammatory process. During this $NF{\kappa}B$-dependent inflammation process, inducible nitric oxide synthase (iNOS) are expressed in the inflammatory cells. Our previous data indicated that a specific septapeptide (GVAWWMY) from the soybean extract fermented by Bacillus licheniformis B1 inhibited iNOS mRNA expression and NO production in cultured macrophage cells. Our further experiments revealed that treatment of same septapeptide resulted in inhibition of LPS-induced $NF{\kappa}B$ activation by reversing degradation of $I{\kappa}B{\alpha}$, an inhibitory protein for $NF{\kappa}B$. The molecular docking indicated that the septapeptide binds to $I{\kappa}B$ kinase ${\beta}$ ($IKK{\beta}$), and thus it can inhibit phosphorylation of $I{\kappa}B{\alpha}$. Supporting this, the binding site for the septapeptide has the highest affinity (-8.7 kcal/mol) and the site was located at the kinase domain (KD) of $IKK{\beta}$, which can significantly affect the kinase activity of $IKK{\beta}$.

Cold Pasteurization of Frozen Crushed Garlics using Electron Beam Irradiation and the Stability of Bioactive Components (냉동다진마늘에 대한 전자선 살균 효과와 생리활성 성분의 안정성)

  • Recently, the market for ready-to-use vegetables has increased largely due to consumer demands, which led to the production of minimally-processed frozen crushed garlic products. This study was designed to determine the effect of electron beam irradiation (0-7 kGy) on microbial decontamination, bioactive compounds, and antimicrobial activities of frozen crushed garlic obtained from Korea and China. The microbial counts (total bacteria, yeasts & molds) were reduced from log 3-4 CFU/g to non-detectable levels as a result of irradiation at 4 kGy. Irradiation treatment at 4 kGy did not affect the amount of alliin, allicin, total pyruvate, and total thiosulfinate of crushed garlic, however, this dose induced insignificant changes in antibacterial activities against pathogenic bacteria including Bacillus subtilis, Staphylococcus aureus, Salmonella enteritidis, Vibrio parahaemolyticus, and Yersinia enterocolitica. Therefore, electron beam irradiation less than 4 kGy can be considered suitable to improve the microbial decontamination without altering the biological activity of frozen garlics.

Synthesis of Na Compounds from Sodium Concentrated Solution Using Carbonation and Cryo-crystallization (탄산화 및 저온 결정화를 통한 나트륨 농축수로부터 나트륨 화합물 합성)

  • Carbonation (step I) and cryo-crystallization (crystallization at low temperature) (step II) were performed to synthesize Na compounds from sodium concentrated solution. In the step 1, the solubility and pH of carbon dioxide (95 wt.%) affecting carbonation could be changed by the variation of reaction temperature. The step II was performed at 2 ℃ after carbonation. The injection of carbon dioxide was carried out twice for the stable production and the saturated solubility of carbonate ions in solution. Firstly, we tried to inject CO2 for controlling the solubility of CO2 by changing the reaction temperature from 35 ℃ to 10 ℃, and the second injection was aimed at 10 ℃ for inducing nucleation of Na compound through carbonation after NaCl solution addition. In the cryo-crystallization step, the crystal growth of Na compounds could be induced by slowing the carbonation rate through reaction temperature change from 10 ℃ to 2 ℃. In this study, the effect on NaOH concentration was examined and the purity of Na compound was increased when 2M NaOH was used. In addition, the synthesized Na compounds were mostly rod-shaped and consisted of sodium carbonate or sodium carbonate with monohydrate.

Effect of Electron Beam Irradiation on the Anti-allergy Activity of β-Glucan (전자선 조사가 베타글루칸의 항알레르기 활성에 미치는 영향)

  • This study evaluated the change in anti-allergy activity of ${\beta}-glucan$ by electron beam irradiation. ${\beta}-Glucan$ was irradiated at dose of 50 kGy and then orally pre-treated with electron beam irradiated and non irradiated ${\beta}-Glucan$ for 7 days. After pre-treatment, allergy was induced by injection of ovalbumin (OVA). Serum total immunoglobulin E (IgE) and OVA-specific IgE levels in the allergic mice was significantly increased but the mice pre-treated 50 kGy electron beam irradiated ${\beta}-glucan$ was significantly decreased the levels of total IgE and OVA-specific IgE, respectively. Moreover, cytokine production (interleukin-4) was also decreased in the 50 kGy electron beam irradiated ${\beta}-Glucan$ pre-treated mice. These results indicate that pre-treatment of 50 kGy electron beam irradiated ${\beta}-glucan$ may elevate the anti-allergy activity. Therefore, electron beam-irradiated ${\beta}-glucan$ could be used for nutraceutical foods in food industry.

Effect of IAA and Kinetin on Induction of Protonemal Gemmae and Gametophytes of Hypnum plumaeforme Wilson (털깃털이끼(Hypnum plumaeforme Wilson) 원사체 무성아와 배우체의 형성에 미치는 IAA와 Kinetin의 영향)

  • This study was conducted to examine the effects of IAA and kinetin on induction of gemmae and gametophytes in Hypnum plumaeforme moss during tissue culture. Explants were obtained from sterilized gametophyte tips cultured on solid basal medium containing Knop's major salts and Nitsch and Nitsch's trace elements. After culture, inoculated gametophyte tip produced protonema firstly, changed to new gametophytes after four weeks. Aseptic gametophytes were chopped and inoculated on the same media containing 0.01, 0.1, 1 and $10{\mu}M$ of IAA and kinetin. As a result, secondary protonemata as well as protonemal gemmae were formed from gametophytes. But protonemal gemmae formation was varied according to the concentration of IAA and kinetin. Lower concentration of IAA and kinetin promoted gemma formation and bud induction. Especially, $0.01{\mu}M$ of kinetin showed the highest frequency of bud and gemma production. All the materials, obtained from $0.01{\mu}M$ kinetin medium, were subcultured to media supplemented with 0.01, 0.1, 1 and $10{\mu}M$ of IAA and kinetin again to induce gametophytes. After subculture, protonema and calli were developed from secondary protonema, and induced gametophytes finally. IAA regulated induction and growth of gametophytes, and kinetin influenced gemma formation and gametophyte induction also. All aspects of development of this moss species were governed by the external growth regulators.
