Inhibition of Oxidative Stress and Enhancement of Cellular Activity by Mushroom Lectins in Arsenic Induced Carcinogenesis

  • Chronic arsenicosis is a major environmental health hazard throughout the world, including India. Animals and human beings are affected due to drinking of arsenic contaminated ground water, due to natural mineral deposits, arsenical pesticides or improperly disposed arsenical chemicals. Arsenic causes cancer with production of free radicals and reactive oxygen species (ROS) that are neutralized by an elaborate antioxidant defense system consisting of enzymes and numerous non-enzymatic antioxidants. Dietary antioxidant supplements are useful to counteract the carcinogenesis effects of arsenic. Oyster mushroom lectins can be regarded as ingredients of popular foods with biopharmaceutical properties. A variety of compounds have been isolated from mushrooms, which include polysaccharides and polysaccharopeptides with immune-enhancing effects. Lectins are beneficial in reducing arsenic toxicity due to anticarcinogenetic roles and may have therapeutic application in people suffering from chronic exposure to arsenic from natural sources, a global problem that is especially relevant to millions of people on the Indian subcontinent.

  • King Oyster(Pleurotus eryngii) is one of the most promising mushrooms produced on the domestic farms. The quality as well as quantity of King oyster is sensitively affected by micro climate factors such as temperature, relative humidity, $CO_2$ concentration, and light intensity. To safely produce high-quality King oysters year round, it is required that the environmental factors be carefully controlled by well designed structures equipped with various facilities and control systems. In this study, we are focusing on carrying out growing experiment to find out reasonable range of each environmental factor together with economic and safe structures influencing on the optimal productivity of king oyster mushroom. The optimal productivity will be evaluated by considering the quality and quantity of mushroom production, energy requirements, facility construction and management cost, etc.

  • The purpose of the cultivated crops have been changes in the aim of improving quality production. In recent years, as people's attention on health, the demand for healthy crops such as mushrooms gradually increased. Farmers use plastic greenhouse cultivation mode more and more in order to reduce the impact of outdoor environment on crop cultivation, which requires farmers to adjust the greenhouse temperature at any time. But the majority of farmers still use a thermometer to measure temperature. This paper constructs an environment that can automatically adjust the temperature, so as to measuring temperature in real time, improving the efficiency of the farm work, and reducing unnecessary labor.

  • Certain European truffles including Tuber melanosporum (Perigord black truffle) and T. aestivum(Burgundy truffle), are valuable food commodities, selling for hundreds of dollars per kilogram worldwide. These underground(hypogeous) mushrooms may become profitable agroforestry crops in the south-central United States. Native to Europe, T. melanosporum and T. aestivum are being cultivated to a limited extent and with mixed results in New Zealand and the United States. Tuber species grow mutualistically as ectomycorrhizae with the roots of compatible host trees including white and evergreen oaks(Quercus spp.) and hazels(Corylus spp.). Tuber melanosporum and T. aestivum have environmental requirements compatible with conductive sites in the south central US. Climatic and edaphic conditions for cultivating T. melanosporum are more constraining, but T. melanosporum also commands a much higher price.

Secretory Production of the Hericium erinaceus Laccase from Saccharomyces cerevisiae

  • Mushroom laccases play a crucial role in lignin depolymerization, one of the most critical challenges in lignin utilization. Importantly, laccases can utilize a wide range of substrates, such as toxicants and antibiotics. This study isolated a novel laccase, named HeLac4c, from endophytic white-rot fungi Hericium erinaceus mushrooms. The cDNAs for this enzyme were 1569 bp in length and encoded a protein of 523 amino acids, including a 20 amino-acid signal peptide. Active extracellular production of glycosylated laccases from Saccharomyces cerevisiae was successfully achieved by selecting an optimal translational fusion partner. We observed that 5 and 10 mM Ca2+, Zn2+, and K+ increased laccase activity, whereas 5 mM Fe2+ and Al3+ inhibited laccase activity. The laccase activity was inhibited by the addition of low concentrations of sodium azide and ⳑ-cysteine. The optimal pH for the 2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt was 4.4. Guaiacylglycerol-β-guaiacyl ether, a lignin model compound, was polymerized by the HeLac4c enzyme. These results indicated that HeLac4c is a novel oxidase biocatalyst for the bioconversion of lignin into value-added products for environmental biotechnological applications.

In related to the methods of mushroom cultivation, the recent technologies apply the new technology such as sensors and IT convergence services. And then cultivating mushroom is managed effectively. Novel Antihypertension Mechanism of 𝛽-Glucan by Corin and ANP-Mediated Natriuresis in Mice

  • Many of the 𝛽-glucans are known to have antihypertensive activities, but, except for angiotensin-converting enzyme II inhibition, the underlying mechanisms remain unclear. Corin is an atrial natriuretic peptide (ANP)-converting enzyme. Activated corin cleaves pro-ANP to ANP, which regulates water-sodium balance and lowers blood pressure. Here, we reported a novel antihypertensive mechanism of 𝛽-glucans, involved with corin and ANP in mice. We showed that multiple oral administrations of 𝛽-glucan induced the expression of corin and ANP, and also increased natriuresis in mice. Microarray analysis showed that corin gene expression was only upregulated in mice liver by multiple, not single, oral administrations of the 𝛽-glucan fraction of Phellinus baumii (BGF). Corin was induced in liver and kidney tissues by the 𝛽-glucans from zymosan and barley, as well as by BGF. In addition to P. baumii, 𝛽-glucans from two other mushrooms, Phellinus linteus and Ganoderma lucidum, also induced corin mRNA expression in mouse liver. ELISA immunoassays showed that ANP production was increased in liver tissue by all the 𝛽-glucans tested, but not in the heart and kidney. Urinary sodium excretion was significantly increased by treatment with 𝛽-glucans in the order of BGF, zymosan, and barley, both in 1% normal and 10% high-sodium diets. In conclusion, we found that the oral administration of 𝛽-glucans could induce corin expression, ANP production, and sodium excretion in mice. Our findings will be helpful for investigations of 𝛽-glucans in corin and ANP-related fields, including blood pressure, salt-water balance, and circulation.

Log Cultivation of the Birch Fungus Piptoporus betulinus using Betula davurica (물박달나무를 이용한 자작나무버섯의 원목재배)

  • A birch fungus, Piptoporus betulinus, is judged the mushroom with economical value that produces the fruiting body on Betula trees, but the mushroom rarely occurs in Korean birch forest. So, the fungus was applied to log cultivation for the production of fruiting body on Betula davurica. The fungus produce the fruiting bodies in the early August of the next year after inoculation in April and its mushroom matured in October. The mushroom produced 212 g to 1,298 g fresh weight in one or two mushrooms per a log. Thus, it is considered that log cultivation of the fungus has a potential to mass production of fruiting body and is expected to introduce to more researches to develop the use of this mushroom.

  • To increase production of Tricholoma matsutake, experiments were carried out in a mountain located in Keumma 2-ri, Joocheon-myun, Yongwol-gun, Kangwon-do, Korea during the period from April to November 1979. The area of the experiment showed pH 6.55 of soil acidity and the rock was found to be granite. Among its vegetation, the major growths were Pinus densiflora Siebold et Zuccarini, Rhododendron mucronulatum Turcz., Rhododendron schlippenhachii Max., Lespedeza bicolor and Pteridium aquilinum (Linne) Kuhn var. japonicum Nakai. Of two equally divided areas of A and B in which pine mushrooms grew, artificial methods such as thinning the upper portion of land raking and water-spraying were applied to area B only, where 21% increase of the production was obtained as compared with the control area A.

The Production of Mushroom Mycelium Using Rancid Oil of Fried Chicken (통닭튀김 산패유를 이용한 버섯균사체의 생산)

  • The conditions for mycelial production of mushroom rising rancid oil of fried chicken were investigated. Pleurotus ostreatus Nonggi 202 which showed the greatest mycelial growth among mushrooms was chosen for the investigation. The optimum temperature and pH for mycelial growth were $25^{\circ}C$ and 5.5 to 6.5, respectively. Tryptone was better effective than any other nitrogen sources on the mycelial growth. The mycelial growth was enhanced with addition of 0.3% $KH_2PO_4$. and 0.02% $MgSO_4$, respectively. Among the vitamins, thiamine was the most effective on the mycelial growth.
