Production of the Wild Entomopathogenic Fungi, Cordyceps militaris, in the Silkworm, Bombyx mori

  • The wild entomopathogenic fungi, Cordyceps militaris, were collected at the Whawang mountain, Korea. The pupae of the silkworm, Bombyx mori, were used as infecting hosts for the production of the silkworm-mili-taris dongchunhacho, silkworm vegetable wasps and plant worms with C. militaris. Three inoculation methods in terms of injection, spray and immersion were tested against the silkworm pupae. The three inocu1ation methods revealed 100% infectivity to the silkworm pupae tested. Of the three inoculation methods, the injection method was highly effective in the reduction of the period required for the endosclerotium and the completion of fruiting body formation. These results indicate that the silkworm pupae are very effective host insects for the production of C. militaris.

Effect of Salinity on Orobanche cernua Seed Germination

  • Seeds of broomrape (Orobanche cernua) were exposed to 0, 25, 50, 75, and 100 mM NaCl solutions during their preconditioning period (14 days of moisture) under laboratory conditions and induced to germinate by synthetic germination stimulant (GR24). There was significant reduction in seed germination with increased salt concentration as shown in 35.2, 32.5, 23.6, 14.3, and 9.2% germination, respectively. Exposure of Orobanche cernua seeds to 0.0, 1.0, 1.25, and 1.5 M levels of NaCl for 9 hours resulted in 29.4, 21.3, 20.5, and 17.4% germination, respectively. Water preconditioned seeds showed Heavier protein profile bands of 6.5-14.2 KDa than those of dry seeds. Seeds treated with 0.75 M NaCl showed profile similar with that of water preconditioned ones, plus an extra band at 29-36 KDa. The protein profiles of 1.0 and 1.5 M NaCl treated seeds showed weaker bands with the absence of 29-36 KDa band.

An Analysis of the Impact of US Beef Import Tariff Rate Changes on the Korean Beef Cattle Market (미국산 쇠고기 수입관세율 변화가 한육우 시장에 미치는 영향분석)

  • Korea-US FTA amendment became effective January 1, 2019 through several trade negotiations between the two countries. These amendments did not include changes in the agricultural sector. However, given the policy direction of the Trump administration, it is difficult to be certain that the existing Korea-US FTA on the agricultural sector will remain unchanged. This study examines the potential impact of changes in the US beef import tariff rates under the Korea-US FTA, which is progressively eliminated until 2026 using a dynamic partial equilibrium model. The modelling system is simulated with 100% decreases of tariff rates over 2020~2026 period and then compared to the baseline which is developed based on the current Korea-US FTA tariff rates. According to the scenario analyses results, 100% decreases of US beef tariff rate lowered Korean beef cattle production value up to 4.23%. Looking at this change in terms of absolute value rather than percentage, the total production value over 2020~2026 is expected to decrease by 815 billion won compared to Baseline. This reduction in production value in dynamic analysis is 67 billion won higher than the comparative static analysis.

Antifungal Effect of Tanshinone from Salvia miltiorrhiza against Disseminated Candidiasis (Tanshinone 단삼성분의 전신성 캔디다증에 대한 항균효과)

  • The aim of this present study was to investigate the antifungal effect of tanshinones isolated from Salvia miltiorrhiza against Candida ablicans, a polymorphic fungus. For the work, tanshinone IIA (TSN), cryptotanshinone (CTS), and dihydrotanshinone I (DTS) were chosen. Initially, their antifungal effect was analyzed by in-vitro susceptibility test. Data from the susceptibility test showed that while all of these three compounds had antifungal activity, DTS was the most potent. At $100{\mu}g$ DTS/ml, there was about 80% CFU (colony forming unit) reduction as compared to DTS-untreated C. albicans yeast cells (P<0.05). Thus, DTS was selected to determine its antifungal activity in a murine model of disseminated candidiasis due to C. albicans. Results showed that DTS enhanced resistance of mice against disseminated candidiasis. During the entire period of 30-day observation, 60% of DTS-given mice groups survived whereas control animals all died within 14 days (P<0.05). Moreover, DTS inhibited the hyphal production, one of the virulence factors of this fungus, from the blastoconidial form of the fungus. Therefore, the tanshinone appears to have antifungal activity specific for C. albicans infection, which could possibly be mediated by the blockage of hyphal production.

Antifungal Effect of Obacunone on Candida albicans (Obacunone 황백성분의 Candida albicans에 대한 항진균효과)

  • In the present study, we determined the antifungal effect of obacunone isolated from Phellodendri Cortex against Candida ablicans, a pathogenic fungus. The antifungal effect was analyzed by an in-vitro susceptibility test and in a murine model of disseminated candidiasis. Possible mechanism of the antifungal activity was also examined. Analyses of data resulting from the susceptibility test revealed that the compound inhibited C. albicans growth. At 25 ${\mu}g$ obacunone/ml, there was app. 45% reduction of CFUs (colony forming units) as compared to obacunone-untreated C. albicans yeast cells (P<0.01). In the murine model of disseminated candidiasis due to C. albicans, obacunone enhanced resistance of mice against disseminated candidiasis. During an entire period of 30-day observation, control animals all died within 14 days, whereas 60% of obacunone-treated mice survived (P<0.05). In addition, obacunone inhibited the hyphal production, a major virulence factor of C. albicans, from the blastoconidial form. Thus, obacunone appears to have antifungal activity for C. albicans infection. This may possibly be mediated by the blockage of hyphal production.

An Analysis of Potential Environmental Impact Reduction for Combined Sewer Overflow Project using a LCA Methodology (LCA 기법을 활용한 합류식 하수도 월류수 사업의 잠재적 환경영향 저감효과 분석)

  • In this study, LCA(Life Cycle Assessment) on 'Saemangum CSO Project' was carried out to evaluate environmental impact which occurred during the construction and operation periods and the potential environmental impact reduction was analyzed by comparing production and reduction level of pollution loads. LCA was conducted out according to the procedure of ISO14040 which suggested Goal and Scope Definition, Life Cycle Inventory Analysis, Life Cycle Impact Assessment and Interpretation. In the Goal and Scope Definition, the functional unit was 1 m3 of CSO, the system boundary was construction and operation phases, and the operation period was 20 years. For the data collection and inventory analysis, input energies and materials from civil, architecture, mechanical and electric fields are collected from design sheet but the landscape architecture field is excepted. LCIA(Life Cycle Impact Assessment) was performed following the procedure of Eco-Labelling Type III under 6 categories which were resource depletion, eutrophication, global warming, ozone-layer destruction, and photochemical oxide formation. In the result of LCA, 83.4% of environmental impact occurred in the construction phase and 16.6% in the operation phase. Especially 78% of environmental impact occurred in civil works. The Global warming category showed the highest contribution level in the environmental impact categories. For the analysis on potential environmental impact reduction, the reduction and increased of environmental impact which occurred on construction and operation phases were compared. In the case of considering only the operation phase, the result of the comparison showed that 78% of environmental impact is reduced. On the other hand, when considering both the construction and operation phases, 50% of environmental impact is increase. Therefore, this study showed that eco-friendly material and construction method should be used for reduction of environmental impact during life cycle, and it is strongly necessary to develop technology and skills to reduce environmental impact such as renewable energies.

FACTORS INFLUENCING FEED EFFICIENCY AND BACKFAT THICKNESS IN STATION TESTED BEEF BULLS

  • Records taken on 372 young beef bulls tested at the Ellerslie Bull Test Station, Alberta, Canada from November 1981 to April 1987 were analyzed to quantify the effects of age of dam, on-test age, on-test liveweight and herd of origin of bull on feed efficiency (feed/gain, kg/kg) in the test period (n = 231) and ultrasonic measurement of bakcfat thickness (mm) at the end of the test (n = 372). The reduction in $R^2$ due to each influencing factor (i.e. the variation accounted for by the factor) was used to indicate the importance of the influencing factor. Age of dam and on-test age of bull were not important factors on feed/gain and ultrasonic backfat thickness, as they accounted for less than 0.5% of the variation in feed/gain and ultrasonic backfat thickness, respectively (p > 0.1). On-test liveweight had some influence on feed/gain and ultrasonic backfat thickness, accounting for 3.5% (p < 0.01) and 0.4% (p < 0.05) of the total variation, respectively. The regression coefficients of feed/gain and ultrasonic backfat thickness on on-test liveweight were 0.016 (kg/kg)/kg and .013 mm/kg, respectively, both being significant (p < 0.05), indicating that lighter bulls entering the test were generally more efficient in feed utilization in the test period and had less backfat at the end of the test than heavier entering bulls. Herd of origin of bull accounted for a substantial amount of the total variation (> 16%) in feed/gain and ultrasonic backfat thickness (p = 0.08), indicating that a prolonged aqjustment period was needed to reduce the influence of herd of origin when assessing aggregate genetic merit of beef bulls for growth rate, feed efficiency and lean meat production using a central station performance testing program.

Influence of Cooking, Storage Period, and Re-heating on Production of Cholesterol Oxides in Chicken Meat

  • The objective of present study was to investigate the effect of cooking and their combinations with re-heating methods on the formation of cholesterol oxidation products (COPs) in stored chicken thigh meat. Pan roasting, steaming, oven grilling, charcoal grilling, and microwaving were used for cooking. Re-heating of samples was done using the same cooking methods or microwaving after 3 and 6 d of refrigerated storage. Cooking and re-heating resulted in reduction of crude fat and cholesterol contents of chicken thigh meat depending on storage period before re-heating. Cooking and storage period had no influence on the total amount of COPs. The highest total amount of COPs was observed in meat samples cooked by steaming and reheated by microwaving after 6 d of storage, which showed similar value to raw chicken meat stored for 6 days. However, different re-heating methods formed different types of COPs depending on storage period before re-heating. The high amount (p<0.05) of 25-hydroxycholesterol or ${\alpha}-epoxide$ was detected in meat samples reheated by steaming or microwaving at 3 or 6 d of storage after steamed cooking, respectively. As a result, the combination of steaming and re-heating with microwaving could increase the total amount of COPs in chicken thigh meat and different cooking/re-heating methods could form different types of COPs, even though no significant difference in the total amount of COPs depending on storage period.

Breeding and Production Research Direction for Soybean Self-Sufficiency Improvement in Korea

  • Recently, soybean production and market price are unstable, even if demand of soybean is maintained. Diverse conditions such as climate change, a decrease in rural population, and consuming affect food industry. In this situation, food security is soaring as important key-word again, and MAFRA is promoting policies for improving soybean self-sufficiency with the goal of 40% until 2030. The point of policy is to extend a production and stabilize a demand for soybean with supporting large-scale soybean paddy-field complex. According to the background, soybean breeding and production research in NICS are proceeded with three parts. First, production improvement with soybean cultivation land enlargement and high-yield cultivar development. Various growth period soybean cultivars for double cropping, irrigation management technologies in paddy field, and hyper-yield and specific-region adaptable cultivar development. Second, reduction of production expense with mechanized cultivation and digital-based field management technologies. Third, consumer-friendly and high quality soybeans with high protein cultivar for alternative protein usage and high food process-ability for soy milk, tofu, soybean sprouts, and grain usage. Each part need to be combined and advanced to improve soybean industry and soybean self-sufficiency.

Nutritive Evaluation of Some Fodder Tree Species during the Dry Season in Central Sudan

  • The potential nutritive value was studied on leaves of seven fodder trees in Central Sudan during dry season at two distinct periods, the early dry and the late. The chemical composition, mineral concentration, in vitro organic matter digestibility (IVOMD), in situ OM or nitrogen degradability and estimated metabolizable energy showed a wide variation among fodder tree species and between different periods of the dry season. Crude protein (CP) ranged from 285 to 197 g/kg DM at early dry season, with a significant reduction in late dry season. Ziziphus spina-christi and Balanites aegyptiaca showed the least reduction in CP content. The NDF, ADF and lignin were about 200, 160 and 19 g/kg DM, respectively at the early period, and significantly increased at the late period of the dry season, except for lignin of Z. spina-christi. For mineral concentration, all fodder tree leaves were rich in calcium but poor in phosphorus. In situ OM degradability significantly decreased at the late period of dry season, but values remained as high as over 600 g/kg OM. At both periods, Z. spina-christi showed the highest value, while the lowest was recorded in Acacia seyal. The IVOMD showed a similar trend to those of in situ OM degradability, except for A. seyal. The nitrogen degradability was highest in B. aegyptiaca and lowest in Z. spina-christi at both periods. A significant and positive correlation had existed between CP and IVOMD or in situ OM degradability (r=0.68, p<0.05; r=0.77, p<0.05, respectively). Also, a significant but negative correlation was found between condensed tannins and nitrogen degradability (r=-0.87, p<0.01). Results demonstrated that Z. spina-christi potentially has a good nutritive value as dry season feed or supplement, while A. seyal tends to be less promising. A. nubica and B. aegyptiaca may be a useful source for degradable protein, even though it may have a limited supply of energy to animals. A. tortilis, A. mellifera and A. ehrenbergiana may have potential value for a supplementation of energy or protein, if they were harvested in the early dry season or in wet season as preserved feed. It is highly recommended to supplement with an appropriate amount of phosphorus when these fodder trees were used.
