  • In the course of studies for anti-plaque agents, novel procyanidin structure isolated from Artocarpus heterophyllus folium was established by thiolysis and spectroscopic analysis. The chemical structure was identified for $(-)-epiafzelecin-(4{\beta}{\rightarrow}8)-afzelecin-(4{\alpha}-8)-catechin$ containing the trimeric flavan-3-ols and molecular weight was 833[M-H] by FAB-MS negative ion method. The inhibitory effect on the glucosyltransferase activity was investigated, novel compound showed complete inhibition at 1.0 mM and inhibited on the glucosyltransferase noncompetitively.

  • Phytochemical examination of the barks of Pinus densiflora Siebold et Zuccarini has led to the isolation of one phenylpropanoid, one lignan, one flavonoid, one flavan 3-ol and two procyanidins : 4-O-$\beta$-D-glucopyranosyl-p-coumaric acid (1), 2,3-dihydro-2-(4-methoxy)-7-hydroxy-3-hydroxymethyl-5-(3-hydroxy propyl)-benzofuran 3-O-$\alpha$-D-glucopyranoside (2), taxifolin 3'-O-$\beta$-D-glucopyranoside (3), (+)-catechin (4), procyanidin B1 (5) and epicatechin-($4{\beta}$-8)-catechin-($4{\alpha}$-8)-catechin (6). Among them, Compound 4, 5 and 6 showed potent anti-oxidative activities and these anti-oxidative activities were significantly different compared with ascorbic acid as positive control.

  • Effect of polyphenolic compounds from green tea leaves and surfactant micelles on lipid oxidation in corn oil-in-water emulsion (O/W) wag determined. Concentrations of polyphenolic compound and surfactant in continuous phase of O/W were measured. Particle size of O/W with 17 mM Brij 700 and 5% corn oil increased with increasing concentration of polyphenolic compound (100-200 ppm). Concentration of surfactant in the continuous phase was lower than that of control. Lipid oxidation rates, as determined by the formation of lipid hydroperoxides and headspace hexanal, in O/W emulsions containing polyphenolic compounds decreased with increasing concentration of polyphenolic compounds (100-200 ppm). Inhibition of hydroperoxide and headspace hexanal produced via lipid oxidation by polyphenolic compounds in O/W was BHT>procyanidin B3-3-O-gallate>(+)-gallocatechin >(+)-catechin.

  • 감잎으로부터 통풍치료, 미백효과, 고혈압 억제효과의 개발목적으로 9종의 flavan-3-ol 화합물을 분리하였고 기기분석에 의해 화학구조를 자혔다. 각 화합물은 (+)-catechin (+)-gallocatechin procyanidinB-l, pyrocyanidin C-1, prodelphinidin B-3, gallocatechin-(4$\alpha$$\longrightarrow$8)-catechin과 감나무 잎에서 새로운물질인pocyanidin B-7-3-0-gallate, procyanidin C-1-3'-3" -3.'S _0-trigallate, (-)-epigallocatechin-(4 $\beta$ -18)-epigallocatechin-(4$\beta$-'8)-catechin 3종류를 발견하였다. 감잎으로부터 순수 분리한 polyphenol류의ACE 저해활성측정을 실험한 결과 pocyanidin B-7-3-0-gallate는 100rM농도에서 94%의 저해효과를 나타내었으며 epigallocatechin-(4$\beta$$\longrightarrow$8)-epigallo-catechin-(4$\beta$$\longrightarrow$8)-catechia procyanidin C-1-3'-3" -3f'.-0-trigallate는 각각 90.69, 80.90% 저해를 하였다. Xanthine oxidase 저해활성측정을 조사한 결과pocyanidin B-7-3-0-galtate와 pocyanidin C-1-3'-3" -3/'S _0-trigallate 즉, gallate가 붙은 호합물에서100rM의 농도에서 66%와 63%의 강한 저해효과를 나타났다. Tyrosinase 저해활성을 측정한 결과는pocyanidin C-1-3'-3" -3.'S _0-trigallate는 100rM에서 70%의 강한 저해효과를 나타냈으며,epigallocatechin-(4$\beta$$\longrightarrow$8)-epigallo-catechin-(4$\beta$$\longrightarrow$8)-catechin는 51%의 저해효과를 나타내었다. 산업적응용을 위해 분획한 폴리페놀군은 미백효과 검증실험인 tyrosinase 저해율 측정평가에서 폴리페놀 함량이 가장 높은 Fraction 111의 경우 Sooppm에서 74.2%의 높은 저해율을 나타내었다. 항산화력 실험에서는500pw1이상에서 강한 활성능을 보인 SOD 유사활성능을 제외한 나머지 DPPH와 xanthine oxidase 저해효과에서는 Fraction II와III 모두가50ppm이상에서 80% 이상의 높은 유리라디칼 소거능력을 나타내었다. 그리고 각 Fraction별 항균력 측정 결과 Fraction 르와 111이 우수하게 나타났고 항균활성은 그람음성균보다 그람양성균에서 효과적이었으며, 농도별 항균력시험 결과 농도가 증가할수록 비례하여 저해율도 증가함을 알 수 있었다. 첨가농도를 달리하여 미생물의 생육도를 측정한 결과, fraction II磎꼭\ulcorner경우 그람양성균에 대해 500 ppm 이상에서 뚜렷한 증식억제효과를 나타내었다.서 뚜렷한 증식억제효과를 나타내었다.

Illumina MiSeq sequencing reveals the effects of grape seed procyanidin on rumen archaeal communities in vitro

  • Objective: The present study explored the effects of grape seed procyanidin extract (GSPE) on rumen fermentation, methane production and archaeal communities in vitro. Methods: A completely randomized experiment was conducted with in vitro incubation in a control group (CON, no GSPE addition; n = 9) and the treatment group (GSPE, 1 mg/bottle GSPE, 2 g/kg dry matter; n = 9). The methane and volatile fatty acid concentrations were determined using gas chromatography. To explore methane inhibition after fermentation and the response of the ruminal microbiota to GSPE, archaeal 16S rRNA genes were sequenced by MiSeq high-throughput sequencing. Results: The results showed that supplementation with GSPE could significantly inhibit gas production and methane production. In addition, GSPE treatment significantly increased the proportion of propionate, while the acetate/propionate ratio was significantly decreased. At the genus level, the relative abundance of Methanomassiliicoccus was significantly increased, while the relative abundance of Methanobrevibacter decreased significantly in the GSPE group. Conclusion: In conclusion, GSPE is a plant extract that can reduce methane production by affecting the structures of archaeal communities, which was achieved by a substitution of Methanobrevibacter with Methanomassiliicoccus.

Quantitative Analysis of Taxifolin, (+)-Catechin and Procyanidin B1 from the Preparation of Pinus densiflora (PineXol®) (적송제제(PineXol®)로 부터 Taxifolin과 (+)-Catechin, Procyanidin B1의 함량분석)

  • Pinus densiflora contained diverse phenol compounds like flavonoid, phenylpropanoid and tannin. PineXol$^{(R)}$ is nutraceutical preparation which was treated from bark of Pinus densiflora. Validation and contents determination of taxifolin, (+)-catechin and procyanidion B1 for the preparation of Pinus densiflora (PineXol$^{(R)}$) were confirmed using High-Performance Liquid Chromatography (HPLC). As a result, content of taxifolin, (+)-catechin and procyanidin B1 were, respectively 4.90%, 2.35% and 8.19%. These analysis method and results could be used as important basic data for the preparation of Pinus densiflora.

Extractives of the Bark of Ash and Elm as Medicinal Hardwood Tree Species (약용 활엽수종인 물푸레나무와 느릅나무 수피의 추출성분)

  • The bark of ash(Fraxinus rhynchophylla) and elm(Ulmus davidiana var. japonica) trees were collected, extracted with acetone-$H_2O$(7:3, v/v), fractionated with hexane, chloroform and ethylacetate, and freeze dried to give some dark brown powder. Each fraction of the powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluents. The ash bark contained a large amount of coumarin derivatives such as aesculetin and aesculin in addition to trace amount of ligstroside and oleuropein. Most of the elm bark extractive were (+)-catechin and its glycosides such as (+)-catechin-7-O-xylopyranose and (+)-catechin-7-O-apiofuranose in addition to a small amount of procyanidin B-3, a dimeric (+)-catechin. NMR and FAB-MS spectrometric analyses were performed to characterize the structures of isolated phenolic compounds.

Skin Protection Effect of Grape Pruning Stem Extract on UVB-induced Connective Tissue Injury (포도전정가지 추출물이 UVB로 유도된 결합 조직 손상에 미치는 피부 보호 효과)

  • This study aimed to analyze the contents of rutin, procyanidin B3, quercetin, and kaempferol, known to have antioxidant, anti-inflammatory, and anti-carcinogenic effects, among the polyphenol types contained in grape pruning stem extracts (GPSE). It utilized grape stems discarded after harvest to measure the effects of GPSE on skin moisture, inhibition of skin cell proliferation, and anti-inflammatory activity on the damaged skin of HR-1 mice induced with ultraviolet B (UVB), and to verify the applicability of GPSE as a material for functional food and functional cosmetics. The polyphenol was extracted from grape pruning stems with 80% EtOH, and then the extract was used while storing at $-20^{\circ}C$, after filtering, concentrating, and freeze-drying it. The content of an active ingredient of GPSE was analyzed using high performance liquid chromatography (HPLC). From 53 kg of the grape pruning stem specimen, 2.34 kg of the EtOH fraction extracts were extracted to achieve a 4.42% yield ratio. Analysis of the active ingredients showed 0.28 mg/g of procyanidin B3, 12.81 mg/g of rutin, 0.51 mg/g of quercetin, and 8.24 mg/g of kaempferol. After UVB irradiation on the dermis, to confirm the degree of inhibition of collagen synthesis, we examined the protein expression of MMP-9 using immunohistochemical staining. The results of this study confirm the existence of active polyphenol types, such as rutin, kaempferol, quercetin, and procyanidin B3, in GPSE. Moreover, the study found that GPSE has anti-collagenase effects and it decreases the effects of UV damage on skin barrier function. GPSE is a functional ingredient with a potential for skin protection effects, and it has high utilization potential as an ingredient for functional cosmetics.

Functional Activities of Cacao Nibs and Couvertures according to Process Conditions (카카오닙과 커버춰의 가공 조건에 따른 기능성 분석)

  • This study was conducted to provide information regarding changes in antioxidant activity in response to conching temperatures, conching times, and cacao mass content (CMC) in dark chocolate. The radical scavenging activities and functional components of cacao nibs were highest for raw cacao nib (R0) under all conditions. Moreover, antioxidant activities and functional compounds increased during roasting for 25 min. As the conching temperature increased, the radical scavenging activities and functional components increased. Quantitative analysis of major catechin-derived compounds by HPLC revealed that R0 had the highest value for other roasted cacao nibs in all aspects (P<0.05). The content of procyanidin B2, catechin, and epicatechin increased during roasting for 25 min. Finally, evaluation of couvertures revealed that procyanidin B1 content increased as conching time increased to 48 h, except for 70% CMC and conched at $60^{\circ}C$ (HH) and 70% CMC and conched at $50^{\circ}C$. Overall, HH48 contained the richest catechin-derived components.
