Enhancement of Spermidine Content and Antioxidant Capacity by Modulating Ginseng Spermidine synthase in Response to Abiotic and Biotic Stresses

  • Polyamines (putrescine, spermidine and spermine) play pivotal roles in plant defense to different abiotic and biotic stresses. In order to understand the function of ginseng spermidine synthase gene, a key gene involved in biosynthesis of polyamines, transgenic plant was generated in Arabidopsis. The transgenic plants exhibited high levels of polyamines compared to the untransformed control plants. We investigated the tolerance capacity of transgenic plants to abiotic stresses such as salinity and copper stress. In addition, transgenic plants also showed increased resistance against one of the important fungal pathogens of ginseng, the wilt causing Fusarium oxysporum and one of important bacteria, bacterial blight causing Pseudomonas syringae. However, an activity of the polyamine catabolic enzyme, diamine oxidase (DAO) was increased significantly in F. oxysporum and P. syringae infected transgenic plant. Polyamine catabolic enzymes which may trigger the hypersensitive response (HR) by producing hydrogen peroxide ($H_2O_2$) seem act as an inducer of PR proteins, peroxidase and phenyl ammonium lyase activity. The transgenic plants also contained higher antioxidant enzyme activities, less MDA and $H_2O_2$ under salt and copper stress than the wild type, implying it suffered from less injury. These results strongly suggest an important role of spermidine as a signaling regulator in stress signaling pathways, leading to build-up of stress tolerance mechanisms.

원자력발전소의 출력감발모드를 위한 경보축약 규칙 (Practical Alarm Suppression Rules and their Implementation for Nuclear Power Plants)

  • It is necessary to adopt some logical techniques and methods of alarm processing for a large complex plant such as nuclear power plants in order to present the occurred alarm messages properly and concisely. Among such alarm processing techniques, the alarm suppressing function is a strong tool to avoid alarm flooding during the sudden transients of plant output power such as turbine trips, reactor trips and other incidents. Unless any suppression or representation technologies are used in an alarm message listing system, it cannot provide quick assistance to plant operators or supervisors during plant upsets because too many alarm messages are presented in an alarm list window. This paper presents the key suppression methods and analysis processes developed for implementing a suppressed alarm message listing function of an integrated alarm system called LogACTs which has been applied to a CANDU nuclear power plant. A simulation testing of the suppressing function conducted with the real plant alarm message list data has demonstrated an effective performance of the developed logics with the high suppression rate.

  • A fossil power plant can be modeled by a lot of algebraic equations and differential equations. When we simulate a large, complicated fossil power plant by a computer such as workstation or PC, it takes much time until overall equations are completely calculated. Therefore, new processing systems which have high computing speed is ultimately needed to develope real-time simulators. Vital points of real-time simulators are accuracy, computing speed, and deadline observing. In this paper, we present a enhanced strategy in which we can provide powerful computing power by parallel processing of DSP processors with communication links. We designed general purpose DSP modules, and a VME interface module. Because the DSP module is designed for general purpose, we can easily expand the parallel system by just connecting new DSP modules to the system. Additionally we propose methods about downloading programs, initial data to each DSP module via VME bus, DPRAM and processing sequences about computing and updating values between DSP modules and CPU30 board when the simulator is working.

Isolation and Identification of Fungal Species from the Insect Pest Tribolium castaneum in Rice Processing Complexes in Korea

  • The red flour beetle, Tribolium castaneum, is one of the most common and economically important pests of stored cereal products worldwide. Furthermore, these beetles can act as vectors for several fungal post-harvest diseases. In this study, we collected T. castaneum from 49 rice processing complexes (RPCs) nationwide during 2016-2017 and identified contaminating fungal species on the surface of the beetles. Five beetles from each region were placed on potato dextrose agar media or Fusarium selection media after wet processing with 100% relative humidity at $27^{\circ}C$ for one week. A total of 142 fungal isolates were thus collected. By sequence analysis of the internal transcribed spacer region, 23 fungal genera including one unidentified taxon were found to be associated with T. castaneum. The genus Aspergillus spp. (28.9%) was the most frequently present, followed by Cladosporium spp. (12.0%), Hyphopichia burtonii (9.2%), Penicillium spp. (8.5%), Mucor spp. (6.3%), Rhizopus spp. (5.6%), Cephaliophora spp. (3.5%), Alternaria alternata (2.8%) and Monascus sp. (2.8%). Less commonly identified were genera Fusarium, Nigrospora, Beauveria, Chaetomium, Coprinellus, Irpex, Lichtheimia, Trichoderma, Byssochlamys, Cochliobolus, Cunninghamella, Mortierella, Polyporales, Rhizomucor and Talaromyces. Among the isolates, two known mycotoxin-producing fungi, Aspergillus flavus and Fusarium spp. were also identified. This result is consistent with previous studies that surveyed fungal and mycotoxin contamination in rice from RPCs. Our study indicates that the storage pest, T. castaneum, would play an important role in spreading fungal contaminants and consequently increasing mycotoxin contamination in stored rice.

  • A rice mill pilot plant was designed and developed in the basis of the simulation results on the mill plants. The performance of the developed rice mill plant was evaluated, and the simulation model on the mill system was validated with the experimental data in the mill plant. The results of this study were as followings : 1. A rice mill pilot plant with the capacity of 0.5 t/h was designed and developed. 2. The hulled ratio of the mill plant was 87.3%, and the milled rice recovery and the head rice recovery of the cleaned rice were 74% and 87% , respectively. The degree of milling of the cleaned rice was 10.6% with a high polish. The intensity of the cleaned rice appeared high compared with that of the milled rice in the analysis of whiteness test using an image processing system. 3. The bottleneck, processing time, and production amount of the developed mill system almost coincided with those of the simulation of the rice mill plant. The developed simulation model of the rice mill plant was proven to be applicable to the design of a rice mill plant through experiments.

In vitro propagation of Coleus forskohlii, an important medicinal plant

  • For mass multiplication of an important medicinal plant Coleus forskohlii, a procedure for the high frequency regeneration of Coleus forskohlii has been developed using leaf explants via callus culture. Callus formation occurred in MS medium supplemented with 1-2 mg/L each of NAA and BAP. A large number of shoots were formed on MS + 1 mg/L BAP from 50-60 days old greenish calli. Rooting of healthy shoots occurred on 0.1-0.4 mg/L NAA. The protocol described could be useful in future for genetic manipulation of this plant species.

High Frequency of Plant Regeneration through Cyclic Secondary Somatic Embryogenesis in Panax ginseng

  • Somatic embryogenesis is one of good examples of the basic research for plant embryo development as well as an important technique for plant biotechnology such as medicinally important plants. Single embryos develop into normal plantlets with shoots and roots. Therefore, direct single embryogenesis derived from single cells is highly important for normal plant regeneration. Here we demonstrate that the cyclic secondary somatic embryogenesis in Panax ginseng Meyer is a permanent source of embryogenic material that can be used for genetic manipulations. Secondary somatic embryos were originated directly from the primary somatic embryos on hormone-free Murashige and Skoog medium, and proliferated further in a cyclic manner. EM medium (one third of modified MS medium [MS medium containing half amount of NH4NO3 and KNO3] with 2% to 3% sucrose) favored further development of proliferated secondary somatic embryos into plantlets with root system. The plantlets developed into plants with well-developed taproots in half-strength Schenk and Hildebrandt basal medium supplemented with 0.5% activated charcoal.

Molecular identification of oriental medicinal plant Schizonepeta tenuifolia bunge (Hyung-Gae) by multiplex PCR

  • Schizonepeta tenuifolia (Korean name "Hyung- Gae") is an oriental medicinal plant that is widely used in Korea, China and Japan. S. tenuifolia (Hyung-Gae) has many pharmacological activities and is mostly used for many medicinal preparations. The dried aerial part (spikes and stems) of three oriental medicinal plants, S. tenuifolia (Hyung-Gae), Agastache rugosa (Kwhak-Hyang) and Elsholtzia ciliata (Hyang-Yoo) belonging to the same family, mint family Labiaceae, have such similar shape and smell that it is difficult to differentiate between them. The trnL-F regions of chloroplast DNA of the three medicinal plants were sequenced and used as targets in multiplex PCR reaction to identify S. tenuifolia. After alignment of trnL-F sequences of the authenticated plant samples, one single nucleotide polymorphism (SNP) specific to S. tenuifolia was found. Based on this SNP, a new primer was designed that specifically amplifies the trnL-F region of S. tenuifolia. The established multiplex-PCR was proven to be effective in the differentiation of commercial S. tenuifolia samples from A. rugosa and E. ciliata. This rapid and accurate molecular method is highly promising for use in the food industry.
