  • Redundancies in the computational procedure of the FFT by which the DFT is calculated are analized and a modified algorithm to reduce redundancies is proposed. An amended FFT fortran program adopting the proposed algorithm shows that the time necessary for DFT computation is decreased about one-tenth. The proposed method can be applied to the most of other transformation algorithms.

  • Optimized conditions for Agrobacterium-mediated lisianthus pollen transformation were adjusted using various factors such as temperature, pH and sucrose concentration. Pollen tube growth was successfully achieved in a medium (pollen germination medium; PGM) containing $7-15\%$ sucrose with pH in the range of 5.5-7.0 at temperature of $20-27^{\circ}C$. Lisianthus pollen was vacuum-infiltrated with Agrobacterium cell suspension for 20 min, and transformed pollen was confirmed by GUS histochemistry and Southern hybridization following RT-PCR. Transgenic pollen system may be utilized for establishing an area of plant transient expression systems based on the convenient pollen transformation procedure presented in here.

  • Genetic transformation of a mushroom-forming fungus Coprinellus congregatus to antibiotic resistance gene had been successfully carried out by electroporation to oidia instead of protoplasts. Since there was no protoplast generation step which required not only cell wall degrading enzymes but many skillful procedures, commercial herbicide (basta) could be used without any difficulty with simple procedure. The transformation yield was 10-20 $transformants/{\mu}g$ DNA, and the transformants were very stable even after 10 consecutive transfers through the non-selective medium.

  • XML is becoming a de facto standard for the representation and exchange of data, and XML itself is well structured to store and manage data, but it is difficult to understand the document structure at a glance due to its diverse and complex expressions. Thus, it is not appropriate to use it as a tool of conceptual model. In this paper, we propose a method to transform XML schema document structure into conceptual model. To do this, we describe CMXML as a conceptual modeling tool and present transformation rules and data structures for transforming an XML schema structure into CMXML diagram. In the transformation procedure, the XML schema document is input and the CMXML diagram is generated by applying the transformation rules.

  • The fundamental objective of this paper has been to develop a means for incoporating the concept of the linear fractional transformation more generally and easily into multivariable feedback design procedure. When we design a continuous system, generally, we are constrained by design methods which arise specifically for the system. Also, in the design of descrete systems, it is the same concept. But the approach developed in this paper is very flexible in the view that in spite of being the continuous or discrete, the design can be done using a well known design method in both cases. That is, when we design a contnuous system or discrete system, the design can be done by a standard design method of continuous systmes or discrete ones, depending on the choice of the linear fractional transformation. Therefore, it is noted that this concept has broken the unflexibility of the conventional design rules for multivariable control system. In essence, the concept shows that if a given system is controllable, some desirable design, for examples, pole assignment within prespecified region, optimal controllers with poles within prespecified region etc., could be done easily by transforming a desirable region into a standard region, such as the complex left-half plane or the unit disk, by the chosen linear fractional transformation, and then by designing the transformed system using the well known standard results.

  • In order to transform the underground utility data into the 3-D spatial objects, this thesis defined the type of the 3-D spatial objects and the storage structure of objects, and developed the spatial data transformation system that transforms the 2-D underground utility data into the pre-defined 3-D spatial objects through the Booch Method, an object-oriented development method. For this sake, the relational data model of ARC/INFO and the storage structure are analyzed, as well as the pre-requisites, required for the 3-D visualization of the underground utilities. Also, the 2-D underground utility data, saved in ARC/INFO, were transformed into the ASCII files through the internal pre-processing procedure, then used as the input data of the transformation system. In addition, to develop the transformation system, the object-oriented development methods are studied first and, then, applied to the system analysis and the design procedure with the Booch Method as a development method. Finally, the results of analysis and design procesure are implemented through AML, a ARC/INFO macro language, and C++, an object-oriented programing language. As a result of this study, the 3-D spatial objects that can visualize the 2-D utilities in 3-D, are acquired, and the adaptation of object-oriented development method to the system development procedure enabled an effective development prodedure.

Optimization of filling process in RTM using genetic algorithm

  • In resin transfer molding (RTM) process, preplaced fiber mat is set up in a mold and thermoset resin is injected into the mold. An important interest in RTM process is to minimize cycle time without sacrificing part quality or increasing cost. In this study, the numerical simulation and optimization process in filling stage were conducted in order to determine the optimum gate locations. Control volume finite element method (CVFEM) was used in this numerical analysis with the coordinate transformation method to analyze the complex 3-dimensional structure. Experiments were performed to monitor the flow front to validate simulation results. The results of numerical simulation predicted well the experimental results with every single, simultaneous and sequential injection procedure. We performed the optimization analysis for the sequential injection procedure to minimize fill time. The complex geometry of an automobile bumper core was chosen. Genetic algorithm was used in order to determine the optimum gate locations with regard to 3-step sequential injection case. These results could provide the information of the optimum gate locations in each injection step and could predict fill time and flow front.

A Rapid and Effective Colony PCR Procedure for Screening Transformants in Several Common Mushrooms

  • In the post-genomic era, gene function analysis has attracted much attention. Transformation is often needed to investigate gene function. In this study, an easy, rapid, reliable, and cost-effective colony polymerase chain reaction (PCR) method for screening mushroom transformants was developed: picking up a suitable amount of transformant's tissue ($1-10{\mu}g$) to $20{\mu}l$ 0.25% Lywallzyme solution, and vortexing for 10 s followed by incubation at $34^{\circ}C$ for 15 min. Finally, $2{\mu}l$ of the suspension was used as templates to perform PCR and single target bands were successfully amplified from respective transformants of Tremella fuciformis, Pleurotus ostreatus, and Pleurotus tuber-regium. This procedure could be widely employed for screening transformants in mushroom transformation experiments.

  • In this paper, we studied on the design of elliptic digital filters using the bilinear z transformation method, and proposed a design procedure satisfying prescribed specifications. The magnitude characteristics of digital filters are compared with its of analog filiters by computer simulation. Finaly we considered the quantization effects of digital filters. In cascader realization of fixed-point digital filters under dynamic range constraints, the outpout noise for IIR digital filters depends on the pole-zero pairing and ordering of the second order sections. Therfore an optmization procedure to finding a good ordering and pairing is very desirable. Thus, we proposed a sub-optimization procedure for finding "near optimal" solution.
