Finger millet (Eleusine coracana) is widely cultivated in tropical regions worldwide owing to its high nutritional value. Finger millet is more tolerant against biotic and abiotic stresses such as pests, drought, and salt than other millet crops; therefore, it was proposed as a candidate crop to adapt to climate change in Korea. In 2019, we used expressed sequence tag simple sequence repeat (EST-SSR) markers to evaluate the genetic diversity and structure of 102 finger millet accessions from two geographical regions (Africa and South Asia) to identify appropriate accessions and enhance crop diversity in Korea. In total, 40 primers produced 116 alleles, ranging in size from 135 to 457 bp, with a mean polymorphism information content (PIC) of 0.18225. Polymorphism was detected among the 40 primers, and 13 primers were found to have PIC values > 0.3. Principal coordinate and phylogenetic analyses, based on the combined data of both markers, grouped the finger millet accessions according to their respective collection areas.Therefore, the 102 accessions were classified into two groups, one from Asia and the other from Africa. We have conducted an in-depth study on the finger millet landrace pedigree. By sorting out and using the molecular characteristics of each pedigree, it will be useful for the management and accession identification of the plant resource. The novel SSR markers developed in this study will aid in future genetic analyses of E. coracana.
For effective water quality management, it is necessary to secure reliable water quality information. There are many variables that need to be included in a comprehensive practical monitoring network : representative sampling locations, suitable sampling frequencies, water quality variable selection, and budgetary and logistical constraints are examples, especially sampling location is considered to be the most important issues. Until now, monitoring network design for water quality management was set according to the qualitative judgments, which is a problem of representativeness. In this paper, we propose network design system for optimal water quality monitoring using the scientific statistical techniques. Network design system is made based on the SAS program of version 9.2 and configured with simple input system and user friendly outputs considering the convenience of users. It applies to Excel data format for ease to use and all data of sampling location is distinguished to sheet base. In this system, time plots, dendrogram, and scatter plots are shown as follows: Time plots of water quality variables are graphed for identifying variables to classify sampling locations significantly. Similarities of sampling locations are calculated using euclidean distances of principal component variables and dimension coordinate of multidimensional scaling method are calculated and dendrogram by clustering analysis is represented and used for users to choose an appropriate number of clusters. Scatter plots of principle component variables are shown for clustering information with sampling locations and representative location.
An Aquifer test was carried out on five boreholes to determine the hydrologic anisotropy and the major groundwater flow direction in the aquifer system of the study area. With an assumption of the aquifer's anisotropy and homogeneity, the major transmissivity(T(equation omitted)), the minor transmissivity( $T_{ηη}$ ), and primary tensor direction ($\theta$) for each borehole were determined from the test. Besides the boreholes BH-1, BH-4 and BH-5, the anisotropy transmissivity tensor values of BH-2 and BH-3 did not correspond with the assumption. Thereafter the values were plotted on the polar coordinate, and showed that the tensor values were out of the anisotropy ellipsoid due to the high heterogeneity of BH-2 and BH-3 comparing with the other boreholes. Therefore. the anisotropy of the aquifer was examined from BH-1, BH-4. and BH-5. In BH-1, T(equation omitted) is 171.9 $\m^2$/day. $T_{ηη}$ is $71.01\m^2$/day, and the principal tensor direction is Nl5.39$^{\circ}$E. In BH-4. T(equation omitted) is $268.2 \m^2$/day, $T_{ηη}$ / is $28.75\m^2$/day and the principal tensor direction is N7.55$^{\circ}$E. In BH-5, T(equation omitted) is $168.4\m^2$/day, $T_{ηη}$ is 66.80 $\m^2$/day, and the principal tensor direction is $N76.59^{\circ}$E. On the basis of teleview logging performed on each borehole. the principal fracture directions were revealed as $N0^{\circ}$~4$^{\circ}$E/$30^{\circ}$~$50^{\circ}$SE and $N30^{\circ}$~$80^{\circ}$W/$20^{\circ}$~$50^{\circ}$NE that are the most frequently occurred sets as well as that correspond well with the calculated transmissivity tensor.
Objective: RNA epigenetic modifications play an important role in regulating immune response of mammals. Bovine mastitis induced by Staphylococcus aureus (S. aureus) is a threat to the health of dairy cattle. There are numerous RNA modifications, and how these modification-associated enzymes systematically coordinate their immunomodulatory effects during bovine mastitis is not well reported. Therefore, the role of common RNA modification-related genes (RMRGs) in bovine S. aureus mastitis was investigated in this study. Methods: In total, 80 RMRGs were selected for this study. Four public RNA-seq data sets about bovine S. aureus mastitis were collected and one additional RNA-seq data set was generated by this study. Firstly, quantitative trait locus (QTL) database, transcriptome-wide association studies (TWAS) database and differential expression analyses were employed to characterize the potential functions of selected enzyme genes in bovine S. aureus mastitis. Correlation analysis and weighted gene co-expression network analysis (WGCNA) were used to further investigate the relationships of RMRGs from different types at the mRNA expression level. Interference experiments targeting the m6 A demethylase FTO and utilizing public MeRIP-seq dataset from bovine Mac-T cells were used to investigate the potential interaction mechanisms among various RNA modifications. Results: Bovine QTL and TWAS database in cattle revealed associations between RMRGs and immune-related complex traits. S. aureus challenged and control groups were effectively distinguished by principal component analysis based on the expression of selected RMRGs. WGCNA and correlation analysis identified modules grouping different RMRGs, with highly correlated mRNA expression. The m6 A modification gene FTO showed significant effects on the expression of m6 A and other RMRGs (such as NSUN2, CPSF2, and METTLE), indicating complex co-expression relationships among different RNA modifications in the regulation of bovine S. aureus mastitis. Conclusion: RNA epigenetic modification genes play important immunoregulatory roles in bovine S. aureus mastitis, and there are extensive interactions of mRNA expression among different RMRGs. It is necessary to investigate the interactions between RNA modification genes regulating complex traits in the future.

Saemangeum reclaimed land is a part of Saemangeum Development Project. Most of the persistent problems of Saemangeum reclaimed land remain to be related to soil salinity. Soil salinity is a major abiotic factor related to microbial community structure and also fungi have been reported to be more sensitive to salinity stress than bacteria. The aim of this study was conducted to investigate the effect of soil salinity levels on the microbial communities in Saemangeum reclaimed land using 454 pyrosequencing analysis. Soil samples was collected from 12 sites of in Saemangeum reclaimed land. For pyrosequencing, 27F/518R (bacteria) and ITS3/ITS4 (fungi) primers were used containing the Roche 454 pyrosequencing adaptor-key-linker (underlined) and unique barcodes (X). Pyrosequencing was performed by Chun's Lab (Seoul, Korea) using the standard shotgun sequencing reagents and a 454 GS FLX Titanium sequencing System (Roche, Inc.). In the soil samples, Proteobacteria (bacteria) and Ascomycota (fungi) shows the highest relative abundance in all the soil sample sites. Proteobacteria, Bacteroidetes, Plantomycetes, Gemmatimonadetes and Parcubacteria were shown to have significantly higher abundance in high salinity level soils than low salinity level soils, while Acidobacteria and Nitrospirae has significantly higher relative abundance in low salinity level soils. The abundance of fungal, Ascomycota has the highest relative abundance in soil samples, followed by Basidiomycota, Chlorophyta, Zygomycota and Chytridiomycota. Basidiomycota, Zygomycota, Glomeromycota and Cerozoa were show significantly higher relative abundance in low salinity level soils. The principal coordinate analysis (PCoA) and correlation analysis shown to salinity-related soil parameters such as ECe, Na+, SAR and EPS were affected to bacterial and fungal community structure. Proteobacteria, Bacteroidetes, Plantomycetes exhibited significantly positive correlation with soil salinity, while Acidobacteria exhibited significantly negative correlation. In the case of fungal community, Basidiomycota and Zygomycota were seen show significantly negative correlation with salinity related soil parameters. These results suggest that provide understanding effect of soil salinity on microbial community structure and correlation of microbial community with soil parameters in Saemangeum reclaimed land.

This study investigated dietary habits and intra- and inter-specific food resource partitioning of co-occurring walleye pollock (Gadus chalcogrammus) and Pacific cod (G. macrocephalus) from the waters off the north-eastern coast of South Korea using stomach contents and stable isotopes (δ13C and δ15N) analyses. Both species are mesopelagic carnivores that consumed mainly benthopelagic crustaceans, but teleosts were also abundant in the diet of Pacific cod. Non-metric multidimensional scaling (nMDS) ordination and permutational multivariate analysis of variance (PERMANOVA) of dietary data revealed significant intra- and inter-specific dietary differences, i.e., food resource partitioning. Nitrogen stable isotope values (δ15N) were similar between walleye pollock and Pacific cod, but carbon stable isotope values (δ13C) were significant different, suggesting different trophic positioning. Canonical analysis of principal coordinate (CAP) ordination plot further demonstrated that differences in the type and range of prey ingested by the two species contributed such an inter-specific difference in the diet compositions. Ontogenetic changes in diet compositions were evident. As walleye pollock, they preyed more upon carid shrimps and cephalopods, but no such trend was observed in the diets of Pacific cod. While stable isotope values indicated that large-sized specimens of both species were significantly enriched in 15N relative to smaller conspecifics thus supporting these data. Consequently, in this study, both methodologies, i.e., stomach contents and stable isotope analyses, provided evidence of inter- and/or intra-specific dietary segregations and trophic niche partitioning between co-occurring walleye pollock and Pacific cod off eastern Korean waters.

We evaluated the influence of sampling technique (cannulation vs. stomach tube) and site (dorsal sac vs. ventral sac) on the rumen microbiome and fermentation parameters in Hanwoo steers. Rumen samples were collected from three cannulated Hanwoo steers via both a stomach tube and cannulation, and 16S rRNA gene amplicons were sequenced on the MiSeq platform to investigate the rumen microbiome composition among samples obtained via 1) the stomach tube, 2) dorsal sac via rumen cannulation, and 3) ventral sac via rumen cannulation. A total of 722,001 high-quality 16S rRNA gene sequences were obtained from the three groups and subjected to phylogenetic analysis. There was no significant difference in the composition of the major taxa or alpha diversity among the three groups (p>0.05). Bacteroidetes and Firmicutes represented the first and second most dominant phyla, respectively, and their abundances did not differ among the three groups (p>0.05). Beta diversity principal coordinate analysis also did not separate the rumen microbiome based on the three sample groups. Moreover, there was no effect of sampling site or method on fermentation parameters, including pH and volatile fatty acids (p>0.05). Overall, this study demonstrates that the rumen microbiome and fermentation parameters are not affected by different sampling techniques and sampling sites. Therefore, a stomach tube can be a feasible alternative method to collect representative rumen samples rather than the standard and more invasive method of rumen cannulation in Hanwoo steers.

The genetic diversity and structure of P. tongkangensis in 5 populations from 3 regions was investigated using 56 markers derived from 6 ISSR primers. Genetic diversity at the species level (P=94.6, SI=0.377, h=0.240) was substantial considering the limited distribution and small size of populations. Genetic differentiation among regions (12%) and among populations (13%) in the region was not clearly evident, which suggested a moderate level of gene flow among adjacent populations. The Mantel test revealed a significant correlation between genetic differentiation (${\Phi}_{ST}$) and geographic distance among populations. This was supported by cluster analysis and principal coordinate analysis (PCoA). The significant difference in marker band frequency at many loci and their fixation in opposite directions in the smallest and most isolated population SC were considered due to genetic drift. Therefore, the genetic diversity of P. tongkangensis could be compromised if the distribution area or the size of the population was further reduced. In particular, small and isolated populations could be at great risk of extinction. Considering this, the unique habitats of P. tongkangensis should be protected and the reduction of population size should be closely monitored. Conservation efforts including the seeding and planting of seedlings should be done carefully based on their genetic and ecological traits. Our data support the argument that establishing an integrated management system for the efficient conservation of P. tongkangensis is critical.
The sensory characteristics of nine rice-based distilled soju were determined using check-all-that-apply (CATA) profiling. A total of 53 consumers evaluated the soju for two appearance attributes, nine aroma attributes, nine flavor/taste attributes, four mouth-feel related sensory attributes, and overall desirability. The total sum of CATA terms indicated that 14 characteristics showed frequency differences of over 10 and that there were significant differences among nine samples for eleven sensory attributes as determined using Cochran's q test (p<0.05). Based on correspondence analysis of CATA data, the samples were primarily separated by the first dimension, which accounted for 89% of the total variance among samples. The "brown color," "fruit taste," and "grain aroma" characteristics had higher frequencies than those for the "white color," "acetone aroma," and 'alcohol taste" characteristics. Overall, there was a higher preference for oak-aged samples than for samples aged in other containers. "sweet aroma', 'fruit aroma," and "sweet taste" seemed to positively affect consumer preferences, while "bitter taste," "alcohol taste," and "acetone aroma" appeared to negatively affect consumer preferences as determined by principal coordinate analysis.
Dietary niches can support the coexistence of sympatric species in marine ecosystems, which can lead to the presence of greater abundances of those species. Stomach contents of silver croaker (Pennahia argentata) and Japanese sillago (Sillago japonica) inhabiting the Nakdong River estuary, Korea were analyzed to determine diet compositions and the presence of any size-related and inter-specific feeding relationships. These species were bottom-feeding predators that consumed mainly benthic crustaceans and mollusks, but polychaetes were also important in the S. japonica diets. Non-metric multidimensional scaling (nMDS) ordination and multivariate analyses based on weight contributions of the different prey taxa to stomach contents revealed significant inter-specific dietary differences. Size-related change was also significant for P. argentata diets, but not for S. japonica. Canonical analysis of principal coordinate (CAP) ordination plot further demonstrated that differences in the type and range of prey ingested by the two species contributed such an inter-specific difference in the diet compositions. The results from this study provide clear evidence of niche segregation between co-occurring P. argentata and S. japonica in the Nakdong River estuary, which would reduce the likelihood of inter-specific competition for food resources.
