Sing1e or Combined Treatment Effect of Priming, Gibberellin and Prechilling on Germination of Gourd Seeds

  • 현재 수박접목묘로서 가장 많이 이용되고 있는 박 종자의 발아율을 높이기 위하여 Priming, $GA_3$, 저온의 단일처리 효과와 저온처리 전후에 가하여지는 priming과 $GA_3$ 의 복합처리 효과를 조사하였던 바 그 결과를 요약하면 다음 과 같다. $3^{\circ}C$에서 1주간 행하여지는 저온처리, 100 mM $KNO_3$에 1일간 행한 priming, 0.01 mM에 1일간 행한 $GA_3$의 단일처리에 대한 발아율을 조사한 결과 저온처리에서 가장 높게 나타났다. 그러나 발아율은 저온처리 후의 priming 처리는 무처리보다는 낮았고 처리기간이 길어질수록 발아율이 현저히 감소되었다. 저온처리 전에 행한 priming과 $GA_3$ 처리는 저온처리 전의 $GA_3$ 처리에서와 같이 농도와 처리기간에 따른 차이가 없었다. 따라서 박의 종자 발아율 향상을 위한 종자처리로써 여러 개의 단일처리를 조합한 복합처리는 처리조합과 순서에 따라 발아율이 영향을 받는 것으로 사료된다.

Pre-Sowing Seed Treatment: Effect of Deep Sea Water Priming on Germination of Wild Vegetables; Gondalbi (Cirsium setidens), Jandae (Adenophora triphylla var. Joponica Hara) and Deoduck (Codonopsis lanceolata Trautv.)

  • This experiment was conducted on wild vegetables; Gondalbi (Cirsium setidens), Deoduck (Codonopsis lanceolata Trautv.), and Jandae (Adenophora triphylla var. Joponica Hara) seed to study whether priming with deep sea water results in enhancement of seed germination and identify the optimum concentration of the priming solution, and duration of priming. Seeds were primed with 5 various concentrations (5%, 10%, 15%, 20% and 30%) of deep sea water (DSW) in 12 hours, 24 hours, and 48 hours at $24^{\circ}C$. Since Jandae had seed dormancy, it was kept for four weeks in refrigerator at $2^{\circ}C$ after priming treatment. In Deoduck, 5 percentage DSW priming significantly improved the early germination percentage, radicle length, and plumule emergence percentage. Among the priming period of treatments, 24 hours priming showed better performance in this treatment whereas, in Jandae, 12 hours priming with 10 percentages DSW significantly improved the germination percentage and germination rate. This treatment had increased the final germination percentage by 54%, 15% and 40% compared with control, plain water and $KNO_3$ priming respectively. But in Gondalbi, priming did not improve the germination of seed. However, among the priming treatments, 12 hours priming with 3% $KNO_3$ and 20% DSW gave better performance. In both the wild vegetables; Deoduck and Jandae, priming in deep sea water had improved the germination percentage and germination rate as compare to plain water, $KNO_3$, and without priming treatment. Hence the best seed priming treatment on Deoduck and Jandae are 24 hours with 5% DSW and 12 hours with 10% DSW respectively.

Effects of Sonication, Osmotic Priming and Modified Drum Priming on the Germination of Tomato Seeds

  • In order to increase the germination speed and uniformity of tomato seeds, sonication and modified drum priming treatments were investigated to produce high quality seeds for export. Sonication treatment was performed for 5, 10 and 20minutes at an intensity of 5.2, 10.4 and 15.7kHz in water at $15^{\circ}C$. After sonication treatment, seeds were primed with water or 100mM $KNO_3$ for 4days. 40, 50 and 60% seed moisture content (SMC) of hydrated seeds were incubated for 60, 72 and 84h in a container with a relative humidity of 99% at 26rpm for a modified drum priming treatments. Germination speed were highly improved by sonication with osmotic priming. The seed treatment of osmotic priming or hydro priming after sonication or sonication without priming enhanced germination percentage (GP) on the $2^{rd}$day after sowing to 46%, 43% and 28%, respectively, while untreated seeds resulted in only 1% GP. These treatments also highly improved mean germination time (MGT) to 1.4, 1.8 and 2.6days, respectively, when compared to 3.5days MGT of untreated seeds. The modified drum priming treatment (72h incubation after 60% SMC hydration) significantly improved results of 74% GP(on the $3^{rd}$day after sowing), 2.6days MGT and $39%{\cdot}day^{-1}$ germination rate (GR), however, untreated seeds showed 19% GP, 4.1 MGT, and $25%{\cdot}day^{-1}$ GR. Although osmotic priming after sonication, hydro priming showed similar improved germination characteristics, however, modified drum priming is considered as an industrially promising treatment methods considering the shortening of the treatment period and environment-friendly aspects.

Priming 처리가 한지형 잔디류의 발아 증진에 미치는 영향 (Effect of Seed Priming on the Enhancement of Seed Germination in Cool Season Turfgrass)

  • Kentucky bluegrass와 Tall fescue에서는 발아력을 극대화시킬 수 있는 priming 최적 처리 조건은 100mM의 $K_3PO_4$ 용액으로 $25^{\circ}C$에서 1일간 처리였다. Priming 처리된 종자는 무처리에 비해 높은 발아율을 보였고, $T_{50}$과 MDG가 단축되어 신속한 발아를 유도하였다. 반면 NaOH나 KOH는 처리농도가 높아지고 처리기간이 길어질수록 발아력이 급격하게 저하되어 priming 화학제로는 적당하지 않았다. Creeping bentgrass 및 Bentgrass에서는 priming 처리에 관계없이 발아율이 10% 미만으로 낮았다. 그러나 200mM의 $KNO_3$ 로 3일간 priming 처리하면 발아율을 6%까지 향상시킬 수 있었으나, 현저한 수준은 아니었다. Kentucky bluegrass와 Tall fescue에서 priming 처리 기간은 1일 처리가 발아율 향상에 가장 좋았고, 처리 기간이 6일로 길어질수록 발아율이 저하되었고 발아 속도는 지연되었다. 또한 priming 화학제의 처리 농도는 100 mM 처리가 발아력 증진에 좋았으나, 200 mM과 300 mM로 처리농도가 높아지면 발아율이 감소하였다.

Effects of Osmotic and Non-Osmotic Priming Methods for Enhancing Germination of Pepper Seed

  • This study was conducted to investigate the effects of osmotic priming, liquid smoke (LS), sonication and modified drum priming treatments to improve the germination speed and uniformity of pepper seeds for high quality export seeds. Seeds were treated in 0.5, 1.0, 5.0, 10% LS solution only or with 100mM $K_2SO_4$ solution for 6days at $15^{\circ}C$. Sonication treatment was performed for 5, 10, and 20 minutes at an intensity of 5.2, 10.4, 15.7, 21.0, 26.1kHz in water at $15^{\circ}C$. After sonication treatment, seeds were primed with water or 100mM $K_2SO_4$ for 4days. 40, 50 and 60% seed moisture content (SMC) of hydrated seeds were incubated for 84, 96 and 108h in a container with a relative humidity of 99% at 26rpm for a modified drum priming treatments. 0.5% LS treatment with water or 100mM $K_2SO_4$ priming showed significant effects with 71% GP (9.0days MGT) and 66% GP (7.4days MGT), respective, while untreated seeds resulted in only 61% GP and 10days MGT. The healthy seed % was increased by 24% by $K_2SO_4$ priming with LS treatment compared to untreated seeds. However, sonication treatment showed less germination or no difference compared to untreated seed, regardless of intensity and treatment time. The modified drum priming treatment (108h incubation after 50% SMC hydration) significantly improved the GP (on the $4^{th}$days after sowing), germination rate (GR), and MGT to 65%, $28%{\cdot}day^{-1}$, and 3.6days, whereas untreated seed resulted only 7% GP, $17%{\cdot}day^{-1}$ GR and 5.9days MGT. Thus, modified drum priming treatment effectively improved seed germination and is considered as an industrially promising treatment methods considering the shortening of the treatment period and environment-friendly aspects.

Effect of Seed Priming Treatment on the germination of Sesame

  • This experiment was conducted to find out optimum priming treatment conditions to the sesame seed as a preliminary study for enhancing sesame germination properties. Effective priming agents and concentrations for sesame seed were $K_3PO_4$, 200 mM and PEG6000, -1.0 MPa respectively. Optimum priming temperature and duration were $15^{\circ}C$, 4 days in view of germination speed and germination percentage. PEG6000 with -1.0 MPa was selected as an efficient priming treatment condition at $15^{\circ}C$, 4 days. This study suggested that priming treatment to sesame seed would be an effective technique enhancing sesame seed germination and shortening time to the $T_{50}$ at the field condition, but the efficiency of priming treatments to the sesame seed would be strongly dependent on individual or integrated conditions of priming agent, concentration, temperature and duration etc.

Effect of ultrasonication, light and liquid smoke treatment on germination of lettuce seeds

  • Seed priming leading to improved emergences, germination speed and uniformity under unfavorable conditions without loss of longevity are the best level of techniques. We studied the possibility to obtain primed seed with reduced mean germination time (MGT) and uniformity but with simple, fast and effective methods. The experiments were carried out at $15^{\circ}C$ for 16 hours with priming treatment. To compare the effects of each treatment, we used 200mM $CaCl_2$ priming or hydro priming or no imbibition after ultrasonication or red light treatment. The light treatment was performed by irradiating 2000 lux of red light for 15, 30, 60, and 120 minutes. Addition, in order to investigate the effect of the priming solution, treated at 200mM $CaCl_2$ for 8 hours. Ultrasonication treatment was performed for 5, 10, and 20 minutes at exposures of 13.0, 17.3, and 21.6 kHz during priming. For liquid smoke (LS) treatment, seed soaked in 0%, 0.5%, 1.0%, 5.0% and 10.0% of diluted water and 200mM $CaCl_2$ solution. After each treatment, the seeds were dried to moisture content ranged 5-8% at $25^{\circ}C$ for 24 hours. The effect of seed treatment was evaluated with germination percentage (GP), MGT, germination index(GI), germination rate(GR), Germination Uniformity(GU) and heath seed percentage(HS). For several factors tested, we found that the desired germination improvement could be obtained by treating the seed with ultrasonication at 17.3 kHz for 5 minutes in water or red light exposure at 2000 lux for 120 min in water that resulted very similar to those used to 200mM $CaCl_2$ priming for 16 hrs. However, LS treatment showed no improvement in all diluted solution. Therefore, the methods applied ultrasonication and red light treatment showed high potential for fast and easy treatment avoiding pollution of salt solution waste.

Effect of ultrasonication, salt solution and liquid smoke treatment on germination of Setaria italica seeds

  • The preference for domestic cereal crop of Setaria Italica has been increased with the recent interest in healthy foods. However, the productivity of Korean domestic produce, which has been decreasing due to the lack of cultivation technology and the low rate of mechanization during cultivation. Increase of germination ability will have a positive effect on the cultivation by reducing the labor of the manpower consuming and weeding work. Therefore, red light, ultrasonication, liquid priming and liquid smoke treatment that are effective for the germination of the seeds evaluated. The seeds of 1.4mm or more were used for the experiment. The priming solution used in the experiment was 1% $KH_2PO_4$ (74mM). During the priming, the light treated seeds at 2000 lux for 15, 30, 60 and 120 minutes. Ultrasonicationd treatment was performed for 5, 10, and 20 minutes at exposures of 60%, 80%, and 100% of ultrasound up to 21.6 kHz during priming. Light or ultrasound treated seeds transferred to priming treatment at $15^{\circ}C$ for 24 hours. The treatment of the liquid smoke was divided into the treatment of the liquid smoke alone and the treatment of the liquid smoke with the priming. The liquid smoke alone was diluted with distilled water without priming solution and the treatment of the liquid smoke was diluted with the salt priming solution. Both treatments were performed at 0.0%, 0.5%, 1.0%, 5.0%, and 10.0% of the liquid smoke (pH7) concentration at $15^{\circ}C$ for 6 hours. After each treatment, the seeds were dried to moisture content ranged 5-8% at $25^{\circ}C$ for 24 hours. All treatments showed better results than the non-treated control. Light treatment for 120 minutes improved for germination percentage (GP), Germination uniformity (GU) and heath seed percentage (HS). Ultrasonication treatment was most effective when treated with ultrasound at 21.6 kHz for 5 minutes in all germination characteristics. Ten % of the liquid smoke increase in 92% GP, 1.8 days MGT, $54%{\cdot}day^{-1}GR$, 0.76 GU and 88% HS comparing to non-treated control (72% GP, 2.3 days MGT, $45%{\cdot}day^{-1}GR$, 1.48 GU, and 63 % HS). This study showed that it is possible to obtain high germination by adding liquid smoke treatment to the seeds supplied to the farmers. The efficacy of light, ultrasonication, inorganic salt priming, and liquid smoke treatment on the seeds found in the experiment will be a positive alternative to labor force problems in the cultivation by improving germination.

Osmotic Priming 및 Solid Matrix Priming 처리에 의한 당근과 양파종자의 발아증진과 초기생육에 미치는 영향 (Effect of Osmotic and Solid Matrix Priming to Improve Germination and Early Growth of Carrot and Onion Seed)

  • 본 연구는 당근과 양파에서 발아력 증진에 미치는 osmotic priming과 SMP효과를 비교하기 위해 수행되었다. Osmotic priming과 SMP 처리과정중 작물별 수분흡수율은 처리 1시간 후 급속한 수분흡수가 이루어졌고, 두 작물 모두 osmotic priming이 SMP보다 수분흡수 속도가 빨랐다. 이러한 경향은 처리 4시간까지 유지되었다. 처리최종일의 수분흡수율은 당근과 양파에서 osmotic priming이 SMP보다 2%높았다. Osmotic priming과 SMP 처리는 두 작물 모두 발아율을 향상시키지 못했지만 $T_{50}$ 및 MDG는 단축되어 조기발아 하였으며, 이러한 경향은 저온인 $15^{\circ}C$에서 더욱 현저하였다. Osmotic priming과 SMP 처리 후 초기함수율로 재건조하면 당근에서는 발아속도에 큰 차이가 없었으나, 양파에서는 표면건조에 비해 발아속도가 지연되었다. Osmotic priming과 SMP 상호처리간 발아력을 비교한 결과 두 작물 모두 SMP 처리가 osmotic priming보다 발아촉진에 효과적이었다. 당근과 양파종자를 osmotic priming과 SMP 처리하면 묘출현율도 향상되었고, 묘출현에 소요되는 일수를 단축시켜 신속한 묘출현을 유도하였다. 그러나 묘출현에 이은 유묘의 초기생육에는 통계적인 유의성은 인정되지는 않았으나 osmotic priming과 SMP 처리된 종자는 무처리에 비해 향상되었다.

Effect of Deep Sea Water Seed Priming on the Growth of Rice (Oryza sativa L.) Seedlings

  • This experiment was conducted on rice (cv. 2005 Thaoi) seeds to study whether priming with deep sea water (DSW) results in enhancement of seed emergence and seedling growth and to identify the optimum concentration of Deep Sea Water (DSW) for priming. Two experiments were conducted subsequently. In experiment 1, four concentrations of the DSW (10%, 20%, 30% and 40%), and in experiment 2, five concentrations of DSW (10%, 15%, 20%, 25% and 30%) were prepared and seeds were primed for 24 hours duration at $25^{\circ}C$. Beside this, hydro priming with plain water was also included as a control. Experiments were laid out in Completely Randomized Design (CRD) with three replications. Result showed that 20% DSW seed priming treatment had improved the emergence, seedling height, number of roots and root length as compare to other with DSW or without DSW treatments. Beyond 20% DSW priming (i.e. 25%, 30% and 40%) were not suitable for priming the seed. On the basis of seedlings growth parameters; emergence, seedling height, root number and length, and shoot root ratio, 20% DSW priming was the best priming treatment.
