고율 조류 생세포체 배양지에서 조사 조건으로 본 조류 배양 특성 (Algae Culture Characteristics Viewed with Continuous and Cyclic Irradiation in High Rate Algae Biomass Culture Pond)

  • The utilization methods of algae biomass have been studied constantly in whole world. These are $\circled1$the wastewater treatment if waste stabilization pond and oxidation ditch etc. and $\circled2$the biosorption of heavy metals and recovery of strategic' precious metals and $\circled3$the single-celled protein production and the production of chemicals like coloring agent and $\circled4$the production of electric energy through methane gasification. The culture system also has been developed constantly in relation with such utilization method developments. In the result of experimental operation under continuous and cyclic irradiation of light, using high rate algae biomass culture pond(HRABCP), which had been made so as to be an association system with the various items which had been managed to have high efficiency for algae culture, the algae production of the 12 hours-irradiance pond was 41.48 Chlorophyll-a ${\mu}g/L$ only in spite of having the more chance of $CO_2$ synthesis to algae cell than the 24 hours-irradiance pond. This means that the energy supply required for dark-reaction of photosynthesis is very important like this. The difference of algae production between continuous and cyclc irradiation explains that the dark-reaction of photosynthesis acts on algae production as the biggest primary factor. The continuous irradiance on HRABCP made the good algae-production($1403.97{\;}{\mu}g$ Chlorophyll-a/mg) and the good oxygen-production(5.8 mg $O_2/L$) and the good solid-liquid seperation. especially, DO concentration through the oxygen-production was enough to fishes' survival.

공유관리와 간호단위문화에 대한 조사;세개 종합병원 대상 (A Comparison of Shared Governance and Nursing Unit Culture in Three Hospitals)

  • The primary purpose this descriptive study was to identify, describe, and compare the patterns of shared governance and nursing unit culture in the hospital settings. The secondary purpose of the research was to identify, describe whether the participation style or responsibility style of nursing management activities shared through nurses in a consistent way. Methodology included survey and in-depth interviews with a total 145 members of 15 nursing units in 3 hospitals. One was a national hospital, another was a corporation hospital and the other was a teaching hospitals. Conclusions from this research included the following: 1. The degree of shared governance in nursing management activities was the highest in the corporation hospital. 2. In the participation style of nursing management activities, 'all participation' was the highest in the corporation hospital. 'Nursing administration only' was the highest in the national hospital. 3. Distribution of responsibility style differed from that of participation style. Three hospitals showed high in nursing management activities such as 'nursing administration only' and 'head nurse only' style. 4. Five experts surveyed showed that the ideal level of nurses' participation in nursing management activities was a traditional nursing governance pattern. 5. There was a distinct difference in the nursing unit culture throughout the institutions in the professional growth. 6. There was no significant difference in the same nursing units of three hospitals in nursing unit culture. According to these results, the following implications can be made; 1. In nursing administration, there should be an emphasis on preparing staff nurses' potential decision-making ability through continuing education so that staff nurses' autonomy and responsibility will be developed and increased. 2. It is necessary to develop a strategic nursing unit for improving nursing quality in hospital setting. 3. The relationship of shared governance, nursing unit culture and nursing outcome should be researched further.

Establishment and characterization of gastric surface mucous cell lines (GSM06 and GSM10) from transgenic mice harboring temperature-sensitive simian virus 40 large T-antigen gene

  • In the present study, in order to make an in vitro model of gastric mucosa for physiological and pharmacological studies, we established two immortalized gastric surface mucous cell lines (GSM06 and GSM10), which produce periodic acid-Schiff (PAS)-and concanavalin A (Con A)-positive glycoproteins, from a primary culture of gastric fundic mucosal cells of adult transgenic mice harboring a temperature-sensitive simian virus 40 large T-antigen gene 〔1]. Gastric fundic mucosal cells were isolated as a modification of a previously described method for rats by Schepp et al. (2). The isolated gastric fundic mucosal cells were cultured in DME/F12 medium supplemented with 2% fetal bovine serum (FBS), 1% ITES (consisting of 2 mg/1 insulin, 2 mgg/1 transferrin, 0.122 mg/1 ethanolamine and 0.00914 mg/1 sodium selenite) and 10 ng/ml recombinant epidermal growth factor (EGF) in a collagen-coated culture dish. To remove fibroblastic cells from the culture, gastric mucosal cells were incubated in the culture medium containing dispase (25 U/ml) for 24 h. The cells, uncontaminated with fibroblastic cells, were then cloned by colony formation. In our series of three attempts, two cell lines (GSM06 and GSM10) have been established at last. The cells proliferated, attached to the dish ana grew until confluent monolayers were formed, and maintained tight contact with neighboring cells. Alteration of Ryanodine-receptors in Cultured Rat Aortic Smooth Muscle Cells

  • Vascular smooth muscle cells can obtain a proliferative function in environments such as atherosclerosis in vivo or primary culture in vitro. Proliferation of vascular smooth muscle cells is accompanied by changes in ryanodine receptors (RyRs). In several studies, the cytosolic $Ca^{2+}$ response to caffeine is decreased during smooth muscle cell culture. Although caffeine is commonly used to investigate RyR function because it is difficult to measure $Ca^{2+}$ release from the sarcoplasmic reticulum (SR) directly, caffeine has additional off-target effects, including blocking inositol trisphosphate receptors and store-operated $Ca^{2+}$ entry. Using freshly dissociated rat aortic smooth muscle cells (RASMCs) and cultured RASMCs, we sought to provide direct evidence for the operation of RyRs through the $Ca^{2+}$- induced $Ca^{2+}$ -release pathway by directly measuring $Ca^{2+}$ release from SR in permeabilized cells. An additional goal was to elucidate alterations of RyRs that occurred during culture. Perfusion of permeabilized, freshly dissociated RASMCs with $Ca^{2+}$ stimulated $Ca^{2+}$ release from the SR. Caffeine and ryanodine also induced $Ca^{2+}$ release from the SR in dissociated RASMCs. In contrast, ryanodine, caffeine and $Ca^{2+}$ failed to trigger $Ca^{2+}$ release in cultured RASMCs. These results are consistent with results obtained by immunocytochemistry, which showed that RyRs were expressed in dissociated RASMCs, but not in cultured RASMCs. This study is the first to demonstrate $Ca^{2+}$ release from the SR by cytosolic $Ca^{2+}$ elevation in vascular smooth muscle cells, and also supports previous studies on the alterations of RyRs in vascular smooth muscle cells associated with culture.

요하네스 베르메르의 작품을 통해 본 17세기 네덜란드 여성 시민복과 시민문화 (The Civil Culture and the Civil Costume of Netherlands Women in the 17th Century through the works of Johannes Vermeer)

  • This thesis aims to investigate the characteristics of the civil costume in Netherlands women and the way how the civil culture was reflected on these by analyzing the women's costume depicted in the works of the Johannes Vermeer regarded as the representative painter of Netherlands in 17th century. The method of study was to select 24 pieces of Vermeer's works among the 30 pieces, and were analyzed in detail. These were approved to be common civil costumes by researching the works of other painters in that era on the other hand. The result shows that the civil costume of Netherlands women from 1653 until 1675 was in the simple form of two pieces dress, and minimal decorations with simple hair style and headdress would take the constitution of the frugality, chastity and practicality as the mainstream, along with using the red, yellow and blue as the primary three colors on to the costumes. These characteristics might be ascribed to the Netherlands civil culture influenced by the Calvinism that emphasized the frugality and chastity, denouncing the luxury with supporting the religion and morality. This trend was also noted in the men's costume, giving evidence of the intimate relationship between the costume, religion and civil culture. This thesis might be a help to elucidate the relationship between the costume and cultural society, and be a affordable tools to study the contemporary costume.

조선 중기 두류 수급 현황과 콩 음식 문화 고찰 - 오희문(吳希文)의 『Shaemirok (쇄미록(瑣尾錄))』을 중심으로 - (Study on the Consumption Status of Beans and the Soybean Food Culture in the Mid-Joseon Period According to Shamirok)

  • This study researched the food culture and bean economy of the Joseon dynasty during the 16th century and according to the primary lifestyle reference "Shaemirok (?尾錄)". The research analyzed the textual contents of the "Shaemirok (?尾錄)". It is clear that the people of the Joseon dynasty produced more beans than grain, at a ratio of 41 to 50, respectively. The soy bean sauce consumption was split into family consumption and non-family consumption. It was evident that there was more family consumption compared to that of non-family consumption at a ratio of 7 to 3, respectively. People of the Joseon dynasty annually recorded their way of making soy sauce from 1595 to 1600. The Joseon writers edited the record six times for making meju and four times for making soy sauce. The recorded ratio displays the ingredients of soy sauce, which were: 6 Du of Mal Jang and 2 Du of salt. Mal Jang and salt had a three to one ratio, respectively. The most mentioned food was Tofu during the mid-Joseon period with fifty six mentions. The Joseon people regarded making Tofu in a Buddhist temple as a family-bonding experience. Porridge was the second most prominent food next to Tofu, among the bean-related food. Porridge appears thirty five times. There were 3 types of porridge named: bean porridge, bean powder porridge and mung bean porridge.

Screening of Endophytic Fungal Isolates Against Raffaelea quercus-mongolicae Causing Oak Wilt Disease in Korea

  • Oak wilt disease caused by Raffaelea quercus-mongolicae has emerged obviously in Korea. We selected antifungal isolates against R. quercus-mongolicae among 368 endophytic fungal isolates from different parts of oak and pine trees. The experiment was conducted in the primary and secondary screenings by dual culture test. The antifungal activity of the selected isolates was assessed in culture filtrate test based on the inhibition rates in mycelial growth, sporulation, and spore germination of oak wilt fungus. Five isolates, E089, E199, E282, E409 and E415, showed strong antifungal activity in culture filtrate test, and their antifungal activity decreased on the culture media supplemented with heated culture filtrate. Higher mycelial growth inhibitions on the unheated media were recorded in E409 (Colletotrichum acutatum), E089 (Daldinia childiae), E415 (Alternaria alternata) and E199 (Daldinia childiae) with the inhibition rates of 79.0%, 70.1%, 68.9% and 64.5%, respectively. These isolates also had the higher sporulation inhibitions on unheated media with the rates of 96.8%, 84.2%, 82.8% and 80.5%, respectively. The spore germination of the oak wilt fungus was completely inhibited by E282 (Nectria balsamea) on both unheated and heated media. These results showed that a higher number of potent antifungal isolates against oak wilt fungus was isolated from the petiole compared to the other parts. This study could contribute to the development of biological control approaches for the management of oak wilt disease caused by R. quercus-mongolicae.

Training of Future Specialists in Modern Conditions: Cultural Aspects

  • An increasing number of students from different cultures study in higher primary schools. This trend is due to: 1) the government's discourse on increasing the level of participation of foreign students in national educational programs and the need for internationalization; 2) the need of employers for professionals who are trained to work in a multicultural environment to meet the needs of different markets and customers. Methodology. This study is based on the results of the OECD (2018) structured survey of 1,093 teachers at universities in Australia, Colombia, the Czech Republic, Denmark, Georgia, Malta, Vietnam, Turkey, and Argentina, examined policy, the practice of cultural characteristics in training specialists, and teachers' attitudes to cultural diversity. Results. The attitude and perception of cultural features by teachers does not determine the practice of forming a cultural environment and managing this environment to ensure quality education of students of different nations. The main culturological aspects of training are self-expression of cultural and ethnic identities, expression of cultural characteristics and their value through multicultural activities in universities, teaching students to combat ethnic or cultural discrimination. Therefore, the formation of a multicultural environment in higher education occurs through the activities of students and teachers, which complement each other. The practical value lies in identifying two important components of the formation of cultural diversity among students, such as self-expression of ethnic and cultural identity and the expression of cultural differences by teachers in the course of educational activities.

Development of a Family Nursing Model for Prevention of Cancer and Other Noncommunicable Diseases through an Appreciative Inquiry

  • Background: Cancer and non-communicable diseases are a major issue not only for the developed but also developing countries. Public health and primary care nursing offer great potential for primary and secondary prevention of these diseases through community and family-based approaches. Within Thailand there are related established educational curricula but less is known about how graduate practitioners enact ideas in practice and how these can influence policy at local levels. Aim: The aim of this inquiry was to develop family nursing practice in primary care settings in the Isaan region or Northeastern Thailand and to distill what worked well into a nursing model to guide practice. Materials and Methods: An appreciative inquiry approach involving analysis of written reports, focus group discussions and individual interviews was used to synthesize what worked well for fourteen family nurses involved in primary care delivery and to build the related model. Results: Three main strategies were seen to offer a basis for optimal care delivery, namely: enacting a participatory action approach mobilizing families' social capital; using family nursing process; and implementing action strategies within communities. These were distilled into a new conceptual model. Conclusions: The model has some features in common with related community partnership models and the World Health Organization Europe Family Health Nurse model, but highlights practical strategies for family nursing enactment. The model offers a basis not only for planning and implementing family care to help prevent cancer and other diseases but also for education of nurses and health care providers working in communities. This articulation of what works in this culture also offers possible transference to different contexts internationally, with related potential to inform health and social care policies, and international development of care models.

배양세포의 성염색질에 관한 세포학적 연구 1. 배양기간을 달리한 고양이, 몰못트, 닭의 배양세포에 있어서의 성염색질에 대하여

  • The frequency and pattern of sex chromatin in primary cultured cells of kidney cortex of cats and guinea-pigs, and muscle of chick embroys were examined and compared to those of in vivo condition, with special reference to the various cultured stages. 1. In cat, the frequencies of sex chromatin positive I of peripheral position were average 62.7% in female, and 15% in male, whereas those of non-peripherla position were 5.8% in female and 0.1% in male. The incident proportion between them showed a marked difference-approximately 10 times higher in female than male. These results failry indicated that a distinct nuclear dimorphism with regard to the sex chromatin positive I was established in cultured cells. The position of sex chormatin was usually peripheral location. The tendency of frequencies , with reference to the cultured stages, was low count in primary extracted and initial stage cells , but it showed a peaked frequency in 10-13 days after primary culture, and after that the frequencies were decreased gradually. Compared between I vitro and in vivo condition of the same tissues, the cells in vivo exhibited the sex chromatin in high frequency at the peak showed stage. 2. In guinea-pig , the frequencyies of peripheral positive I were 36.8% in female and 6.3% in male, while non-peripheral positions were 6.1% in female and 3.5% inmale. Its incident was a rate of nearly 4 times higher in female than male. The nuclear dimorphism was also established in cultrued cells of guinea-pig. The position and the incident frquency showed a similar pattern as in cat except the primary extracted cells. 3. In chick embryo, the frequencies of sex chromatin positive I of peripheral position were 38.2% in female, and 18.3% in male, non-peripheral position, however, was hardly to find. These results suggest that the definite sexual dimorphism was unable to find in chick embryo cultured cells. The position and the incident tendency were a similar pattern as in above mammals and the frequency was higher in vitro cells.
