Analysis of Total Bacteria, Enteric Members of γ-proteobacteria and Microbial Communities in Seawater as Indirect Indicators for Quantifying Biofouling

  • In this study, total bacteria, enteric members of the $\gamma$-proteobacteria, and microbial communities in seawater were analyzed as indirect indicators for quantifying biofouling. Biomass in seawater can significantly affect feed water pretreatment and membrane biofouling of reverse osmosis desalination processes. The purpose of this paper is to investigate microbiological quantity and quality of seawater at the potential intake of a desalination plant. For this analysis, the total direct cell count (TDC) using 4'-6-diamidino-2-phenylindole (DAPI)-staining and DNA-based real-time PCR were used to quantify the total bacteria and relative content of enteric members of $\gamma$-proteobacteria in seawater, respectively. In addition, microbial communities were examined using 16S rRNA gene cloning and bacterial isolation to identify the most abundant bacteria for a further biofouling study. The experimental results of this study identified about $10^6$ cells/mL of (total) bacteria, $10^5$ 16S rRNA gene copies/mL of enteric $\gamma$-proteobacteria, and the presence of more than 20 groups of bacteria.

Regulation of AQP-4 Water Channel Expression in the Brain during Development and by Ischemia

  • Water transport is mediated by two distinct pathways, diffusional and channel-mediated water transport. The first molecular water channel was identified from human erythrocytes in 1992. Genetically-related proteins from other mammalian tissues have subsequently been identified to transport water, and the group is referred to as th "Aquaporins". Aquaporin-4 (AQP4) is most abundant in the brain, which may be involved in CSF reabsorption and osmoregulation. However, ontogeny and regulatory mechanisms of AQP4 channels have not been reported. Northern blot analysis showed that AQP4 mRNA began to be expressed in the brain just before birth and that its expression gradually increased by PN7 and then decreased at adult level. AQP4 was expressed predominantly in the ependymal cells of ventricles in newborn rats. And then its expression decreased in ependymal cells and increased gradually in other regions including supraoptic and paraventricular nuclei. AQP4 is also expressed in the subfornical organ, in which the expression level is not changed after birth. Cryogenic brain injury did not affect expression of AQP4 mRNA, while ischemic brain injury decreased it. Osmotic water permeability of AQP4 channel expressed in Xenopus oocytes was inhibited by the pretreatment of BAPTA/AM and calmidazolium, a $Ca^{2+}/Calmodulin$ kinase inhibitor, in a dose-dependent manner. These results indicate that the expression and the function of AQP4 channel are regulated by developmental processes and various pathophysiological conditions. These results will contribute to the understanding of fluid balance in the central nervous system and the osmoregulatory mechanisms of the body.

Recovery of Xylo-oligomer and Lignin Liquors from Rice Straw by Two 2-step Processes Using Aqueous Ammonia Followed by Hot-water or Sulfuric Acid

  • A two-step process was investigated for pretreatment and fractionation of rice straw. The two-step fractionation process involves first, soaking rice straw in aqueous ammonia (SAA) in a batch reactor to recover lignin-rich hydrolysate. This is followed by a second-step treatment in a fixed-bed flow-through column reactor to recover xylo-oligomer-rich hydrolysate. The remaining glucan-rich solid cake is then subjected to an enzymatic process. In the first variant, SAA treatment in the first step dissolves lignin at moderate temperature (60 and $80^{\circ}C$), while in the second step, hot-water treatment is used for xylan removal at higher temperatures ($150{\sim}210^{\circ}C$). Under optimal conditions ($190^{\circ}C$ reaction temperature, 30 min reaction time, 5.0 ml/min flow rate, and 2.3 MPa reaction pressure), the SAA-hot-water fractionation removed 79.2% of the lignin and 63.4% of the xylan. In the second variant, SAA was followed by treatment with dilute sulfuric acid. With this process, optimal treatment conditions for effective fractionation of xylo-oligomer were found to be $80^{\circ}C$, 12 h reaction time, solid-to-liquid ratio of 1:12 in the first step; and 5.0 ml $H_2SO_4/min$, $170^{\circ}C$, and 2.3 MPa in the second step. After this two-step fractionation process, 85.4% lignin removal and 78.9% xylan removal (26.8% xylan recovery) were achieved. Use of the optimized second variant of the two-step fractionation process (SAA and $H_2SO_4$) resulted in enhanced enzymatic digestibility of the treated solid (99% glucan digestibility) with 15 FPU (filter paper unit) of CTec2 (cellulase)/g-glucan of enzyme loading, which was higher than 92% in the two-step fractionation process (SAA and hot-water).

AE-SOM을 이용한 EVA 생산 공정 이상 검출 및 진단 (Fault Detection and Diagnosis for EVA Production Processes Using AE-SOM)

  • 본 연구에서는 auto-encoder와 self-organizing map을 결합한 auto-encoder with self-organizing map(AE-SOM) 기법을 이용하여 EVA 생산공정의 이상을 검출 및 진단하였고, Granger의 인과분석을 통해 이상 검출 데이터의 이상 전파 방향을 확인하였다. 분석 데이터는 1년 7개월 간의 조업데이터를 이용하였으며, autoclave 반응기의 조업 변수를 주로 분석하였다. 데이터 전처리 과정에서 데이터의 표준화를 먼저 진행하고, 조업의 각 grade의 sample 수를 동일하게 200개 임의로 추출하였다. 이후 AE-SOM을 적용하여 각 grade의 best matching unit (BMU)를 도출하였다. 각각의 BMU를 기준으로 조업 데이터가 얼마나 벗어났는지를 기준으로 데이터의 이상을 판별하였다. 공정 이상이 발견될 시 이상원인을 contribution plot을 이용하여 확인하였고 이상원인 변수의 인과성을 Granger의 인과분석을 통해 분석하였다. 그 결과 조업 시 발생한 2번의 셧다운의 전조를 모두 검출하였으며 이상이 발생한 원인변수에서 기인한 공정 이상의 전파 방향을 분석하였다.

미세결정셀룰로오스의 제조를 위한 억새 바이오매스의 처리 및 특성연구 (Study of Preparation and Characterization of Microcrystalline cellulose from Miscanthus sinensis)

  • Microcrystalline cellulose (MCC) was prepared from Miscanthus in this study. Two pulping methods, soda pulping and alkaline sulfite pulping were applied as a pretreatment process. After pulping, two different bleaching processes such as $ClO_2$ treatment followed by $H_2O_2$ treatment and $O_3$ treatment followed by $H_2O_2$ treatment were carried out. Two concentration of $H_2SO_4$, 47% and 57% were applied to the purified Miscanthus cellulose as a acid hydrolysis process to make MCC. The crystallinity index and morphological properties of the produced MCC were evaluated with X-ray diffractometer and scanning electron microscopy. The MCC originated from the soda pulping sample showed the higher crystallinity index than that originated from the alkaline sulfite pulping sample. The two stages of treatmen twith $O_3$ and $H_2O_2$ resulted in the higher purified cellulose products.

FGF-2 inhibits TNF-α mediated apoptosis through up-regulation of Bcl2-A1 and Bcl-xL in ATDC5 cells

  • FGF-2 is involved in cell survival, proliferation, apoptosis, and angiogenesis in a wide variety of cells. FRGRs, PI3K and MAP kinases are well known mediators of FGF signaling. Despite its known roles during many developmental processes, including osteogenesis, there are few known targets of FGF-2. In the present study, we identified Bcl2-A1 and Bcl-xL as two prominent targets involved in promoting cell survival. Pretreatment of ATDC5 cells with FGF-2 increased cell survival, while siRNAs specific for Bcl2-A1 and Bcl-xL compromised the anti-apoptotic effect of FGF-2, sensitized the cells to apoptosis triggered by TNF-${\alpha}$. Chemical inhibition of FGFR, NFkB, and PI3K activity by PD173074, pyrrolidine dithiocarbamate, and LY294002 respectively abrogated the FGF-2-mediated induction of Bcl2-A1 and Bcl-xL expression. Taken together, our data demonstrate that a subset of Bcl2 family proteins are the targets of FGF-2 signaling that promotes the survival of ATDC5 cells.

PCC 제조를 위한 칼슘이온 추출 조건에 관한 연구 (A Study on the Calcium Ion Extraction for PCC Production)

  • 본 연구에서는 PCC (precipitated calcium carbonate) 제조에 있어 칼슘이온 추출 최적화를 위하여 추출 용제, 용제의 농도, 추출원의 투입량, 전처리 공정 등 다양한 추출 조건에 따른 실험을 수행하였다. 칼슘추출원으로 CaO를 사용하였으며, 칼슘이온의 추출량과 CaO의 입자크기를 확인하기 위하여 ICP와 SEM 분석을 수행하였다. 그 결과 2 M의 hydrochloric acid를 용제로 사용한 경우 100%의 칼슘이온이 추출되었으며, 추출원의 최적 투입량은 6 g으로 확인하였다. 반면 반응시간, 반응온도, 입자 분쇄 및 열처리 공정은 칼슘이온 추출량에 큰 영향이 없음을 확인하였다.

Quercetin Ameliorates NO Production via Down-regulation of iNOS Expression, $NF{\kappa}B$ Activation and Oxidative Stress in LPS-Stimulated Macrophages

  • Effect of quercetin on NO production and regulation mode of quercetin on oxidative stress, $NF{\kappa}B$ activation, and iNOS expression, possible mechanisms of NO suppression in LPS-stimulated macrophages were investigated. Treatment of RAW 264.7 cells with quercetin significantly reduced lipopolysaccharide (LPS)-stimulated nitric oxide (NO) production dose-dependently ($IC_{50}$, $9.2\;{\mu}M$). Expression of iNOS and specific DNA binding activities of nuclear factor kB ($NF{\kappa}B$) were significantly suppressed by quercetin pretreatment. Quercetin reduced thiobarbituric acid-reactive substances (TBARS) accumulation, enhancing GSH level and antioxidant activities of enzymes, such as superoxide dismutase (SOD) and catalase. These results demonstrate quercetin may ameliorate inflammatory diseases by suppressing NO production through inhibition of iNOS expression, $NF{\kappa}B$ transactivation, and oxidative stress, which may be mediated partially by antioxidative effect of quercetin. Thus, quercetin appears to be used as a potential therapeutic agent for treating LPS-induced inflammatory processes.

자연수 중의 미량금속 분석을 위한 청결기술 (Clean techniques for trace metal analysis in natural waters)

  • Al, Ag, Au, Cu, Cd, Co, Fe, Ni, Ph, Zn 등과 밀접한 금속은 일반적인 원소이며, 지속적인 오염물질에 속한다. 금속은 생명체의 생리활동에 필수원소인 경우도 있으나 경우도 있으나 그 중의 일부는 정해진 한계값을 초과할 때에는 독성을 가진다. 자연수 중의 미량금속 측정은 정확도와 정밀도에 있어서 큰 오차를 가지고 있음이 장기간 인식되어 왔다. 미국과 유럽에서는 1975 년 이후 미량금속의 해수 중 농도가 과거에 인지된 농도보다 $1/10{\sim}1/1,000$ 정도 낮아졌으며, 수직 분포는 알려진 생물, 물리, 지화학적인 과정들과 일치하고 있음이 밝혀졌다. 이와 같은 결과는 대부분 미량원소에 대한 분석방법과 기기의 발전에 기인하며, 시료 채취, 보관 및 분석 과정에 발생할 수 있는 오염을 제거해야 하는 세심한 주의 요구되어 진다.

Preventive Effect of Hwangryunhaedok-tang on Inflammatory Responses in PHA-stimulated Peripheral Blood Mononuclear Cells from Cerebral Infarction Patients

  • Objectives: Hwangryunhaedok-tang (HRHDT), a prescription composed of four herbs, has been wi dely used in Oriental Medicine for the treatment of cerebral infarction. However, the mechanisms by which the herbal formula affects on the production of pro- and anti-inflammatory cytokines in cerebral infarction patients remain unknown yet. Methods: The levels of pro- and anti-inflammatory cytokines, including tumor necrosis factor (TNF)-a, interleukin (IL)-1b, and IL-6, IL-10, and TGF-${\beta}1$ were determined in peripheral blood mononuclear cells (PBMCs) from cerebral infarction patients under our experimental conditions. Results: The secretory levels of pro- and anti-inflammatory cytokines, including tumor necrosis factor (TNF)-a, interleukin (IL)-1b, and IL-6, and IL-10 were significantly increased in phytohemagglutinin (PHA)-stimulated peripheral blood mononuclear cells (PBMCs) from cerebral infarction patients. However, pretreatment with HRHDT significantly inhibited the secretion of pro- and anti-inflammatory in PBMCs. Also, HRHDT induced a significant increase of transforming growth factor (TGF)-b1 in PBMCs. Conclusions: These data indicate that HRHDT may be beneficial in the suppression of inflammatory processes of cerebral infarct through suppression of TNF-$\alpha$, IL-$1{\beta}$, IL-6, and IL-10 and induction of TGF-${\beta}1$.
