Optimization of Analytical Method for Annatto Pigment in Foods (식품 중 안나토색소 분석법 최적화 연구)

  • In this study we sought to develop a simultaneous analysis method for cis-bixin and cis-norbixin, the main components, to detect annatto pigment in food. To establish the optimal test method, the HPLC analysis methods of the European Food Safety Authority (EFSA), Japan's Ministry of Health, Labor and Welfare (MHLW), and National Institute of Food and Drug Safety Evaluation (NIFDS) were compared and reviewed. In addition, a new pretreatment method applicable to various foods was developed after selecting conditions for simultaneous high-performance liquid chromatography (HPLC) analysis in consideration of linearity, limit of detection (LOD), limit of quantification (LOQ), and analysis time. The HPLC analysis method of NIFDS showed the best linearity (R2 ≥ 0.999), exhibiting low detection and quantification limits for cis-norbixin and cis-bixin as 0.03, 0.05 ㎍/mL, and 0.097, 0.16 ㎍/mL, respectively. All previously reported pretreatment methods had limitations in various food applications. However, the new pretreatment method showed a high recovery rate for all three main food groups of fish meat and meat products, processed cheese and beverages. This method showed an excellent simultaneous recovery rate of 98% or more for cis-bixin and cis-norbixin. The HPLC analysis method with a new pretreatment method showed high linearity with a coefficient of determination (R2) of 1 for both substances, and the accuracy (recovery rate) and precision (%RSD) were 98% and between 0.4-7.9, respectively. From this result, the optimized analytical method was considered to be very suitable for the simultaneous analysis of cis-bixin and cis-norbixin, two main components of annatto pigment in food.

Synchronized Synergism Using Ethanol, L-lysine and $NaBH_4$ Glutaraldehyde Treated Porcine Pericardium (글루타르알데하이드 고정 돼지 심낭에서 Ethanol, L-lysine, $NaBH_4$ 병합 처치시 상승효과)

  • Background: Calcification is the most frequent cause of clinical failure of bioprosthetic tissues that are fabricated from Glutaraldehyde (GA)-fixed porcine valve or bovine pericardium. We recently used a multi-factorial approach of employing different mechanisms to investigate how to reduce the calcification of bioprosthetic tissues. The purpose of the present study was to evaluate the synchronized synergism using ethanol, L-lysine and $NaBH_4$ in glutaraldehyde treated porcine pericardium from the standpoint of calcification and tissue elasticity. Material and Method: Porcine pericardium was fixed with 0.625% GA (commercial fixation). An interim step of ethanol (80%; 1 day at room temperature) or L-lysine (0.1 M; 2 days at $37^{\circ}C$) or $NaBH_4$ (0.1 M; 2 days at room temperature) was followed by completion of the GA fixation (2 days at $4^{\circ}C$ and 7 days at room temperature). The tensile strength and thickness of the samples were measured. The treated pericardiums were implanted subcutaneously into three-week old Sprague-Dawley rats for 8 weeks. The calcium content was assessed by atomic absorption spectroscopy and the histology of the samples. Result: The amount of calcium in the pericardium pretreated with ethanol (13.6${\pm}$10.0 ug/mg, p=0.008), L-lysine (15.3${\pm}$1.0 ug/mg, p=0.002) and both (16.1${\pm}$11.1 ug/mg, p=0.012) was significantly reduced compared with the control (51.2${\pm}$8.5 ug/mg). However, $NaBH_4$ pretreatment (65.7${\pm}$61.8 ug/mg, p=0.653) and combined pretreatment that including ethanol, L-lysine and $NaBH_4$ (92.9${\pm}$58.3 ug/mg, p=0.288) were not significantly different from the controls(51.2${\pm}$8.5 ug/mg). Both the combined pretreatment using ethanol and L-lysine (7.60${\pm}$1.55, p=0.76) and the combined pretreatment that included ethanol, L-lysine and $NaBH_4$ (7.47${\pm}$1.85, p=0.33) increased the tensile strength/thickness ratio compared with that of the controls (4.75${\pm}$1.88). Conclusion: The combined pretreatment using ethanol and L-lysine seemed to decrease the calcification of porcine pericardium fixed with glutaraldehyde, as compared to single pretreatment, and it increase the tissue elasticity, but to the degree that showed synchronized synergism. $NaBH_4$ pretreatment seemed to increase the calcification of porcine pericardium, irrespective of whether single or combined pretreatment was used.

Comparison of the saccharide content of spent mushroom (Pleurotus ostreatus, Pleurotus eryngii, and Flammulina velutipes) substrates under various pretreatment conditions (전처리 방법에 따른 느타리, 큰느타리 및 팽이버섯 수확후 배지의 당함량 비교분석)

  • A new method to utilize spent mushroom substrates (SMS) for ethanol production was investigated. Analysis of the chemical properties of SMS revealed that they were decomposed by the mushrooms during cultivation. In particular, the free sugar content in SMS was reduced to half of that in mushrooms. Of the tested SMS, the Pleurotus eryngii SMS was determined to be suitable for saccharification. Upon pretreatment with a 1% alkaline solution, Pleurotus eryngii SMS achieved 80.7% of its maximum saccharification ratio. The optimum pretreatment conditions for enzyme saccharification were 1% NaOH solution at $120^{\circ}$ for 60 min. Further studies are required to determine ethanol production using Pleurotus eryngii SMS.

  • Liberation of fermentable sugars from lignocellulosic biomass is one of the key challenges in production of cellulosic ethanol. Aqueous ammonia cleaves ether and ester bonds in lignin carbohydrate complexes. It is an effective swelling reagent for lignocellulosic biomass. The aqueous ammonia pretreatment selectively reduces the lignin content of biomass. However, at high temperatures, this process solubilizes more than 50% of the hemicellulose in the biomass. Here we conducted a SAA(Soaking in Aqueous Ammonia) process by moderate reaction temperatures at atmospheric pressure using various lignocellulosicbiomass. The optimum condition of this process was 15 wt% of aqueous ammonia at 50 of reaction time during 72 hr. The delignification was up to 60% basis on initial biomass and the enzymatic digestibility was 60-90% for agricultural biomass, respectively.

Involvement of Adenosine in Cardioprotective Effect of Catecholamine Preconditioning in Ischemia-Reperfused Heart of Rat

  • Preconditioning of a heart with small doses of catecholamines induces a tolerance against the subsequent lethal ischemia. The present study was performed to find a specific receptor pathway involved with the catecholamine preconditioning and to test if adenosine plays a role in this cardioprotective effect. Isolated rat hearts, pretreated with small doses of ${\alpha}-\;or\;{\beta}-adrenergic$ agonists/antagonists, were subjected to 20 minutes ischemia and 20 minutes reperfusion by Langendorff perfusion method. Cardiac mechanical functions, lactate dehydrogenase and adenosine release from the hearts were measured before and after the drug treatments and ischemia. In another series of experiments, adenosine $A_1\;or\;A_2$ receptor blockers were treated prior to administration of adrenergic agonists. Pretreatments of a ${\beta}-agonist,\;isoproterenol(10^{-9}{\sim}10^{-7}\;M)$ markedly improved the post-ischemic mechanical function and reduced the lactate dehydrogenase release. Similar cardioprotective effect was observed with an ?-agonist, phenylephrine pretreatment, but much higher $concentration(10^{-4}\;M)$ was needed to achieve the same degree of cardioprotection. The cardioprotective effects of isoproterenol and phenylephrine pretreatments were blocked by a ${\beta}_1-adrenergic$ receptor antagonist, atenolol, but not by an ${\alpha}_1-antagonist,$ prazosin. Adenosine release from the heart was increased by isoproterenol, and the increase was also blocked by atenolol, but not by prazosin. A selective $A_1-adenosine$ receptor antagonist, 1,3-dipropyl-8-cyclopentyl xanthine (DPCPX) blocked the cardioprotection by isoproterenol pretreatment. These results suggest that catecholamine pretreatment protects rat myocardium against ischemia and reperfusion injury by mediation of ${\beta}_1-adrenergic$ receptor pathway, and that adenosine is involved in this cardioprotective effect.

Ganoderma lucidum균 전처리를 이용한 볏짚의 상압.소다펄프화

  • This study was carried out to develop the biochemical pulping method to enhance energy saving and decrease the capital cost through the soda pulping under atmospheric pressure (100%). Nonwood substrates, rice straw, were pretreated by white-rot fungi, Ganoderma Iucidum. It has acquired several basic data that can be applied in bio-soda pulping. The results of this study were as follow. Without any nutrients or with glucose, N and glucose+N the weight losses of rice straws inoculated by Canoderma Iucidum were 8.5~29.8%, 9.3~32% 11.8~30.1% and 11.8~24.4% respectively for 5, 10, 15, 20, 25 and 30 days incubation. The more extending incubation periods, the more increasing weight losses. The yield of untreated rice saw was 54.8% after pulping. When any nutrients was not added or glucose, N and glucose+N were added for the pretreatment, the total yields were ranged 50.6~38.5%, 48.6~34.4%, 47.2~38.4% and 49.5~42.6% respectively for 5, 10, 15, 20, 25 and 30 days incubation. The yields were gradually decreased based on extending of incubation. The physical properties of rice straw soda pulps without fungal treatment, the density, breaking length, burst index, tear index and folding endurance were 0.24 g/$\textrm{cm}^3$, 2.32 km, 0.91 kPa $m^2$/g, 56.7 mN.$m^2$/g and 35 times, respectively. After pretreatment without any nutrients or with glucose, N and glucose+N as nutrients the density was 0.27~0.30 g/$\textrm{cm}^3$, the breaking length 3.14~5.25 km, burst index 1.42~2.78 kPa.$\textrm{m}^2$/g, tear index45.8~64.5 mN.$\textrm{m}^2$/g and folding endurance 47~288 times at all incubating periods when pulping was done. The physical properties were increased with the increasing incubation periods. However, when glucose+N was added, the physical properties were shown superior results each incubating duration.

The effect of denture base surface pretreatments on bond strengths of two long term resilient liners

  • PURPOSE. Purpose of this study was to evaluate effect of two surface treatments, sandblasting and monomer treatment, on tensile bond strength between two long term resilient liners and poly (methyl methacrylate) denture base resin. MATERIALS AND METHODS. Two resilient liners Super-Soft and Molloplast-B were selected. Sixty acrylic resin (Trevalon) specimens with cross sectional area of $10{\times}10$ mm were prepared and divided into two groups of 30 specimens each. Each group was surface treated (n = 10) by sandblasting (250 ${\mu}$ alumina particles), monomer treatment (for 180 sec) and control (no surface treatment). Resilient liners were processed between 2 poly(methyl methacrylate) surfaces, in the dimensions of $10{\times}10{\times}3$ mm. Tensile strength was determined with Instron Universal testing machine, at a crosshead speed of 5 mm/min; and the modes of failure (adhesive, cohesive or mixed) were recorded. The data were analyzed using one-way ANOVA, followed by Tukey HSD test (${\alpha}$= 0.05). RESULTS. Monomer pretreatment of acrylic resin produced significantly higher bond strengths when compared to sandblasting and control for both resilient liners (P < .001). Sandblasting significantly decreased the bond strength for both the liners when compared to monomer pretreatment and control (P < .001). Mean bond strength of Super-Soft lined specimens was significantly higher than Molloplast-B in various surface treatment groups (P < .05). CONCLUSION. Surface pretreatment of the acrylic resin with monomer prior to resilient liner application is an effective method to increase bond strength between the base and soft liner. Sandblasting, on the contrary, is not recommended as it weakens the bond between the two.

Adsorption of Heavy Metal Ions on Bark(I) (수피에 의한 중금속 흡착(I))

  • The pretreatment of bark powder with sodium hydroxide and formalin showed the most excellent adsorption ratio, but this method could not practically be used because of the occurrence of dark-colored pigments in filtrates during pretreatment. Instead, acid and formalin were the most affirmative and effective among the pretreatment methods tested, and could be used for this purpose. Among tested species, Quercus acutissima and Robinia pseudo-accacia showed the largest amount of metal adsorption, and $Pb^{2+}$ was the best(83 to 96%) among the four heavy metals tested. The order of adsorption ratios other metals was as follows; $Cu^{2+}$ > $Zn^{2+}$ > $Cd^{2+}$, and the ratio was approximately 45 to 55%. In addition, as the substrate amount increased, the amount of adsorbed heavy metals in subtrates gradually increased, but the adsorbed amount was not proportional to the substrate amount. The order of heavy metal adsorption was as follows; $Pb^{2+}$ > $Cu^{2+}$ > $Cd^{2+}$ > $Zn^{2+}$. Depending on flow rate and column size, pine bark power adsorbed more heavy metals in the 5ml/min flow rate and 3.5cm column size rather than the 10ml/min and 2.0cm. However, oak bark power showed contrary results compared with pine bark powder. The adsorption of $Pb^{2+}$ occurred rapidly in the incipient stagte. Even though bark powders were repeatedly used three times, there was no change in the adsorption ratio(45%), but after four times, the adsorption ratio was significantly reduced to 35%.

A Study on the Media Treatment Technology of the High-Coloured Digital Textile Printing (고발색 디지털 프린팅을 위한 미디어 전처리 기술)

  • In recent years, the application of digital textile printing has increased. The benefits of using this method include the ease of sampling and the production of printed textiles. However, the production process of digital textile printing differs from that of conventional printing. For successful digital textile printing by ink-jet technology, the pretreatment of fabrics is very important in order to overcome the following problems. Low viscosity ink can spread easily on the textile surface leading to poor resolution. As a result, the combination of ink and pretreatment chemicals is still impractical and consequently most fabrics used in digital textile printing will require a pre treated coating in order to prevent the ink colours from bleeding on the fabric. Research presented in this paper shows some preliminary attempts to establish the relationship between the pre treatment and the digital textile printing quality. Various cotton fabrics were treated with pre treatment agents including ingredients like thickener, alkali and humectant, and then ink spread effect and colour yield of printed fabrics by reactive ink were analysed by using an optical microscope and K/S value. The results show that digital textile printing quality on cotton fabrics can be optimized with appropriate pre treatments.

Effects of Oldenlandiae Diffusae Herba Methanol Extract on Acetaminophen Induced Acute Liver Injury in Mice (백화사설초 메탄올 추출물이 acetaminophen으로 유도된 생쥐의 급성 간손상에 대한 효능 연구)

  • Objectives : Oldenlandiae Diffusae herba has been used as a natural drug for tumor, inflammation and liver disease in traditional medicine. This study was performed in order to investigate the antioxidative effects of Oldenlandiae Diffusae herba methanol extract(ODHM) on acetaminophen induced acute liver injury in mice. Methods : In order to investigate the protective effect of ODHM on acute hepatic injury in vivo, ICR mice were pretreated with ODHM, and then treated with acetaminophen(500mg/kg). And the levels of LPO and glutathione(GSH), antioxidative enzyme activities were measured. The levels of LPO were measured by TBA method. And catalase activity was measured as the decrease in hydrogen peroxide absorbance at 240nm on spectrophotometer using 30mM hydrogen peroxide. Superoxide dismutase(SOD) was assayed by recording the inhibition of nitro blue tetrazolium reduction with xanthine and xanthine oxidase. Glutathione peroxidase(GPX) activity was determined by the modified coupled assay developed by Paglia and Lawrence. The reaction was started by addition of 2.2mM hydrogen peroxide as substrate. The change in absorbance at 340nm was measured for 1min on spectrophotometer. Glutathione-S-transferase(GST) activity was assayed with CDNB as substrate and enzyme activity of GST towards the glutathione conjugation of CDNB. And Total SH and GSH levels were measured. Results : In vivo study, LPO levels of acetaminophen treatment group were significantly higher than other groups. This increased level was significantly reduced by ODHM pretreatment. The acetaminophen treatment resulted in a decrease of catalase, GPX, SOD and GST activities. By contrast, ODHM pretreatment markedly increased compare to those of untreated groups. Total SH and GSH levels were reduced by of acetaminophen treatment, and ODHM pretreatment significantly increased GSH levels.
