Collagen 유발(誘發) 관절염(關節炎) 동물모델에 대(對)한 전침자극(電鍼刺戟)의 진통효과(鎭痛效果) 및 그 기전(機轉)에 관(關)한 연구(硏究) - serotonergic receptor(5-HT1, 5-HT2)와 관련(關聯)된 기전연구(機轉硏究) - (The Study on the Analgesic Effect and its Serotonergic Mechanism of Electroacupuncture in the Rat Model of Collagen-induced Arthritis)

  • Objective: To investigate the analgesic effect and its serotonergic mechanism, especially related with 5-HTI and 5-HT2 receptor, of electroacupuncture(EA) in the rat model of collagen-induced arthritis(CIA). Methods : Immunization of male Sprague-Dawley rats with bovine type II (C II) collagen emulsified in Freund's incomplete adjuvant, followed by a booster injection 14 days later, leads to development of arthritis in more than 70% of rats by 21 days postinjection. After three weeks of first immunization, EA stimulation(2 Hz, 0.07 mA, 0.3 ms) was delivered into Jogsamni($ST_{36}$) for 30 minutes. Analgesic effect was evaluated by tail flick latency(TFL). We compared the analgesic effect of EA with TFLs between pretreatment of normal saline and pretreatment of spiroxatrine (5-HT1 receptor antagonist, 1mg/kg, intraperitoneal) and spiperone (5-HT2 receptor antagonist, 1 mg/kg, intraperitoneal) in CIA. Results : 1. TFLs were gradually decreased in CIA as increasing severity of arthritis. 2. Jogsamni($ST_{36}$) EA stimulation in CIA increased TFLs and the effect lasted for 60 minutes. 3. Increased TFLs with Jogsamni($ST_{36}$) EA stimulation were inhibited by pretreatment of spiroxatrine and spiperone in CIA. Conclusions : Jogsamni($ST_{36}$) EA showed analgesic effects in CIA The analgesic effects of Jogsamni($ST_{36}$) EA were inhibited by spiroxatrine and spiperone pretreatment. These observations suggest that 5-HT1 and 5-HT2 serotonergic receptor, which involve the release of serotonin neurotransmitter, play an important roles in analgesic mechanism of EA stimulation.

흰쥐에 있어서 Bromobenzene전처치가 간조직 중 Glutathione 및 Glutathione S-transferase활성에 미치는 영향 (Effect of Bromobenzene Pretreatment on the Hepatic Glutathione Content and Glutathione S-transferase Activity in Bromobenzene Treated Rats)

  • To evaluate the effect of bromobenzene pretreatment on the bromobenzene metabolism, the animal group was induced the stage of slight liver damage with 7 times bromobenzene injection every two days (400 mg/kg body wt. i.p.). In the present experimental animal model, the single dose of bromobenzene(400 mg/kg body wt. i.p.) was injected to the bromobenzene-pretreated rats and the hepatic aniline hydroxylase(AH) activity, glutathione(GSH) content and glutathione S-transferase (GST) activity were determined at the intervals of 2, 4, 8, 24 hours throughout 24 hr. The activities of hepatic AH and GST were generally higher in bromobenzene-pretreated rats than those in normal group throughout the whole course of experiment. Furthermore, the decreasing rate of hepatic GSH content was also higher in bromobenzene pretreated rats than in normal rats. Moreover, the value of V$_{max}$ in hepatic GST was higher in bromobenzene pretreated rats than that in the normal rats. In conclusion, these results indicate that the pretreatment of bromobenzene may rather enhance the bromobenzene metabolism.

Effect of Temperature-increase Rate and Terminal Temperature on the Solubilization of Sewage Sludge using Microwave Irradiation

  • Solubilization of sewage sludge creates favorable conditions for anaerobic microorganisms to produce biogas. In this paper, we quantify the effect of heating pretreatment on the degree of solubilization of sewage sludge. The pretreatment process was carried out using a lab-scale industrial microwave unit (2450 MHz frequency). Response surface analysis was applied to determine the combination of temperature-increase rate (ramp rate) (2.9 to 17.1 ${^{\circ}C}$/min) and terminal temperature (52 to 108${^{\circ}C}$). Both ramp rate and temperature significantly affected the solubilization degree of sludge. Within the design boundaries, the conditions predicted to maximize the solubilization degree of 15.8% were determined to be 2.9 ${^{\circ}C}$/min and 104${^{\circ}C}$.

흰쥐에 toluene 투여가 백혈구 xanthine oxidase 및 superoxide dismutase 활성에 미치는 영향 (Effect of Toluene Treatment on the Xanthine Oxidase and Superoxide Dismutase Activities in Leukocyte of Bacterial Infected Rats)

  • This study was designed to observe the effect of tohiene pretreatment on leukocyte variation in whole blood and the oxygen free radical generating, scavenging enzyme activities in neutrophil of bacteria infected rats. Toluene was administered 7 times intraperitoneally at levels of 9.45 mM/kg body weight to the rats and then infected with S. aureus $2\times10^7$ cfu/ml. The toluene treated-rats showed the significantly decreased numbers of lymphocyte and monocytes, but the similiar numbers of neutrophils with the control. Furthermore the increased neutrophils in blood of bacteria infected rats were reduced by the toluene pretreatment. Concomitantly the increased activities of xanthine oxidase and superoxide dismutase in neutrophil of bacteria infected rats were also decreased by the toluene pretreatment. On the other hand, injection of benzaldehyde to rats also led to similiar results in the count of leukocytes, xanthine oxidase and superoxide dismutase activities of neutrophil with those of toluene treated rats. These data suggest that toluene and its intermediate metabolite, benzaldehyde influence on the phagocytosis and defence mechanism of neutrophil.

흰쥐에 Toluene과 Alcohol의 병행투여가 간 손상에 미치는 영향 (Effect of Acute Pretreatment of Ethanol on the Liver Damage in Toluene Treated Rats)

  • To evaluate an effect of acute ethanol pretreatment on the liver injury in toluene-treated rats, toluene(50% in olive oil) was intraperitoneally given four times at 0.3ml/100g body weight at interval of one day to the ethanol-pretreated rats(0.3ml of 50%/100g body weight). The increasing rate of liver weitht per body weight(%), serum levels of alanine aminotransferase (ALT) activity, liver lipid peroxide content was higher in toluene treated rats pretreated with ehtanol than those treated with toluene alone. Concomitantly the hepatic glucose-6-phosphatase activity was increased whereas glutathione content was decreased by ehtanol pretreatment before toluene administration in rats. In case of direct administration of acetaldhyde or benzaldehyde to the rats, the liver weitht per body weight(%) and serum levels of ALT activity were almost higher than the control group. There results indicate that the toluene treated rats showed the reversible injury of liver and intensified. however, by the ethanol treatment.

사염화탄소(四鹽化炭素) 간장장해(肝臟障害) 동물(動物)에서의 Sulfisoxazole의 흡수(吸收)와 배설(排泄)에 관(關)한 연구(硏究) (The Effects of Pretreatment with Carbon Tetrachloride on the Absorption and Excretion of Sulfisoxazole)

  • This paper attempted to investigate the effect of pretreatment with carbon tetrachloride on absorption, excretion, protein binding, and biological half-life of sulfisoxazol from rats and rabbits. Absorption of sulfisoxazol was found to decrease in severe damage rats, compared with that of normal rats, but in mild rats, absorption of sulfisoxazol was similar to that of nomal rats. Absorption of sulfisoxazol was decreased significantly in severe damage rabbit pretreated with carbon tetrachloride but in mild damage rabbit, absorption of sulfisoxazol was not influenced significantly. Pretreatment with carbon tetrachloride gave the effect on clearance of sulfisoxazol in part but protein binding percent of sulfisoxazol was not influenced by concentration of carbon tetrachloride.

  • The fragrant fabrics are prepared by the attaching microcapsules using acrylic binder. To improve the handle of fragrant fabrics, pretreatment by softener is suggested. The durability of fragrant fabrics and the change of their handle are investigated. It is carried out One-way ANOVA and Duncan test to determine bending properties, shear properties, surface properties of untreated, SO-MC and BI-MC by Kawabata system. BI-MC shows significant differences in B and 2HB. SO-MC shows significant differences in G and 2HG. It turned out the pretreatment by softener mitigated the effect of binder and makes fabrics softer than untreated fabrics.

  • In this study, the adhesive effect, blistering behavior and electrochemical polarization behavior of red lead film by the pretreatment work of sandblasting and potassium chromate($K_2CrO_4$) were investigated to prevent the corrosion of gas storage tank under marine environment. The main results obtained are as follows : 1) The adhesive effect of red lead film by the pretreatment work of sandblasting and potassium chromate is more improved than that of red lead film. 2) The blistering occurrence of red lead film by the pretreatment work of sandblasting and potassium chromate is more delayed than that of red lead film. 3) As the anodic potential is increased, corrosion current density of red lead film by the pre treatment work of sandblasting and potassium chromate is lower drained than that of red lead film.

흰쥐에서 플루바스타틴이 딜티아젬 및 그 대사체인 데스아세틸딜티아젬의 약물동태에 미치는 영향 (Effect of Fluvastatin on the Pharmacokinetics of Diltiazem and its Metabolite, Desacetyldiltiazem in Rats)

  • The aim of this study was to investigate the effect of fluvastatin on the pharmacokinetics of diltiazem and its active metabolite, desacetyldiltiazem, in rats. Pharmacokinetic parameters of diltiazem and desacetyldiltiazem were deter-mined after an oral administration of diltiazem (15 mg/kg) to rats pretreated with fluvastatin (0.5 and 1.5 mg/kg). Compared with the control (given diltiazem alone), the pretreatment of fluvastatin significantly (p<0.05) increased the area under the plasma concentration (AUC), peak plasma concentration $(C_{max})\;and\;K_a$ of diltiazem. Relative bioavailability $(RB\%)$ of diltiazem increased from 1.36- to 1.55-fold. However there were no significant changes in $t_{max},\;K_{el}\;and\;t_{1/2}$ of diltiazem. The pretreatment of fluvastatin also altered the pharmacokintic parameters of desacetyldiltiazem. The pretreatment of fluvastatin (1.5 mg/kg) significantly (p<0.05) increased the AUC of desacetyldiltiazem, whereas the metabolite parent ratio (MR) of desacetyldiatlazem was decreased significantly (p<0.05), suggesting that fluvastatin might inhibit the metabolism of diltiazem. The pretreatment of fluvastatin enhanced the bioavailability of diltiazem in a dose dependent manner at doses ranging from 0.5 to 1.5 mg/kg. further studies for the drug Interaction will be needed in the clinical trials when dilitazem is administrated concomitantly with fluvastatin in humans.

테트라메톡시프라본이 파크리탁셀의 약물동태에 미치는 영향 (The Effect of Tetramethoxyflavone on the Pharmacokinetics of Paclitaxel in Rats)

  • The pharmacokinetics of orally administered paclitlxel (50 mg/kg) was studied in six rabbits after 1hr pretreatment (2.0 mg/kg and 10 mg/kg) of tetramethoxyflavone or coadministration of (2.0 mg/kg, 10 mg/kg and 20 mg/kg) tetramethoxyflavone. The area under the plasma concentration-tine curve (AUC) and plasma concentration of paclitaxe1 coadministered with tetramethoxyflavone (10 mglkg) were increased significantly (p<0.05) compared with control. However, coadministration of tetramethoxyflavone (2 and 20 mg/kg) showed no significant effect on the pharmacokinetic parameters of paclitaxel. Pretreatment with tetramethoxyflavone significantly (p<0.05) increased the plasma concentration of paclitaxel. The area under the plasma concentration-time curve (AUC) and the peak concentration (C$_{max}$) of paclitaxel pretreated with tetramethoxyflavone were increased significantly (p<0.01, p<0.05) compared with control. The terminal half. life of paclitaxel pretreated with tetramethoxyflavone (2 mg/kg and 10 mg/kg) was significantly (p<0.05) prolonged compared with control. Pretreatment with tetramethoxyflavone (2.0 mg/kg, 10 mg/kg) significantly (p<0.01, p<0.05) increased the absolute bioavailability of paclitaxel compared with the control (154∼179%). On the basis of the results, it might be considered that tetramethoxyflavone may inhibit cytochrome P450 or P-glycoprotein efflux pump which are engaged in paclitaxel metabolism, result in increased AUC and t$_{1}$2/ of paclitaxel. However, further study should be conducted to clarify the roles of cytochrome P450 and P-glycoprotein on paclitaxel bioavailability with/or without tetramethoxyflavone.
