This study was performed to investigate the antimicrobial effect against food-borne pathogens of Sanguisorbae officinalis L. ethanol extract. The antimicrobial activity of the ethanol extract was determined using a paper disc-diffusion method and the diameter of the clear zone was measured. The diameters of the clear zone in the presence of 10 mg of the ethanol extract were the maximum against Staphylococcus aureus among the tested 4 gram-positive bacteria and Pseudomonas aeruginosa among the tested 7 gram-negative bacteria. Analysis of the minimum inhibition concentrations (MIC) showed that the ethanol extract exhibited a similar efficacy as sorbic acid, well-known chemical preservatives. The growth inhibitory effects of the ethanol extract in the concentrations of 250, 500, 1,000 and 2,000 mg/L on food-borne pathogens were determined against Staphylococcus aureus, Bacillus subtilis, Salmonella Typhimurium and Pseudomonas aeruginosa. The growth of the microorganisms was significantly (p<0.05) inhibited by the ethanol extract in the concentrations higher than 250 mg/L. Thus, the results of the present study demonstrate that the ethanol extract exhibits antimicrobial effects against food-borne pathogens, suggesting that Sanguisorbae officinalis L. could be used as natural antibacterial agent in food.
Human papillomavirus (HPV) is the major cause of cervical cancer, a deadly threat to millions of females. The early oncogene product (E7) of the high-risk HPV16 is the primary agent associated with HPV-related cervical cancers. In order to understand how E7 contributes to the transforming activity, we investigated the structural features of the flexible N-terminal region (46 residues) of E7 by carrying out N-15 heteronuclear NMR experiments and replica exchange molecular dynamics simulations. Several NMR parameters as well as simulation ensemble structures indicate that this intrinsically disordered region of E7 contains two transient (10-20% populated) helical pre-structured motifs that overlap with important target binding moieties such as an E2F-mimic motif and a pRb-binding LXCXE segment. Presence of such target-binding motifs in HPV16 E7 provides a reasonable explanation for its promiscuous target-binding behavior associated with its transforming activity.
In this study, the colorants of Amur cork tree were extracted with boiling water. Chitosan crosslinked cotton fabrics have been dyed with aqueous extract of Amur cork tree and their dyeabilities on the fabrics were studied. Additionally the fastness to washing and light were also investigated. Cotton fabrics were treated with a crosslinking agent epichlorohydrin in the presence of chitosan to provide the cotton fabrics the dyeing properties of natural dye(Amur cork tree) by the chemical linking of chitosan to the cellulose structure. This process was applied by means of the conventional mercerizing process. The chitosan finishing and durable press finishing of the cotton fabrics occurred simultaneously in the mercerization bath. On the surface color change, the fabric of no-chitosan finished and no-mordanted has greenish yellow. The more crosslinked chitosan on cotton fabrics has the more turned down greenish on the surface color, as increasing the concentration of chitosan, greenish color turn down to the yellow close the 90o hue angle. In all sorts of fabrics, dyeability(K/S) is slightly affected by the number of manufacturing process and the concentration of chitosan. But only mercerized cotton fabric has higher dyeability (K/S) than mordant treated cotton fabrics. Wash fastness has little different results by each condition, but almost similar values. Light fastness was improved with chitosan treatment on cotton fabric.
We isolated Streptomyces spry. from Korean soil, which showed high cytotoxicity against tumor cell lines, L1210 and P388Dl. Among 30 strains, three strains (DKM 104, DKM 128, DKM 409) were appeared to possess plasmid. Strain DKM 104 and DKM 128 had two CCC(Covalently Closed Circular) form plasmid, about 20 Kb in size and about 1 Kb compared with λ Hind III DNA size marker. And strain DKM 409 had three plasmids, among which two plasmic were CCC form about 20 Kb and about 20 Kb compared with same size marker. To find out whether plasmid involved in production of antitumor agent or not, we performed to curing experiment. Comparing cytotoxicity between culture filtrate of plasmid-containing strains and cured strains, we knew that only the cytotoxic activity of the strain DKM 128 was involved in plasmid.
In the present study, we intended to investigate whether polycationic peptides including poly-L-lysine (PLL) and poly-L-arginine (PLA) specifically inhibit the mucin release and do not affect significantly the release of the other releasable glycoproteins with less molecular weight than mucin's from cultured airway goblet cells. Confluent primary hamster tracheal surface epithelial (HTSE) cells were metabolically radiolabeled with 3H-glucosamine for 24 hr and chased for 30 min in the presence of varying concentrations of either poly-L-arginine (PLA) or poly-L-lysine (PLL) to assess the effects on 3H-mucin release and on the total elution profile of the treated culture medium. The results were as follows : (1) PLL 78,000, PLL 9,600 and PLA 8,900 inhibited mucin release in a dose-dependent manner; (2) These polycationic peptides did not inhibit the release of the other releasable glycoproteins with less molecular weights than mucin's. We conclude that these polycationic peptides 'specifically'inhibit mucin release from airway goblet cells. This finding suggests that these polycationic peptides might be used as a specific airway mucin-regulating agent.
Loss of response to anti-tumor necrosis factor (anti-TNF) agents in the treatment of inflammatory bowel disease (IBD) is a major consideration to maintain sustained response. Reversal of immunogenicity can re-establish response and increase the durability of these agents. Strategies to reverse immunogenicity include dose-intensification and/or the addition of an immunomodulator. However, there is a relative paucity of data on the efficacy of such interventions in pediatric IBD patients. Available reports have not strictly utilized homogenous mobility shift assay, which reports on anti-drug antibodies even in the presence of detectable drug, whereas prior studies have been confounded by the use of drug sensitive assays. We report four pediatric inflammatory bowel disease patients with successful reversal of immunogenicity on an anti-TNF agent using dose intensification and/or addition of an immunomodulator.
Plant pathogenic oomycetes, such as Phytophthora spp., are the causal agent of the most devastating plant diseases. During infection, these pathogens accomplish parasitic colonization of plants by modulating host defenses through an array of disease effector proteins. These effectors are classified in two classes based on their target sites in the host plant. Apoplastic effectors are secreted into the plant extracellular space, and cytoplasmic effectors are translocated inside the plant cell, through the haustoria that enter inside living host cell. Recent characterization of some oomycete Avr genes showed that they encode effector protein with general modular structure including N-terminal conserved RXLR-DEER motif. More detailed evidences suggest that these AVR effectors are secreted by the pathogenic oomycetes and then translocated into the host plant cell during infection. Recent findings indicated that one of the P. infestans effector, Avrblb2, specifically induces hypersensitive response (HR) in the presence of Solanum bulbocastanum late blight resistance genes Rpi-blb2. On the other hand, another secreted RXLR protein PexRD8 originated from P. infestans suppressed the HCD triggered by the elicitin INF1. In this review, we described recent progress in characterized RXLR effectors in Phytophthora spp. and their dual functions as modulators of host plant immunity.
The tin (IV) oxide nanoparticles are prepared by controlled precipitation method and calcined at temperatures of $200-600^{\circ}C$. The prepared $SnO_2$ nanoparticles characterized by XRD patterns, TEM image, IR and UV-Vis spectra. The XRD patterns and TEM image show the tetragonal structure and spherical morphology for $SnO_2$ nanoparticles, respectively. The photocatalytic activity of the prepared $SnO_2$ nanoparticles studied in degradation reaction of methylene blue (MB). The results show the size of nanoparticles, band-gap energy and photocatalytic activity of $SnO_2$ depends on the calcinations temperature. The $SnO_2$ nanoparticles calcined at $500^{\circ}C$ indicated the highest photoreactivity. Also, the zero-valent tin (ZVT) nanoparticles with tetragonal structure are prepared by a reducing agent and used as a catalyst in degradation of MB. In basic pH of 11, the degradation >95% of MB at time 150 min obtained at presence of ZVT nanoparticles.
Avian pneumovirus(APV), also known as avian rhinotracheitis(ARTV), affects both turkeys and chickens and is known to be the primary causative agent of turkey rhinotracheitis(TRT). The aim of this study was to establish the presence or absence of antibodies to APV by an enzyme-linked immunosorbent assay(ELISA) and confirm APV by a reverse transcriptase-polymerase chain reaction(RT-PCR). The tested serum and feces were collected from laying hens in Gyeongbuk province. The positive farms with antibody against APV by ELISA were 90(96.7%) of 93 and positive serum samples were 433(93.1%) of 465 different sera. By regional group, sera from Uiseong, Cheongsong and Bonghwa were noted as 100% positive and positive rates of samples from Yeongju, Andong and Yeongyang were 93.3%, 85.7% and 50%, respectively. However, APV was not detected in feces samples by RT-PCR.
The aim of the present study was to develop a sensitive and specific assay for the diagnosis of alcelaphine herpesvirus 1 (AlHV-1) which is a cause agent of malignant catarrhal fever in ruminants. A1HV-1 is a gamma herpesvirus, which is frequent latent, and it is often difficult to detect its antigens or specific nucleic acids because of its low genomic copies in the infected tissues. In this study, polymerase chain reaction (PCR)-dot blot hybridization (DBH) assay for detecting AlHV-1 DNA was developed and evaluated for its sensitivity and specificity as comparison with PCR and DBH alone. The developed PCR-DBH was more sensitive than PCR or DBH alone and also very specific. The results showed that the sensitivity of PCR-DBH were higher and stronger than those of PCR and DBH alone. This PCR-DBH assay can be applied efficiently to confirm the presence of AlHV-1 virus on clinical samples and to differentiate specifically between AlHV-1 infection and other viral infections.
