계작지모탕(桂芍知母湯)의 소염.진통작용 및 Alcohol성 고뇨산혈증(高尿酸血症)에 미치는 영향 (Antiinflammatory, Analgesic and Antihyperuricemic Effects of 'Gyejakjimo-Tang' in Rats)

  • 'Gyejakjimo-Tang(桂芍知母湯)', which is composed of Cinnamon bark(桂枝), Peony root(芍藥), Anemarrhena root(知母) and other seven herbs, is described as antipyretic, diuretic and analgesic prescription in traditional medical literatures including Geumgwe-Yoryak(金櫃要略). So it is being used in the treatment of gout clinically in oriental region. As the results of the pharmacological and biochemical trials of the prescription, it was found to have antiinflammatory and significant analgesic effects indicated by carrageenin edema, dye permeabilities and writhing behavior. And it potentiated the output of uric acid in blood and the increase of uric acid in urine of alcohol-toxicated rats, which mean the decrease of uric acid level only by excretion procedure. But the prescription didn't show any effects on the activities of adenine/guanine deaminase, purine nucleoside phosphorylase, xanthine oxidase or uricase, which are related with formation and metabolism of uric acid.

허혈/재관류 환경하에서 청폐사간탕의 염증 관련 반응 억제 효과 (Inhibitory Effects of Chungpesagan-Tang on Ischemia/Reperfusion-Induced Inflammatory Responses In vitro)

  • Chungpesagan-Tang (CST), a Korean traditional prescription composed of oriental medical herbs, has been used successfully to improve human health and regimen. This study was designed to examine the inhibitory effects of CST against in vitro ischemia/reperfusion-induced inflammatory response. We have characterized the production of prostaglandin $E_2$ and arachidonic acid during ischemia/reperfusion in the human neuroblastoma SK-N-MC and human monocytic macrophage U937 cells and the inhibitory effect of CST on these inflammation-related substance formation has been found out in this study. This result suggested that CST used in this experiment reinforced antiinflammatory potentials and protected cells against ischemia/reperfusion-induced inflammatory resopnse.

대식세포의 oxLDL 생성에 미치는 강활속단탕의 영향 (Effects of KanghwalSokdantang(KS) on LDL Oxidation in Macrophage Cell)

  • Objective : As a link in chain of research to confirm the oriental medical prescription which has the anti-atherosclerosis effects, this research evaluated the effects on the macrophage-related factors by using KanghwalSokdantang(KS). Methods : In order to perform this research, we have evaluated the effects on the oxLDL formation from the macrophages, the nitric oxide formation, and the oxidation of macrophages. Thus, with this evaluation, we have investigated the applicapability on the artherosclerosis. Results : KanghwalSokdantang has showed a noticeable reduction of protein oxidation in the process of oxLDL formation, has remarkably restrained phospholipid peroxidation, an index to estimated the phospholipid oxidation and reduction that are formed in the process of macrophage's oxLDL formation, and has increased the nitrite concentration noticeably in the LDL-dealing macrophages. By increasing the survival rate of macrophages, KanghwalSokdantang has restrained the cellular damages. KanghwalSokdantang is ineffective on the LDH outflow from damaged cells. $1{\mu}g/ml$ KanghwalSokdantang sample has increased acid phosphatase activity remarkably. Conclusion : KanghwalSokdantang has the possibility to be used in the prevention and treatment of atherosclerosis, which is formed by the oxLDL formation of macrophages.

The Radioprotective Effects of Bu-Zhong-Yi-Qi-Tang as a Prescriptions of Traditional Chinese Medicine in Irradiated Mice

  • We performed this study to determine the effect of Bu-Zhong-Yi-Qi-Tang, as a prescription of traditional Oriental medicine, and its major ingredients on jejunal crypt survival, endogenous spleen colony formation, and apoptosis in jejunal crypt cells of mice irradiated with high and low dose of r-radiation. Bu-Zhong-Yi-Qi-Tang administration before irradiation protected the jejunal crypts (p<0.0001), increased the formation of endogenous spleen colony (p<0.05) and reduced the frequency of radiation-induced apoptosis (p<0.05). 삼백초(三白草) 에탄올추출물의 항산화능 및 피부 미백작용 연구 (A Study on the Antioxidative and Depigmentation Activities of the Ethanol Extract of Saururus Herba)

  • Objectives : The purpose of this study is to investigate the etahnol extract of Saururus Herba and their dermal boiactivity properties related to cosmeceuticals such as depigmentation and radical scavenging effect. Methods : Using B16/F10 Melanin cells, we measured depigmentation effect of the etahnol extract of Saururus Herba. Antioxidant activities of the etahnol extract of Saururus Herba were determined by DPPH radical scavenging activity. Results : The etahnol extract of Saururus Herba showed considerable radical scavenging activity($SC_{50}$ : 25 ${\mu}g$/ml). Antioxidant activity is in a dose dependant manner. The etahnol extract of Saururus Herba down-regulated melanin synthesis effectively. B16 cells incubated with the etahnol extract of Saururus Herba showed reduces melanin formation and tyrosinase activity. The etahnol extract of Saururus Herba alone markedly suppressed melanin content and tyrosinase activity. Conclusions : These results suggest that the ethanol extract of Saururus Herba is a potent depigmentaion agent and it may be a candidate for antioxidant agent.

월국환(越鞠丸) 물 추출물이 Acetaminophen으로 유도된 마우스의 급성 간손상에 미치는 효과 (Effects of Wolguk-whan Water Extract on Acute Oxidative Liver Injury Induced by Acetaminophen)

  • Objectives : Wolguk-whan has been used as a prescription of natural drug for the treatment of stress digestive system disease. Recently, we reported that Wolguk-whan methnol extract (WGWM) exerted a significant protective effect against oxidative damage to the liver of ICR mice. This study was purposed to investigate the effects of Wolguk-whan water extract (WGWW) on liver injury induced by oxidative stress. Methods : In order to investigate the effects of WGWW on acute liver injury, ICR mice were pretreated with WGWW for 6days, starved for 24hrs, and administerated acetamirtophen(500mg/kg, i.p.). In the liver homogenates, lipid peroxide and glutathione(GSH) levels were measured. In addition, activities of hepatic enzyme, such as catalase, glutathione peroxidase(GSH-Px), glutathione S-transferase(GST) were measured in the hepatic mitochondrial and cytosolic fractions. Results : In vivo administeration of WGWW showed effective inhibition of acetaminophen induced lipid peroxidation, and showed elevations of GSH level, catalase, GSH-Px, GST activities. Conclusions : These results suggested that WGWW might suppress the formation of oxidative metabolites, and prevent acetaminophen induced hepatotoxicity.

시호사물탕이 t-BHP로 유도된 간세포의 산화적 손상 및 자유기에 의한 지질과산화 반응에 미치는 영향 (Preventive Effects of Sihosamultang Extract on Lipid Peroxidation by Free Radicals and Oxidative Damage of Hepatocytes by tert-Butyl Hydroperoxide)

  • Sihosamultang(SST) has been used for the treatment of puerperal fever, liver disease in traditional medicine. The present study was carried out to evaluate the antioxidant effects of SST extract in vitro. The inhibitory effect of SST extract on lipid peroxidant was examined in the linoleic acid autoxidation system. In this test, SST extract significantly inhibited the time course of the lipid peroxidation. And SST extract showed about 73% scavenging effect on DPPH radical. And this extract inhibited not only the lipid peroxide formation induced by hydroxyl radical derived from $ H_{2}O_{2}-Fe^{2+}$ in the rat liver homogenate, but also the superoxide generation from xanthine-xanthine oxidase system in a dose-dependent manner. In addition, SST extract protected the hepatic cell death induced by tert-butyl hydroperoxide. These data indicated that SST might play a protective role against oxidative injury by free radicals.

대시호탕(大柴胡湯)이 태음인(太陰人) 병증용약(病證用藥)에 끼친 영향에 대한 고찰 (A Study of the Influence of Daesihog-tang on Taeeum-in's Patholoy and Prescription)

  • Objectives The purpose of this study is to understand the influence of Daesihog-tang on Taeeum-in's patholoy and prescription in "Donguisusebowon" of Sasang Constitutional Medicine. Methods The pathologic condition in which Daesihog-tang used and the relevant herbs and prescriptions in "Sanghanron", "Donguibogam", "Dongmuyoogo haedong", "Chobongwon", and "Donguisusebowon". Results and Conclusions 1. Daesiho-tang in "Sanghanron" was used to the symptom of Yangmyung disease merging with Soyang disease. In "Donguibogam", its indications were mainly used to the severe fever-symptom of Yangmyung disease and expanded to Onbyeong and so on. 2. The concept of Daesiho-tang in "Donguibogam" was changed into Hwanggeumdaehwang-tang in "Chobongwon", which can be used to treat Onbyeong with heat-dysenteric diarrhea, and severe fever of Taeeum-in. 3. The notion of combination between Daehwang and hwanggeum used in "Dongmuyoogo haedong" and "Chobongwon" was connected to Chungmongsukgondamhwan or Jogakhwanggeumdaehwang-tang in "Chobongwon". Especially, Jogakhwanggeumdaehwang-tang for Yeokkigammo and severe constipation was formed while the concept of Onbyeong in Hwanggeumdaehwang-tang' merged with the usage of Jogak and Daehwang or Yisunggugo-hwan. 4. Jogakhwanggeumdaehwang-tang influenced on the usage of Jogakdaehwang-tang for Yangdok and Onbyeong, while the usage of Chungmongsukgondamhwan was not found in "Donguisusebowon". 5. As a result, Daesiho-tang may be presented without usages for a specific patholoy in "Donguisusebowon" because it indirectly influenced on the concept of Yangdok and Onbyeong of Taeeum-in, and the formation of Jogakdaehwang-tang in "Donguisusebowon".

Effects of Curcumin, the Active Ingredient of Turmeric(Curcuma longa), on Regulation of Glutamate-induced Toxicity and Activation of the Mitogen-activated Protein Kinase Phosphatase-1 (MKP-1) in HT22 Neuronal Cell

  • Glutamate causes neurotoxicity through formation of reactive oxygen species and activation of mitogen-activated protein kinase (MAPK) pathways. MAPK phosphatase-1 (MKP-1) is one of the phosphatases responsible for dephosphorylation/deactivation of three MAPK families: the extracellular signal-regulated kinase-1/2 (ERK-1/2), the c-Jun N-terminal kinase-1/2 (JNK-1/2), and the p38 MAPK. In this report, the potential involvement of MKP-1 in neuroprotective effects of curcumin, the active ingredient of turmeric (Curcuma longa), was examined using HT22 cells. Glutamate caused cell death and activation of ERK-1/2 but not p38 MAPK or JNK-1/2. Blockage of ERK-1/2 by its inhibitor protected HT22 cells against glutamate-induced toxicity. Curcumin attenuated glutamate-induced cell death and ERK-1/2 activation. Interestingly, curcumin induced MKP-1 activation. In HT22 cells transiently transfected with small interfering RNA against MKP-1, curcumin failed to inhibit glutamate-induced ERK-1/2 activation and to protect HT22 cells from glutamate-induced toxicity. These results suggest that curcumin can attenuate glutamate-induced neurotoxicity by activating MKP-1 which acts as the negative regulator of ERK-1/2. This novel pathway may contribute to and explain at least one of the neuroprotective actions of curcumin.

보정방암탕 에타놀층의 혈관형성 저해작용에 관한 연구 (Study on the Anti-angiogenic Activity of Ethanol Extract of Bojungbangam-tang)

  • The anti-angiogenic activity of ethanol extract of Bojungbangam-tang, a new herbal prescription composed of nine crude drugs, was evaluated in human umbilical vein endothelial cells (HUVECs). HPLC profile revealed that five major compunds such as apioliquiritin, narirutin, hesperidin, liquiritin and glycyrrhizin. Ethanol extract of Bojungbangam-tang (EBJT) did not showed any significant cytotoxicity against HUVECs up to 200 ug/ml. EBJT significantly inhibited basic fibroblast growth factor (bFGF)-induced HUVECs proliferation to 69% at 200 ${\mu}g/ml$. Migration using window scraping method and tube formation in bFGF stimulated HUVECs were also significantly suppressed by EBJT in a dose-dependent manner. Taken together, these results suggest that Bojungbangam-tang can be a potent prescription for angiogenesis related disease.
