Developing a Molecular Prognostic Predictor of a Cancer based on a Small Sample

  • One Important problem in a cancer microarray study is to identify a set of genes from which a molecular prognostic indicator can be developed. In parallel with this problem is to validate the chosen set of genes. We develop in this note a K-fold cross validation procedure by combining a 'pre-validation' technique and a bootstrap resampling procedure in the Cox regression . The pre-validation technique predicts the microarray predictor of a case without having seen the true class level of the case. It was suggested by Tibshirani and Efron (2002) to avoid the possible over-fitting in the regression in which a microarray based predictor is employed. The bootstrap resampling procedure for the Cox regression was proposed by Sauerbrei and Schumacher (1992) as a means of overcoming the instability of a stepwise selection procedure. We apply this K-fold cross validation to the microarray data of 92 gastric cancers of which the experiment was conducted at Cancer Metastasis Research Center, Yonsei University. We also share some of our experience on the 'false positive' result due to the information leak.

충돌사고 재구성 해석을 위한 차량 블랙박스의 개발 (Development of an Automobile Black Box for Reconstruction Analysis of Collision Accidents)

  • This paper presents design concepts, specifications and performances of a newly developed Black Box, the reconstruction analysis tool with the records, and results of validation tests. The Black Box can detect crash accidents automatically, and record the vehicle's motion and driver's maneuvers during a pre-defined time period before and after the accident. The items of the Black Box included the acceleration, yaw-rate, vehicle speed, engine RPM, braking application, steering and several digital inputs for recording driver's maneuvers. To detect the accident-related-crash, it is important to understand characteristics of the crash signal, which are much different from those of normal driving. Therefore, analytical considerations should be taken in designing pre-filtering circuits and selecting appropriate parameters for identifying crash accidents. And, it is necessary to select proper combination of motion sensors and design proper pre-filtering circuits in order to describe the vehicle's motion. The analysis algorithms were developed and implemented which can perform accurate detection of crash accidents, simulating pre-crash trajectories, and calculating parameters for reconstruction analysis of crash accidents. The developed Black Box was installed on passenger cars and several types of validation tests were conducted. Through the tests, the accuracy of the recorded data and usefulness of the analysis tool for reconstruction have been validated.

  • In this paper, we propose Class Diagram With Constraints (CDWC) as an object oriented modeling technique which makes validation possible in software development. CDWC is a simple and basic model for the object oriented analysis, and has a reasonable strictness for software developers. CDWC consists of class diagrams and constraints (invariant and pre/post conditions), using UML and a subset of OCL.. We introduce a method of validation of CDWC using the verification technique of algebraic formal specification language CafeOBJ.

과학학습의 정의적 영역에서 사전-사후 통합 검사 설계의 타당화 연구: 과학영재를 대상으로 (A Validation Study of Retrospective Pre-post Testin the Affective Domain in Science Learning:for Scientifically Gifted Elementary Students)

  • In this study, the reliability and validity of the retrospective pre-post test were analyzed in order to solve the problem of traditional pre-post test including response shift bias. Samples of the study were 162 elementary school students who are studying at the S university gifted education center in Seoul. Before completion of the field trip, we conducted pre test of science-related attitudes. After completion of the field trip, respondents were asked to compare their responses of pre and post science-related attitudes to quantitatively analyze the commonalities and differences of the two tests. To find out more characteristics, qualitative data such as daily records and interview were also gathered and analyzed. The major results of the study are as follows. First, for the paired t-test, there was no statistically significant difference between separate pre-test scores and retrospective pre-test. There was a very high correlation between the separate pre-test scores and the retrospective pre-test. Second, there were significant differences in all seven sub-factors of science-related attitudes between the retrospective pre-test and the post-test. Third, the separate pre-test scores showed a slightly higher tendency than the retrospective pre-test scores. This suggests that the response shift bias appears when it is performed the separate pre-test in affective domain. As a result of the interview, it was found that the evaluation standards of separate pre-test did not match with those of post-test. Forth, internal consistency reliability of the retrospective pre-test was higher than that of the separate pre-test. However, there were significant differences in six factors of science-related attitudes excluding the 'social implications of science' between the separate pre-test and the post-test. Based on these results, the retrospective pre-post test design provides simplicity and convenience to both respondents and investigators, as it is done with one test. The retrospective pre-post test design can be regarded as a valid design for the self-report measurement of affective domain on a single experimental group.

  • The Ministry of Health and Welfare of Japan has set up six research groups concerning the endocrine disrupting chemicals. One of these projects was "A study on development of testing methodology for health effects due to exposure of environmental endocrine disruptors". In this paper, three topics are described. In OECD collaboration for pre-validation of uterotrophic assay, the most sensitive response to ethnyl estradiol was noted in the ovarectomized rats treated subcutaneously for 7 days. Secondly, it was suggested that changes of the serum $\alpha_{2u}$-globulin level may be a sensitive parameter for detecting the estrogenic activities of chemicals. Finally, development of the sexually dimorphic nucleus of preoptic area in the brain oj male rats was inhibited by the treatment with estrogenic chemicals, and their masculine behaviors and reproductive abilities were impaired after sexual maturation. In conclusion, these parameters are considered to be sensitive endpoints for testing estrogenic chemicals.chemicals.

예비유아교사 교사효능감 척도(PS-TES) 타당화 (The Validation of Pre-service Teacher Efficacy Scale for Preschool Teachers)

  • 본 연구의 목적은 Ohio State Teacher Efficacy Scale(OSTES)을 활용하여 예비유아교사 교사효능감 척도(PS-TES)를 타당화 하는데 있다. 이를 위한 본 연구는 G지역에 소재한 3년제 대학 유아교육과 3학년 재학생 428명을 대상으로 설문지를 실시하였다. 척도의 타당도와 신뢰도를 확인하기 위해 탐색적 요인분석과 신뢰도(Cronbach's ${\alpha}$)계수, 상관분석 및 확인적 요인분석(CFA)을 실시하였다. 본 연구의 결과를 연구문제를 중심으로 살펴보면 다음과 같다. 첫째, 예비유아교사 교사효능감 척도(PS-TES)의 하위요인을 살펴보기 위해 탐색적 요인분석 결과 3개의 하위요인(교수전략 효능감, 학생참여 효능감, 교실관리 효능감)으로 나타났으며 총 문항은 12문항으로 이루어졌다. 예비유아교사 교사효능감 척도(PS-TES)의 하위요인 신뢰도는 '교수전략 효능감' .82, '학생참여 효능감' .72, '교실관리 효능감' .76으로 나타났으며 전체 신뢰도는 .87로 나타났다. 둘째, 예비유아교사 교사효능감 척도(PS-TES)의 타당도를 살펴보기 위해 확인적 요인분석(CFA)을 실시한 결과 모델 적합도는 CMIN/DF = 1.59, RMSEA = .05, SRMR = .05, NNFI = .96, CFI = .97로 나타나 예비 유아교사 교사효능감 척도(PS-TES)가 적합한 측정 도구임이 확인되었다.

  • The cost-effective reduction of greenhouse gas(GHG) emission to avert the most severe impacts of climate change remains one of the widely accepted priorities for global action. In order to facilitate cost-effective abatement strategies, the Kyoto Protocol introduced three mechanisms, or flexible instruments, the Emissions Trading(ET), the Joint Implementation(JI) and the Clean Development Mechanism(CDM). The CDM enables Annex I countries to the Kyoto Protocol to partially meet cost-effectively their emission reduction commitments by undertaking GHG mitigation Projects in developing countries, which do not have any GHG abatement obligations and where the emission reductions are cheaper. One of the major barriers hampering the wide spread implementation of CDM is the high transaction costs associated with the initial identification of promising CDM projects. This paper presents development of a pre-validation program of CDM. The developed program may provide a useful aid to potential investors and project developers as a supportive pre-evaluation tool, and may become an effective tool for the promotion of renewable energy and fuel switching projects.

Different Real Time PCR Approaches for the Fine Quantification of SNP's Alleles in DNA Pools: Assays Development, Characterization and Pre-validation

  • Single nucleotide polymorphisms (SNPs) are becoming the most common type of markers used in genetic analysis. In the present report a SNP has been chosen to test the applicability of Real Time PCR to discriminate and quantify SNPs alleles on DNA pools. Amplification Refractory Mutation System (ARMS) and Mismatch Amplification Mutation Assay (MAMA) has been applied. Each assay has been pre-validated testing specificity and performances (linearity, PCR efficiency, interference limit, limit of detection, limit of quantification, precision and accuracy). Both the approaches achieve a precise and accurate estimation of the allele frequencies on pooled DNA samples in the range from 5% to 95% and don't require standard curves or calibrators. The lowest measurement that could be significantly distinguished from the background noise has been determined around the 1% for both the approaches, allowing to extend the range of quantifications from 1% to 99%. Furthermore applicability of Real Time PCR assays for general diagnostic purposes is discussed.

근전도 기반의 Spider Chart와 딥러닝을 활용한 일상생활 잡기 손동작 분류 (Classification of Gripping Movement in Daily Life Using EMG-based Spider Chart and Deep Learning)

  • In this paper, we propose a pre-processing method that converts to Spider Chart image data for classification of gripping movement using EMG (electromyography) sensors and Convolution Neural Networks (CNN) deep learning. First, raw data for six hand gestures are extracted from five test subjects using an 8-channel armband and converted into Spider Chart data of octagonal shapes, which are divided into several sliding windows and are learned. In classifying six hand gestures, the classification performance is compared with the proposed pre-processing method and the existing methods. Deep learning was performed on the dataset by dividing 70% of the total into training, 15% as testing, and 15% as validation. For system performance evaluation, five cross-validations were applied by dividing 80% of the entire dataset by training and 20% by testing. The proposed method generates 97% and 94.54% in cross-validation and general tests, respectively, using the Spider Chart preprocessing, which was better results than the conventional methods.
