본 연구에서는 우리나라 전통 발효 된장 내 바이오제닉 아민 함량을 측정하고 이들 아민의 축적을 억제할 수 있는 프로바이오틱 바실러스균을 분리하였다. 된장 내 세균수, pH, 적정산도, 염도 및 바이오제닉 아민 함량은 시료마다 유의한 차이가 있었다. 된장에서 분리된 바실러스균 중에서 Bacillus (B.) licheniformis DB102, B. subtilis DB203, B. stearothermophilus DB206, Bacillus sp. DB209, Bacillus sp. DB310, B. coagulans DB311, B. cereus DB313, B. amyloliquefaciens DB714, Bacillus sp. DB917, B. cereus DB 915, B. subtilis DB1020 및 Bacillus sp. DB1022는 바이오제닉 아민 생성능이 있는 것으로 확인되었다. 반면, 바이오제닉 아민 분해균은 Bacillus sp. DB403, Bacillus sp. DB407, B. subtilis DB517, B. licheniformis DB612 및 B. subtilis DB821로 동정되었다. 특히, Bacillus sp. DB407과 B. subtilis DB821은 인공 소화액에 대한 저항성, 장관 상피세포에 대한 부착능, 항생제에 대한 내성 및 바이오제닉 아민 생성균에 대한 항균 활성 등의 프로바이오틱 특성을 나타내었다. 결론적으로 이들 두 프로바이오틱 바실러스균은 바이오제닉 아민이 낮은 대두 발효 식품 제조에 적합한 스타터로 사료된다.
본 연구는 고온기 복합생균제(L. plantarum, B. subtilis, Saccharomyces cerevisiae) 급여가 육계의 항산화 방어작용에 미치는 영향을 조사하기 위하여 대조군(CON), 0.25% 생균제 급여군(LPB) 및 0.5% 생균제 급여군(HPB) 등 3 처리군으로 설정하여 혈장, 소장 점막 및 간 조직에서 항산화 지표를 분석하였다. 실험 결과, 혈장 total protein, albumin, bloodurea nitrogen(BUN) 및 glutathione(GSH) 수준 등 질소화합물은 처리군 간 차이가 없었다. 혈장에서 조사한 항산화 효소 superoxide dismutase(SOD), glutathione peroxidase(GPX)와 glutathione S-transferase(GST) 및 지질과산화(MDA) 역시 생균제 급여에 따른 유의적 차이가 없었다. 소장점막조직에서 SOD 활성도는 0.5% 생균제 급여군(HPB)에서 대조군과 LPB군 보다 유의하게(P<0.05) 증가하였다. 소장 점막조직의 MDA 수준은 대조군보다 HPB군에서 현저하게(P<0.05) 감소하였다. 그러나 소장 점막조직의 GPX와 GST 활성도는 생균제 급여에 따른 차이는 없었다. 간 조직에 존재하는 SOD, GPX, GST 활성도 및 MDA 수준은 생균제 급여와 급여 수준에 따른 영향을 받지 않았고 모든 군에서 비슷한 수준을 보였다. 따라서 HPB군의 소장 점막조직에서 SOD 활성도가 증가하고 지질과산화도가 감소하는 결과로 보아 고온기에 0.5% 생균제 급여는 육계 소장 점막조직의 항산화 방어작용에 긍정적인 효과를 미치는 것으로 나타났다.

S-Adenosyl-L-methionine (SAM) exerts several beneficial effects on depression, chronic diseases, and cognitive impairments. Bifidobactrium bifidum BGN4 reportedly produces higher amounts of SAM than any other lactic acid bacterium used in yogurt. The aim of this study was to develop a SAM-reinforced probiotic yogurt using Bifidobacterium. The sensory aspects of the yogurt via response surface methodology (RSM) and the texture and SAM content of the yogurt were assessed. Based on the sensory assessments for sweetness, sourness, and thickness evaluated by 48 panelists, the optimized conditions for preparation of SAM-reinforced yogurt were 4.0-4.4%(w/w) sugar, 3.2-3.5%(w/w) skim milk, and a pH of 4.7-4.8. The SAM content of the developed yogurt was 0.05 nmol/mL. In conclusion, SAM-reinforced probiotic yogurt may provide a vehicle for the potential exploitation of the benefits of increased dietary SAM.

We characterized the probiotic properties of Lactobacillus helveticus strains KII13 and KHI1 isolated from fermented cow milk by in vitro and in vivo studies. The strains exhibited tolerance to simulated orogastrointestinal condition, adherence to Caco-2 cells, and antimicrobial activity. Both L. helveticus strains produced bioactive tripeptides, isoleucylprolyl-proline and valyl-prolyl-proline, during fermentation of milk. KII13 showed higher in vitro cholesterol-lowering activity (47%) compared with KHI1 (28%) and L. helveticus ATCC 15009 (22%), and hence, it was selected for in vivo study of cholesterol-lowering activity in atherogenic diet-fed hypercholesterolemic mice. For the study, mice were divided into four groups (viz., normal diet control group, atherogenic diet control group (HCD), KII13-atherogenic diet group (HCD-KII13), and Lactobacillus acidophilus ATCC 43121-atherogenic diet group (HCD-L.ac) as positive control). The serum total cholesterol level was significantly decreased by 8.6% and 7.78% in the HCD-KII13 and HCD-L.ac groups (p < 0.05), respectively, compared with the HCD group. Low-density lipoprotein cholesterol levels in both HCD-KII13 and HCD-L.ac groups were decreased by 13% and 11%, respectively, compared with the HCD group (both, p < 0.05). Analysis of cholesterol metabolism-related gene expression in mice liver showed increased expression of LDLR and SREBF2 genes in mice fed with KII13. By comparing all the results, we conclude that L. helveticus KII13 could be used as a potential probiotic strain to produce antihypertensive peptides and reduce serum cholesterol.
As probiotics play an important role in maintaining a healthy gut flora environment through antitoxin activity and inhibition of pathogen colonization, they have been of interest to the medical research community for quite some time now. Probiotic bacteria such as Lactobacillus plantarum, which can be found in fermented food, are of particular interest given their easy accessibility. We performed whole-genome sequencing and genomic analysis on a GB-LP1 strain of L. plantarum isolated from Korean traditional fermented food; this strain is well known for its functions in immune response, suppression of pathogen growth, and antitoxin effects. The complete genome sequence of GB-LP1 is a single chromosome of 3,040,388 bp with 2,899 predicted open reading frames. Genomic analysis of GB-LP1 revealed two CRISPR regions and genes showing accelerated evolution, which may have antibiotic and antitoxin functions. The aim of the present study was to predict strain specific-genomic characteristics and assess the potential of this new strain as lactic acid bacteria at the genomic level using in silico analysis. These results provide insight into the L. plantarum species as well as confirm the possibility of its utility as a candidate probiotic.
Ulcerative colitis (UC), a chronic inflammatory bowel disease, substantially impacts patients' health-related quality of life. In this study, an effective strategy for discovering high-efficiency probiotics has been developed. First, in order to survive in the conditions of the stomach and intestine, high bile salt-resistant and strong acid-resistant strains were screened out from the fecal flora of healthy adults. Next, the probiotic candidates were rescreened by examining the induction ability of IL-10 (anti-inflammatory factor) production in dextran sodium sulfate (DSS)-induced colitis mice, and Lactobacillus sakei 07 (L07) was identified and selected as probiotic P. In the end, fourteen bifidobacterium strains isolated from stools of healthy males were examined for their antimicrobial activity. Bifidobacterium bifidum B10 (73.75% inhibition rate) was selected as probiotic B. Moreover, the colonic IL-6 and $TNF-{\alpha}$ expression of the DSS-induced colitis mice treated with L. sakei 07 (L07) - B. bifidum B10 combination (PB) significantly decreased and the IL-10 expression was up-regulated by PB compared to the DSS group. Furthermore, Bacteroidetes and Actinobacteria decreased and Firmicutes increased in the DSS group mice, significantly. More interestingly, the intestinal flora biodiversity of DSS colitis mice was increased by PB. Of those, the level of B. bifidum increased significantly. The Bacteriodetes/Firmicutes (B/F) ratio increased, and the concentration of homocysteine and LPS in plasma was down-regulated by PB in the DSS-induced colitis mice. Upon administration of PB, the intestinal permeability of the the DSS-induced colitis mice was decreased by approximately 2.01-fold. This method is expected to be used in high-throughput screening of the probiotics against colitis. In addition, the L. sakei 07 - B. bifidum B10 combination holds potential in UC remission by immunomodulatory and gut microbiota modulation.
In this study, the survival capacity (acid and bile salt tolerance, and adhesion to gut epithelial cells) and probiotic properties (enzyme activity-inhibition and anti-inflammatory activities, inhibition of adipogenesis, and stress hormone level reduction) of Lactiplantibacillus plantarum LRCC5314, isolated from kimchi (Korean traditional fermented cabbage), were investigated. LRCC5314 exhibited very stable survival at ph 2.0 and in 0.2% bile acid with 89.9% adhesion to Caco-2 intestinal epithelial cells after treatment for 2 h. LRCC5314 also inhibited the activities of α-amylase and α-glucosidase, which are involved in elevating postprandial blood glucose levels, by approximately 72.9% and 51.2%, respectively. Treatment of lipopolysaccharide (LPS)-stimulated RAW 264.7 cells with the LRCC5314 lysate decreased the levels of the inflammatory factors nitric oxide, tumor necrosis factor (TNF-α), interleukin (IL)-1β, and interferon-γ by 88.5%, 49.3%, 97.2%, and 99.8%, respectively, relative to those of the cells treated with LPS alone. LRCC5314 also inhibited adipogenesis in differentiating preadipocytes (3T3-L1 cells), showing a 14.7% decrease in lipid droplet levels and a 74.0% decrease in triglyceride levels, as well as distinct reductions in the mRNA expression levels of adiponectin, FAS, PPAR/γ, C/EBPα, TNF-α, and IL-6. Moreover, LRCC5314 reduced the level of cortisol, a hormone with important effect on stress, by approximately 35.6% in H295R cells. L. plantarum LRCC5314 is identified as a new probiotic with excellent in vitro multifunctional properties. Subsequent in vivo studies may further demonstrate its potential as a functional food or pharmabiotic.
Fermentation is an effective process for providing various beneficial effects in functional beverages. Lactic acid bacteria and yeast fermentation-based biotransformation contribute to enhancement of nutritional value and digestibility, including lactose intolerance reduction and control of infections. In this study, the probiotic fermented fruit juice (PFJ) was produced by Lactobacillus plantarum TISTR 1465, Lactobacillus salivarius TISTR 1112, and Saccharomyces boulardii CNCM I-745 while mixed fruit juice (MFJ) was used as the basic medium for microorganism growth. The potential function, the anti-salmonella activity of PFJ, was found to be effective at 250 mg/ml of MIC and 500 mg/ml of MBC. Biofilm inhibition was performed using the PFJ samples and showed at least 70% reduction in cell attachment at the MIC concentration of Salmonella Typhi DMST 22842. The antioxidant activities of PFJ were determined and the results revealed that FSB.25 exhibited 78.40 ± 0.51 mM TE/ml by FRAP assay, while FPSB.25 exhibited 3.44 ± 0.10 mM TE/ml by DPPH assay. The volatile compounds of PFJ were characterized by GC-MS, which identified alcohol, aldehyde, acid, ester, ketone, phenol, and terpene. The most abundant organic acid and alcohol detected in PFJ were acetic acid and 2-phenylethanol, and the most represented terpene was β-damascenone. The sensory attributes showed scores higher than 7 on a 9-point hedonic scale for the FPB.25, illustrating that it was well accepted by panelists. Taken together, our results showed that PFJ could meet current consumer demand regarding natural and functional, fruit-based fermented beverages.
Probiotics로서의 사용을 위한 우수 젖산균주 선발을 위해 김치 및 발효유 제품으로부터 분리한 젖산균과 공시 젖산균주를 대상으로 내산성, 내담즙성, 장내 점착능, 병원성균 억제능 등의 probiotics 특성과 장관면역활성, mitogenic activity 및 대식세포 활성화 등의 면역활성을 검토하였다. 내산성 실험의 결과, Lactobacillus acidophilus DDS-1, Lb. acidophilus B-3208, Lb. plantarum과 Leuconostoc mesenteroides ssp. mesenteroides ATCC 8293의 경우 생존률이 50% 이상으로 나타났으며, 특히 Lb. acidophilus DDS-1과 Lb. acidophilus B-3208의 경우에는 70% 정도의 높은 생존률을 나타내었다. 또한 젖산균주의 담즙산 내성에서는 Lactococcus lactis ssp. lactis ATCC 7962를 제외한 모든 균주가 3% oxgall이 함유된 MRS배지에서도 성장이 가능한 것으로 관찰되었다. 젖산균주의 장내 점착능 면에서 볼 때, Lb. acidophilus DDS-1, Lb. acidophilus B-3208과 Bifidobacterium bifidum KCTC 3357의 장내 점착능이 실험에 사용된 다른 젖산균주와 비교하여 우수하다고 관찰되었으며, 특히 Lb. acidophilus DDS-1과 B-3208의 장내점착능이 높게 나타났다. 젖산균주의 병원성균 억제능 실험의 결과, Lb. acidophilus DDS-1, Lb. acidophilus B-3208 및 B. bifidum KCTC 3357은 Staphylococcus aureus를 제외한 Escherichia coli O157:H7 ATCC 43895, Listeria monocytogenes ATCC 59414, 그리고 Salmonella enteritidis ATCC 49313에 대해 높은 증식억제능을 나타내었다. 젖산균의 세포질 획분과 세포벽 획분을 대상으로 면역활성 실험을 실시한 결과, 장관면역 활성은 젖산균의 세포질 획분에서 양성 대조군으로 사용한 LPS와 같은 수준의 면역활성을 나타내었고, 특히 Lb. acidophilus속 균주들의 세포벽 획분의 장관 면역활성이 다른 균주보다 높게 나타났다. 한편 젖산균주의 세포질과 세포벽 성분에 대한 비장 림프구의 증식능은 대조군과 비슷한 수준으로 낮게 관찰되었으나, 이들의 대식 세포증식능은 세포벽 및 세포질 모두의 획분에서 대조군보다 높았으며 특히 세포벽 획분의 경우에는 양성대조군인 LPS 보다 높거나 유사한 정도의 높은 활성을 확인할 수 있었다. 이상의 결과로부터 시험 균주 중에서 Lb. acidophilus 균주인 DDS-1과 B-3208이 프로바이오틱스로서 요구되는 조건을 충족시킨다는 것을 알 수 있었으며, 이들 균주의 상업적 이용 가능성을 재차 확인할 수 있었다.
The beneficial effects of probiotics on human health have been extensively studied. Strains of Lactobacillus and Bifidobacterium have gained considerable attention, as they are major groups of probiotic bacteria that improve gastrointestinal health. Recently, probiotics have been found to have positive effects on obesity and type 2 diabetes mellitus (T2D) that go beyond the improvement of gut health. Probiotics can alter the secretion of substances such as leptin and adiponectin from white adipose tissue via certain mechanisms. Additionally, probiotics can modulate the adaptive immune system against low-grade inflammation caused by obesity. The potential role of probiotics in the treatment of T2D has garnered interest because of increasing evidence of the anti-diabetic effect of probiotics. In this review, we discuss the results of in vivo studies that have examined the use of probiotics to prevent obesity and T2D, while primarily focusing on the studies that explore the cellular and molecular mechanisms underlying the activity of promising probiotic strains. The conclusions of this review could be of help to researchers who are investigating the effects of probiotics on obesity or T2D, and to patients with these diseases.

제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
