Chloroplast genome sequence and PCR-based markers for S. cardiophyllum (감자 근연야생종 Solanum cardiophyllum의 엽록체 전장유전체 구명 및 이를 이용한 S. cardiophyllum 특이적 분자마커의 개발)

  • The diploid Solanum cardiophyllum, a wild tuberbearing species from Mexico is one of the relatives to potato, S. tuberosum. It has been identified as a source of resistance to crucial pathogens and insects such as Phytophthora infestans, Potato virus Y, Colorado potato beetle, etc. and is widely used for potato breeding. However, the sexual hybridization between S. cardiophyllum and S. tuberosum is limited due to their incompatibility. Therefore, somatic hybridization can introduce beneficial traits from this wild species into the potato. After somatic hybridization, selecting fusion products using molecular markers is essential. In the current study, the chloroplast genome of S. cardiophyllum was sequenced by next-generation sequencing technology and compared with those of other Solanum species to develop S. cardiophyllum-specific markers. The total length of the S. cardiophyllum chloroplast genome was 155,570 bp and its size, gene content, order and orientation were similar to those of the other Solanum species. Phylogenic analysis with 32 other Solanaceae species revealed that S. cardiophyllum was expectedly grouped with other Solanum species and most closely located with S. bulbocastanum. Through detailed comparisons of the chloroplast genome sequences of eight Solanum species, we identified 13 SNPs specific to S. cardiophyllum. Further, four SNP-specific PCR markers were developed for discriminating S. cardiophyllum from other Solanum species. The results obtained in this study would help to explore the evolutionary aspects of Solanum species and accelerate breeding using S. cardiophyllum.

  • Calmodulin (CaM) mediates cellular Ca2+ signals in the defense responses of plants. We previously reported that GmCaM-4 and 5 are involved in salicylic acid-independent activation of disease resistance responses in soybean (Glycine max). Here, we generated a GmCaM-4 cDNA construct under the control of the cauliflower mosaic virus (CaMV) 35S promoter and transformed this construct into potato (Solanum tuberosum L.). The constitutive over-expression of GmCaM-4 in potato induced high-level expression of pathogenesis-related (PR) genes, such as PR-2, PR-3, PR-5, phenylalanine ammonia-lyase (PAL), and proteinase inhibitorII (pinII). In addition, the transgenic potato plants exhibited enhanced resistance against a bacterial pathogen, Erwinia carotovora ssp. Carotovora (ECC), that causes soft rot disease and showed spontaneous lesion phenotypes on their leaves. These results strongly suggest that a CaM protein in soybean, GmCaM-4, plays an important role in the response of potato plants to pathogen defense signaling.

  • 거의 모든 국가에서 생산된 씨감자는 감자바이러스 이병율을 검사해서 합격품만을 농민에 분배하고 있다. 우리나라는 딴 씨감자 생산국과는 사정이 달라 고령지에서 씨감자를 생산하고 있는 관계로 감자밭의 필수는 전국적으로 막대한 수에 달한다. 우리나라에서의 감자바이러스 김정은 원원종에 한하고 있으나 농민에 분배되는 씨감자도 바이러스 검정을해서 합격품만을 나누어주어야 국가의 체면이 서겠다. 그러나 현재의 시설로는 이를 감당 할 수 없어 이에 대치 될 만한 실제적 방법의 하나로 매개진딧물 밀도와 감자바이러스 이병율간의 상관관계를 밝혀 번잡한 바이러스 검정법 대신 매개진딧물의 공중밀도를 조사하여 해지점의 감자바이러스 이병율을 추산 할 목적으로 본 시험에 착수하였다. 본 보고에서는 진딧물조사 결과만을 발표하고 18개소에서 행한 포장시험결과는 1970년에 행할 바이러스 검정을 마치면 종합결과를 발표 할 예정이다. 표 1에서와 같이 서해안, 내륙, 및 대관령에서의 매개진딧물과 일반진딧물의 수는 각각 8/31, 179/680 및 50/237이었으며 필자가 행한 1967, 1968년의 조사성적을 뒷받침하는 결과를 얻었다. 이에 필자는 씨감자 부족량 약 60,000톤의 조속한 충족을 위해 씨감자의 증식 단계에서는 서해안 일모작 답을 환용 할 것을 다시 한번 강조하는 바이다.

Incidence and Occurrence Pattern of Viruses on Peppers Growing in Fields in Korea (국내 노지재배 고추의 바이러스 발생률 및 발병 현황)

  • Field surveys to investigate the incidence and occurrence pattern of viruses in red pepper were conducted during 2015-2016 in Korea. A total of 424 samples in 2015 and 368 samples in 2016 were collected based on selection of plants showing symptoms from farmer's field from every June to September. Reverse transcription-Polymerase chain reaction was used to test all samples for the presence of one or more of following viruses: Cucumber mosaic virus (CMV), Broad bean wilt virus 2 (BBWV2), Tomato spotted wilt virus (TSWV), Beet western yellows virus (BWYV), Pepper mottle virus (PepMoV), Potato virus Y (PVY) and Pepper mild mottle virus (PMMoV). The average disease incidence was 91.7% in 2015 and 98% in 2016 and the all seven viruses were found although there were different kinds of regions. The percent virus incidence in collected samples during 2015 was as follows: CMV, 73.8%; BBWV2, 68.3%; BWYV, 46.9%; PMMoV, 14.6%; TSWV, 12.7%; PepMoV, 6.6% and PVY, 3.3%. For 2016, incidence was as follows: CMV, 73.3%; BBWV2, 71.4%; BWYV, 34.7%; TSWV, 27.9%, PMMoV, 19.2%; PepMoV, 13.5% and PVY, 3.5%. Mixed infections were prevalent over single infections and infection rate was 83% and 86.7% in 2015 and 2016, respectively.

Classification of Potato Virus Y Strains based on Reactions of Differential Plants (감자바이러스 Y 계통분류를 위한 판별 식물)

  • Four varieties of tobacco, Nicotiana tabacum var. NC 2326. NC 95, NC 744 and Havana. and two plant species, Physalis floridana Rydb., N. repanda Wild. were used for classification of ten isolates of potato virus Y(PVY) obtained Iron potato or tabacco plants showing various symptoms. Each of the 10 isolates could be distinguished by the host reactions showing necrosis, vein banding and/or mottling, or no symptom on these hosts. Five isolates of PVY, PVY-VN, PVY-N, PVY-NSNR, PVY-Chile, and PVY-Argentina, showed necrotic symptom on NC 2326, but others showed vein handing and/or mottling symptom. On NC95, each of the isolates tested showed similar symptons as on NC2326, except necrotic symptom by the isolate PVY-MSNR. Havana showed mottling reaction to the PVY-NSNR, PVY-MSNR, PVY-MSMR, PVY-VB, and necrosis to PVY-Chile, PVY-Argentina, but showed no symptom to the others. NC744 showed mottling symptom to the PVY-MSNR and PVY-MSMR, necrosis to the PVY-Chile and PVY-Argentina, and no symptom to the others. On N. repanda, necrotic reaction to PVY-Argentina. no symptom to PVY-VN and PVY-C, and nettling to others were observed.

Characterization of Tobacco mosaic virus Isolated fromSolanum tuberosum ‘Chubak’ in Korea (감자 '추백' 에 발생한 Tobacco mosaic virus 의 특성)

  • An isolate of Tobacco mosaic virus (TMV) was isolated from potato cultivar ‘Chubak’ showing vein clearing and mild mosaic at Namhae in Korea. This isolate, TMV-St, was differentiated from other tobamoviruses based on biological properties, serological relationships and nucleotide sequence analyses of coat protein genes. TMV-St caused typical symptoms on four indicator plants as compared to the tobamovirus of TMV-U1, Pepper mild mottle virus (PMMoV), and Tomato mosaic virus (ToMV), which caused economic losses in Solanaceous vegetables, tomato, pepper, and eggplant. Remarkably, the TMV-St induced distinctly different symptom of systemic chlorotic spots on Chenophodium murale. On C. murale, Gomphorena globosa, and Nic-otiana rustica, the four viruses were classed by the virulence of systemic or local infections. In serological test TMV-St antiserum showed a precipitation line with each other tabamovirus. The CP gene of TMV-St contain 477 nucleotides, and the nucleotides sequence was the most similar to that of TMV-U1.

Effects of Light-emitting Diodes on In Vitro Growth of Virus-free Sweet Potato Plantlets (LED가 고구마 바이러스 무병묘의 기내 생장에 미치는 영향)

  • The in vitro growth of virus-free sweet potato [Ipomoea batatas (L.) Lam.] plantlets was investigated under different light sources: fluorescent lamp (control); red (660 nm), blue (460 nm), white light-emitting diodes (LED), and two mixtures of blue and red LED (R:B = 8:2, and 7:3). Single node explants (10 mm) of three cultivars ('Matnami', 'Shincheonmi', and 'Yeonhwangmi') were cultured on Murashige and Skoog medium supplemented with $0.2mg{\cdot}L^{-1}$ 6-benzyladenine for 4 weeks. Explants were exposed to $150{\pm}5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ photosynthetic photon flux at a distance of 20 cm, constant temperature of $25^{\circ}C$, and under 16/8-h (day/night) photoperiod. Using the same method, the in vitro growth of 10 cultivars under red LED was also compared. After 3 weeks, vine length was highest in plantlets cultured under red LED, and lowest in plantlets cultured under blue LED. Fresh and dry weights were also greatest in plantlets cultured under red LED. Compared to the control, vine thickness was significantly higher in plantlets grown under white LED and the 7:3 R:B LED mixture. Significant differences were observed among the 10 cultivars grown under red LED. 'Matnami', 'Shincheonmi', and 'Shinhwangmi' all had excellent vine lengths, and fresh and dry weights. Compared to the control, vine elongation of sweet potato plantlets was most effective under red LED, and culture duration was about 1 week shorter.

Effects of Cultural Conditions on the In Vitro Propagation of Plantlets derived from Apical Meristem in Ipomoea batatas L. (고구마 정단분열조직 유래 식물체의 기내 증식에 미치는 배양조건의 영향)

  • Sweet potato is a crop vegetatively propagated by vine cuttings, an ineffective method for maintaining pathogene-free stock plants. As an alternative method, single-node cultures of virus-free plantlets derived from apical meristem in sweet potato (cv. Yulmi) was examined. Effective pH range, sugar concentration and nodal order were investigated to establish an in vitro mass propagation system with high quality virus-free stock plantlets to farmhouse. Although the plantlets grew at wide range of pH, the most effective pH of the medium was 4.8 in single-node cultures. High sugar concentration of 60∼80 g/L resulted in increased growth response in shoot length, root length, number of node, leaf area and fresh and dry weight of shoot and root, whereas reducing sugar contents below 6% was showed reduced growth response. The first node including meristem tip was the best for the rapid growth of plantlets and the other nodes also showed a very similar growth response. Uniform plantlet can be obtained massively at the same time by culture of single node except for the first node including meristem tip. In conclusion, the most effective pH range and sugar concentration of medium for the growth of plantlets via single-node cultures was 4.8, 60∼80 g/L respectively. The first node was the best for the rapid propagation of plantlets in nodal order.

Breeding of Burley Tobacco KB 110 Resistant to PVY and Black Shank and its Agromomic Characteristics (감자바이러스 Y 및 역병 저항성 연초 버어리종 신품종 KB 110의 육성 및 농경적 특성)

  • The vein-necrosis strain or potato virus Y (PVY-Vff) and black shank (Phytophlhora parasitica roar. nicotianae) causes severe damage on burley tobacco(Wicotiana tabacum L.) in Korea, A new burley tobacco resistance to PVY and black shank, KB 110, was developed by Korea Ginseng and Tobacco Research Institute. It was developed from the cross of Burley 21 with TC 591 in 1990, and was backrossed to Burley 21 in the following season. TC 591 has resistance to PVY and moderate resistance to race 0 of black shank, but it is susceptible to tobacco mosaic vim (TMV). KB 110 was evaluated for its resistance to PVY, TMV and black shank in the greenhouse and at fields for preliminary and performance trials. KB 110 which has secreting glandular trichomes was resistant to PVY-VN, TW and black shank. It had an erect growth habit and two more leaves per plant than that of Burley 21, and matures two to three days later. It yielded approximately 3 percent more cured leaf than the standard cultivar Burley 21, but other plant characteristics were very similar to those of Burley 21. It had acceptable standards for chemical and physical characteristics of lured leaf on regional farm test in 1995-1997. KB 110 produced average yields of good quality tobaccos and was appeared to be resistant to PVY inwhere occurrence of the virus are severe chronic at burley growing area.

Incidence of Viral Diseases on Red Pepper in Yeongyang-Gun, Gyeongbuk Provice (경북 영양지역 노지고추의 바이러스병 발생양상)

  • Incidence of virus diseases in red pepper of open field in Yeongyang-Gun, Gyeongbuk Province was investigated by reverse transcription-polymerase chain reaction in 2012-2016. The infection rate of Cucumber mosaic virus (CMV), Broad bean wilt virus 2 (BBWV2), Pepper mottle virus, Potato virus Y, Pepper mild mottle virus and Tomato spotted wilt virus was 46.1%, 41.5%, 2.0%, 2.0%, 4.4% and 0.1% respectively. Incidence rate of single and mixed infection was 31.2% and 62.6%. Most of single infections were CMV and BBWV2. Among mixed infections, the incidence rate of CMV+BBWV2 mixed infection was the highest as 49.3% and most of mixed infections of triplex and tetraplex included CMV+BBWV2 mixed infection. CMV single infection caused mosaic, chlorosis, yellowing and vein necrosis and BBWV2 single infection induced cholosis and mosaic. CMV+BBWV2 mixed infection caused severe mosaic with chlorosis or malformation.
