Characteristics of Absorption and Accumulation of Inorganic Germanium in Panax ginseng C. A. Meyer

  • The characteristics of absorption and accumulation of inorganic germanium in Panax ginseng C. A. Meyer were examined. In 4-year-old P. ginseng, the germanium content of the field soil increased with increased amounts and frequencies of inorganic germanium application, while chemical components of the soil, such as available phosphate and exchangeable calcium, potassium, and magnesium, decreased with the increased inorganic germanium application. In the 4-year-old P. ginseng, the germanium content was highest in the rhizome and increased in the order of stem, leaf, lateral root, and main root, suggesting that inorganic germanium was absorbed from the root and translocated to the stem and leaf via the rhizome. As for changes in ginsenosides in 4-year-old P. ginseng rhizomes, the contents of ginsenosides $Rb_1$, $Rb_2$, Re, and Rf decreased as the germanium content in soil increased. Ginsenosides $Rb_1$, $Rb_2$, Rc, Re, and Rf in the main root also decreased with increasing germanium content in the main root. The results suggest that inorganic germanium treatment may increase organic germanium in harvested P. ginseng, thus enhancing the medicinal effi cacy of ginseng products.

Physicochemical and Functional Properties of Turnip (순무의 이화학적 및 기능적 특성)

  • This study was conducted to investigate physicochemical and functional properties of turnip. The concentrations of antocyanin in the rind of root was 3.24mg%, which was about 3.5 times higher than those in the flesh. Among free amino acids in each part of turnip, glutamic acid showed the highest concentrations(132.5mg%) in the root followed by alanine(25.0mg%), valine (23.3mg%), and serine (20.7mg%). Contents of minerals in the root of turnip were 395.7mg% for potassium, 187mg% for calcium, 53.6mg% for phosphate, and 40.7mg% for magnesium. According to dynamic heasdspace analysis, dimethyl disulfide was the most abundant flavor components from the juice of fresh turnip and other volatile components such as dimethyl trisulfide, 1 hexanal, 2 methyl 1 butamine, and 1 penten 3 ol were also identified. From the blanched turnip, however, 3 isothiocyanato 1 propene and 4 isothiocyanato 1 butene were mainly detected. Fresh and blanched turnips were extracted with water or 70% acetone to investigate percent yield. The concentrations of total polyphenols in the extracts of fresh turnip were higher than those of blanched turnip. The nitrite scavenging effects of water and 70% acetone extracts from fresh or blanched turnip were decreased as pH was increased. Between two water extracts, the 30oC water extracts showed the higher values of electron donating ability than 95$^{\circ}C$ water extracts.

Simulation of a Six-zone Simulated Moving Bed Chromatographic Process for NPK Fertilizer Production (NPK 비료 생산을 위한 6구역 모사이동층의 공정모사)

  • Using strong cation exchange simulated moving bed (SMB) chromatography, a nitrogen-phosphate-potassium (NPK) fertilizer is produced in a cost-effective manner. The SMB process operated in a non-traditional way is divided into production and regeneration sections for exclusion of undesirable ions, and composed of six zones including two wash-water zones. This paper addresses modeling and simulation studies on the ion-exchange SMB process, comparing simulation results with experimental data obtained both from a pilot plant and an industrial plant. The simulation results show a good agreement with in situ experimental data obtained in the two plants. The model equation validated by the experiments will be applicable for optimization problems to obtain optimum operating conditions of the process.

Conditions for protoplast formation and fusion of the killer yeast (Killer 효모의 원형질체 형성 및 융합조건)

  • Auxotrophic mutant were isolated from wild types by the treatment with NTG as a mutagen, and the conditions of protoplast formation for them were established. The protoplasts of killer yeast Saccharomyces cerevisiae K52 were formed to the level of above 70% when cells grown for 20 hr in PM medium were treated with 200 unit/ml Lyticase 50,000 at $30^{\circ}C$ for 60 min after pretreatment of 50 mM 2-mercaptoethanol in 10mM potassium phosphate buffer (pH 7.5) containing EDTA and 0.6 M sorbitol for 15 min. Also, the protoplast of the recipient S. cerevisiae S 29 were formed to the level of above 85% as it was cultured to the log phase of 24 hr in PM medium under the same conditions. The fusion frequency between the protoplast of killer yeast S. cerevisiae K 52 and the protoplast of recipient S. cerevisiae S 29 was reached to $8.2\times 10^{-6}$ when the hypertonic regeneration medium embeded with the fused protoplasts after mixing the parental protoplasts to 10$^{8}$ cells/ml in SP buffer containing 20 mM $CaCl_{2}$ and 30% PEG 6,000 for 15 min at $30^{\circ}C$ were incubated.

Enzymatic Properties of Extracellular Cytosine Deaminase (세포외 Cytosine Deaminase의 효소학적 성질)

  • Enzymological proprties of an extracellular cytosine deaminase from Bacellus polymyxa YL 38-3 were investigated. The extracellular enzyme was very stable, and optimum pH and temperature for the enzyme activity were found to be near pH 6.0 in 0.2M potassium phosphate buffer and at $30^{\circ}C$, respectively. 5-Fluorocytosine was converyed to 5-fluorouracil by the enzyme, but 5-methylcytosine was not to thymine by it. The enzyme activity was completely inhibited by some heavy metal ion such as 1mM of $Cd^{2-}$ and $Hg^{2+}$, and by 1mM of p-chloromercuribenzoate, respectively. The enzyme activity was inactivated about 75% by 1mM of o-phenanthroline and monoiodoacetate. But the enzyme activity was stimulated up to 200% by 1mM of 2-mercaptoethanol.

  • In order to find out optimum seed production date according to different defoliation and flooding period of Italian ryegrass, this studies with Tetrone were canied out on the experimental field of Honam Crop Experiment Station from 1986 to 1988. Treatments included cutting date of Nov. 20 and Apr. 10 and flooding period of 5-25 days. In soil after experiment, organic matter, phosphate and silicate content increased, but potassium content decreased 0.16% than that before experiment. Heading and maturing date of Nov. 20 cutting were same with those of non cutting, those of Apr. 10 cutting lates 6 days to heading date and 2 days to maturing date. Plant height and culm length of Nov. 20 cutting were sirniller to those of non cutting, those of Apr. 10 cutting were shorter and panicle length have no difference between non cutting and cutting. Lodging of cutting treatment was reduced than that of non cutting. Lodgin was increased as flooding period was long, also loding of all treatment occured at 30 days after heading. Two cutting times of Nov. 20 and Apr. 10 have the most fresh yield, while non cutting have the most dry matter yield. Optimun seed productin date was considered to suitable when 35 days after heading (Jun. 14), at this time, seed production was 1,640 to 2,640 kg/ha. Also if flooding j u r y have, seed production was good between 10 days and 15 days after flooding.

  • The optimal treatments of carcinoma in situ of glottis include radiotherapy, laser surgery and vertical partial laryngectomy. Conventional surgical treatments need general anesthesia and radiotherapy has several complications. Recently, the effectiveness of 532 nm potassium titanyl phosphate (KTP) laser has been proven and widely used in vocal fold diseases even some cases of vocal fold dysplasia. A patient with difficult laryngeal exposure underwent fiberoptic laryngeal laser surgery using KTP laser under local anesthesia, showed improved voice outcome and the glottic lesion was removed successfully without local recurrence and regional metastasis 18 months after surgery.

  • Currently, the potentiometric titration and the high pressure liquid chromatography (HPLC) method were utilized in Korean Pharmacopoeia XII (KP XII) as well as other pharmacopoeias (USP, EP, BP) for determination of loperamide hydrochloride in raw materials and capsules, respectively. The research objective is to overcome the remaining drawbacks from current methods such as solubility of mobile phase (KP XII), less scientific approach (USP 43) or using paired-ion chromatography reagent which shows some limitations (BP2017 and other formulation monographs). The proposed method was optimized by Design of Experiment (DoE) tool to obtain the satisfied method for determination of loperamide hydrochloride. The optimal condition was performed on the common C18 column (150 mm × 4.6 mm; 5 ㎛) using isocratic elution with the mobile phase containing 40 mM of potassium phosphate monobasic (pH 3.0) and acetonitrile (56:44), at a flow rate of 0.7 mL/min. The optimized method was validated and met the requirements of the International Conference on Harmonization. The developed method was applied to determine loperamide hydrochloride in capsules and can be used to update the current monograph in KP XII.

Amplification of Glutathione Production in E. coli Cells Using Recombinant DNA Techniques

  • Conditions for glutathione production in E. coli cells which possess pGH501 (2 gshI+gshII) were studied. In terms of ATP supply for the glutathione synthesis, two different systems have been constructed and compared. When the acetate kinase reaction of E. coli was used for ATP generation, 20 mM of L-cysteine was completely converted to glutathione by toluene-treated E. coli cells (100 mg/ml) harboring pGH501 within 2 h at $37^{\circ}C$. However, considering the economical aspects, the glycolytic pathway of yeast was chosen as a better system for ATP generation. The optimal concentrations of reactants for glutathione production were determined to be as follows; 80 mM L-glutamate, 20 mM L-cysteine, 20 mM glycine, 20 mM $MgCl_2$, 50 mM potassium phosphate buffer (pH 7.5), 400 mM glucose, polyoxyethylene stearylamine ($5\;\mul/ml$), toluene-treated E. coli HB101/pGH501 (100 mg/ml), and dried yeast cells (400 mg/ml). The conversion ratio of L-cysteine to glutathione was 80% (about 5 mg/ml) under optimal condition within 6 h at $37^{\circ}C$.

The growth of large KTP crystal and the study of its optical inhomogeneity (대형 KTP 단결정 성장 및 광학적 불균일성에 관한 연구)

  • Single crystals of Potassium Titanyl Phosphate (KTP) were grown from the flux of $K_6P_4O_13(K_6)$ using a high temperatures solution growth method. To grow the large KTP crystal without inclusion, the temperature gradient in furnace, crystal rotation, orientation of seed crystal, and the cooling rate were controled. The KTP crystals are up to $10(a){\times}28(b){\times}33(c)mm^3$ in size. We investigated the optical inhomogeneity in this KTP crystal by the SHG power measurement and TEM analysis.
