Effect of Slurry Composting Biofiltration (SCB) Liquid Manure on Shoot Growth and Fruit Qualities of Peach (Prunus persica L.) and Soil Chemical Properties in Orchard

  • This study was carried out to investigate the effect of slurry composting and biofiltration (SCB) liquid manure application on shoot growth, fruit qualities and soil chemical properties in peach orchard. SCB liquid manure was fertigated ten times from April to October in SCB plot, whereas chemical fertilizer was treated two times as basal and additional fertilizers in control plot. The shoot growth, leaf nitrogen and potassium content, soil exchangeable K, fruit weight and yield were higher in SCB plot than in control. Soluble solid content and acidity, soil organic matter, soil available phosphate and soil exchangeable Mg showed no significant difference between treatments, and the leaf calcium and magnesium content were lower in SCB plot than in control. In conclusion, fertigating SCB liquid manure in peach orchard has positive effects on fruit weight and yield, and it is suggested that periodical soil testing is needed because of the possibility of K accumulation in SCB liquid manure treated soil when the orchard is fertigated based on the soil nitrogen content.

  • Leuconostoc citreum is one of the representative strains of Leuconostoc spp. that show fast growth rates in fermented vegetables. Sequential experimental designs including the Plackett-Burman design, fractional factorial design, steepest ascent analysis, central composite design and response surface methodology were introduced tooptimize and improve the medium for Leuconostoc citreum. Fifteen medium ingredients were examined and glucose (20 g/l), yeast extract (12.5 g/l), sodium acetate trihydrate (6.12 g/l), potassium phosphate (42.55 g/l) and dibasic ammonium citrate (4.12 g/l)were chosen as the best components to give a critical and positive effect for cell-growth. The biomass was increased to 2.79 g/l (169%), compared to the 1.65 g/l in MRS medium.

The biomass was increased to ($2.79g/\ell$), (169%), compared to the $1.65g/\ell$ in MRS medium.

Growth Regulators Prolong Bract Longevity of Potted Bougainvillea

  • When bougainvilleas are subjected to indoor low-light conditions, flower bracts regularly abscise. This study elucidates the effects of plant growth regulators on bract longevity of potted bougainvillea. Potted 'Taipei Red' bougainvillea in four different bract development stages were treated with 1-MCP (1-methylcyclopropene), NAA (1-naphthaleneacetic acid), SNA (sodium salt of naphthaleneacetic acid), IBA (indolebutyric acid), BA (6-benzylaminopurine), $KH_2PO_4$ (potassium dihydrogen phosphate), Put (diamine putrescine), SA (salicylic acid), or STS (silver thiosulfate) and were moved to indoor low-light conditions after treatments. Experimental results indicate that 1-MCP, NAA, SNA, BA, Put, and SA prolonged bract longevity, and this effect increased as bract stage increased. The effect of STS was significant in early bract stages and decreased as bract stages increased. Additionally, 1-MCP, NAA, SNA, BA, Put, SA, and STS treatment significantly reduced endogenous ACC (1-aminocyclopropene-1-carboxylate) content and ACC oxidase activity, suggesting that the inhibition of ethylene production was achieved via physiological metabolism. However, treatment with IBA or $KH_2PO_4$ had no effect on the bract longevity at any stage. In the combined chemical treatments, NAA + STS or NAA + SA were effectively for prolonging bract longevity and contained less protein or chlorophyll degradation, decrease ACC oxidase or ethylene production than the control. In conclusion, we propose that combined chemical treatment significantly prolonged the bract longevity and more effectively than single chemical treatment at any stage.

제주지역 상동나무의 자생지 생육환경 및 식생조사 (Vegetation and Habitat Environment of Sageretia thea in Jeju Island)

  • This study was carried out to investigate the vegetation and habitat environment of Sageretia thea which is distributed in Jeju island, Korea. Sageretia thea were mainly distributed to the west area in Jeju island. Soil pH and EC of Sageretia thea habitat were 5.8 and $0.34dS{\cdot}m^{-1}$. The contents of organic matter, available phosphate, exchangeable potassium, exchangeable sodium, exchangeable magnesium and exchangeable calcium were 15.27%, $13.6mgkg^{-1}$, $0.27cmol_+kg^{-1}$, $1.3cmol_+kg^{-1}$, $1.7cmol_+kg^{-1}$, $4.9cmol_+kg^{-1}$, respectively. Thirty one taxa including 25 families, 31 genus, 27 species and 4 varieties were identified surrounding Sageretia thea habitat. Specific plant species were 1 taxon in III grade, 1 taxon in I grade and 3 taxa in I grade.

  • Super water-absorbent resins were prepared by inverse suspension copolymerization of sodium acrylate, acrylamide and 2-hydroxyethyl acrylate using N, N'-methylene-bis-acrylamide as cross-linker. For the suspension copolymerization, monohexadecyl phosphate was employed as the dispersing agent, cyclohexane as the dispersing medium and potassium persulfate as the initiator. The dependence of water-absorption capacity on the amount of crosslinking agent, oil/water ratio, degree of neutralization and the composition of the copolymer were systematically investigated. Furthermore, the swelling kinetics of the super water-absorbent copolymer was carried out. The absorption of the resins is more than 1800 g/g for deionized water and 100 g/g for 0.9% NaCl solution, respectively. The copolymers showed an increased salt resistance and enhanced water retention of soil.

Development of Analytical Method of Biotin in Complex Drugs and Dietary Supplements Using HPLC-UV

  • Recently, Korean Food and Drug Administration (KFDA) has focused on developing quality control guidelines for all commercial products in Korea to enforce regulations, improve the quality control, and protect consumers by developing prevalently used and efficient analytical tools to determine and quantify target compounds. Because the Korean Pharmacopeia (KP) presents microbiological assays for biotin, which is laborious and time-consuming, this study is focused on applying HPLC-UV to detect and quantify biotin in complex drugs and dietary supplements like multi-vitamin. Biotin in complex drugs was extracted from methanol and analyzed using mobile phase with 10 mM potassium phosphate (monobasic, pH=3.0) in distilled water and acetonitrile. Gradient condition was used to successfully detect and quantify biotin within 20 minutes. Validation result for linearity was significant that average $r^2$ was 0.999 (n=3) and its relative standard deviation (RSD) was 0.0578% which was less than 2%. Using this method, quantification of biotin in complex drugs was completed successfully and recovery tests were finished that recovery percentage greater than 95% with relative standard deviation less than 2%.

Hypolipidemic Activities of Dietary Pleurotus ostreatus in Hypercholesterolemic Rats

  • This work was conducted to investigate dietary supplementation of oyster mushroom fruiting bodies on biochemical and histological changes in hyper and normocholesterolemic rats. Six-week old female Sprague-Dawley albino rats were divided into three groups of 10 rats each. Feeding a diet containing a 5% powder of Pleurotus ostreatus fruiting bodies to hypercholesterolemic rats reduced plasma total cholesterol, triglyceride, low-density lipoprotein (LDL), total lipid, phospholipids, and LDL/high-density lipoprotein ratio by 30.18, 52.75, 59.62, 34.15, 23.89, and 50%, respectively. Feeding oyster mushrooms also significantly reduced body weight in hypercholesterolemic rats. However, it had no adverse effects on plasma albumin, total bilirubin, direct bilirubin, creatinin, blood urea nitrogen, uric acid, glucose, total protein, calcium, sodium, potassium, chloride, inorganic phosphate, magnesium, or enzyme profiles. Feeding mushroom increased total lipid and cholesterol excretion in feces. The plasma lipoprotein fraction, separated by agarose gel electrophoresis, indicated that P. ostreatus significantly reduced plasma ${\beta}$ and pre-${\beta}$-lipoprotein but increased ${\alpha}$-lipoprotein. A histological study of hepatic cells by conventional hematoxylin-eosin and oil red O staining revealed normal findings for mushroom-fed hypercholesterolemic rats. These results suggest that a 5% P. ostreatus diet supplement provided health benefits by acting on the atherogenic lipid profile in hypercholesterolemic rats.

Optimazation of Submerged Culture Conditions for Exo-Biopolymer Production by Paecilomyces japonica

  • Optimization of submerged culture conditions for the production of exo-biopolymer from Paecilomyces japonica ws studied. Maltose, yeast extract, and potassium phosphate were the most suitable sources of carbon, nitrogen, and inorganic salt, respectively, for both production of the exo-biopolymer and mycelial growth. The optimal culture conditions in a flask culture were pH 5.0, $25^{\circ}C$, and 150 rpm in a medium containing (as in g/l) 30 maltose, 6 yeast extruct, 2 polypeptone, $0.5{\;}K_3HPO_4,{\;}0.2{\;}KH_2PO_4,{\;}0.2{\;}MnSO_4{\cdot}5H_2O,{\;}0.2{\;}MgSO_4{\cdot}7H_2O$. Exo-biopolymer production and mycelial growth in the above suggested medium were significantly increased in a 2.5-1 jar fermentor, where the maximum biopolymer concentration was 8 g/l. The morphological changes of the mycelium in the submerged culture were observed within pH ranges from 4.0 to 9.0; i.e., growth of the filamentous form was optimal at culture pHs of 5.0 and 6.0, whereas pellet was formed at other pHs.
