Longitudinal Study of Iron Concentration in Korean Preterm Human Milk

  • 목적: 미숙아에게 필요한 영양성분을 알기 위해서는 그들의 모유 성분을 분석하는 것이 도움이 된다. 저자들은 미숙아 모유의 장기적인 철분함량의 변화를 관찰하고 만삭아 모유와 비교하였으며, 이를 토대로 모유수유만 하는 미숙아에서 철분 보충의 필요성과 지침에 대해 알아보고자 하였다. 방법: 재태연령 34주 미만이거나 출생체중 1,800 g 미만의 미숙아를 출산한 산모 103명으로부터 생후 1, 2, 4, 6, 8, 12주의 모유를 수집하였고, 만삭아의 모유는 24명의 산모로부터 생후 1주째에 수집하여 철분함량을 분석하였다. 결과: 생후 2주에서 8주 사이의 미숙아 모유내의 철분함량은 유의한 차이가 없었으며(36.3${\pm}$23.1~45.8${\pm}$26.0 $\mu$g/dL), 이는 생후 1주째의 미숙아모유 (23.1${\pm}$14.6 $\mu$g/dL)나 만삭아 모유(25.2${\pm}$7.55 $\mu$g/dL)의 철분함량보다는 유의하게 높았다. 교정나이로 만삭이 된 미숙아의 모유(42.2${\pm}$19.4 $\mu$g/dL)는 만삭아의 모유보다 유의하게 높은 철분함량을 나타내었다. 결론: 미숙아 모유의 철분함량은 생후 3개월까지 큰 차이가 없었으며, 이는 필요량보다 적어 모유수유만 할 경우 AAPCON(2004)에서 권유하는 대로 2 mg/kg/day의 철분을 보충하는 것이 좋겠다.

우리나라 중남부지역 젖소목장에서 이등유 발생 조사 (Investigation of the incidence rate of second grade milk in dairy farms on the central-southern region of Korea)

  • The incidence of second-grade milk production in 9 dairy farms of South Korea was investigated from May 2011 to March 2012, and the serum composition of cows producing first- and second-grade milk in 14 farms including the 9 farms was analyzed. The incidence rate of second-grade milk production of 402 cows in nine dairy farms located in the central and southwestern regions of Korea was 15.4% with the highest rate being 34.4%. Seasonal morbidity was higher during late winter (February) and early summer (June) with the highest rate observed in February (32.6%) followed by November (33.3%). Second-grade milk was most frequently found within one month postpartum (34.1%) while only 3.5% was found during the first 60~90 days of lactating period (n=785, 5 herds). The morbidity increased thereafter (P<0.05) with the highest observed between 270~300 days of lactation (36.1%). The acidity was not significantly different between second-grade ($0.159{\pm}0.026%$) and first-grade milk ($0.158{\pm}0.027%$). Blood serum analysis of 371 cows in the 14 dairy farms indicated that aspartate aminotransferase (AST) level was significantly higher (P<0.001) in cows producing second-grade milk while albumin was significantly lower (P<0.001) than cows producing first-grade milk. Total protein and triglyceride was also significantly low along with glucose, non-esterified fatty acid and blood urea nitrogen in cows producing second-grade milk. Statistical analysis including sensitivity, specificity and positive/negative prediction values showed that lactating cows with high AST, low albumin, total protein and triglyceride levels in the serum tended to produce second-grade milk. It was concluded that serological parameters, especially live functional and metabolic-related serum compositions (AST, albumin, total protein and triglyceride), were significantly influenced in cows producing second-grade milk.

Influence of a Single Dose of Fe Dextran Administration with Organic Trace Mineral Supplementation on the Performance of Piglets

  • This study was designed to evaluate the influence of a single or double dose of Fe dextran with organic trace mineral supplementation on the performance of piglets from dams fed diets with either inorganic (ITM) or organic trace minerals (OTM). It also determined the effect of the source of the trace minerals on the reproductive performance of sows. The trace mineral premixes were prepared using metal proteinates and the corresponding inorganic salts for the OTM and the ITM, respectively. Each mineral premix provided 100 ppm Fe/175 ppm Fe, 35 ppm Cu/170 ppm Cu, 90 ppm Zn/120 ppm Zn, and 40 ppm Mn/35 ppm Mn when added at 0.20% in sows /weaned pigs' diets, respectively. The first dose of Fe dextran was administered to piglets at 3 d and the second dose at 10 d after birth. One dose of Fe dextran supplied 100 mg of Fe. A total of 16 gestating sows (Landrace${\times}$Yorkshire) in parities 2 to 4 were randomly allocated to four treatments: 1) diet with ITM/one dose of Fe dextran to piglets, 2) diet with ITM/two doses of Fe dextran to piglets, 3) diet with OTM/one dose of Fe dextran to piglets, and 4) diet with OTM/two doses of Fe dextran to piglets. The total born alive, weaned, body weight at birth and at weaning were not affected by the sow's dietary treatment. Although organic trace mineral supplementation tended to increase the milk Fe content (p<0.10) at 7 d postpartum, piglets in all treatments performed equally from birth to weaning. The double doses of Fe dextran neither improved the average daily gain (ADG) nor influenced the survival of piglets from birth to weaning (21 d). Results suggest that a single dose of Fe dextran given to suckling pigs is adequate to sustain their needs for growth throughout the lactation period (21 d). Furthermore, there was a 21% improvement in both the ADG and the average daily feed intake (ADFI) (p<0.05) in weaned pigs fed diets with OTM. Cu and Fe in the liver (p<0.01), and Zn in both the bone (p<0.01) and the serum (p<0.01) were higher in piglets fed OTM than in those fed ITM. It would be concluded that single dose of Fe dextran administration with organic trace mineral supplementation show similar growth performance compared to 2 dose Fe dextran administration with inorganic mineral supplementation in young pigs.

출산 후 경과한 날에 따른 한국인 산모의 모유 단백체 분석 (Proteomic analysis of Korean mothers' human milk at different lactation stages; postpartum 1, 3, and 6 weeks)

  • 이 연구는 출산 후 1, 3, 6주가 경과한 산모에서 얻은 모유의 단백체 발현 양상과 과 발현 단백질을 검출하는 것을 목적으로 하였다. 샷 건 정량 단백체 분석법을 이용하여 모유 중의 단백질을 동정하였고, 각 수유단계 간에 정량적 비교를 하였다. 각 주의 모유 샘플은 두 명의 산모로부터 얻어진 모유를 혼합하였고, 각 샘플 마다 3회 반복 실험을 하였다. Casein은 모유 내에 가장 많이 존재하는 단백질로서 실험의 정확성을 위하여 제거하였고, 트립신을 이용한 절편 화로 모유 단백질들을 펩타이드로 변환하였다. 처리된 펩타이드들은 역상 C18 미세관 크로마토그래피 및 이온-트랩 질량분석기를 이용하여 분석하였으며, Spectra Counting으로 단백질의 정량적 비교를 하였다. 각 샘플 당, 80-109 개의 단백질을 중복 제거한 후 동정하였다. 당화 단백질, metabolic enzyme, 및 lactoferrin, Carboxylic ester hydrolase, Clusterin을 포함하는 chaperon 효소들이 주로 검출되었다. 각 반복실험에서 재현성 있게 검출되는 63개의 단백질에 대한 정량적 비교분석 결과 25개의 단백질이 통계적으로 유의하게 수유단계에 따라 변화하는 것을 확인할 수 있었고, 특히 Ig lambda-7 chain C region과 Tenascin은 시간에 따라 현저하게 감소하였다. 향후 이와 같은 수유 단계에 따른 모유 내 단백의 변화가 생리적으로 가지는 의미에 관하여 추가적인 연구가 필요하다 생각된다.

수유기간별 모유의 주요 무기질 및 미량원소 함량 변화 (Changes in the Contents of Major Minerals and Trace Elements of Human Milk During the Breast-Feeding)

  • 한국인 수유부 21명의 모유에 함유된 무기질(Na, K, Ca, P, Mg)과 일부 미량원소(Mn, Mo, Ni)의 함량을 경시적으로 초유부터 12주의 성숙유에 이르기 까지 일정한 간격으로 세분하여 비교 분석하였다. 모유에 함유된 5종의 무기질과 8종의 미량원소들의 함량은 수유기간에 따라 유의적으로 감소하였다. 모유의 평균 나트륨 함량은 초유에서 293.01$\mu\textrm{g}$/$m\ell$, 이행유에서는 $251.24$\mu\textrm{g}$/m\ell$ 그리고 성숙유에서 $199.17$\mu\textrm{g}$/m\ell이었으며$ 칼륨농도는 각각 $436.18$\mu\textrm{g}$/m\ell,458.90,$ $390.10$\mu\textrm{g}$/m\ell이었다.$ 초유, 이행유 및 성숙유의 평균 칼슘 농도는 각각 $378.88$\mu\textrm{g}$/m\ell,$ $379.41$\mu\textrm{g}$/m\ell$ 그리고 $338.58$\mu\textrm{g}$/m\ell로$ 측정되었고 모유의 Ca/P의 비는 각각 3.39, 2.81과 2.45이었다. 또한 마그네슘 함량은 초유와 이행유에서 각각 $37~38$\mu\textrm{g}$/m\ell이었고$ 성숙유에서 $33.37$\mu\textrm{g}$/m\ell로$ 분석되었다. 모유의 평균 망간 함량은 초유와 이행유에서 각각 $0.024$\mu\textrm{g}$/m\ell,$ 성숙유에서 $0.014$\mu\textrm{g}$/m\ell,$ 믈리브덴의 평균 농도는 각각 $0.035$\mu\textrm{g}$/m\ell,$ $0.044$\mu\textrm{g}$/m\ell$ 그리고 $0.044$\mu\textrm{g}$/m\ell이었고$ 니켈 함량은 초유에서 $0.062$\mu\textrm{g}$/m\ell이행유에서$ $0.022$\mu\textrm{g}$/m\ell,$ 성숙유에서 $0.017$\mu\textrm{g}$/m\ell이었다.$ 수유기간에 따른 모유내 무기질과 미량원소 함량의 감소가 영아의 성장발달과 어떻해 관련 되는가에 관한 연구와 함께 유선조직에서 유즙으로의 무기질과 미량원소의 분비기전에 관한 생리학적 연구가 병행되어야 하겠으며, 본 연구결과가 앞으로 우수한 조제분유나 기타 영아식 개발에 활용되고, preterm milk의 무기질 성분을 비교하는데 이용될 수 있기를 희망한다.

Effect of Undegradable Dietary Protein Level and Plane of Nutrition on Lactation Performance in Crossbred Cattle

  • An experiment was conducted in order to assess the effect of level of RDP:UDP ratio and level of feeding concentrate on milk yield, milk composition and nutrient utilization in lactating crossbred cattle. Twenty four medium producing (-10 kg/d, 45 to 135 days postpartum) lactating crossbred cows were divided into four groups of six animals each in a 2${\times}$2 factorial completely randomized design. The cows in group 1 were fed concentrate mixture I containing 59:41 RDP:UDP ratio (low UDP) at normal plane (LUDP+NP), in group 2 were fed low UDP ration at 115% of NRC (1989) requirements (LUDP+HP), whereas cows in group 3 were fed concentrate mixture II containing 52:48 RDP:UDP ratio (high UDP) at normal plane (HUDP+NP) and in group 4 were fed high UDP ration at 115% of NRC (1989) requirements (HUDP+HP). Green jowar was fed ad libitum as the sole roughage to all the animals. The experimental feeding trial lasted for 105 days. The total dry matter intake (DMI), DMI/100 kg body weight, DMI/kg $W^{0.75}$, digestibilities of DM, OM, CP, CF, EE and NFE and intakes of TDN and DCP did not differ significantly among the different groups and also due to both UDP level and plane of nutrition and also due to their interaction. The total dry matter intake varied from 145 g in group 1 (LUDP +NP) to 152.57 g/kg $W^{0.75}$ in group 2 (LUDP+HP) diet. However, increase in milk yield with increased UDP level and also with increased plane of nutrition was observed consistently throughout the experimental period. The average milk yield was 7.66, 8.15, 8.64 and 9.35 kg in groups 1, 2, 3 and 4, respectively and there was no significant difference in milk yield among different groups of cows. The overall daily average milk yields in cows fed with low and high UDP diets were 7.91 and 8.99 kg, respectively and at normal and higher plane of feeding the milk yields were 8.15 and 8.75 kg/day, respectively. Thus, there was 13.65% increase in milk yield due to high UDP level and 7.36% due to higher plane of feeding. The daily 4% FCM yields were 9.20 kg for low UDP diet and 10.28 kg for high UDP diet, whereas it was 9.11 kg at normal plane of feeding and 10.37 kg at higher plane of feeding. Fat yields for the corresponding treatment groups were 0.37, 0.43, 0.41 and 0.48 kg, respectively. The 4% FCM yield and also fat yield did not differ significantly among different dietary treatments and also due to UDP level and plane of nutrition, however, 4% FCM yield was increased by 11.74% with high UDP level and 13.83% with higher plane of feeding. The values for total solids, fat, lactose, solids-not-fat and gross energy contents in milk differed significantly (p<0.05) among the different groups and were significantly (p<0.05) higher in milk of cows fed LUDP+HP diet followed by HUDP+HP diet. Total solids (14.65 and 13.83%), lactose (5.44 and 4.92%), solids-not-fat (9.44 and 8.83%) and gross energy (887 and 838 kcal/kg) of milk decreased significantly (p<0.05) with increased UDP level while total solids (13.84 and 14.64), fat (4.84 and 5.36%) and gross energy (832 and 894 kcal/kg) increased significantly (p<0.05) with increase in plane of feeding. Gross and net energetic efficiencies and also gross and net efficiencies of nitrogen utilization for milk production were not significantly different among different groups and also were not affected significantly due to either UDP levels or plane of feeding. Results of the present study suggest that, increasing UDP level from 41% to 48% of CP in concentrate mixture and also increasing plane of feeding from normal (100%) to 115% of NRC requirements maintain a consistently higher milk production.
