Reversible histoarchitecture study of testis and cauda epididymis and changes in cauda epididymal epithelial cell types on treatment with benzene extract of Ocimum sanctum leaves in albino rats

  • In the present study, an attempt has been made to assess whether the effect of benzene extract of Ocimum sanctum leaves on the ultrastructural changes in the epithelial cells of the cauda epididymis, its subsequent recovery in the seminiferous epithelium and fertility of male albino rats. Wistar strain male albino rats were orally administered benzene extract of 250 mg/kg body weight of O. sanctum leaves followed by subsequent recovery maintaining suitable controls for 48 days. Results indicate decrease in the weights of testis, epididymis and seminal vesicles. Other accessory organs were not affected. Total count, cell and nuclei diameters of germ cells and Leydig cells were reduced. Cauda epididymis exhibited significant reduction in epithelial height and nuclei diameter of epithelial cells. Cells showed vacuolization with exhibit of signs of degeneration. Ultra study revealed that, in general, the cauda epididymis was affected and in particular, the principal, clear and basal cells were highly disturbed. Further, there was decrease in the size of lipid droplets, mitochondria, Golgi complex, endoplasmic reticulum and accumulation of lysosomal bodies. Fertility performance test showed no implantation in female rats mated with O. sanctum treated rats. Moreover, their recovery after withdrawal of treatment was observed suggesting that the effect of the treatment is transient and reversible. A recovery period resulted in normal spermatogenesis and fertility, suggesting reversible antispermatogenic and antifertility effects of the plant.

Effects of 5-Aminolevulinic Acid on Growth and Inhibition of Various Plant Species

  • The purpose of this study is to investigate the positive or negative effects of 5-aminolevulinic acid(ALA) on the growth of several crops and weeds, by applying a seed soaking treatment, foliar treatment, and application timing, while comparing biological activity between ALA produced by chemical synthesis (Synthetic-ALA) and extracellularly-accumulated ALA by overexpressing the hemeA gene isolated from Bradyrhizobium japonicum(Bio-ALA). Seed soaking treatment of ALA in barley (five cultivars) and wheat (five cultivars) had not shown positive effects at lower concentrations, 0.05 to 0.5 mM as well as negative effects at higher concentrations, 1 to 30 mM. In rice, there also was no positive effect by seed soaking treatment of ALA at lower concentrations, although the rice became damaged by an application of 5 and 10 mM ALA. Growth in barley cultivars, Ganghossalbori, Naehanssalbori, Songhakbori, Saessalbori, and Daehossalbori were increased up to 14%, 19-51 %, 17-64%, 18-23%, and 22-38% by ALA foliar application at lower concentrations, 0.05 to 0.5 mM, respectively. On the other hand, the growth in barley cultivars was inhibited by ALA foliar application at higher concentrations. Barley responded more positively to ALA foliar application than wheat and rice. The growth stimulation caused by ALA seed soaking treatment was less than by ALA foliar treatment. ALA treatment at the 1.5-leaf stage increased growth of barley by 19-58%, while pretreatment to seeds, post-emergence treatment at 3 days after seeding, 3-leaf stages, and 5-leaf stages had not shown positive effects. Thus, the positive effects of ALA on barley were dependent greatly upon the timing of application and its concentration. Monocots weeds were more sensitive to ALA foliar treatment than dicotyledonous weeds. A monocot weed, Setaria viridis L. was the most susceptible plant to ALA while a dicotyledonous weed, Plantago asiatica L. was the most tolerant. No significant difference in biological activity between bio-ALA and synthetic ALA on barley, wheat, rice, and weed, Ixeris dentate tested was observed. Thus, ALA produced by microorganisms would be a potent substance to be used effectively in agricultural production.

Mulching Methods and Removing Dates of Mulch Affects Growth and Post Harvest Quality of Garlic (Allium sativum L.) cv. Uiseong

  • This experiment was conducted to elucidate the effects of mulching materials and removing time of the transparent polyethylene (PE) film on the growth of garlic at Uiseong experimental field, Korea. The experimental mulching materials comprised of transparent polyethylene film (0.025 mm) and net polyethylene (NPE). Plant height and leaf number of garlic were highest at PE treatment when the PE removing date was March 18 and this treatment also promoted the no. of cloves. Length of leaf sheath and bolting rate were highest and bulb weight loss rate was lowest at PE + NPE treatment when the PE removing date was March 18. But clove number was the lowest in this treatment compared to conventional PE film treatment. Conventional mulching method accelerated secondary growth rate but bulb weight loss was vice versa. There were statistically no differences in bulb diameter among treatments but conventional treatment positively focused on bulb diameter. Whenever PE film remove can suppress weeds compared to no mulching treatment but the dry weight of weeds were increasing trends as the removal dates of PE film were delayed. Transparent PE or PE + NPE treatments can be recommended to grow best quality garlic when PE film removing date is March 18.

Biological Control of Apple Anthracnose by Paenibacillus polymyxa APEC128, an Antagonistic Rhizobacterium

  • The present study investigated the suppression of the disease development of anthracnose caused by Colletotrichum gloeosporioides and C. acutatum in harvested apples using an antagonistic rhizobacterium Paenibacillus polymyxa APEC128 (APEC128). Out of 30 bacterial isolates from apple rhizosphere screened for antagonistic activity, the most effective strain was APEC128 as inferred from the size of the inhibition zone. This strain showed a greater growth in brain-heart infusion (BHI) broth compared to other growth media. There was a reduction in anthracnose symptoms caused by the two fungal pathogens in harvested apples after their treatment with APEC128 in comparison with non-treated control. This effect is explained by the increased production of protease and amylase by APEC128, which might have inhibited mycelial growth. In apples treated with different APEC128 suspensions, the disease caused by C. gloeosporioides and C. acutatum was greatly suppressed (by 83.6% and 79%, respectively) in treatments with the concentration of $1{\times}10^8$ colony forming units (cfu)/ml compared to other lower dosages, suggesting that the suppression of anthracnose development on harvested apples is dose-dependent. These results indicated that APEC128 is one of the promising agents in the biocontrol of apple anthracnose, which might help to increase the shelf-life of apple fruit during the post-harvest period.

Antifungal Activity of Nor-securinine Against Some Phytopathogenic Fungi

  • Crude extracts and active principles from medicinal plants have shown potential role in controlling plant diseases in glasshouses as well as in fields as one of the safest and ecofriendly methods. The effect of nor-securinine (an alkaloid) isolated from Phyllanthus amarus has been seen against spore germination of some fungi (Alternaria brassicae, A. solani, Curvularia pennisetti, Curvularia sp., Erysiphe pisi, Helminthosporium frumentacei) as well as pea powdery mildew (Erysiphe pisi) under glasshouse conditions. The sensitivity of fungi to nor-securinine varied considerably. Nor-securinine was effective against most of the fungi. H. frumentacei was more sensitive even at the lowest concentration ($1,000\;{\mu}g/ml$). Likewise conidia of E. pisi were also inhibited in partially or completely appressorium formation. Pre-inoculation treatment showed greater efficacy than post-inoculation in inhibiting powdery mildew development on pea plants in a glasshouse. Maximum inhibition occurred at $2000\;{\mu}g/ml$.

Symptomatology, Interacton and Management of Rhizome Rot of Ginger by Xenobiotics (생강 뿌리썩음증상의 병징학, 기생체간 상호작용 및 약제방제)

  • Three diffeerent types of symptoms were observed according to the pathogen associated with the rhizomes. The maximum rotting was observed in case when Pythium aphanidermatum was inoculated first followed by Fusarium solani. There was no interaction in case of root knot nematode Meloidogyne incognita and Pythium aphanidermatum. Average per cent germination of the rhizomes were increased significantly in each treatment and maximum in case of Alliette(.25%). The per cent pre & post drenching rotting was minimum in case of Alliette, Burgandy mixture, Dithane-M 45 and Difolatan. These fungicides also increase the yield of rhizome significantly.

  • This paper presents failure analysis on weld-joint of the waterwall tubes in USC boilers. Visual inspections were performed to find out the characteristics of the fracture. Additionally both microscopic characteristics and hardness test were carried out on failed tube samples. Failures seem to happen mainly because the welding process such as preheating and PWHT(post-weld heat treatment) was not conducted strictly. Thus, this paper has the purpose to describe the main cause of the poor welding process and to explain how to prevent similar failures in those weld-joints.

Removal of nitrogen and phosphorus of the secondary effluent by electro-coagulation (전기응집을 이용한 2차 유출수의 질소.인 제거 공정 연구)

  • To reduce extensive energy costs of the internal recycling for the purpose of denitrification in the advanced wastewater treatment, a post-treatment process using an electro-coagulation to treat nitrate in the secondary effluents is evaluated in this study. Removals of phosphorus and organics in the secondary effluents by the electro-coagulation were also evaluated to propose an alternative advanced wastewatert treatment process. A series of experiments of the electro-coagulation were carried out with the following 4 different samples: synthetic solution containing nitrate only, synthetic solution containing nitrate as well as phosphorus, secondary effluents from activated sludge cultivated in laboratory, and secondary effluents from real wastewater treatment plants. Removals of nitrate and phosphorus in the synthetic solution were 30 and 97 % respectively, which verified the feasibility of the process. Removals of nitrate, phosphorus and COD in the secondary effluents from the cultivated sludge in laboratory were 49, 90 and 19 % respectively. Removal efficiency of the total nitrogen, nitrrate, phosphorus and COD in the secondary effluent from real wastewater treatment plant were 50, 61, 98 and 80 % respectively. The removal of the total nitrogen was less than the nitrate as expected, which is due to the formation of ammonia nitrogen in the cathode. But the proposed scheme could be an energy saving and alternative process for the advanced wastewater treatment if further studies for the process optimization are carried out.

Antimycotic Activity of Allium Sativum Against Beauveria Bassiana, Pathogenic Fungus of White Muscardine Disease in Silkworm, Bombyx mori L. (Lepidoptera: Bombycidae)

  • White Muscardine is the most common fungal disease of silkworm, Bombyx mori L. caused by the pathogenic fungus, Beauveria bassiana (Balsamo) Vuillemin. In the present investigation, an attempt has been made to screen locally available medicinal/ weed plants against Beauveria bassiana. Among the plant extracts (PE) tested, 5% aqueous crude extract of the bulb of Allium sativum (Garlic) has been found to be most effective against Beauveria bassiana. The radial growth of Beauveria bassiana in vitro was inhibited to the tune of 54.9% in aqueous extract and 54.4% in ethanolic extract of Allium sativum and correspondingly mycelial dry weight gave rise to 110.7 mg and 108.7 mg against 201.7 mg in control 15 days post treatment. Similarly, silkworm larvae topically inoculated with the Beauveria bassiana conidia ($1.8{\times}10^6/ml$) registered survival up to 53.0% against 0.0% in control after treatment with aqueous extract of Allium sativum. Simultaneously, as a preventive measure, silkworm larvae were put to rear in conidia contaminated seat paper instantly treated with aqueous extract of Allium sativum that also increased survival up to 61.0% against 4.6% in control. It is also observed that the plant extract is absolutely innocuous to silkworm.

Evaluation of the Nutrient Removal Performance of the Pilot-scale KNR (Kwon's Nutrient Removal) System with Dual Sludge for Small Sewage Treatment (소규모 하수처리를 위한 파일럿 규모 이중슬러지 KNR® (Kwon's nutrient removal) 시스템의 영얌염류 제거성능 평가)

  • A simple dual sludge process, called as $KNR^{(R)}$ (Kwon's Nutrient Removal) system, was developed for small sewage treatment. It is a hybrid system that consists of an UMBR (Upflow multi-layer bioreactor) as anaerobic and anoxic reactor with suspended denitrifier and a post aerobic biofilm reactor, filled with pellet-like media, with attached nitrifier. To evaluate the stability and performance of this system for small sewage treatment, the pilot-scale $KNR^{(R)}$ plant with a treatment capacity of $50m^3/d$ was practically applied to the actual sewage treatment plant, which was under retrofit construction during pilot plant operation, with a capacity of $50m^3/d$ in a small rural community. The HRTs of a UMBR and a post aerobic biofilm reactor were about 4.7 h and 7.2 h, respectively. The temperature in the reactor varied from $18.1^{\circ}C$ to $28.1^{\circ}C$. The pilot plant showed stable performance even though the pilot plant had been the severe fluctuation of influent flow rate and BOD/N ratio. During a whole period of this study, average concentrations of $COD_{cr}$, $COD_{Mn}$, $BOD_5$, TN, and TP in the final effluent obtained from this system were 11.0 mg/L, 8.8 mg/L, 4.2 mg/L, 3.5 mg/L, 9.8 mg/L, and 0.87/0.17 mg/L (with/without poly aluminium chloride(PAC)), which corresponded to a removal efficiency of 95.3%, 87.6%, 96.3%, 96.5%, 68.2%, and 55.4/90.3%, respectively. Excess sludge production rates were $0.026kg-DS/m^3$-sewage and 0.220 kg-DS/kg-BOD lower 1.9 to 3.8 times than those in activated sludge based system such as $A_2O$ and Bardenpho.
