Model Plants in Marine Biotechnology

  • The genus Porphyra, consisting of 133 species includes several economically valuable species (i.e. P. yezoensis, P. tenera, P. pseudolinearis etc.). They are predominantly consumed and cultivated in East Asian countries such as Japan, Korea and China, and they are regarded as a big commercial market today. In addition to the industrial importance, P. yezoensis is currently regarded as a feasible candidate for a model plant in marine biotechnology, therefore there are a wide range of studies being undertaken: strain-preservation, development of mutant strains and genetic analysis and exhaustive molecular analysis including EST and macro/micro array. Focusing on the activities of our research group, current situation and future perspectives in marine biotechnological studies using P. yezoensis will be discussed in this mini review.

  • 김 (Porphyra species)은 양식기술의 진보와 더불어 생산량이 급격히 증가되었으나, 지나치게 과도한 저 품질의 생산과 품종간 교잡으로 인해 점차 열성화 되는 현상이 나타나고 있다. 따라서 우리나라 고유의 맛과 향을 지니고 있으며, 지역특성에 적합한 한국 고유종의 개발과 품종개량이 절실히 요구된다. 김 속 식물은 세포층수, 세포당 엽록체수, 정자낭반의 형태, 정자낭 및 과포자낭 분열형식, 무성포자의 형성유무, 각포자체의 형태 및 생태적 특성 등을 종합하여 분류하였으며 (Kurogi 1972) 거치상돌기의 유무도 김의 주요 식별형질로 다루어져 왔다 (Miura 1988). Effects of Salinity on Chlorophyll Fluorescence from Porphyra Thalli and Comparison of Species with Different Intertidal Distribution

  • Characteristic changes in chlorophyll fluorescence from thalli of red alga, Porphyra, under high salt stress and during subsequent recovery were investigated, and the differences in the sensitivity to the stress among four species of Porphyra with different intertidal distributions were compared. By the treatment of NaCl with $9\%$ or higher concentrations, photochemical efficiency (Fv/Fm) decreased in a biphasic pattern: a rapid decrease was observed within 1­3 h and followed by a slow decline. The decrease of Fv/Fm was mainly due to the increase of Fo without significant increases of Fm. When the thalli treated with $15\%$ NaCl for 6 h were returned to natural sea water for recovery, the increase of Fv/Fm also showed a biphasic pattern: a rapid increase of Fv/Fm was observed within 2 h and followed by a slow increase. Differences in the sensitivity to salt stress among the four species could be found during recovery after the treatment of severe salt stress. After the treatment of $20\%$ NaCl for 6 h, Fv/Fm decreased below 0.3 in all of the four Porphyra species, and the species living in upper parts of the intertidal zone (P. suborbiculata and P. pseudolinearis) could recover better compared with the species in lower parts of the intertidal zone (P. seriata and P. yezoensis), during recovery for 24 h. The species collected from the coast of the South Sea seemed to be more tolerant than those in the East Sea.

Marine macroalgae of the Aleutian Islands: I. Bangiales

  • We sequenced the rbcL gene in more than 100 collections of foliose Bangiales made in the Aleutian Islands and western Alaska Peninsula during the past 25 years. This work allows us to recognize four previously undescribed species, two in the genus Boreophyllum and two in Pyropia. Boreophyllum aleuticum appears to be endemic to the Aleutian Islands, whereas B. ambiguum is known to occur from the Yakutat area to the tip of the Alaska Peninsula. The two previously undescribed species of Pyropia are more broadly distributed. Pyropia taeniata, which was previously identified under the name Py. pseudolinearis, occurs from northern Southeast Alaska through the Aleutian Islands. Pyropia unabbottiae, which is sister to Py. abbottiae, occurs from southern Vancouver Island to Attu Island. Collections throughout the Aleutian Islands allow us to document the distribution of another dozen species of foliose Bangiales in this region, including Boreophyllum aestivale, Fuscifolium tasa, Pyropia fallax, Py. fucicola, Py. gardneri, Py. kurogii, Py. nereocystis, Py. pseudolanceolata, Py. torta, Wildemania amplissima, W. norrisii, and W. variegata. We were unable to confirm the occurrence of the following species previously recorded from the Aleutian Islands: Porphya ochotensis, Pyropia abbottiae, Py. perforata, Py. pseudolinearis, P. purpurea, P. umbilicalis, Py. yezoensis and Wildemania schizophylla. At least two undescribed filamentous Bangiales also occur in the Aleutian Islands.
