An oligonucleotide microarray system was developed for the simultaneous detection of porcine epidemic diarrhea virus, transmissible gastroenteritis virus, porcine enteric calicivirus, porcine group A and C rotavirus. RNAs of the reference viruses and porcine diarrhea samples were extracted and amplified using one-step multiplex RT-PCR in the presence of cyanine 5-dCTP and hybridized on the microarray chip that spotted the virus-specific oligonucleotides. This system were approximately 10-to 100-fold higher in sensitivity than conventional RT-PCR, and the assay time was less than 3 hours. The relative sensitivity and specificity were 92% and 72.2%, respectively, based on 102 porcine diarrhea samples using RT-PCR as gold standard. These results suggested that the oligonucleotide microarray system in this study be probably more reliable and reproducible means for detecting porcine enteric viruses and that it could be of substantial use in routine diagnostic laboratories.

Intestinal infections are common in growing pigs and can be caused by multiple pathogens, environmental and management factors [1]. Among the most important viruses in swine enteritis are porcine epidemic diarrhea virus (PEDV), transmissible gastroenteritis virus (TGEV), porcine enteric calicivirus (PECV), porcine group A rotavirus (PRV gp A) and bacteria are Escherichia coli and Salmonella spp. and protozoa is Isospora suis [1]. The DNA chip system can serve as a powerful tool that can be utilized for simultaneous detection of specific pathogenic bacteria strains and viruses [2,3]. The combination of PCR and DNA chip technology will provide a novel method for the detection of porcine enteric pathogens thus revolutionize the diagnosis and management of the disease. The aim of this study is to develop DNA chip system for the rapid and reliable detection of five major porcine enteric pathogens based on oligonucleotide DNA chip hybridization. (omitted)
The present study was aimed to investigate the prevalence of porcine group A rotavirus, group C rotavirus and calicivirus from the 46 pig farms located in Incheon area. Group A rotavirus was detected in 16 (5.3%) from 8 farms (17.4%), and group C rotavirus was determined in 17 samples (5.7%) from 6 farms (13.0%). Porcine calicivirus was also detected in fecal samples [11 samples (3.7%) from 2 farms (4.3%)]. Correlation analysis was carried out among porcine enteric viruses and clinical signs, herd size and temperature on the basis of these results. The occurrence of porcine group A rotavirus, group C rotavirus and calicivirus infections in Incheon area was not associated with season and temperature. Especially, group C rotavirus was also detected in the pigs without any clinical symptoms.
From January to June 2006, 54 suckling pigs had been submitted in virology lab., College of Veterinary Medicine, Seoul National University. All pigs had suffered from various symptoms such as respiratory sign, enteric signs, neurologic signs, etc. Among 54 pigs, 24 pigs (44.4%) were positive for porcine hemagglutinating encephalomyelitis virus (HEV) through reverse transcription-nested polymerase chain reaction. According to this result, HEV infections seemed to be prevalent and widespread in Korean swine farms, and the infection is associated with respiratory signs and neurologic signs more than enteric signs. The HEV positive pigs showing respiratory signs were co-infected with viruses such as PRRSV, and PCV2, or bacteria such as Pasteurella spp. The single infection may subclinically have an influence on outbreak of other respiratory pathogens in suckling pigs.
Porcine circovirus type 2 (PCV2) and porcine reproductive and respiratory syndrome virus (PRRSV) are known as significant immunosuppressive viruses in pigs. In this study, we investigated the correlation of PCV2 and PRRSV in enteric lesions of porcine salmonellosis. A total of 64 cases were classified into four pig groups as group A (24 cases, S. Typhimurium), group B (11 cases, S. Typhimurium+PCV2), group C (16 cases, S. Typhimurium+PRRSV) and group D (13 cases, S. Typhimurium+PCV2+PRRSV). Comparing with group A, ulcerative enteritis in large intestine was little more prevalent in the PCV2 infected pigs in group B and D. And lymphoid depletion in gut-associated lymphatic tissue (GALT) of large intestine was also detected in PCV2 positive group B (36.4%) and D (30.8%). According to the results of immunohistochemistry (IHC), PCV2 antigens (83.3%) were more prevalently distributed in the intestinal lesions of porcine salmonellosis than PRRSV antigens (10.3%). PCV2 were also detected in the lymphoid depleted GALT of the large intestine from 7 of the 8 pigs (87.5%), but PRRSV were not found in all cases. It may explain that PCV2 can play a certain immunological role to enhance secondary bacterial infection in porcine alimentary tracts.
돼지 장염 바이러스를 동시에 감별 진단할 수 있는 새로운 진단법인 oligonicleotide microarray 기법을 돼지 설사 분변을 대상으로 바이러스 감염을 진단하고 기존의 진단법으로 널리 사용되고 있는 RT-PCR과 진단기법의 민감도와 특이도를 비교하고자 하였다. 32개 양돈장, 102 예의 포유 및 이유자돈의 설사분변에서 microarray를 이용한 검사결과 Transmissible gastroenteritis virus (TGEV) 9.8%, porcine epidemic diarrhea virus (PEDV) 28%, (porcine enteric calicivirus (PECV) 18.6%, porcine rotavirus (PRV) group A 6.9%, PRY group C 1% 의 검출률을 확인하였다. 또한 RT-PCR 기법과의 비교에서도 100%의 민감도와 72.2%의 특이도를 보였으며 agreement는 85.3%, kappa value 0.71로 우수한 진단기법임을 확인하였다. 이를 통해 microarray 진단법은 RT-PCR 후의 전기영동 과정과 민감도를 높이기 위해 수행되는 nested PCR 수행의 번거러움을 없애면서 정확한 감별진단을 수행할 수 있는 진단 기법임을 확인하였다.
돼지의 주요 장염 유발 바이러스인 돼지 전염성 위장염 바이러스 (transmissible gastroenteritis virus; TGEV), 돼지 유행성 설사증 바이러스 (porcine epidemic diarrhea virus; PEDV), 돼지 칼리시 바이러스 (porcine enteric calicivirus; PECV), 돼지 로타바이러스 A 형과 C 형 (porcine rotavirus; PRY, group A and C)을 동시에 감별 진단 할 수 있는 신속하고 정확한 oligonucleotide microarray 진단법을 개발하였다. 이 진단법은 유리슬라이드에 각각의 바이러스에 특이적인 부위에서 제작된 oligonucleotide probe를 찍은 DNA chip을 제작하여 여기에 각각의 바이러스를 역전사하고 cy5-dCTP를 포함한 multiplex PCR을 수행한 다음 hybridization 하였다. 이후 hybridization 결과는 fluorescence scanner를 이용하여 확인하였다. 이 새로운 microarray system은 RT-PCR과 같은 기존의 진단방법보다 소량의 바이러스를 민감하게 검사할 수 있을 뿐 아니라 hybridization을 통해 검사결과의 정확성을 확인할 수 있었다. 따라서 본 연구에서 개발한 microarray system은 돼지의 설사 유발 바이러스를 진단하는데 매우 유용한 진단 방법임을 확인하였다.
Porcine epidemic diarrhea (PED) is a highly contagious enteric viral disease of pigs with watery diarrhea in piglets, which ultimately results in huge economic losses in the swine industry. The spike (S) protein plays an important role in viral pathogenicity, tissue tropism, infection, dissemination and the trypsin-dependent proliferation of the PED virus (PEDV). In the present study, we determined the full-length spike (S) gene sequences of twenty PEDV field strains detected in Jeonbuk province in 2022. Phylogenetic analysis showed that the twenty PEDV field strains were classified into G2b group and shared 98.6~100% of nucleotide homology and 97.4~100% of amino acid homology with each other. Mutations of amino acid sequences on the neutralizing epitope of S protein were observed in the twenty field strains compared to the previous vaccine strain SM-98-1 (G1a group). Therefore, these amino acid mutations in the PEDV S protein may result in a new genotype of the virus and highly pathogenic virus, so continuous monitoring is required.
Interferon (IFN)-λ plays an essential role in mucosal cells which exhibit strong antiviral activity. Lactobacillus plantarum (L. plantarum) has substantial application potential in the food and medical industries because of its probiotic properties. Alphacoronaviruses, especially porcine epidemic diarrhea virus (PEDV) and transmissible gastroenteritis virus (TGEV), cause high morbidity and mortality in piglets resulting in economic loss. Co-infection by these two viruses is becoming increasingly frequent. Therefore, it is particularly important to develop a new drug to prevent diarrhea infected with mixed viruses in piglets. In this study, we first constructed an anchored expression vector with CWA (C-terminal cell wall anchor) on L. plantarum. Second, we constructed two recombinant L. plantarum strains that anchored IFN-λ3 via pgsA (N-terminal transmembrane anchor) and CWA. Third, we demonstrated that both recombinant strains possess strong antiviral effects against coronavirus infection in the intestinal porcine epithelial cell line J2 (IPEC-J2). However, recombinant L. plantarum with the CWA anchor exhibited a more powerful antiviral effect than recombinant L. plantarum with pgsA. Consistent with this finding, Lb.plantarum-pSIP-409-IFN-λ3-CWA enhanced the expression levels of IFN-stimulated genes (ISGs) (ISG15, OASL, and Mx1) in IPEC-J2 cells more than did recombinant Lb.plantarum-pSIP-409-pgsA'-IFN-λ3. Our study verifies that recombinant L. plantarum inhibits PEDV and TGEV infection in IPEC-J2 cells, which may offer great potential for use as a novel oral antiviral agent in therapeutic applications for combating porcine epidemic diarrhea and transmissible gastroenteritis. This study is the first to show that recombinant L. plantarum suppresses PEDV and TGEV infection of IPEC-J2 cells.
돼지 설사유발 칼리시 바이러스 (Porcine enteric calicivirus: PECV)는 자돈에서 설사를 일으키지만, 사람에서도 위장염을 일으키는 원인체인 Sapporo-like calicivirus와 형태학적으로나 유전학적으로 유사하다고 이미 알려졌다. 본 연구는 국내에서 발생하고 있는 PECV의 RNA dependent RNA polymerase (RDRP) 부위와 capsid 부위 염기서열과 아미노산 서열을 기존에 보고되었던 것과 비교하여 분류하였다. 연구 결과 국내 분리주는 기존의 PECV RDRP 부위 (염기서열: 90%, 아미노산: 97%) 와 유사성이 아주 높았으며 capsid 부위(염기서열: 83%, 아미노산: 81%)는 다소 낮았다. 또한 이 바이러스는 모든 칼리시 바이러스의 RDRP 부위에 특이적으로 존재하는 GLPSG와 YGDD 아미노산 배열이 존재하였으며 capsid 부위에서는 국내에서 발생한 PECV 에서만 "TAA" 염기서열이 삽입되어 있었다. 본 연구를 통하여 국내에서 발생한 PECV는 porcine sapporo-like calicivirus와 유사하며 그 외 caliciviridae과인 Norwalk-like virus, Vesivirus, Lagovirus와는 상이하다는 것이 규명되었다.
