Development and Characterization of a Novel Anti-idiotypic Monoclonal Antibody to Growth Hormone, Which Can Mimic Physiological Functions of Growth Hormone in Primary Porcine Hepatocytes

  • B-32 is one of a panel of monoclonal anti-idiotypic antibodies to growth hormone (GH) that we developed. To characterize and identify its potential role as a novel growth hormone receptor (GHR) agonist, we determined that B-32 behaved as a typical $Ab2{\beta}$ based on a series of enzyme-linked immunosorbent assay assays. The results of fluorescence-activated cell sorting, indirect immunofluorescence and competitive receptor binding assays demonstrated that B-32 specifically binds to the GHR expressed on target cells. Next, we examined the resulting signal transduction pathways triggered by this antibody in primary porcine hepatocytes. We found that B-32 can activate the GHR and Janus kinase (2)/signal transducers and activators of transcription (JAK2/STAT5) signalling pathways. The phosphorylation kinetics of JAK2/STAT5 induced by either GH or B-32 were analysed in dose-response and time course experiments. In addition, B32 could also stimulate porcine hepatocytes to secrete insulin-like growth factors-1. Our work indicates that a monoclonal anti-idiotypic antibody to GH (B-32) can serve as a GHR agonist or GH mimic and has application potential in domestic animal (pig) production.

Preparation and Characterization of an Antibody Antagonist That Targets the Porcine Growth Hormone Receptor

  • A series of antagonists specifically targeting growth hormone receptors (GHR) in different species, such as humans, rats, bovines, and mice, have been designed; however, there are currently no antagonists that target the porcine growth hormone (GH). Therefore, in this study, we developed and characterized a porcine GHR (pGHR) antibody antagonist (denoted by AN98) via the hybridoma technique. The results from enzyme-linked immunosorbent assay, fluorescence activated cell sorter, indirect immunoinfluscent assay, and competitive receptor binding analysis showed that AN98 could specifically recognize pGHR, and further experiments indicated that AN98 could effectively inhibit pGH-induced signalling in CHO-pGHR cells and porcine hepatocytes. In addition, AN98 also inhibited GH-induced insulin-like growth factor-1 (IGF-1) secretion in porcine hepatocytes. In summary, these findings indicated that AN98, as a pGHR-specific antagonist, has potential applications in pGH-pGHR-related research on domestic pigs.

Determination of Optimum Aggregates of Porcine Hepatocytes As a Cell Source of a Bioartificial Liver

  • Large quantities of porcine hepatocyte aggregates with various degrees of aggregation (DA) could be obtained by controlling the suspension periods (0,9,24, and 48 h), and by entrapping the hepatocyte aggregates in model materials of encapsulation such as Ca-alginate and type-I collagen gels. The effects of DA on liver-specific functions of hepatocytes were evaluated in order to obtain optimum DA for the cell source of bioartificial liver (BAL) systems. Irregular rugged aggregates (size $75 \pm 28$ $\mu\textrm{m}$) farmed by 24 h of suspension culturing showed peak viability and hepatic functions such as ammonia removal and albumin secretion in the two types of entrapment systems, thus offering themselves as a stable cell source of a BAL system for hepatic functions and scale-up.

In vitro functional assenssment of bioartificial liver system using immobilized porcine hepatocyte spheroids

  • To treat fulminant hepatic failure (FHF) patients, various extracorporeal bioartificial liver (BAL) systems have been developed. Several requirements should be met for the development of BAL systems: hepatocytes should be cultured in a sufficiently high density; their metabolic functions should be of a sufficiently high level and duration; and the BAL systems module should permit scaling-up and aseptic handling. Several investigators have found that freshly isolated primary hepatocytes can be cultured into three dimensional, tightly packed, freely suspended, multicellular aggregates, or spheroids. These specialized cell structures exhibited enhanced liver specific functions and a prolonged differentiated state compared to cells maintained in a monolayer culture. Cells in spheroids appear to mimic the morphology and ultrastructure of the in vivo liver lobule. The ability of hepatocytes to organize into three-dimensional structures was hypothesized to contribute to their enhanced liver-specific activities. In this study, the ammonia removal rate and urea secretion rate of pig hepatocytes spheroids encapsulated in Ca-alginate bead were determined. A packed-bed bioreactor with encapsulated pig hepatocytes was devised as BAL support system. The efficacy of the system was evaluated in vitro.

Enterovirus 감염 돼지의 간에서 virus의 형태학적 관찰 (Ultrastructure of virus particles in the liver of piglets infected with porcine enterovirus serotype 3)

  • Enterovirus감염 돼지의 감염초기 virus의 동태를 규명하고자 초유를 섭취하지 않는 1~2일령의 돼지에 돼지 enterovirus 3형을 경구감염시킨후 간조직을 전자현미경으로 관찰하였던 바 다음과 같은 결과를 얻었다. 감염후 3일째 부검한 돼지의 간에서 간세포의 심한 공포변성이 인정되었고 간 동양혈관 내피세포 및 Kupffer세포에서 picornavirus의 특징적인 virus결정이 출현하였으며 확장된 ER은 virus 결정과 밀접하게 관련되어 있었다. 또한 동양혈관 세포와 간세포에서는 미숙한 virus모양의 입자들로 구성된 세포질내 봉입체가 관찰되기도 하였다. 이상의 소견으로 보아 돼지 enterovirus 3형은 신경친화성 뿐만 아니라 감염초기 간에 친화성이 큰 것으로 생각된다.

Effect of Low Temperature Preservation and Cell Density on Metabolic Function in a Bioartificial Live

  • Difficulties associated with bioartificial liver (BAL) preservation limit not only the commercialization of BAL, but also its clinical trials. In this study, the possibility of cold preservation of BAL cartridges containing porcine hepatocytes was examined at 4$^{\circ}C$. In an in vitro perfusion culture System, BAL cartridges maintained cytochrome P450 metabolic function for at least 50 days. However, all BAL cartridges completely lost their ammonia eliminating ability when stored at 4$^{\circ}C$. We a1so studied the effect of cell density on the maintenance of BAL liver function In a highly differentiated and healthy state. As expected, BALs containing a larger number of hepatocytes demonstrated higher metabolic functions. When metabolic functions were compared per gram of hepatotytes, no large differences were observed between devices containing different densities of hepatocytes. Decreased cell density did not Successfully prolong BAL function. The viability and function of isolated hepatotytes highly depend on the culture conditions, such as cell density, substrata, culture media, and additives to the culture media. Perfusion culture of BAL cartridges at 4$^{\circ}C$ gave a promosing result with respect to the maintenance of P450 activity. However, as indicated by the rapid loss of ammonia metabolic activity, many factors still remain to be optimized for preservation of BAL keeping high metabolic functions for a longer time.

  • PAU is the block copolymer consists of a small amount of a small amount of poly(${\gamma}-methyl-L-glutamate$) (PMLG) and the polyurethane. The urethane segments are hydrophobic and then strongly interact with the other hydrophobic materials such as PTFE, and the PMLG segments with the ${\alpha}-helix$ structure possess the cytocompatibility. Therefore, PAU can be easily coated onto the PTFE fiber and acts as an artificial extracellular matrix with the high cytocompatibility Results shows, the immobilization, cultured and functions of porcine hepatocytes is greatly improved.

제주지역 양돈장 자돈에서 발생한 이유후 전신성 소모성 증후군의 증례 (Occurrence of postweaning multisystemic wasting syndrome in pigs in Jeju)

  • Multiplex PCR and immunohistochemistry were used to detect and differentiate between porcine circovirus (PCV) type-I and the PCV associated with postweaning multisystemic wasting syndrome (PMWS). Unique DNA product to PCV type-II was confirmed the some organs including lymph nodes, tonsil and spleen from eight pigs in Jeju by multiplex PCR. In this study, the samples were tested by a multiplex PCR assay to determine the type of PCV in each case; all cases were PCV type-II positive. PCV type-II was identified not only in typical PMWS cases, but also in field cases submitted with various clinical histories, some of which were not suggestive of PMWS. Immunohistochemically PCV type-II antigen was detected in macrophage-like cells in the tonsil, liver, lymph nodes and spleen, while some hepatocytes and renal tubular epithelial cells were also positive to the virus. This study suggested that PCV type-II is one of the causative agents of PMWS as well as the major type of PCV in the affected pigs in Jeju.
