Herein we analyzed the nuclear DNA content and the histological characteristics of the triploid of the 'Hyunsasi' (Populus alba ${\times}$ P. glandulosa $F_1$) which were developed for biomass production and molecular breeding research. The flow cytometric analysis showed that the nuclear DNA content of the 3 triploids were 1.6 times greater than those of the diploid. In terms of histological characteristics, the cross-section area of the stem of 'Line-18' was 1.6 times larger than that of the diploid. The area of pith, and cortex and phloem of the stem of 'Line-18' was also 1.6 and 2.0 times larger than that of the diploid, respectively. Moreover, the length and area of guard cell of 'Line-18' was 1.2 times larger than that of the diploid. These results helps to understand the cytological characteristics of the triploid poplar clones, and further investigations in the growth rate and wood properties of the triploids growing in the field will determine whether the triploid poplars are good candidates for molecular breeding programs and for the improvement of industrial biomass productivity.
Populus species have been as a model species in tree breeding and we have enormous varieties resulting from the poplar breeding because of their fast growth performance and short rotation age. New varieties developed in Korea are common italian poplar(P euramericana, I-214, I-476), P euramericana“Eco 28”(Italian poplar No.1) and p. deltoides“Lux”(Italian poplar No.2), which were introduced from foreign countries. As hybrid polars, Hyun-Sasi(p. alba ${\times}$ P. glandulosa No.1, No.2, No.3, No4.), P. nigra x P. maximowiczii and P. koreana x P. nigra val. italica, were developed, and P. davidiana was selected as the result of selection breeding The total plantation areas covered with the new varieties are 935,162ha that include 745,773ha of P. euramericana, 184,636ha of P. alba x P. glandulosa, and other new varieties are 4,735ha. The new poplars are contributed to increase farmer's income as well as bare land tree-planting in Korea. The technologies associated with the poplar species were developed, such as the determination of optimum site for new the poplar species, the crossing method between incompatible poplar species, and the vegetative mass propagation. In the future, poplar species will be considered for phytoremediation species at contaminated areas such as landfill sites or with lives stock's waste water as well as wood production, a shade tree like road-side tree and public park tree.
Two experiments were conducted to evaluate the nutritional value of common fodder tree leaves with goats fed at 50% of total ration. In experiment 1, leaves from six fodder tree species i.e., Ailanthus aitissima, Elaeagnus angustifolic, Morus alba (Mulberry), Populus spp, Robina pseudoacacia and Salix babylonia were harvested in spring and winter from northern areas of Pakistan. Chemical composition and apparent in situ dry matter digestibility (DMD) of fodder tree leaves were measured. Results showed that crude protein (CP) values were higher (p<0.05) in all the species during spring compared to winter (17.9% vs 12.0%). The concentration of NDF in Elaeagnus and Robinia was higher in spring, whereas no seasonal difference was found in other species. In situ DMD was higher (p<0.05) in Ailanthus and Populus at spring while it was higher (p<0.05) in Elaeagnus, Mulberry and Robinia at winter. There was no (p<0.05) seasonal effect on in situ DMD of Salix. In experiment II, four iso-nitrogenous and iso-caloric rations viz., A, B, C and D were prepared containing 50% (winter harvested) sun dried leaves of Salix, Robinia, Mulberry and Elaeagnus, respectively and 50% concentrate. Dry matter and crude protein intakes were higher (p<0.05) given ration A (Salix) whereas DM and CP digestibility was lowest (p<0.05) given ration B. Nitrogen retention was higher (p<0.05) given ration A. Goats fed on fodder tree leaves and concentrate showed moderate intake and digestibility.
We estimated the biomass productivity and the storage potential of nitrogen, the major contributor of non-point source pollution, with four three-year-old four poplar clones in a riparian woody buffer established in the Anseong River in Anseong, Korea. Stem of Populus alba ${\times}$ P. glandulosa clone 72-31 and Populus deltoides ${\times}$ P. nigra clone Dorskamp showed the highest percentage of aboveground biomass components, followed by branch and leaf. Nitrogen content in aboveground biomass components of two poplar clones was the highest in leaf and the lowest in stem. Nitrogen content in leaf and branch of clone 72-31 was higher than that of clone Dorskamp, while it in stem was lower. Populus deltoides clone Ay48 showed the highest above-ground biomass productivity, which was estimated as $37.5ton\;ha^{-1}$ at age 3. However, clone 72-31 was the lowest in above-ground biomass productivity. Nitrogen storage potential in aboveground biomass of 3-year-old poplar clones was high in order of aboveground biomass. Clone Ay48 showed the highest nitrogen storage potential in aboveground biomass, which was estimated as $218.3kg\;ha^{-1}$ at age 3.
Dehydrins are group II, late embryogenesis abundant proteins that act putatively as chaperones in stressed plants. To elucidate the function of dehydrins in poplar, we isolated the $SK_2$-type dehydrin gene Podhn from Populus alba $\times$ P. tremula var. glandulosa suspension cells and analyzed its expression following treatments of abiotic stress, wounding and plant growth regulator. Sequence homology and phylogenetic analyses indicate Podhn encodes an acidic dehydrin (pI 5.14, 277 amino acids, predicted size 25.6 kDa) containing two lysine-rich "K-segments" and a 7-serine residue "S-segment", both characteristic of $SK_2$-type dehydrins. Southern blots show Podhn genes form a small gene family in poplar. Podhn was expressed in all tissues examined under unstressed conditions, but most strongly in cell suspensions (especially in the stationary phase). Drought, salt, cold and exogenous abscisic acid (ABA) treatments enhanced Podhn expression, while wounding and jasmonic acid caused its reduction. Therefore, Podhn might be involved in ABA or stress response.
아임계 및 초임계수에 의한 목질바이오매스의 당화특성을 분석하기 위하여 분해공정 동안 압력을 23MPa(물의 임계압력)로 고정하고 물의 아임계 온도$(325^{\circ}C,\;350^{\circ}C)$와 초임계 온도$(380^{\circ}C,\;400^{\circ}C,\;425^{\circ}C)$에서 현사시나무 목분을 각각 60초 동안 처리하였다. 생성된 현사시나무의 분해산물에는 액상과 고형분의 분해산물이 섞여 있었다. 각 처리조건에 따른 목질바이오매스의 분해율은 온도가 상승함에 따라 증가하였으며 초임계 온도인 $425^{\circ}C$에서 최고 83.1%의 분해율을 나타냈다. 아임계 및 초임계수에 의해서 생성된 단당류는 고성능 음이온 교환 크로바토그래프(HPAEC)를 이용하여 분석하였다. 목질바이오매스의 초임계수 분해과정에서 처리 온도가 높아지면서 단당류 수율은 증가하는 경향을 보였으며, $425^{\circ}C$에서 가장 높은 7.3%의 단당류 수율을 나타내었다. 아임계 온도 범위에서는 현사시나무의 섬유소 성분 중에서 자일란이 우선적으로 분해되어 자일로스의 생성비율이 비교적 높았으며, 처리온도가 높아지면서 셀롤로오스의 분해에 의한 글루코오스 생성율이 급격히 상승하였다. 이렇게 생성된 단당류 성분들은 고온의 반응조건하에서 열분해 반응에 의해서 더욱 분해되어 퓨란계 화합물로 변형되었다.
This study was carried out to investigate the effects of Zn and Pb concentration on seed germination and plant growth in water and soil culture, and the frequency of chlorosis invegetation and the relationship between plants and soil in the Sambo mine. The inhibition of germination were observed in 1000ppm of Zn, 10ppm of Pb and 5000ppm of Zn + Pb, but germination was more stimulated in 10ppm of Zn than control. The symptoms of chlorosis and abnormality were occurred in plant leaves grown to the soils treated with more than 1000ppm of Pb. Reasons of chlorosis were considered as an antagonistic effect of other metals towards uptake of iron by the plant in Zn treatment. The contents of Zn and Pb in fruits were lower than those of leaves, and that was remarked in case of Pb. With increasing rate of Zn and Pb treatment, chemical components of soils in pot culture were accompanied by slight decrease in pH, total nitorgen and exchangeable K. Chlorotic individuals of 10 species were shown in the areas of the Sambo mine. Chlorotic symptoms were especially extensive and severe in Sophora angustifolia, Populus alba, Spiraea prunifolia, Amorpha fruticota, Lespedeza bicolor and Salix dependens. Plants in the investigated areas grew in soils containing Zn of 311ppm and Pb of 151ppm on an average, and accumulated Zn of 2084ppm and Pb of 49ppm.

A cell culture system of poplar (Populus alba x P.glandulosa) was established to test four different methods for evaluation of cellular stresses. Two different kinds of stresses were given to the cultures by adding either Pb(NO$_3$)$_2$ or paraquat and the cellular responses were monitored during a week period. While fresh weight reduction was observable in two days after the treatment of Pb(NO$_3$)$_2$, such changes were apparent only in later stage in paraquat treated cultures. Cells in paraquat treated cultures in the first 3 days showed no alteration in fresh weight as compared to untreated cultures, but had their MTT reducing activities completely inhibited. Neither Evans blue staining nor ion conductivity of the medium was consistent with fresh weight changes of the cultures. Overall, cell clumps formed during suspension culture appeared to interfere with staining and washing reactions and thus cause the assays unreliable. Among the four methods examined, fresh weight changes and MTT reducing activity appeared to be the most reliable and consistent.
A study was conducted to determine the kiln drying schedule for hydrid aspen, This kiln drying schedule was found by oven drying method and developed by pilot testing of 2.0cm, 2.5cm, 3.0cm-thick green lumber. The results of this study were as follows: 1. Kiln drying schedule developed by oven-drying method was $T_{12}-E_5$ for 2.5cm-thick green lumber. 2. Drying times for 2.0cm-thick green lumber(127.0 percent moisture content) to 7.3 percent moisture content, for 2.5cm-thick green lumber(95.0 percent moisture content) to 9.7 percent moisture content, and for 3.0cm-thick green lumber(118.5 percent moisture content) to 10.0 percent moisture content were 45 hours, 45 hours, and 54 hours, respectively. 3. Drying rate from 90 to 10 percent moisture content of 2.5cm-thick lumber was about 0.7 times faster than that of 2.0cm-thick lumber and about 1.1 times faster than that of 3.0em-thick lumber. 4. End checks for 2.0cm-, 2.5cm-, and 3.0cm-thick green lumber occurred 95, 74 and 100 percent moisture content, respectively and reached maximum amount in 53, 41, and 60 percent moisture content, respectively. 5. No surface check and no honeycomb occurred. 6. Cupping and collapse slightly occurred and the quality of dry lumber was first.

In order to elucidate the relationship among the several forest types in the green-sbelt nearby Seoul, the vascular plant species data collected preferentially from 52 stands were subjected to DCA (detrended correspondence analysis). Eight types of forests were arranged on the I/II plane fo DCA stand ordination, Quercus mongolica, Alnus hirsuta, Populus alba $\times$ glandulosa, Robinia pseudo-acacia, Pinus densiflora, Pinus koraiensis, Larix leptolepis and pinus rigida forest. Correlation analysis between the stand scores of DCA and environmental factors revealed that the axis I of DCA stand ordination implied the gradient of altitude, organic matter, total nitrogen and depth of A horizon. The axis II implied the gradient of total nitrogen and soluble phosphorus. The results of PCA (principal components analysis) by environmental data were in good agreement with that of DCA by floristic data. The recruitments of Quercus spp. Occurred in all kinds of forest types in the green-belt and their recruitments were closely related with altitutde: Quercus dentata in the foot. Quercus serrata in the midslope and Quercus mongolica in the uperslope of the mountain.
