Growth Characteristics of Polyporales Mushrooms for the Mycelial Mat Formation

  • Mushroom strains of Polyporales from the genera Coriolus, Trametes, Pycnoporus, Ganoderma, and Formitella were explored in terms of mycelial growth characteristics for the application of mushroom mycelia as alternative sources of materials replacing fossil fuel-based materials. Among the 64 strains of Polyporales, G. lucidum LBS5496GL was selected as the best candidate because it showed fast mycelial growth with high mycelial strength in both the sawdust-based solid medium and the potato dextrose liquid plate medium. Some of the Polyporales in this study have shown good mycelial growth, however, they mostly formed mycelial mat of weak physical strength. The higher physical strength of mycelial mat by G. lucidum LBS5496GL was attributed to its thick hyphae with the diameter of 13 mm as revealed by scanning electron microscopic analysis whereas the hyphae of others exhibited less than 2 mm. Glycerol and skim milk supported the best mycelial growth of LBS5496GL as a carbon and a nitrogen source, respectively.

Two Unrecorded Wood Decay Fungi (Polyporales) in Dadohaehaesang National Park, Korea

  • Dadohaehaesang National Park is a well-known habitat for a variety of indigenous animals and plants; however, there are no studies on the diversity of macrofungi. To undertake a basic survey of the macrofungi diversity in an unexploited area, we focused on surveying the macrofungi in Dadohaehaesang National Park, Korea, during 2017 and discovered two unrecorded species belonging to the order Polyporales. The two species were identified as Cinereomyces lindbladii and Steccherinum bourdotii based on a combination of morphological characteristics and sequence analysis of the internal transcribed spacer region. We provide macro- and micro-morphological descriptions for these two unrecorded species in Korea.

Identification of three wood decay fungi in Yeoninsan Provincial Park, Korea

  • Though several wood decay fungi have been reported in the world-wide, only about 600 wood decay fungi have been reported in Korea to date. Thus, the objective of this study was to secure resources for the wood decay fungi in Korea. We investigated wood decay fungi in Yeoninsan Provincial Park, Korea, and the collected specimens were identified based on ITS sequence analysis. Two species were unrecorded species in Korea: Postia hirsuta (Polyporales, Basidiomycota) and Hyphodontia reticulata (Hymenochaetales, Basidiomycota). Another species was previously reported without detailed description: Ceriporia alachuana (Polyporales, Basidiomycota). Here, we provided additional detailed microscopic features and phylogenetic analysis of these species.

  • The wood decay fungi are a diverse taxonomic group that plays a pivotal role in forest carbon cycling. Wood decay fungi use various enzymatic pathways to digest dead or living wood in order to obtain carbon and other nutrients and these enzymatic systems have been exploited for both industrial and medical applications. Over 600 wood decay fungi species have been described in Korea; however, the recent application of molecular markers has dramatically altered the taxonomy of many of these wood decay fungi at both the genus and species levels. By combining molecular methods, specifically sequences of the internal transcribed spacer region, with traditional morphological characters, this study identified five new species records for Korea in five genera: Aurantiporus, Favolus, Neofavolus, Loweomyces, and Hymenochaetopsis. Three of these genera (Aurantiporus, Favolus, and Loweomyces) were previously unknown in Korea. The relatively simple morphology of the wood decay fungi often leads to ambiguous taxonomic assignment. Therefore, molecular markers are a necessary component of any taxonomic or evolutionary study of wood decay fungi. Our study highlights the need for a more robust and multifaceted approach in investigating new wood decay fungi in Korea.

국내 기수역 환경의 균류 다양성 (Diversity of Fungi in Brackish Water in Korea)

  • 본 연구에서는 국내 기수 환경의 물과 토양에서 균류의 다양성을 확인하기 위해 을숙도, 금강하구둑, 순천만습지, 대호방조제, 신두리사구 및 부남방조제 총 6지점의 현장조사 및 시료를 채집하였다. 현장에서 여과한 담수시료와 담수퇴적물로부터 곰팡이를 순수분리하여 PDA배지에 접종한 후 25℃에서 배양하였다. 기수역 시료에서 순수분리된 균주는 총 173균주로 ITS 부분을 분석하여 속 수준으로 동정하였다. 분리 동정된 균주를 다양성 지수를 이용하여 분석하였다. 분리된 균류는 Pleosporales, Eurotiales, Capnodiales, Hypocreales, Polyporales, Saccharomycetales, Agaricales, Glomerellales, Mucorales, Dothideales, Russulales, Xylariales, Sordariales, Myrmecridiales, Tubeufiales, Onygenales, Cantharellales, Amphisphaeriales 목에 속하며 Cladosporium속이 20%로 가장 우점하며, 그 다음은 Penicillium (19%), Fusarium (5%)의 순서로 많이 분포하였다. Sarocladium kiliense NNIBRFG3280와 Fusicolla merismoides NNIBRFG23708를 국내 기수환경에서 발굴하였으며, 이를 국내 미기록종으로 보고하고자 한다.

Molecular Identification of Endophytic Fungi Isolated from Needle Leaves of Conifers in Bohyeon Mountain, Korea

  • Fungal endophytes are microfungi that live in plants without causing apparent symptoms of infection. This study was conducted to identify endophytic fungi isolated from leaves of coniferous trees in Bohyeon Mountain of Korea. We collected leaves of two species of coniferous trees, Pinus densiflora and Pinus koraiensis, from 11 sites in the study area. A total 58 isolates were obtained and identified using molecular and morphological characteristics. Four species of endophytic fungi were isolated from P. densiflora: Lophodermium conigenum, Leotiomycetes sp., Septoria pini-thunbergii, and Polyporales sp., while two fungal species were isolated from P. koraiensis: Eurotiomycetes sp. and Rhytismataceae sp. The most frequently isolated species were L. conigenum and S. pini-thunbergii.

Molecular Taxonomy of Ganoderma cupreum from Southern India Inferred from ITS rDNA Sequences Analysis

  • Ganoderma is a cosmopolitan wood-rot basidiomycete that has been extensively studied for its pathogencity and medicinal properties. Identification of Ganoderma based on macro-microscopic features led to large number of synonyms which resulted in 250 taxonomic names. A Ganoderma species collected from Courtallam, Tamil Nadu was identified as G. cupreum. Phylogenetic analysis inferred from internal transcribed spacer rDNA region resolved the Indian isolate MYC1 as Ganoderma cupreum which clustered with Australian and Asian "cupreum" clade with 85% bootstrap support BS and shared 99% and 98% nucleotide similarity with Malaysian and Australian 'cupreum' respectively. This study represents the first molecular evidence of G. cupreum from Asian origin.

변산반도 국립공원 고등균류의 자원이용적 특성 (Resources Use Characteristics of Higher Fungi in Byeonsanbando National Park)

  • 2009년부터 2011년, 2015년까지 변산반도 국립공원의 고등균류를 조사한 결과, 총 2문 6강 18목 61과 157속 323종이 조사 되었으며, 주름버섯목(Agaricales)이 23과 67속 153종, 그물버섯목(Boletales) 6과 27속 45종, 무당버섯목(Russulales) 3과 4속 40종 및 구멍장이버섯목(Polyporales) 6과 21속 28종으로 4목의 종수가 총 266종으로 전체 종수의 82.4%로 대부분을 차지한 것으로 나타났다. 가장 많은 종수가 조사된 균류는 그물버섯과로 37종이었으며, 무당버섯과(36종), 주름버섯과(28종) 및 광대버섯과(25종) 순으로 우점하고 있는 것으로 나타났다. 서식환경별에서는 외생균근성 버섯은 15과 38속 130종(40.2%)으로 독성 버섯은 46종, 식 약용 버섯 51종 및 식 독 불명 버섯 26종 등이 조사되었으며 낙엽 및 목재부후균은 33과 72속 114종(35.3%)이었고 이 중 독성 버섯은 10종, 식 약용 버섯 52종 및 식 독 불명 버섯 46종 등이, 지상균은 24과 47속 72종(22.3%)이었고 독성 버섯은 8종, 식 약용 버섯 31종 및 식 독 불명 버섯 29종 등인 것으로 나타났다. 월별 발생에서는 대부분의 독성 버섯, 식 약용 버섯 및 식 독 불명 버섯 등은 7월과 8월에 집중적으로 발생하는 것으로 나타났다. 고도별에서는 대부분의 독성 버섯, 식 약용 버섯 및 식 독 불명 버섯 등은 1~199m 지역에서 많이 발생되고 있는 것으로 나타났으며 200m 이상 지역에서는 현저히 낮아지는 것으로 조사되었다. 독성 버섯, 식 약용 버섯 및 식 독 불명 버섯 등은 평균온도에서는 $24.0{\sim}25.9^{\circ}C$, 최고온도 $28.0{\sim}29.9^{\circ}C$, 최저온도 $20.0{\sim}21.9^{\circ}C$, 상대습도 77.0~79.9% 및 강수량 300.0~499.9mm일 때 발생이 높은 것으로 나타났다.

신두리 해안사구에 자생하는 사구식물 내생진균의 다양성 분석 (Endophytic Fungal Diversity Associated with the Roots of Coastal Sand-dune Plants in the Sindu-ri Coastal Sand Dune, Korea)

  • 98주의 내생진균은 신두리 해안사구에 자생하고 있는 해안사구식물의 뿌리로부터 분리하였다. 8종의 해안사구식물 샘플은 모래지치(Argusia sibirica), 갯메꽃(Calystegia soldanella), 갯그령(Elymus mollis), 반디지치(Lithospermum zollingeri), 갯무(Raphanus sativus), 솔장다리(Salsola collina), 왕잔디(Zoysia macrostachya) 및 갯잔디(Zoysia sinica)이며, 신두리 해안사구로부터 채집되었다. 그리고 분리된 내생진균들은 ITS1, 5.8S와 ITS2를 포함하는 ITS-rDNA 영역에 의해 분석되었다. 해안사구식물로부터 분리된 내생진균에 대하여 다양한 지수를 적용하여 분석하였다. 해안사구식물로부터 분리된 모든 내생진균은 Capnodiales (3.09%), Eurotiales (70.10%), Glomerellales (1.03%), Helotiales (3.09%), Hypocreales (9.28%), Mortierellales (2.06%), Onygenales (1.03%), Ophiostomatales (1.03%), Pleosporales (1.03%), Polyporales (1.03%), Russulales (1.03%), Saccharomycetales (2.06%), Xylariales (1.03%)로 13개 목과 분류체계가 명확하지 않은 Incertae sedis (3.09%)으로 확인되었다. 본 연구에서는 8종의 식물로부터 내생진균을 분석한 결과 Eurotiales 목과 Hypocreales 목의 Penicillium 속(59.18%)과 Fusarium속 (5.10%)이 가장 많이 분포하는 것으로 나타났다. 갯메꽃으로부터 분리된 내생진균이 다른 해안사구식물들로부터 분리된 내생진균의 다양성 보다 높은 것으로 확인되었다.

Re-evaluation of the Genus Antrodia (Polyporales, Basidiomycota) in Korea

  • The wood decay fungi Antrodia P. Karst. play important ecological roles and have significant industrial and economic impacts as both wood degraders and sources of pharmaceutical and biotechnological products. Although each Antrodia species has distinct morphological characteristics, the misidentification rate is especially high due to their simple morphological characters. A combination of morphological and internal transcribed spacer region sequence analyses revealed that 27 of 89 specimens previously identified by morphology alone were correct, whereas 35 of these specimens were misidentified as other Antrodia species. We report here that seven Antrodia species exist in Korea (A. albida, A. heteromorpha, A. malicola, A. serialis, A. sinuosa, A. sitchensis, and A. xantha) and based on these specimens, we provide taxonomic descriptions of these species, except for A. serialis, which was only confirmed by isolate.
