Antioxidative Activities and Contents of Polyphenolic Compound of Medicinal Herb Extracts (한약재 추출물의 폴리페놀 화합물과 항산화 활성)

  • The antioxidative activity was measured on the substances of water and ethanol soluble extract from Astragalus membranaceus Bunge, Chrysanthemum morifolium Ramat, Lycium chinensis Miller, Glycyrrhiza uralensis Fischer, Angelica gigas Nakai, Zizyphus jojoba Miller, Paeonia lactiflora Pallas, Cnidium officinale Makino by four different in vitro experimental models of DPPH (a,a'-diphenyl-$\beta$-picrylhydrazyl) method, superoxide dismutase like activity, thiocyanate method, and TBARS (thiobarbituric acid reactive substances) method. The Lycium chinensis Miller contained the highest amount of polyphenolic compounds. The electron donating ability of water extract from Glycyrrhiza uralensis Fischer and ethanol extract from Chrysanthemum were higher than those of the others. The superoxide dismutase-like activity of water extract from Astragalus membranaceus Bunge was the highest among those of all the others. The water extract from Zizyphus jujuba Miller showed the highest antioxidative activity determined by TBARS method. Compared to the control, the inducing period associated with the oxidation degree was delayed up to 8 days in both the water extract from Chrysanthemum, Lycium chinensis Miller, Glycyrrhiza uralensis Fischer, and Paeonia lactiflora Pallas and the in ethanol extract from Chrysanthemum and Glycyrrhiz uralensis Fischer. These results support that water and ethanol extracts from 8 kinds of medicinal herbs contain antioxidative compounds.

Studies on Antioxidative Activity of Green Tea Extracts in Medilite-Extraction Water (Medilite 침출수에 의한 녹차 추출물의 항산화 활성연구)

  • The chemical compositions and antioxidant activity of green tea extracts in medilite-extraction water were compared to that of distilled water(DW). Antioxidant activity was fetermined by the formation of thiobarbituric acid reactive substances (TBARS) in rat liver homogenates and microsomes and the scavenging activity of free radicals by DPPH ($\alpha$, $\alpha$'-diphenyl-$\beta$-picrylhydrazyl). The order of total polyphenolic compounds and extracted yield by extracts was medilite 325 mesh-extraction water, medilite 600 mesh-extraction water and distilled water(DW). The ranges of scavenging activity of green tea extracts in DPPH method were 60.95% - 64.51%. The inhibition ratios of TBARS formation in the rat liver homogenates and microsomal fractions were significantly lower with green tea extracts by DW-extraction than with both medilite 325 mesh and 600 mesh-extraction water. The concentration of iron ion of water containing medilite 325 mesh and green tea extracts and of water containing medilite 600 mesh and green tea extracts were significantly higher compared to DW. Therefore, this result suggested that enhanced concentration of iron ion in green tea extracts by medilite-extraction water containing high iron ion content was associated with enhanced peroxidation of the rat liver microsomal fractions. These results showed that total polyphenolic compounds, the % of yield and mineral compounds of green tea extracts were increased using medilite 325 mesh and 600 mesh-extraction water.

Quantitative analysis of water-soluble vitamins and polyphenolic compounds in tomato varieties (Solanum lycopersicum L.) (토마토(Solanum lycopersicum L.) 품종 간 수용성 비타민과 폴리페놀계 성분 함량 변이 분석)

  • Tomato fruit quality is determined by the contents of various functional metabolites in addition to fruit appearance. To develop tomato cultivars with higher amounts of functional compounds, an efficient quantification method is required to identify the natural variations in the compounds in the tomato germplasm. In this study, we investigated tomato varieties, which included 23 inbred lines and 12 commercial F1 cultivars, for their contents of seven watersoluble vitamins (vitamin C, vitamins B1, B2, B3, B5, B6, and B9) and five polyphenolic compounds (quercetin, rutin, kaempferol, myricetin, and naringenin chalcone). The results of high performance liquid chromatography and liquid chromatography-mass spectrometry showed that vitamin C and naringenin chalcone were the major water-soluble vitamins and polyphenolic compounds, respectively, and their abundance was highly variable depending on the cultivar. By contrast, the contents of vitamin B1, quercetin, and kaempferol were lowest among the cultivars. With regard to the relationship between metabolic compounds and fruit characteristics, a significant association was found in fruit size, indicating that cherry tomato varieties contain higher amounts of the compounds compared to large fresh-type varieties. However, no direct association was detected in fruit color, except for naringenin chalcone. The results of this study provide new insights on the quantification of metabolic compounds and the selection of breeding materials, which are prerequisites for the development of functional tomato varieties.

  • Polyphenolic compounds including flavonoids are wide spread in the plant kingdom, and interested recently because epidemiological studies have suggested correlations between the consumption of polyphenol-rich plant foods and the prevention of chronic diseases. Soy is a main source of isoflavonoids which are high dietary intake for the oriental population. In this study, anti-inflammatory action of sophoricoside, an isoflavone glycoside isolated from immature fruits of Sophora japonica (Leguminosae family), has been demonstrated. (omitted)

  • Polyphenolic compounds have been reported to exhibit a wide range of pharmacological properties. In this study. we investigated the hepatoprotective effect of skin and seed of grape which contain abundant polyphenol compounds on dimethylnitrosamine(DMN)-induced liver damage in rats. Ingestion of skin and seed of grape (10% diet, daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phosphatase, and bilirubin levels.   • Proanthocyanidins, one of the major natural polyphenolic compounds of grape has been reported to exhibit a wide range of pharmacological properties. In this study, we investigated the hepatoprotective effect of proanthocyanidins on the dimethylnitrosamine (DMN)-induced liver damage in rats. Fermentation Characteristics for Extruded Hair of Tissue Cultured Mountain Ginseng

  • Effects of extrusion conditions (barrel temperature and moisture content) and fermentation time on the antioxidant properties of root hair of tissue cultured raw mountain ginseng (MG) were investigated. The barrel temperature/ moisture combinations were: $110^{\circ}C$/25% (MG1), $140^{\circ}C$/25% (MG2), $110^{\circ}C$/35% (MG3) and $140^{\circ}C$/35% (MG4). Red ginseng (RG) was also investigated. The contents of 2,2-diphenyl-1-picrylhydrazyl (DPPH) and polyphenolic increased after fermentation in RG and even more in MG, while extruded ginseng samples exhibited little change. The increases noted with MG and RG occurred during the first 4 days of fermentation. DPPH radical scavenging activity decreased after extrusion and was significantly higher in MG (20.93%) than RG (1.63%) on the first day of fermentation. DPPH radical scavenging activity in the barrel temperature/moisture combinations were 19.01% (MG1), 14.45% (MG2), 20.37% (MG3) and 15.78% (MG4). The content of polyphenolic compounds in ginseng samples displayed a similar trend. Acidic polysaccharide in RG and MG1${\sim}$MG4 were higher than MG, but decreased during fermentation. Crude saponin in RG and MG1${\sim}$MG4 decreased after 15 days of fermentation, while increasing in MG.

Quality and Antioxidant Activity of Granules Containing Herbal Medicine Extracts (약재 추출물을 첨가한 과립의 품질 특성 및 항산화 효과)

  • The aim of this study was to investigate the quality characteristics and antioxidant activities of herbal medicine extract granules for the comparison of commercial powder products such as adlay tea powder and roasted grain powder. The antioxidant activities of samples were estimated using reducing power, 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radical scavenging activities, and total polyphenolic contents. Herbal extract granules contained $126.44{\pm}7.26mg/100g$ of total polyphenolic contents. Interestingly, the highest antioxidant activity was found in 5.5% herbal extract granules, which showed DPPH and ABTS free radical scavenging activities of 71.82% and 85.28%, respectively, at 100 mg/mL and reducing power of O.D 1.25. These results suggested that addition of herbal medicine extract had beneficial effects on the functionality of granules without any affecting on its sensory characteristics.
