Role of Gel to Fluid Transition Temperatures of Polydiacetylene Vesicles with 10,12-Pentacosadiynoic Acid and Cholesterol in Their Thermochromisms

  • This study demonstrates gel-to-fluid transition temperatures of polydiacetylene bilayer vesicles could play important roles in their colorimetric transition temperatures. We prepared five types of polydiaceylene vesicles with 10,12-pentacosadiynoic acid (PCDA) and cholesterol (0-40 mol % of total content). From temperature-dependent observations of the optical signals (colors and UV-vis spectra), the blue-to-red colorimetric transition temperatures of polydiacetylene vesicles were decreased with the cholesterol contents. A further study with microcalorimetry and dynamic light scattering revealed that the polydiacetylene vesicles first underwent gel-to-fluid transitions, which were followed by event(s) responsible for the colorimetric transitions. Energies required for each event were quantified from analysis of the peaks in the microcalorimetry thermograms. The inclusion of cholesterol in the vesicles decreased both the gel-to-fluid and the colorimetric transition temperatures, suggesting that the colorimetric transition of the polydiacetylene vesicles was mediated by the former event although the event was not the direct reason for the color change.

Characteristics of PEGylated Polydiacetylene Liposome and its Inclusion Complex Formation with α-Cyclodextrin

  • Diacetylene lipid monomers possess the capability to self-assemble into vesicles via polymerization under ultraviolet irradiation, resulting in the formation of polydiacetylene (PDA) liposomes. Exposure of the polymerized vesicles to external stimuli is known to induce a unique blue-to-red color transition. The cyclic oligosaccharide ${\alpha}$-cyclodextrin known for its use in many applications, such as drug delivery, purification, and stimulus sensing, is able to form an inclusion complex with poly(ethylene glycol) (PEG) in aqueous solution. In this study, we prepared polymeric liposomes with PEG (PEG-PDA) with the aim of improving the stability of the vesicles and colorimetric response toward ${\alpha}$-cyclodextrin. We demonstrated that PEG-PDA liposome displays unique characteristics compared with native PDA liposome and it also shows apparent chromic properties of the inclusion complex formation with ${\alpha}$-cyclodextrin.

Chromatic Detection of Cholesterol Using Polydiacetylene Vesicles

  • In this study, a new system to determine the concentration of cholesterol using a color change was developed. The system comprised diacetylene vesicles and cholesterol oxidase (ChOx). 10,12-Pentacosadiynoic acid (PCDA) was used as the diacetylene compound, and PCDA vesicles were formed using sonication. The H2O2 produced during the reaction between cholesterol and ChOx was used to initiate the polymerization of the PCDA in the vesicles. During polymerization, the vesicles changed from colorless to blue. Therefore, the cholesterol concentration was proportional to the intensity of the blue color. The absorption at 665 nm indicated that the blue color was directly proportional to the cholesterol concentration. This indicates that the system can be used for cholesterol detection. The minimum cholesterol concentration detected using this system was 1.0 mM.

  • 폴리다이아세틸렌(polydiacetylene: PDA)은 독특한 광학적 특성, 즉 외부자극에 의하여 파란색에서 빨간색으로 색상이 변화하는 동시에 형광이 없던 상태에서 자가형광을 발현하는 특성 때문에 화학, 바이오센서로써 응용하기 위한 많은 연구들이 진행되어 왔다. 특히, 센서의 성능에서 감지하고자 하는 물질에 대한 우수한 민감도는 매우 중요하다. 본 연구에서는 다양한 필터 사이즈를 이용하여 10,12-pentacosadynoic acid(PCDA) 베시클의 크기를 조절함과 동시에 중합온도를 조절하여 ${\alpha}$-사이클로텍스트린(CD)을 검출하여 두 가지 효과가 민감도 향상에 어떤 영향을 미치는지 조사하였다. 필터를 사용하지 않은 베시클과 0.22 ${\mu}m$로 필터한 베시클을 $25^{\circ}C$에서 고분자한 후에 ${\alpha}$-CD(5 mM)와 30분 반응하였을 때 색전이 정도(colorimetric response, CR)가 31.4%에서 74.0%로 증가하였다. 또한, 0.22 ${\mu}m$로 필터한 베시클을 $25^{\circ}C$$5^{\circ}C$에서 고분자한 후에 ${\alpha}$-CD(5 mM)와 30분 반응하였을 때 CR값이 74.0%에서 99.2%로 증가하였다. 이는 폴리다이아세틸렌의 크기와 고분자시 온도를 조절함으로써 민감도를 크게 증가시킬 수 있음을 보여준다. 또한, 폴리다이이아세틸렌은 감도 향상이 매우 중요한 바이오물질을 검출하는데 적용될 수 있을 것이다.

  • 본 연구에서는 폴리다이아세틸렌(polydiacetylene, PDA) 베시클을 이용하여 여러 가지 킬레이트제(chelating agent)를 쉽게 검출할 수 있는 센서 시스템을 개발하였다. 다른 센서들과 비교하여 PDA기반 센서는 많은 장점이 있다. 첫째로, 형광물질의 부착이 필요 없는 무표지 검출(label-free detection)이 가능하여 실험 절차가 간단하고 빠르다. 둘째로, PDA는 청색에서 외부 자극에 의해 적색으로 변화하는 색전이를 일으키므로 육안으로 쉽게 검출을 확인할 수 있었다. 끝으로, 특정 파장에서의 colorimetric response를 측정하여 각각의 킬레이트제의 농도에 따른 정량검출도 가능하다. 본 연구에서는 5가지 종류의 킬레이트제, 즉 EDTA, EGTA, NTA, DCTA, DTPA를 PDA 베시클과 반응시켰으며, 이중에서 EDTA, DCTA는 특히 강한 반응으로 PDA의 색전이를 유도함을 알 수 있었다. 본 연구를 통하여 PDA 베시클을 사용하여 어떠한 기계나 동력을 사용하지 않고 색전이를 이용하여 킬레이트를 성공적으로 검출할 수 있음을 보여주었다.

Detection of ${\alpha}-Cyclodextrin$ and E.coli Cell Using Polydiacetylene Supramolecules

  • We immobilized and patterned PDA vesicles on solid substrate using micro arrayer, which have moieties to react with chemical and biological materials. Immobilized vesicle system was developed since it possesses many advantages in multiple screening, durable stability, and higher sensitivity. We applied polydiacetylene supramolecules to chemical and biological sensors for detection of ${\alpha}-cyclodextrin$ and E.coli cell selectively. This detection method could be applied as DNA chip, protein chip, and cell chip for multiple screening as well as chemical sensor by modifying the functional groups of diacetylene monomer.

  • We prepared amino acid-derived diacetylene monomers using solid-phase organic synthesis. The solid-phase synthetic method allowed for the rapid and efficient preparation of functional diacetylenes. Amino acids having hydrophobic sidechains such as alanine, leucine, and phenylalanine, as well as hydrophilic sidechains such as aspartic acid and lysine, were successfully coupled to the diacetylene lipid. The diacetylene monomers prepared in this way were subjected to routine procedures for the generation of polydiacetylene vesicles. Depending on the nature of the side-chains, pink to blue colored polydiacetylenes were generated.
