Polycyclic Aromatic Hydrocarbons Hazard Assessment of Shellfish due to Oil Spill Accidents (유류 유출 사고 해역에 서식하는 패류의 다환방향족탄화수소(PAHs) 위해도 평가)

  • On 7 December 2007, about 12,547 kL of crude oil spilled from the Hong Kong registered tanker Hebei Spirit along the west coast of the Republic of Korea, including Taean-gun, Chungcheongnamdo Province. This study evaluated the safety of seafood collected from the coastal area polluted by the crude oil. The range of total polycyclic aromatic hydrocarbons (${\sum}PAHs$) at 22 stations was 3.9-37.1 ng/g. The concentration of ${\sum}PAHs$ was higher in oysters, Crassoatrea gias, than that in short-necked clams, Ruditapes philippinarum. Benzo(a)pyrene, a highly toxic PAH, ranged from 0.07-1.47 ng/g, which did not exceed the European Union regulatory limit for benzo(a)pyrene. The toxicity equivalent of benzo(a)pyrene in oysters and short-necked clams was 0.49-1.70 and 0.09-1.01 ng/g, respectively. The estimated life time cancer risk was very low, i.e., $1.31{\times}10^{-8}$ for the oysters and $6.9{\times}10^{-9}$ for the short-necked clams. The body burden of PAHs in bivalves originated mostly from petroleum contamination, but the levels was not sufficiently high to harm human health.

Levels of Polycyclic Aromatic Hydrocarbons in Fish, Shellfish and their Processed Products (국내 유통 어패류 및 가공품 중 Polycyclic Aromatic Hydrocarbons 함량)

  • Cocentrations of PAHs [benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenzo(a,h)anthracene, benzo(g,h.i)perylene, indeno(1,2,3-c,d)pyrene] in fish (n=120), shellfish(n=50) and their products (n=35) were estimated by saponification and extraction with n-hexane, clean-up on Sep-Pak Florisil Cartridges and HPLC/FLD. Overall recoveries for eight PAHs spiked into samples ranged from 90 to 106%. Mean level of benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenzo(a,h)anthracene, benzo(g,h,i)perylene and indeno(1,2,3-c,d)pyrene were not detected, 0.01, 0.04, 0.07, 0.05, 0.004, 0.0008 and 0.06ng/g, respectively, similar to those reported by other countries.

  • Pyrene of natural and anthropogenic sources is one of the toxic, mutagenic polycyclic aromatic hydrocarbons (PAHs) listed as priority pollutants. The objectives of this research are to break down pyrene by using ozonation, identify the intermediates and byproducts of pyrene, and test the biodegradability of intermediates and byproducts of pyrene in the aqueous phase. Since pyrene is non-polar, hexane was chosen as a solvent to effectively dissolve pyrene. Pyrene solutions were treated with ozone, as it has high oxidation capacity and electrophilic characteristic. After different ozonation pretreatment times (2, 3, and 10 minutes), intermediates of pyrene in the form of yellowish solid were collected from the hexane solution using a centrifuge. They were identified by gas chromatography/mass spectrometer (GC/MS). $BOD_5$, COD, and E-coli toxicity tests have been performed to assess the ozonation products.

Analysis of Polycyclic Aromatic Hydrocarbons in Agricultural Waterways in Chungbuk and Gyeongbuk Provinces in Korea

  • An efficient, quick and low-cost extraction and clean up method for the determination of 14 polycyclic aromatic hydrocarbons (PAHs) in the agricultural water samples was optimized using gas chromatography-tandem mass spectrometry (GC-MS/MS). The extraction of the target compounds in water sample was carried out with acetonitrile, followed by partitioning promoted by the addition of salt. As a clean-up procedure, dispersive solid phase extraction was employed to purify the analytes of interest for GC-MS/MS analysis. This method was successfully applied for the quantification of PAHs in real water samples collected for the purpose of monitoring from the waterways located in Chungbuk (15 sites) and Gyeongbuk (6 sites), S. Korea. Phenanthrene (0.54 to $2.53{\mu}gL^{-1}$) was detected in all the water samples collected from both the sites. Fluoranthene was detected in the water samples collected from the two sites in Gyeongbuk province, but other PAHs were not determined in these water sampling sites. Based on these results, the determined PAHs were conducted using an environmental risk assessment. The risk characterization ratios (RCRs) for phenanthrene ranged from 0.37 to 3.21. These RCR values referred to as risk was not controlled because RCR values of some sites were greater than 1. In conclusion, it is proposed that the optimized method in combination with GC-MS/MS could be successfully employed for the determination of PAHs in any environmental samples including water samples.

Determination of Polycyclic Aromatic Hydrocarbons in Smoked Food Products (훈연식품 중 polycyclic aromatic hydrocarbons 함량 분석)

  • This study was accomplished that analysis of seven polycyclic aromatic hydrocarbons (PAHs) in smoked or nonsmoked processing foods by high performance liquid chromatography (HPLC) with fluorescence detection. The calibration line was constructed with injected different levels of standard concentration. Limit of detection (LOD) and limit of quantification(LOQ) showed higher linearity ($r^{2}$=0.998) reasonably, and recovery exhibited 0.033-0.666 $\mu$g/kg, 0.108-2.217 $\mu$g/kg and 69.31-90.14%, respectively. As a result, the samples using smoked tuna as smoked materials contained seven PAHs with different range from 0.256 to 0.486 $\mu$g/kg. The benzo[a]pyrene, indicator of PAHs, was detected to below the LOQ in two samples. Concentrations of benzo[a]pyrene in three samples were below the 2 $\mu$g/kg which is the limit of regulation. Smoked tuna sauces were detected from 0.321 to 0.552 $\mu$g/kg and not detected in drying powders. PAHs of smoked meat products were ranged from 0.720 to 2.027 $\mu$g/kg and are higher than concentration of tuna smoked samples. PAHs were very low in non-smoked foods including mustard, herb, and roasted meats.

Characterization of PAH (Polycyclic Aromatic Hydrocarbon)-Degrading Bacteria Isolated from Commercial Gasoline (상용 휘발유로부터 분리한 다환 방향족 탄화수소(PAH) 분해 세균의 특성)

  • BACKGROUND: Recent studies have described the importance of bacteria that can degrade polycyclic aromatic hydrocarbons (PAHs). Here we screened bacterial isolates from commercial gasoline for PAH degraders and characterized their ability to degrade PAHs, lipids and proteins as well as their enantioselective epoxide hydrolase activity, salt tolerance, and seawater survival. METHODS AND RESULTS: One hundred two bacteria isolates from commercial gasoline were screened for PAH degraders by adding selected PAHs on to the surface of agar plates by the sublimation method. A clear zone was found only around the colonies of PAH degraders, which accounted for 13 isolates. These were identified as belonging to Bacillus sp., Brevibacterium sp., Micrococcus sp., Corynebacterium sp., Arthrobacter sp., and Gordonia sp. based on 16S rRNA sequences. Six isolates belonging to Corynebacterium sp., 3 of Micrococcus sp., Arthrobacter sp. S49, and Gordonia sp. H37 were lipid degraders. Arthrobacter sp. S49 was the only isolate showing high proteolytic activity. Among the PAH-degrading bacteria, Arthrobacter sp. S49, Brevibacterium sp. S47, Corynebacterium sp. SK20, and Gordonia sp. H37 showed enantioselective epoxide hydrolase activity with biocatalytic resolution of racemic styrene oxide. Among these, highest enantioselective hydrolysis activity was seen in Gordonia sp. H37. An intrinsic resistance to kanamycin was observed in most of the isolates and Corynebacterium sp. SK20 showed resistance to additional antibiotics such as tetracycline, ampicillin, and penicillin. CONCLUSION: Of the 13 PAH-degraders isolated from commercial gasoline, Arthrobacter sp. S49 showed the highest lipid and protein degrading activity along with high active epoxide hydrolase activity, which was the highest in Gordonia sp. H37. Our results suggest that bacteria from commercial gasoline may have the potential to degrade PAHs, lipids, and proteins, and may possess enantioselective epoxide hydrolase activity, high salt tolerance, and growth potential in seawater.

Vertical Distribution and Potential Risk of Particulate Polycyclic Aromatic Hydrocarbons in High Buildings of Bangkok, Thailand

  • Vertical variations of polycyclic aromatic hydrocarbon (PAH) concentrations in $PM_{10}$ were investigated in order to assess the factors controlling their behavior in the urban atmosphere of Bangkok City, Thailand. Air samples were collected every three hours for three days at three different levels at Bai-Yok Suit Hotel (site-1 and site-2) and Bai-Yok Sky Hotel (site-3) in February $18^{th}-21^{st}$, 2008. The B[a]P concentration showed a value 0.54 fold, lower than the United Kingdom Expert Panel on Air Quality Standard (UK-EPAQS; i.e. 250 pg $m^{-3}$) at the top level. In contrast, the B[a]P concentrations exhibited, at the ground and middle level, values 1.50 and 1.43 times higher than the UK-EPAQS standard respectively. PAHs displayed a diurnal variation with maximums at night time because of the traffic rush hour coupled with lower nocturnal mixing layer, and the decreased wind speed, which consequently stabilized nocturnal boundary layer and thus enhanced the PAH contents around midnight. By applying Nielsen's technique, the estimated traffic contributions at Site-3 were higher than those of Site-1: about 10% and 22% for Method 1 and Method 2 respectively. These results reflect the more complicated emission sources of PAHs at ground level in comparison with those of higher altitudes. The average values of incremental individual lifetime cancer risk (ILCR) for all sampling sites fell within the range of $10^{-7}-10^{-6}$, being close to the acceptable risk level ($10^{-6}$) but much lower than the priority risk level ($10^{-4}$).

Assessing the Formation of Polycyclic Aromatic Hydrocarbons in Grilled Beef Steak and Beef Patty with Different Charcoals by the Quick, Easy, Cheap, Effective, Rugged, and Safe (QuEChERS) Method with Gas Chromatography-Mass Spectrometry

  • This study investigated the effects of different charcoals on the occurrence of 16 polycyclic aromatic hydrocarbons (PAHs) in grilled beef steaks and beef patties. Seven different charcoals were used as follows: from oak wood (C1), from orange wood (C2), from Valonia oak wood (C3), from Marabu wood (C4), extruded charcoal from beech wood (C5), from coconut shells (C6), and from hazelnut shells (C7). The grilling times for each charcoal type were 6 min for the beef patties and 7 min for the beef steaks, until the internal temperature reached at least 74℃. The total concentration of 16 PAHs (PAH16) in beef steaks grilled with C1 (35.75 ㎍/kg) and C7 (36.39 ㎍/kg) was higher than that of C3 (23.80 ㎍/kg) and C6 (24.48 ㎍/kg; p<0.05). The highest amounts of PAH16 (216.40 ㎍/kg) were determined in the beef patty samples grilled using C5 (p<0.05). The summation of benzo[a]anthracene, chrysene, benzo[b]fluoranthene and benzo[a]pyrene, referred to as PAH4, was not detected in any of the beef steaks, whereas it was determined in the beef patties grilled with C2 (7.72 ㎍/kg) and C5 (22.95 ㎍/kg; p<0.05). The PAH16 concentrations of the beef patty samples in each charcoal group were significantly higher compared to the beef steaks (p<0.05). To avoid the formation of high PAH levels, the use of extruded charcoal and hazelnut shell charcoal should therefore be avoided when charcoal grilling beef steaks and beef patties, and low-fat meat products should be preferred.

Distribution of Polycyclic Aromatic Hydrocarbon at Kongsfjorden in Spitsbergen, Svalbard Islands (북극 스발바드 군도 스피츠베르겐섬 콩스피요르드에서의 다환 방향족 탄화수소화합물의 분포 특성)

  • In order to elucidate the polycyclic aromatic hydrocarbon concentration and its origin in arctic area, four arctic brown algae (Laminaria saccharina, L. digita, Alaria esculenta, Desmarestia aculeata), one marine invertebrate (Echinoidea) and sediments were collected from Kongsfjorden in Spitsbergen from the late July to early August, 2003. In case of macroalgae, the young blade part above growth point and the old stipes and blades beneath growth point were separated and analyzed for polycyclic aromatic hydrocarbons (PAHs) in an attempt to check the mechanism of uptake in macroalgae to accumulate PAH. There was no difference in PAH concentrations between sampling sites (Stations B and C), species, and blades beneath and above growth point. PAH concentrations in all samples collected in this study were relatively higher than those reported in other areas of arctic. Especially, station C, which is known as an unpolluted area, showed 10 times higher PAH concentration (8,765 ng/g) in sediment than station A (694 ng/g) around harbor. In addition high PAH concentration, station C had very higher proportion of methylated PAH to parent PAH in sediment than station A. Source analysis using PAH isomer pair ratios as indicators showed that Kongsfjorden area seemed to be relatively contaminated with PAH derived from direct petroleum input.

Factors Affecting Concentration of Polycyclic Aromatic Hydrocarbons in Oyster, Laver and Rockfish (굴, 김 및 조피볼락에서 다환성방향족탄화수소(Polycyclic Aromatic Hydrocarbons, PAHs)의 잔류에 미치는 요인에 대한 연구)

  • A total of 15 different residues of polycyclic aromatic hydrocarbons (PAHs) in each 20 samples of Pacific oysters, dried laver and rockfish obtained from seafood markets were analyzed. The prevalence of samples in which more than one PAH residues were found was 75% in oyster, 35% in rock fish hepatopancreas, 0% in rockfish muscle and laver, respectively. To estimate factors contributing to this residue level difference among organisms, tissue concentrations were analyzed after exposing three organisms to phenanthrene, a representative PAH, with concentration of 0.01 or $0.1{\mu}g/mL$ for 2 weeks. Phenanthrene levels after exposure were higher in the oyster digestive gland, laver and rockfish hepatopancreas, but were lower in the oyster whole meat or rockfish muscle. This finding disproved that any close relationship between the residue difference of market samples and concentrating properties of PAHs. The second possible factor analyzed was total lipid contents in the three organisms. Although higher lipid level in hepatopancreas of rockfish may contribute accumulation of PAH residues in the rockfish, lipid factor did not affect to PAH levels in other organism samples. Activity of 7-ethyoxyresorufin-O-deethylase (EROD), a kind of cytochrome $P_{450}$ enzyme, was measured to evaluate the eliminated amount of PAHs through metabolism. The higher EROD activity in rockfish, compared to that in oyster, was likely to contribute to the lower PAH residues in the rockfish. More factors, such as different exposure history, organisms' ability to escape, ingestion through prey organisms, and post-harvest loss, should be studied in the future.
