Adrenal hypoplasia congenita (AHC) is a rare cause of adrenal insufficiency during neonatal period. Mutations in the gene coding for DAX1 cause X-linked adrenal hypoplasia. Most affected patients are shown to have salt wasting and hyperpigmentation on the skin during the neonatal period and require intensive medical care. In addition, it is usually associated with hypogonadotropic hypogonadism in adolescence. The DAX1 gene is expressed in the adrenal cortex, pituitary gland, hypothalamus, testis, and ovary. We report on a patient with genetically confirmed AHC whose initial clinical presentations were consistent with congenital adrenal hyperplasia. A point mutation in the DAX1 gene identified in this report resulted in a truncated DAX1 protein. Our patient was diagnosed with AHC.

Bacterial canker is a devastating disease of kiwifruit caused by the bacterium Pseudomonas syringe pv. actinidiae. Canker disease of kiwifruit in Korea has been controlled using streptomycin for more than two decades. Four streptomycin-resistant strains, belonging to biovar 2, which are found only in Korea, were collected between 2013 and 2014 from different orchards located in Jeju, Korea. The genetic background for streptomycin resistance among P. syringe pv. actinidiae strains were determined by examining the presence of strA-strB or aadA, which are genes frequently found in streptomycin-resistant bacteria, and a point mutation at codon 43 in the rpsL gene. All four streptomycin-resistant strains of P. syringe pv. actinidiae investigated in this study contained strA-strB as a resistant determinant. The presence of the aadA gene and a mutation in codon 43 of the rpsL gene was not identified.
Pediatric central nervous system (CNS) vascular malformations are a group of abnormal blood vessel formations within the brain or spinal cord in children. The most crucial point of pediatric CNS vascular malformation is that no golden standard classifications exist. In addition, there is a big gap in knowledge and the viewpoint of clinicians, radiologists, and pathologists. In addition, many genes associated with pediatric CNS vascular malformation, such as Sturge-Weber-Dimitri syndrome with guanine nucleotide-binding protein G(q) subunit alpha (GNAQ) gene mutation, and cavernous malformations with cerebral cavernous malformations 1 (CCM1), CCM2, and CCM3 gene mutation, were recently revealed. For proper therapeutic approaches, we must understand the lesions' characterizations in anatomical, morphological, and functional views. In this review, the author would like to provide basic pediatric CNS vascular malformation concepts with understandable diagrams. Thus, the author hopes that it might be helpful for the proper diagnosis and treatment of CNS pediatric vascular malformations.
This study was conducted to evaluate the mutagenic potential of dimethyl isophthalate (DMIP) using Ames bacterial reverse mutation test, chromosomal aberration test and mouse lymphoma $tk^{+/-}$ gene assay. As results, in Ames bacterial reversion assay, DMIP was tested up to the concentration of 5,000 ${\mu}g$/plate and did not induce mutagenicity in Salmonella typhimurium strains TA98, TA100, TA1535 and TA1537, and Escherichia coli WP2uvrA with or without metabolic activation (S9 mix). Using cytotoxicity test, the maximal doses of DMIP for chromosomal aberration assay were determined at 1,250 ${\mu}g/mL$, which was a minimum precipitation concentration ($IC_{50}>1,940\;{\mu}g/mL$ or 10 mM) and at 155 ${\mu}g/mL$ ($IC_{50}:155\;{\mu}g/mL$) in the presence and the absence, respectively, of S9 mix. DMIP in the presence of S9 mix induced statistically significant (P<0.001) increases in the number of cells with chromosome aberrations at the dose levels of over 250 ${\mu}g/mL$, when compared with the negative control. However, DMIP in the absence of S9 mix did not caused significant induction in chromosomal aberrant cells. In MLA, DMIP at the dose range of 242.5-1,940 ${\mu}g/mL$ in the presence of S9 mix induced statistically significant increases in mutation frequencies related to small colony growth, whereas any significant mutation frequency was not observed in absence of S9 mix. From these results, it is conclusively suggested that dimethyl isophthalate may be a clastogen rather than a point mutagen.
We have utilized the PCR-RFLP method to detect the ryanodine receptor(RYR1) gene mutation and to estimate the genotype frequencies of the RYR1 gene in commercial crossbred pig population. The exon region(659bp) including point mutation(C ${\rightarrow}$T; Arg ${\rightarrow}$Cys) in the porcine ryanodine receptor gene, which is a causal mutation for PSS, was amplified by PCR and digested with Cfo I restriction enzyme. The RYR1 gene was classified into three genotypes by agarose gel electrophoresis. The normal homozygous(NN) individuals showed two DNA fragments consisted of 493 and 166bp. The mutant homozygous(nn) individuals showed only one DNA fragment of 659bp. Also, all three fragments(659, 493 and 166bp) were showed in heterozygous(Nn) carrier animals. The proportions of normal, carrier and PSS pigs within crossbred population of pigs were 81%, 15% and 4%, respectively. According to the results of analysis of variance for the association of genotypes of RYR1 of pigs at 30kg, day age at 90kg and average daily gains, the RYR1 nn genotype was very higher than RYR1 NN genotype for day age at 30kg with 5% level of significant difference, but no significant difference for association of any other genotypes with day age at 90kg and average daily gain in crossbred pigs. Therefore, DNA diagnosis by using PCR-RFLP analysis for the PSS gene was useful for large-scale screening of commercial pigs in the swine industry.
본 연구는 319두의 서로 다른 품종에서 PSE육을 생산하는 PSS 돼지 출현빈도를 조사하였다(Yorkshire 150; Landrace 89 and Duroc 80). PCR-RFLP법을 이용하여 돼지의 모근을 DNA sample로 사용하여, PCR로 증폭된 유전자는 Cfo I 제한 효소로 절단하여 종돈에 존재하는 ryanodine receptor (RYR 1) 돌연변이 유전자의 출현빈도를 조사한 결과를 요약하면 다음과 같다. 모근에서 추출한 DNA를 주형으로 한 Primary PCR을 수행한 결과 ryanodine receptor 유전자 중 659bp의 증폭산물을 얻었으며, second PCR을 수행한 결과에서는 522 bp의 증폭산물을 얻었다. 이 증폭산물은 porcine ryanodine receptor 유전자의 exon 영역 중 PSS를 유발하는 point mutation(C\longrightarrowT; Arg\longrightarrowCys) 부분을 포함하고 있으므로 Cfo I 제한효소에 의해 분석될 수 있으며, agarose gel 전기영동에 의하여 세 가지의 유전자형으로 분류할 수 있다. 정상 homotype(NN)은 두 개의 DNA band(439, 83bp)로 나타나며, 열성 homotype(nn)은 552 bp의 단일 밴드로 출현한다. 그리고 세 개의 밴드(522, 439 그리고, 83 bp)는 heterotype(Nn)의 잠재성 돼지로 표현된다. Yorkshire종에서는 정상돼지가 98.00%로 나타났으며, hetero 돼지는 2.00% 그리고, PSS돼지는 출현하지 않았다. Landrace 돼지에서는 정상돼지가 87.64%로 나타났으며, hetero 돼지와 PSS패지가 각각 11.24와 1.12%로 나타났으나, Duroc종에서는 정장돼지(NN)만이 출현하였다. 대립 유전자 빈도는 Yorkshire종은 정상 N유전자가 0.990의 비율로 나타났으며, 열성 n 유전자는 0.010의 비율로 출현하였으며, Landrace종에서는 N유전자와 n유전자가 각각 0.933과 0.067의 빈도로 출현하였으며, Duroc종에서는 N 유전자의 빈도가 1.000의 빈도로 나타났으나, n유전자의 빈도는 0.000의 빈도로 나타났다. 3품종 집단 모두에서 Hardy-Weinberg 법칙과 일치하여 유전적 평형을 이루고 있었다.
VT2(Verotoxin-2)의 효소활성 영역에 해당되는 두 영역의 아미노산들에 대하여, 첫번째 보존영역의 Glu167을 conservative point mutation 시키고, 두 번째 보존영역의 구성 아미노산 5개 전부를 deletion mutation 시켜, 각 변이주에서 독성의 감소 정도를 wild type과 비교한 결과 다음과 같은 성적을 얻었다. 1. pKSC101을 Eco RI과 Pst I으로 절단하여 940bp insert를 통일 제한효소로 절단한 M 13mp19에 삽입하여 pEP19RF를 구축하였다. 이를 이용하여 dU-SSDNA template를 제조하고, mutagenic primer를 annealing 하여 변이를 도입하였으며, 변이가 도입된 insert를 acceptor plasmid에 삽입시켜 각각 발현 플라스미드 pOEX와 pDEX를 구축하였다. 각각의 mutant 단백질을 발현시키기 위하여 pOEX와 pDEX를 JM109에 형질전환시켜 mutant 재조합 균주인 POMUT109와 DEMUT109를 작성하였다. 2. POMUT109와 DEMUT109를 IPTG 유도 발현시킨 배양상층액을 Vero cell에 대하여 세포독성을 시험한 결과 wild type에 비하여 POMUT109의 배양 여액에서는 2000배, DEMUT109의 배양여액에서는 적어도 3000배 이상의 세포독성을 감소시켰다.
old-gold-crimson ($og^c$) 과색 돌연변이는 라이코펜의 함량이 증가된 진붉은색 토마토 과실을 생산한다. 이러한 돌연변이는 토마토의 carotenoid 생합성경로에 관여하여 라이코펜을 ${\beta}$-carotene으로 전환시키는 라이코펜 ${\beta}$-cyclase (Crt-b) 유전자(B)에 point mutation을 일으켜 정상적인 효소생성을 저해한다. 높은 함량의 라이코펜을 생성시키는 토마토 품종개발은 유전자 연관 DNA 마커를 이용한 분자표지이용선발(MAS)을 통해 가속화 될 수 있다. $og^c$ 돌연변이는 라이코펜 ${\beta}$-cyclase(Crt-b) 유전자 내 poly-A 서열반복 지점에서 adenine (A) 단일 뉴클레오티드의 결손에 의한 frame-shift mutation에 의해 일어나며, 이러한 대립유전자의 운영과 검증을 위해 $og^c$ 대립유전자로부터 합성되는 PCR 산물에 Hin fI 제한효소 인식부위가 인위적으로 생성되도록 PCR 프라이머에 단일 뉴클레오티드 mismatch 부위를 만들어 dCAPS 마커를 개발하였다. 본 dCAPS 마커는 유전자 유래의 공우성 PCR 마커로서 고함량 라이코펜 토마토개발을 위한 육종 프로그램의 MAS에 효과적으로 사용될 수 있다.
