Studies on Cellulolytic Enzymes Produced by Pleurotus spp. in Synthetic Medium( I ) -Effects of Carbon and Nitrogen Sources- (합성배지(合成培地)에서 Pleurots속(屬)이 생산(生産)하는 섬유소(纖維素) 분해효소(分解酵素)에 관한 연구(硏究)(제1보)(第1報) -탄소원(炭素源)과 질소원(窒素源)의 영향(影響)-)

  • Among the eight strains, Pleurotus sajor-caju JAFM 1017 was selected as most potent producer of cellulolytic enzymes. The avicelase and CMCase activity reached maximum levels after 10 days, and ${\beta}-glucosidase$ activity reached a maximum level after 19 days. Among the various carbon sources, cellulose powder was most effective for the production of avicelase and ${\beta}-glucosidase$, and Na-CMC (sodium carboxymethyl cellulose) was good for the production of CMCase. The optimum concentration of cellulose powder was 1.0% (w/v), and glucose (1.0%) completely depressed the production of enzymes. Nitrates were effective for the production of enzymes, but nitrites did not support growth. The production of cellulolytic enzymes increased as the concentration of urea increased. The appropriate concentration of urea was 0.054% (w/v).

Selection of Pleurotus sajor-caju as Suitable Species for Cultivation under Summer Climatic Conditions in Korea (하기(夏期) 재배(栽培)에 알맞는 여름 느타리 버섯의 선발(選拔))

  • This experiment was performed to select the most suitable species of oyster mush­room for cultivation during summer season in Korea. The selection was made based upon mycelial growth on the ricestraw substrates, fruiting under hot weather conditions during the summer season in Korea and potential high productivity using six cultures from South-east Asia. The newly selected strain of Pleurotus sajor-caju (ASI 2070) grew well on the rice straw substrates and produced the higher yields of sporophores than other cultures tested. The primordia of the mushroom were formed between 10 to $35^{\circ}C$. The optimal temperature for fruiting ranged from 20 to $25^{\circ}C$.

Development of Suhan Strain-specific SCAR Marker in Pleurotus ostreatus (느타리 버섯에서 수한 품종 특이 SCAR marker 개발)

  • In this study, 81 commercial strains of Pleurotus species cultivated in South Korea were analyzed with randomly amplified polymorphic DNA (RAPD) technique. Sequence characterized amplified region (SCAR) markers were developed by designing from one RAPD polymorhic band specific to Suhan strain. The SCAR primer pair 'S-OPA13-1' amplified a 590-bp fragment in the varieties originated from Suhan strain. The Blast search of S-OPA13-1 showed high homology to the POMFBO1 P. ostreatus cDNA clone MFB02-A05 and Laccaria bicolor S238N-H82. The results showed that this SCAR marker can clearly distinguish Suhan strains from Pleurotus spp.

Effects of Incompatibility on Protoplast Fusion between intra-and inter Species in Basidiomycete, Pleurotus spp. (느타리버섯의 불화합성(不和合性)이 종내(種內) 및 종간(種間) 원형질체(原形質體) 융합(融合)에 미치는 영향(影響))

  • Effects of incompatibility existing between intra-and interspecies in Pleurotus spp. on protoplast fusion, clamp formation of their fusants and fruitbody production were investigated. Protoplast fusion between intra-and interspecies of the fungus was achieved by Poly ethylene glycol treatment. The fusion frequency between intraspecies was a little higher than that of interspecies. Fusion frequency between interspecies was not correlated with their similarities based on isozyme patterns. In case of protoplast fusion between intra-and interspecies, the fusants from the compatible isolates produced normal fruit bodies, while those from the incompatible isolates did not produce clamp connections and fruit bodies except those of a few isolates presumed mutants.

Mulching Cultivation Method and Harvesting Implement of Pleurotus ostreatus (느타리버섯 멀칭재배 방법과 수확도구)

  • The study which was conducted to determine the effects of the polyethylene film on the culture of Pleurotus ostreatus is summarized in the following: 1) The fresh weight of Pleurotus ostreatus cultured by the box culture method mulching with black polyethylene film was 2,237g/box and, 2,028g/box by white polyethylene film mulching, and 1,695g/box at the conventional culture which was by 24.2% and 16.4% higher than that of the conventional culture.2) The fresh weight of P. ostreatus cultured by the shelf culture method mulching with the black polyethylene film was 17.4kg/m$^2$ at the white polyethylene film culture, 14.6kg/m$^2$ at the conventional culture which was by 16.2%, 7.5% higher than that of the conventional culture. 3) The best shape appearance in terms of the diameter in P. ostreatus was 5~30mm and the intervals were 10cm respectively. 4) The black polyethylene film mulching in P. ostreatus observed in good protection against Pseudomonas spp., Trichoderma, or mushroom flies. 5) The black polyethylene film mulching method for the culture of P. ostreatus was much better than that of the shelf-culture, box-culture or the sack culture in terms of total yield and quality. 6) The length of harvesting implement of P. ostreatus was suitable for 45~120cm in order to use on the harvest of the mulching.

The effect of RGB LED lights on oyster mushroom (Pleurotus spp.) fruit-body characteristics (RGB LED 광원이 느타리류의 자실체 특성에 미치는 효과)

  • Light plays an important role in fruit-body development and morphology during Pleurotus spp. cultivation. To understand the effects of light color on fruit-body properties, we evaluated the fruit-body characteristics of Pleurotus spp. Varieties cultivated under blue, red, and purple LED light sources. The main results are as follows: The overall fruit-body shape showed differences depending on the color of the LED light. The fruit-bodies of mushroom cultivated under blue and purple light were generally similar to the mushroom shapes typically produced, while those of mushroom cultivated under green light were abnormally shaped, probably due to the absence of effective light source. The average cap lightness of mushrooms cultivated under blue, green, and purple LED lights was 57.0, 57.4, and 59.4, respectively. The average cap lightness of all varieties except Wonhyeong1ho and Hwang-geumsantari cultivated under the three LED light sources were statistically significantly different (P<0.05). The cap redness varied significantly depending on the LED lighting and variety. Only Gonji7hoM, the cap color mutant of Gonji7ho, showed negative cap redness values under all three LED light sources. Among the eight varieties excluding Gonji7ho, the highest cap redness was observed when cultivated under the blue LED. The average harvest weight of the varieties cultivated under purple, blue, and green LED light were 68.0, 58.3, and 50.1 g, respectively. The yield of Gonji7ho, the mushroom variety with the highest yield, cultivated under blue, green, and purple LED light were 92.8, 77.1, and 98.6 g, respectively. The earliness when grown under the purple, blue, and green LED lights were 5.3, 5.8, and 5.8 days, respectively. Among the varieties, six, three, and two cultivars showed the shortest earliness under the purple, green, and blue LED, respectively. The fruit-body lengths were 66.4, 51.8, and 46.8 mm when cultivated under green, purple, and blue lights, respectively. These results are expected to serve as a foundation for producing mushrooms with traits demanded in the market.

Changes in the Contents of General Compositions and Free Sugars of Oyster Mushrooms (느타리버섯의 일반성분(一般成分) 및 유리당(遊離糖)의 합량변화(合量變化))

  • Chemical composition of the oyster mushroom(Pleurotus spp.) at different cropping stage were analyzed. The contents of crude protein, soluble protein, reducing sugar, total sugar and crude fat in Pleurotus ostreatus 201 were a little higher than those of Pleurotus sajorcaju. The Component contents were not different from the first to the second flush, while the contents were slightly decreased from the third flush. A11 component contents were highest in $3{\sim}5cm$ pileus size, the contents were slightly decreased as pileus size increase to over $3{\sim}5cm$, except for reducing sugar. Crude fiber content in Pleurotus sajor-caju were a little more than Pleurotus ostreatus 201, and in course of increasing cropping time, ash contens of two strains were simmilar, and were decreased after the second flush. These component contents mere increased by the growth of the pileus. The sugars were identified as xylose, glucose, mannitol and trehalose in Pleurotus sajor-caju, and xylose, fructose, glucose, mannitol and trehalose in Pleurotus ostreatus 201, respectively. Mannitol and trehalose contents in Pleurotus sajor-caju were a little more than that in Pleurotus ostreatus 201. Glucose content of Pleurotus sajor-caju and Pleurotus ostreatus 201 was highest in $1{\sim}3cm$ pileus size, while other sugars in $3{\sim}5cm$ pileus size, and the sugar contents were decreased at over the above size of the pileus.

  • Perforated vinyl mulching technique was performed on oyster mushroom beds for controlling mushroom diseases. Mycelium under vinyl sheets were safely protected from outside undesirable microorganisms. One of two mushroom farms showed 75% of disease incidence, the other 40% and National Institute of Agricultural Science and Technology(NIAST) 13% in the conventional growing method, whereas 12%, 14%, and 5% in the vinyl mulching cultivation method. Waterlogging caused mushroom bed worse, and Trichoderma spp. were infested on the conventional mushroom bed. Disease incidence investigated in other case was 25% to 30% in the conventional growing method, whereas 5 to 9% in the vinyl mulching cultivation method. Yields in conventional method were 6.5 to 7.2 $kg/m^2$ and those in vinyl mulching method were 7.6 to 8.1 $kg/m^2$. So it was suggested that vinyl mulching technique was good for prevention from disease and elevation of productivity.

  • The experiment was performed to find out the possibilities to detect virus infection in oyster mushrooms, Pleurotus species by analysis of doublestranded ribonucleic acid (ds RNA). Ds RNA segments were extracted from virus infected isolates which grew abnormally. But virus free isolates didn't show any ds RNA segments. The ds RNA was consisted of one large segment of 8100 base pairs (bp) and 4 smaller segments with 2170, 2120, 1980 and 1984 bp. Whereas, cell free virus particles showed only one larger ds RNA segment. The ds RNA was dissolved by RNase A in low salt, 0.1 M SSC and melted at $85^{\circ}C$. It was possible to use the ds RNA analysis for detecting virus infection directly from the host cells.

Effect of gamma irradiation on post-harvest quality of king oyster mushrooms (Pleurotus eryngii )

  • The consumption of king oyster mushrooms has steadily increased owing to their unique flavor and delicate texture. This study extended the storage period of king oyster mushrooms (Pleurotus eryngii ) via gamma irradiation. Irradiated samples (0, 0.5, 1, 2, and 3 kGy) were stored at 4℃ with 80% relative humidity for up to 28 days, and the experiments were conducted every 7 day. Microbiological analysis revealed a dose-dependent reduction in total aerobic bacteria, Pseudomonas spp., and yeasts and molds, and gamma irradiation above 2 kGy effectively controlled microbial contamination for up to 28 days. At the 28th day of storage, the irradiated king oyster mushrooms exhibited delayed browning through the reduction of tyrosinase activity. Moreover, firmness reduction (%) was 80.59±1.89% for the non-irradiated group and 42.80±1.28, 34.57±1.13, 31.05±3.24, and 39.73±0.94% for the irradiated group (0.5, 1, 2, and 3 kGy), respectively. These results were supported by the scanning electron microscopy photos, which showed smaller pores in the irradiated group than in the non-irradiated group. This study demonstrated that 2 kGy of gamma irradiation effectively reduces microbial contamination and delays the browning and softening of king oyster mushrooms for up to 28 days.
