아위버섯의 성분에 관한 연구 (Stuy on the Composition of Pleurotus ferulae Fruit Body)

  • 아위버섯을 약용과 식용으로 이용하고자 일반성분, 무기질, 구성아미노산, 유리 아미노산, 비타민 등을 분석검토한 결과 조지방이 8.0 g%, 탄수화물이 54.3 g%, 조단백질이 20.2 g%이었다. 무기물은 K가 2,337.0 mg%으로 가장 많이 함유 되어있고, Mg, Na, Zn, Ca, Fe의 순으로 함유되었다. 구성 아미노산은 glutamic acid가 13.65 %mol로 가장 많이 함유되어있고, aspartic acid, glysine, alanine, leucine, arginine, valine, lysine등의 순으로 17종의 아미노산을 함유하고 있으며 필수아미노산은 37,78 %mol이었다. 유리아미노산은 alanine이 547.97 $\mu\textrm{g}$%으로 가장 많이 함유되어 있고, phenylalanine, tyrosine, leucine 등의 순이며, 총 함유량은 2,737.38 $\mu\textrm{g}$%이고, 21종을 함유하고 있기 때문에 아위버섯의 맛이 좋을 것이라 생각되어 진다. 비타민의 경우 vitamin A가 0.12 mg%, vitamin B$_1$은 0.31 mg%, vitamin B$_2$는 0.68 mg%, vitamin C는 7.99 mg%, vitamin E는 316.88 mg%, vitamin D$_3$는 0.29 mg%을 함유하였고, 비타민의 총 함유량은 326.27 mg%이었다. 특히 vitamin C와 vitamin E가 많이 함유되어 노화방지에 좋으리라 생각된다.

몇가지 저장온도와 비천공 Breathable 필름이 아위버섯(Pleurotus ferulae) 저장수명과 품질에 미치는 영향 (Effect of Non-perforated Breathable Films on the Shelf Life and Quality of Ferulae Mushroom (Pleurotus ferulae) during MA Storage at Different Temperatures)

  • 본 실험은 다양한 저장온도 조건에서 아위버섯에 적합한 MAP용 포장재 구명을 위해 수행하였다. 저장중 생체중 감소는 저장온도가 낮을수록 적었는데, 모든 온도 처리(1, 8, $24^{\circ}C$)에서 저장 종료일까지 1.5% 이하로 낮았다. 1저장에서는 산소투과율이 $1,300cc/m^2{\cdot}day{\cdot}atm$ 인 필름 처리구가 가장 큰 저장수명(42일)을 보였으며 저장중 포장내 산소/이산화탄소의 대기조성 조건도 가장 적합한 MA조건인 5%/15% 수준이었으나, 높은 에틸렌 농도와 이취를 보인 반면 3,000cc 처리구는 1,300cc 처리구와 비슷한 대기조성 조건을 갖추고 낮은 에틸렌 농도과 이취를 보였다. 따라서 $1^{\circ}C$에서는 1,300cc와 3,000cc 필름이 적합하였다. 저장중 대기조성이 필름처리간 차이가 없었던 $8^{\circ}C$ 저장에서는 에틸렌 농도가 두 번째로 낮았고 외관상 품질이 가장 높아 저장수명이 19일로 가장 길었던 3,000cc 필름이 적합하였다. 저장 온도가 높아 호흡이 급격히 빨라져 극도의 CA 조건이 만들어졌던 $25^{\circ}C$에서는 외관상 품질에서 가장 양호하였던 1,300cc 필름으로 적합하였다. 또한 온도별 저장수명이 $25^{\circ}C$에 비해 $8^{\circ}C$는 4배, $1^{\circ}C$는 9배나 연장되어 수확 후 저온유통이 반드시 필요하다고 판단되었다.

Optimization of Culture Media for Solid-state Culture of Pleurotus ferulae

  • In order to elucidate the possibility of artificial production of p. ferulae by solid-state culture, the optimization of culture conditions was carried out. When $NH_4H_2PO_4$ and $CaCO_3$ were used in the cultures using test tube with 30 g of Populus sawdust at $25^{\circ}C{\pm}1$ in the dark, the favored mycelial growth was observed with $1\%$ of $NH_4H_2PO_4$ and the production of polysaccharide was 7.85 mg/100 mg of mycelium with $1\%$ of $CaCO_3$. The mixtures of $80\%$ of Populus Sawdust and $20\%$ of rice bran at $60\%$ of water content were determined to be optimal for the production of fruiting bodies in the sawdust culture. When three treatments containing various ratios of garlic powder were conducted, yields of fruiting bodies were drasti[ally higher than those of Synthetic mixture without garlic powder The highest yield (143 g/bag) was obtained with $7\%$ garlic powder. The yield of synthetic mixture containing $7\%$ of garlic powder was $83\%$ higher than that of Sawdust culture. The reason why garlic powder did support growth was not clear but it is possible that garlic powder might contain effective components for the formation of fruiting body. The optimal synthetic mixture composition consisted of cotton seed $77\%$, lime $6.4\%,\;K_2HPO_4\;0.2\%,\;KH_2PO_4\;0.2\%,\;CaHPO_4\;0.2\%$, corn flour $4\%$, wheat flour $5\%$, and garlic pow-der $7\%$.

Effect of Pleurotus ferulae Extracts on Viability of Human Lung Cancer and Cervical Cancer Cell Lines

  • When SiHa cells were incubated for varying periods of time with extracts of PFF and PFM, the cytotoxicity of the ethanol extracts of PFF was higher than those of the other extracts. These results indicated that the extracts from fruiting bodies of p. ferulae contain antitumor Substances. When A549, SiHa and HeLa cells were incubated with different concentrations of PFF and PFM extracts, the ethanol extracts of PFF showed strong cytotoxicity against A549 tells at concentrations over $10{\mu}g/mL$ and against SiHa and HeLa cells at concentrations over $40{\mu}g/mL$. However, the differences in the cytotoxic effects of the hot water and ethanol extracts of PFM and the hot water extracts of PFF on all 3 cancer cells were not significant. Also, the PFF ethanol extracts induced synergistic effects on the TRAIL-induced apoptosis in A549 cells, which were strongly resistant to TRAIL. These results indicated that ethanol extracts of PFF were the most prominent antitumor agents toward lung cancer cells (A549).

Exo-Polysaccharide Production in Liquid Culture of Pleurotus ferulae

  • Batch cultures were carried out to optimize the exo-polysaccharide production by liquid cultures of Pleurotus ferulae. Among the various carbon sources, when $5\%$ of glucose was used, the maximum mycelial growth and exo-polysaccharide concentration reached were 8.78 g/l and 3.59 g/l, respectively. Yeast extract and polypeptone were identified as the most suitable nitrogen sources. In particular, when a mixture of $1\%$ of polypeptone and $0.8\%$ of yeast extract was used, 9.52 g/l of mycelial growth and 4.09 g/l of exo-polysaccharide were obtained. In the case of mineral sources, K$_2$HPO$_4$ and MgSO$_4$$\codt$7H$_2$0 were found to be the best mineral sources for mycelial growth and exo-polysaccharide production. Under the optimized culture conditions, the agitation speed and aeration were investigated for mycelial growth and exo­polysaccharide production in a jar fermentor. The maximum mycelial growth and exo-polysaccharide concentration at 1.5 vvm and 200 rpm obtained were 13.2 g/l and 4.95 g/l, respectively, after 10 days of culture, which were $76\%$ and $79\%$ higher than those of the basal medium. The specific growth rate was decreased with the increase of mycelial growth. However, the specific production rate of the exo-polysaccharide was proportionally increased with the specific growth rate. The proposed model profiles showed good agreement with the experimental results for the mycelial growth and exo-polysaccharide production. The specific production rate using the optimized medium was higher than that of basal medium.
