Characteristics of Culture of Oyster Mushroom (Pleurotus ostreatus) on Addition Rate of Acer tegmentosum (산겨릅나무(Acer tegmentosum) 톱밥의 첨가량에 따른 느타리버섯의 재배적 특성)

  • This study was carried out to investigated optimum mixing ratio of Korean natural Acer tegmentosum for production of functional oyster mushroom. Total nitrogen and carbon sources of Acer tegmentosum were 0.2% and 44.4%, respectively and C/N ratio was 234. Total nitrogen source and pH of substrate mixed with Acer tegmentosum were 2.7~2.9 and 4.8~5.0, respectively. The contents of $P_2O_5$, $K_2O$ and MgO at Acer tegmentosum media were higher than that of the control. Mycerial growth was the fastest at Acer tegmentosum 10%, and slower by increase of Acer tegmentosum substrate. Yields of fruiting body was the highest to 159 g/850 mL at 10% of Acer tegmentosum and dimeter and thick of pileus were the highest, too. The L value of pileus and stipes were decreased by increase of Acer tegmentosum substrate, but there was no significant difference in the a-value and the b-value. The contents of $P_2O_5$ and $K_2O$ of fruiting body were increased at Acer tegmentosum substrate, but there was no significant difference in contents of CaO, MgO and $Na_2O$.

A Short Composting Method by the Single Phase Composter for the Production of Oyster Mushroom (느타리버섯 배지 제조기를 이용한 배지의 제조 연구)

  • A single phase composter was constructed by modifying the conventional mixer of sawdust for the cultivation of oyster mushroom Pleurotus ostreatus. The machine was designed on the basis of 3-phase-1 system which was controlled in prewetting, pasteurization and fermentation processes. In composting 200 kg of straw and cotton waste in the machine, it took 20 minutes in prewetting step and also to hours at $65^{\circ}C$ in pasteurization process. Postfermentation by aerothermophiles was completed by treating the compost at $45^{\circ}C-50^{\circ}C$ for 48 hours which was shorten 24 hours from the conventional method. In the postfermentation at high temperature, forced aeration and/or vigorous mixing process(es) played a great role in the improvement of spawn quality. The growth of mycelium of oyster mushroom was excellent in the culture combinated with 3 parts of surface inoculation and 7 parts of mechanical mixing.

Development of Minimal Processing Technology for Korean Fruit and Vegetables (과실 및 채소의 신선편의 식품화 개발기술에 관한 연구)

  • The purpose of this study was to investigate the effectiveness of various quality preservative treatments for extending shelf life and maintaining good quality of minimally processed fruit and vegetables produced in Korea. To determine the suitable treatments for delaying quality deterioration, fresh Asian pears and Chinese cabbages were sliced and treated with various quality preservatives (1% CaCl$_2$, 1% NaCl, 3% sucrose, 1% Ca-lactate, 1% vitamin C, 0.05% chitosan +1% vitamin C, 0.1% Sporix+1% vitamin C, hot water (60$\^{C}$), 0.2% L-cysteine), packed with polyethylene film (60㎛-thick), and stored at 4$\^{C}$/0$\^{C}$ or 20$\^{C}$. Various biological and sensory tests were performed to evaluate the quality changes in minimally processed products. Results indicated that Chinese cabbages treated with 1% CaCl$_2$ at 4, and 1% CaCl$_2$ and 1% NaCl at 20$\^{C}$ were most effective in maintaining the quality and minimizing the biochemical changes during storage. For sliced Asian pears, 0.2% L-cysteine and 1% NaCl treatments were effective to reduce browning, and 1% CaCl$_2$ treatment was the most effective to prevent softening during storage at 20$\^{C}$ and 0$\^{C}$. Modified atmosphere packaging of Pleurotus ostreatus and Lentinus edodes had a significantly different shelf life depending on packaging material, packaging thickness and storage temperature. Sealed packaging with polyethylene film (60㎛-thick) for two kinds of mushrooms maintained a good quality with an extended shelf life by 30-50% at 20$\^{C}$ and by 30-130% at 0$\^{C}$. To minimize the quality deterioration which appeared in the condition of polyethylene film packaging, quality preservatives such as KMnO$_4$ and KHSO$_2$+K$_2$S$_2$O$\_$5/ for SO$_2$ generation were added inside of mushroom packaging. The best condition for maintaining good quality longer was packaging with polyethylene film+SO$_2$ which showed 5080% extended shelf life for both Pleurotus ostreatus and Lentinus edodes at 20$\^{C}$ and 0$\^{C}$.

Physiological Activities of Extract from Edible Mushrooms (주요 식용버섯 추출물의 생리활성 효과)

  • This study was conducted to investigate the physiological activity of extracts of fresh mushrooms. The components were extracted by hot water; subsequently, the hot-water extract was subjected to 60% ethanol precipitation to yield high-molecular-weight (HMW) and low-molecular-weight (LMW) fractions. Total polyphenol contents, $\beta$-glucan contents, electron-donating ability (EDA), superoxide dismutase (SOD)-like activity, nitrite-scavenging activity, fibrinolytic activity, nitric oxide (NO) production, and inhibition of NO production of the mushroom extracts were measured using lipopolysaccharide (LPS)-stimulated murine macrophages, RAW 264.7 cells. The extracts of Lentinus edodes (Berk.) Singer and Pleurotus ostreatus (Fr.) Kummer contained the highest levels of $\beta$-glucan (33.5% and 25.57%, respectively). Further, the LMW fractions of the Phellinus linteus contained the highest levels of polyphenols (233.23 mg/g). The EDA of LMW fractions (10 mg/mL) of the Phellinus linteus and Agaricus bisporus were 80.74% and 51.35%, respectively. Further, SOD-like activities of the LMW fractions were high as compared to those of the HMW fractions. Nitrite-scavenging activities of the LMW fractions (pH 1.2; concentration, 10 mg/mL) of the Phellinus linteus and Pleurotus ostreatus (Fr.) Kummer were 75.95% and 41.05%, respectively. The fibrinolytic activity of the LMW fractions of all mushrooms showed no enzyme activity by fibrin plate assay. The fibrinolytic activity of the extracts of Tricholoma matsutake was the greatest inhibitory activity at 60.4%. Further study revealed that the mushroom extracts exhibited anti-inflammatory effects on RAW 264.7 cells. The LMW fraction ($500\;{\mu}g/mL$) of the Phellinus linteus considerably inhibited NO production (100%).

Quality Changes in Oyster Mushrooms during Modified Atmosphere Storage as Affected by Temperatures and Packaging Materials (저장 온도와 포장재에 따른 느타리버섯의 MA 저장 중 품질변화)

  • Modified atmosphere packaging was applied to oyster mushrooms (Pleurotus ostreatus) to study the effect of storage temperatures and packaging materialso. Whole mushrooms (200g) were package with polyethylene film $(PE,\;60{\mu}m\;thickness)$, ethylene vinyl acetate (EVA), or ceramic film (containing 5% zeolite) and stored at 0, 5, 10 and $20^{\circ}C$. Weight loss, color, firmness, gas composition $(O_2,\;CO_2)$ inside the film package and ethanol content in the tissue of MA packaged mushrooms were examined. Mushroom that were packed unwrapped in a conventional hardboard box (2 kg) lost marketability at a very early stage of storage due to weight loss, shrinkage, browning, and spore formation. During storage, film packaging prevented or retarded the deterioration of the mushrooms in the aspects of appearance, texture, and discoloration. Firmness slightly decreased with storage time. Total color difference was much higher in the control than in the film-packaged mushroom and rapidly increased at the early of storage. Correlation analysis showed a high correlation between total color difference and b values. These results were characterized by the reduced respiration rate resulting from elevated carbon dioxide and reduced oxygen levels in the package. At all storage temperatures, ethanol content in the tissue increased slightly at the early part of storage and rose considerably towards the end of the storage period. Ethanol content in the oyster mushrooms was higher in the stipe than in pileus tissues. The shelf life of the oyster mushrooms was about $8{\sim}11$ days at $0^{\circ}C$, about $4{\sim}6$ day at $5^{\circ}C$, about $2{\sim}3$ days at $10^{\circ}C$, and about $1{\sim}2$ days at $20^{\circ}C$.

  • This study was carried out to investigated optimum mixing ratio of Korean natural Juglans mandshurica using as functional plants. Total nitrogen and carbon source of J. mandshurica was 0.21% and 46.0%, respectively and C/N ratio was 219. Total nitrogen source and pH of substrate mixed with J. mandshurica was 2.3~2.9 and 5.0, respectively. The contents of CaO, MgO and $Na_2O$ at J. mandshurica media were higher at harvest media than inoculation media. Mycelial growth was the fastest at J. mandshurica 20%, and slower by increase of J. mandshurica substrate. Yields of fruiting body show the highest to 163 g/850 mL of medium which are addition 10% of J. mandshurica sawdust, and diameter and thick of pileus were the highest, too. The L value of pileus and stipes were increased by increase of J. mandshurica substrate, but there was no significant difference in the a-value and the b-value. The contents of $P_2O_5$ and $K_2O$ of fruiting body were increased at J. mandshurica substrate, but there was no significant difference in contents of CaO, MgO and $Na_2O$. The contents of Cu of fruiting body was decreased by increase of J. mandshurica substrate, but Fe, Mn and Zn were increased.

Changes in the characteristics of the fruiting body of oyster mushroom (Pleurotus ostreatus) according to the nitrogen content in sawdust media (느타리버섯 톱밥배지 질소함량에 따른 자실체 특성변화)

  • This study was conducted to elucidate the optimal nitrogen content for each variety of oyster mushroom (Pleurotus ostreatus) by investigating the change in the characteristics of fruiting body according to the nitrogen content of the sawdust media, and to use the results as the basic data for cultivation of high quality oyster mushroom. The nitrogen content was adjusted to 1.3%, 1.6%, 1.8% and 2.3%, and at each condition, mycelial growth was observed, which showed that the nitrogen content did not affect the rate of mycelial growth and density substantially. In the investigation of the characteristics of fruiting body, at the 1.8% nitrogen content, Chunchu No.2 showed the pileus diameter of 27 mm, and the stipe diameter and length of 11 mm and 61 mm, respectively, and the measurement of its color values revealed that the brightness of the pileus and the stipe was 31 and 80, respectively, offering the best commercial value, and yielding 25 valid stipes, which was the largest number. Likewise, at the 1.8% nitrogen content, Heuktari also showed the excellent growth characteristics including the pileus diameter of 29 mm and the stipe diameter and length of 10 mm and 68 mm, respectively, and the superb color values of the brightness of 37 at the pileus and 78 at the stipe. Moreover, the number of valid stipes was also found to be the highest with 19. In terms of morphological characteristics such as the pileus diameter and color, Soltari also produced the best mushrooms at the 1.8% nitrogen content and the number of valid stipes was high with 22. In summary, irrespective of varieties of oyster mushroom, high quality products could be cultivated at the nitrogen content of 1.8%.

Effect of Temperature on the Development of Sciarid fly, Bradysia sp. (Diptera: Sciaridae) (검정날개버섯파리류 1종 Bradysia sp. 의 생육에 미치는 온도의 영향)

  • This study was conducted to investigate the effect of temperature on the development of amushroom-infesting sciarid fly, Bradysia sp. (Diptera: Sciaridae). Egg period was 12.1, 7.0, 4.4, 3.4, and3.2 days, larval period was 38.3, 26.5, 13.4, 13.2, 12.7 days and pupal period was 10.4, 7.1, 4.4, 3.3, 3.2days, and total development period from egg to adult emergence was 60.8,40.6, 22.2, 19.9, and 19.1 daysat 10, 15, 20,25, 28"C, respectively. Development threshold temperature (DT) and effective accumulativetemperatures (ET) were 3.8"C, 74.8DD in eggs, 1.2"C, 321.8DD in larva, and 3.1$^{\circ}$C, 76.5DD in pupa,respectively. The number of eggs laid per female was 107.9, 129.7, 131.8, 86.9, and 82.7 at respectivetemperatures. Preoviposition period was 6.6, 4.4, 2.2, 1.3, 1.8 days, oviposition period 1.5, 1.5, 1.1, 1.1,1.1 days, postoviposition period 2.0, 1.1, 0.9, 0.6, and 0.3 days at th'e temperature of 10, 15, 20, 25 and 28"C, respectively. The longevity of male and female at the temperature was 13.3, 7.8, 5.9,4.1, 3.4 days and10.4, 7.0, 4.2, 3.0, 3.3days, respectively. The optimum temperature for hatchability was estimated to the20$^{\circ}$C and adult emergence was highest at 20$^{\circ}$C. Pupation rate was 50.7, 68.4, 84.3, 86.5, 45.4% at 10, 15,20, 25, and 28"C, respectively. at 10, 15,20, 25, and 28"C, respectively.tively.

Mushroom Production Technology with Crop Residues (농산부산물(農産副産物)을 이용(利用)한 식용(食用)버섯 재배(栽培))

  • The results of series studies on the ratio of supplements, out-door composting and out-door fermentation induced by using the rice straw as a main substrates at the cultivation of Agaricus bisporus, and the cultivation of Pleurotus ostreatus using the rice straw bundles on its compost are as follows; When rice straw for cultivation of A. bisporus was used as the main substrates in synthetic compost as a carbon source, yields were remarkably high. Fermentation was more rapid than that of barley straw or wheat straw and the total nitrogen content was high in rice straw compost. Use of barley straw compost for cultivation of A. bisporus was shown of low yield compared with rice straw, but when a 50% barley straw and 50% rice straw mixture was used, the yield was almost the same as that using only rice straw. The total organic nitrogen on the compost were shown the positive relation to the yield of A. bisporus, but the ammonium nitrogen negative relation to the mycelial growth and yield of A. bisporus. When rice straw was used as the main substrate for compost media, urea was the most suitable source of nitrogen. Poor results were obtained with calcium cyanamide and ammonium sulfate. When urea was applied three separate times, nitrogen loss during composting was decreased and the total nitrogen content of compost was increased. The supplementation of organic nutrient activated compost fermentation and increased yield of A. bisporus. The best sources of organic nutrients selected were as follows: perilla meal, sesame meal, wheat bran and poultry manure, etc. Soybean meal, tobacco powder and glutamic acid fermentation byproducts which were industrial wastes, could be substituted for perilla meal, sesame meal and wheat bran as organic nutrient sources for compost media. During out door composing of rice straw for cultivation of A. bisporus, using of tuner, composter and tunnel system increased up to 13% of its yield, and also cut down 34% of production Cost. The cultivation of P. ostreatus and utilizing of rice straw and wheat straw was established and its yield was high on the rice straw pots. When the substrates 'Rice straw' was heated by steam at $60^{\circ}C$ for 6 hr. mycelial growth of P. ostreatus was moderately rapid and its yield was high.

Effects of NaCl Concentration on Mycelial Growth and Fruiting Body Yields of Oyster Mushrooms, Pleurotus spp. (NaCl의 농도가 병재배 느타리버섯 균사생장 및 자실체수량에 미치는 영향)

  • These studies were investigated for effect of concentration of sodium chloride on occurrence and growth of fruitbody in oyster mushroom, Pleurotus ostreatus. When the mycelia were inoculated and cultured on the PDA plate added with the different concentrations of sodium chloride, the growth of them were not affected at the concentration of 0.5%, started to decrease at 1.0% as compared with the PDA plate without sodium chloride, and they did not grow at 5.0%. When tested at the column filled with sawdust, the tendency was similar, but the decreasing rate of mycelial growth was gentle. In sawdust bottle cultivation sprinkled with the different concentration of sodium chloride solution at the different stage, the productivity and quality of fruitbody started to decrease at 3.0% of the solution and the yield at the treatment of 10.0% solution was only 47% of that in non-treated bottles. Treated at the different stages of mushroom development, treatment just after mushroom sprout did not show any different from that just after scraping spawn from the complete grown bottles. The sprinkle treatment at this stage looked better than that of the soaking with the solution before mushroom sprout. The yield at this treatment with 10% solution showed little damage with that of 90% of non-treated bottle. Sprinkle treatment during mushroom growth stage did not affect and showed almost same to non-treated bottle except the treatment with 10.0% solution. The treatment of this stage showed better results of growing characters than the treatment after and before mushroom sprouting even 10% solution. Morphological characteristics of oyster mushroom do not distinguish to treatment of different stage and concentration of sodium chloride solution. The analyzing data of quality on underwater usually used for oyster mushroom in Inchon, Yeongjong and Kanghwa area showed over-concentration to the standard for edible water and agricultural water. But it was not confirmed whether this water quality could affect to mushroom cultivation.
