  • Some properties of xylanase produced by Pleurotus ostreatus during its gorwth in a rice straw medium were investigated. The results are summarized as follows : 1) The optimum pH of xylanase was 5.0 and the stable pH ranged from 4.5 to 6.0. 2) The optimum temperature for the xylanse was around $50^{\circ}C$ and the xylanse activity was completely lost in 10 minutes at $70^{\circ}C$ 3) The activity of xylanse was inhibited by managanous ion, but was increased by other metalic ions. Especially K, Mg and Ca ions considerably increased the activity.

  • The influences of culture parameters on the decolorization of anthron-type dye, Remazol brilliant blue R(RBBR) by Pleurotus ostreatus were studied in defined media. In the decolorization, 1-10 mM nutrient nitrogen and 40 mM glucose were effective whereas agitation and Tween 80 were not suitable. The decolorization occurred and the activity of extracellular peroxidase was detected during the stationary phase.

  • We found that the cultivated oyster mushroom, Pleurotus ostreatus, was damaged by the phorid fly, Megaselia tamiladuensis Disney (Diptera: Phoridae). This fly is newly recorded as a pest in Korea. Brief morphological characteristics are described.

  • Chemical analysis of both sclerotia and the fruits of Pleurotus tuber-regium showed higher values for such elements as calcium(Ca), iron(Fe), zine(Zn) in the fruits than in the sclerotia. On the contrary magnessium(Mg) was found to be higher in sclerotia than in the fruits. protein and carbohydrate were also found to be more in the fruits. There was no significant difference between the chemical values of old(1 year) and fresh sclerotia. Oil palm fruit fibre substrate produced sporophores with higher values for the minerals, protein and carbohydrate than those on riversand substrate.

  • Cultural conditions and carbon sources affecting the productivity of cellulolytic enzymes of Pleurotus ostreatus 201 were examined in synthetic media. The optimum cultural temperature and initial pH for the production of enzymes were $25^{\circ}C$ and 5.5 in avicelase, and 3$0^{\circ}C$ and 5.0 In CMCase, and 3$0^{\circ}C$ and 6.5 in B-glucosidase. Among carbon sources, cellulose powder was the best for the production of avicelase, and Na-CMC for CMCase, and cellobiose for $\beta$-glucosidase. The optimum concentration of cellulose powder was 1.0% (w/v), and glucose depressed the production of enzymes remarkably.

  • Perforated vinyl mulching technique was performed on Pleurotus sajor-caju beds to assess fruitbody formation. Individual fruitbody of P. sajor-caju was transformed into bunch type on vinyl mulching bed. It was effective to grow the mushroom without waterlogging and abortion of small pins on the beds as well as hygienical bed management. A bunch showed 79 fruitbodies and 225 g of weight. Available site for fruiting was reduced up to 20% in comparison of 100% for conventional bed. The color of fruitbody turned on brownish white from treated vinyl mulching bed.

  • Isozyme comparisons of mycelial extracts from Pleurotus ostreatus were undertaken using isoelectric focusing. Enzyme isozyme patterns were Used to describe the extent of geographical diversity and degree of intraspecific variation in these extracts. A total of 77 bands were resolved from six different enzymes. Cluster analyses were performed using the zymograms for esterase (EST), glucose phosphate isomerase (GPI), leucine aminopeptidase (LAP), malate dehydrogenase(MDH), peroxidase (POX), and phosphoglucomutase (pGM). EST gave multiple banding patterns, while less variability was observed for GPI, MDH, and PGM. Cluster analyses demonstrated that strains of P. ostreatus from geographically different origins are genetically divergent, supporting the idea that there is little or no gene flow between these geographically distant population groups.

  • Extracellular laccase secreted from Pleurotus ostreatus was activated by $Cu^{2+}$ and $Cu^{+}$ . The enzyme was strongly inactivated by 8-hydroxyquinoline, potassium cyanide, sodium azide, sodium bisulfite and 2-mercaptoethanol. The two ionogenic groups, which have pKa values of 5.60-5.70 and 6.70-6.85 respectively, were found to relate with the active site of this enzyme. The oxidation reactions were brought about by initial single electron transfer process on the active site. The enzyme was found to be a metalloprotein which had about 3.9 cupric ions per molecule of protein as a prosthetic group. The enzyme showed a strong peak at 605 nm and a weak shoulder at 330 nm in UV-Visible absorption spectrum. Both signals disappeated upon treatment of the enzyme with 4 electron equivalent ascorbate. These results indicate that type I Cu peak and type III Cu shoulder are present in laccase.
