Antimicrobial Activity of the Essential Oils of Artemsia Princeps var. orientalis (쑥으로부터 추출한 정유의 항균효과)

  • The essential oils of Aritemisia princeps var. orientalis (wonnwood) were tested against the standard cultures &cherichia coli, Bacillus subtilis, Pleurotus oststreatus. Fusarium soiani, Aspergillus niduians. Escherichia coli was not susceptible to the wonnwood essential oil but the growth of Bacillus subtilis, Aspergillus niduians, Fusarium solani and Pleurotus ostreatus was severely inhibited by essential oil. The growth of Bacillus subtilis in 1O~100 ppm was a tenth of the control. The wonnwood essential oil also exhibited strong inhibited of the growth of tested fungi. The growth of Pleurotus ostreatus was fully stopped at 1,000 ppm concentration.

Sensory and Physicochemical Characteristics of Jeungpyun with Pleurotus eryngii Powder (새송이 버섯을 첨가한 증편의 관능적, 물리화학적 품질 특성 변화)

  • This study investigated possible improvements in the sensory and physicochemical characteristics of jeungpyun by the addition of Pleurotus eryngii powder. The volume of the jeungpyun batter prepared with the 1% Pleurotus eryngii powder showed the highest degree of fermentation. As fermentation continued, pH and saccharinity dropped, and then saccharinity increased as more Pleurotus eryngii powder was added. As the Pleurotus eryngii powder ratio increased, the L-value decreased, but the a- and b-values increased. In the texture profile analysis, the 5% Pleurotus eryngii powder showed the highest adhesiveness and the lowest springiness. For the sensory test, the highest springiness and overall desirability were found in the jeungpyun with 1% Pleurotus eryngii powder. Overall, according to the results of these experiments, the 1% and 3% Pleurotus eryngii powders are though to be the optimal concentrations for obtaining improvements in quality.

  • This study conducted an experiment to develop new raw ingredients for natural cosmetics by evaluating the efficacy of cosmetic products containing the extracts of Pleurotus eringii mushrooms grown without agricultural chemicals. To this end, purchased were Pleurotus eringii mushrooms grown without agricultural chemicals in South Gyeongsang province, South Korea in 2016; extraction was conductedwith 80% EeOH at room temperature after dry ing them. The study identified the excellent efficacy of the extracts after evaluating the antioxidant activity using the DPPH assay and ROS assay. According to the evaluation of he categories of moisture, oil, pH and pigment after the application of a cream-type facial mask pack containing 5% of the extracts, there were significant differences in the categories of moisture and oil on the T-zone area, and no statistically significant differences in the categories of oil change on the U-zone, pH and pigment. However as the extracts of Pleurotus eringii mushrooms had excellent efficacy in all categories compared to the control group, the efficacy of the extracts as a cosmetic raw ingredient could be identified. It is expected that this study may further contribute to developing more advanced raw ingredients, given the extract and cultivation conditions by concentration.

Genetic Relationship of Pleurotus ferulae Strains (아위버섯(Pleurotus ferulae) 균주의 유전적 유연관계)

  • This study was carried out to investigate the genetic relationship of Pleurotus ferulae, an edible mushroom found on a medicinal plant, Ferula assa-foetida, in central China. The genetic relationships of 15 Pleurotus species strains, including five P. ferulae strains were analyzed. The strains were divided into seven groups at 80% genetic similarity level according to random amplified polymorphic DNA (RAPD) analysis. Four out of the seven groups consisted of two to four strains, while the other three groups consisted of three strains. In each of the three groups, the three strains were from each of three different Pleurotus species (P. cornucopiae, P. florida and P. sajorcaju). Other strains grouped together for genetic similarity were P. eryngii 26060 and P. fuscus var. ferulae 26065, three strains of P. ostreatus, and four P. ferulae strains (Bakdal, Awi, Cheonsan 1, and Yesan). However, Japanese Seolyi which belongs to P. ferulae and Heukpyung which belongs to P. ostreatus were together in a separate group.

Culture Condition for Biomass of Pleurotus eryngii (새송이버섯 biomass를 위한 최적배양 조건)

  • For the purpose of application for biomass of Pleurotus eryngii, the optimum culture condition were tested. It was found that the optimum culture condition for spot culture of pleurotus eryngii were 24$^{\circ}C$ for 18 days with PDA medium. And the optimum culture condition of bioreactor for biomass were pH 5.5, 18$^{\circ}C$ and 27 days with PDMP broth. It was possible to artificial cultivation of mycelial from Pleurotus eryngii using bioreactor for biomass under the optimum conditions, and it was also possible for Pleurotus eryngii biomass because the forming of fruiting body when Pleurotus eryngii was cultivated using mass artificial cultivated mycelial in the bioreactor.

Application of electric pulsed power on fruit body production of edible and medicinal mushrooms

  • Effect of pulsed power was investigated on fruit body formation of 10 edible mushrooms, Lentinula edodes, Glifola frondosa, Pholiota nameko, Flammulina velutipes, Hypsizygus marmoreus, Pleurotus ostreatus, Pleurotus eryngii, Pleurotus abalonus, Agrocybe cylindracea and Sparassis crispa. Pulsed power of 100-170 kV was directly charged to the substrate just before fructification. The effect of the pulsed power resulted to promote for 10 edible mushrooms fructification. The treatment especially stimulated the fructification on Pleurotus species.

The antibacterial effect of Pleurotus eryngii extracts on oral bacteria (새송이버섯 추출물이 구강세균에 작용하는 항균효과)

  • Objectives: Pleurotus eryngii is used both for edible and medicinal purposes, and has a physiological activity. The purpose of this study is to investigate the antibacterial effect of Pleurotus eryngii against six oral pathogens (Staphylococcus aureus, Streptococcus criceti, Streptococcus mutans, Streptococcus ratti, Streptococcus sobrinus, and Actinomyces viscosus). Methods: The antibacterial activities of various extracts of Pleurotus eryngii were examined by disc diffusion assay and minimum inhibitory concentration (MIC). The disc diffusion assay was performed by putting a paper disc soaked in extracts on plates inoculated bacterial cultures. The MIC of these extracts was determined by using a broth microdilution assay at a concentration ranging between 0.03 mg/ml to 15.00 mg/ml. The growth inhibition effect of extracts was measured at 600 nm for 24 hrs. Results: The antibacterial activity was confirmed against all six tested bacteria at Pleurotus eryngii ethyl acetate extract by the disc diffusion method. Acetone extract showed the antibacterial activity only against 4 strains containing Streptococcus criceti, Streptococcus mutans, Streptococcus ratti, and Actinomyces viscosus. In ethanol extract, no activity was observed against other strains except Staphylococcus aureus. MIC values of ethyl acetate extract were the same, 7.50 mg/ml in all tested bacteria. Conclusions: Pleurotus eryngii exhibited the antibacterial activity against oral pathogens (Staphylococcus aureus, Streptococcus criceti, Streptococcus mutans, Streptococcus ratti, Streptococcus sobrinus, and Actinomyces viscosus). Thus, Pleurotus eryngii may be considered as a natural antibacterial agent for treatment of dental diseases.

Sporocarp development of Pleurotus tuber-regium Fr. under different watering systems

  • Sporocarps of Pleurotus tuber-regium Fr. were subjected to aerial and substrate(soil) watering sytems. Although aerial watering enhanced quick enlargment of pilei, those watered indirectly through the soil developed more naturally pigmented sporophores and with higher overall yield. Thus, substrate(soil) watering after emergence is preferred to direct watering of fruits.

Production, Partial Purification and Physico-Chemical Characteristics of Laccase from Pleurotus ostreatus (Pleurotus ostreatus가 생산하는 Laccase의 부분정제 및 효소적 특성)

  • The production media, partial purification and the enzymatic characteristics of laccase from Pleurotus ostreatus were investigated. Among various media tried the double strength onion media showed much higher production of laccase compared to glucose and/or CMC added media. The laccase from Pleurotus ostreatus has the optimum pH of 5.94 for the activity and appears to be stable at relatively broad pH spectrum (4.7-8.7). The experiments on the temperature stability shows that above 90% activity could be preserved after holding at $30^{\circ}C$ for 40 min., 60% activity was remained after 40 min. at $50^{\circ}C$. The Km value of the laccase from Pleurotus ostreatus was estimated to be 3.209mM and the molecular weight of laccase was estimated to be 55,000 by SDS-Polyacrylamide gel electrophoresis.

Phylogenetic analysis of pleurotus species based on the nuclear SSU rRNA sequences

  • The internal regions of nuclear small subunit rRNA from 6 plaeurotus species and 5 Pleurotus ostreatus strains were amplified by PCR and sequenced. The DNA sequences of 8 Pleurotus strains (P. ostreatus NFFA2, NFFA4501, NFFA4001, KFFA4001, KFCC11635, P florida, P. florida, P. sajor-cuju, P. pulmonarius, and P. spodoleucus) were idential, but P. cornucopiae differed from them in two bases out of 605 bases. However, p[hylogenetic analysis of the sequences by DNA-distance matrix and UPGMA methods showed that P. ostreatus NFFA2m1 and NFFA2m2, known as mutants of P. ostreatus NFFA2, belonged to anther group of Basidiomycotina, which is close to the genus Auricularia. The difference of the SSU rDNA sequences of P. cornucopiae from other Pleurotus species tested corresponds to the difference of mitochondrial plasmid type present in Pleurotus species as observed by Kim et al. (1993, Korean J. Microbiol. 31, 141-147).
