The first case report on Ergasilus sieboldi Nordmann, 1832(Copepoda, Ergasilidae) infection in an aquacultured Shiures asotus in Korea (양식 메기에서의 Ergasilus sieboldi Nordmann, 1832 기생증 1례)

  • Ergasilus sieboldi, a parasitic Copepoda, was detected from the gill of Silurus asotus fingerling(7~10 cm long) cultured at a fish farm in Chungnam Province (Fig. 1 and 2). The infected fish did not show any particular external symptoms except a feeble swimming around the boarder of the pond. The isolated parasite was of 1.07 mm in body length with two egg sacks of 1.05 mm and one pair of hook-shaped secondary tentacle. Bychowsky(1963) described that this parasite infected the gill of Salmonidae, Serranidae, Cyprinidae and Siluridae and that the fish in the Baltic Sea, Black Sea. Caspian Sea. Japanese sea zones, Siberian rivers and Lake Bikal were infectd. Bychowsky further pointed out that heavily infected fish with this parasite become emaciated and slow in growth. and often die. As this parasite infected on the peri-branchial area. the infected fish showed a severe disturbance of respiration. Urawa et ai.(1980) studied on the development of Neoergasilus japonicus. an Ergasilidae, which infects Japanese freshwater fish. In Korea, Chun(1985) reported the identification of Ergasilus sieboldi from imported carp. and Neoergasilus and Pseudogasilus japonicus from bluegills. respectively. Chun also identified Pseudoergasilus zacconis in cultured ayu(Plecoglossus altivelis). However. this report deals with the detection of Ergasilus sieboldi from cultured catfish(Silurus asotus) for the first time in Korea.

Chronological observation on intestinal histopathology and intraepithelial Iymphocytes in the intestine of rats infected with Metagonimus yokogawai (요코가와흡충 감염 흰쥐에 있어서 장병변 및 장상피내 림프구 (IEL) 증감에 대한 경시적 관찰)

  • The relationship between the intestinal histopathology and number and position of' intraepithelial Iymphocytes(IEL) was observed chronologically in the small intestine of rats experimentally infected wiH Metagonimw vokogawci. Fifteen Sprague-Dawley rats were orally infected each with 3,000 metacercariae, and 3 were kept uninfected for controls. Three rats each were sacrificed on the day 5, 10, 15, 24 and 70 post-infection (PI) and samples of the small intestine, 5 cm, 10 cm, 20 cia and 70 cm posterior to the pylonls were taken. The samples were processed routinely and stained with Giemsa. The intestinal histopathology was severe during the day 5-15 PI and characterized by villous atrophy, crypt hyperplasia, and decrease of villus/ciypt height ratio. After the day 24 PI, the intestinal lesions showed some tendency of recovery The number of IEL increased at the early stage of infection, but decreased thereafter to a lower level than that of controls, with progression of the pathological changes. Then, the IEL number began to increase again after the day 24 PI. In control rats, the great majority of the IEL were located at the basal region of the epithelium. During the early stage of infection, however, a considerable proportion of IEL was found to have moved to the intermediate or apical region of the epithelium. From the above results, it is suggested that the change of IEL number and position during the course of M. yokogowoi infection should be closely related to the progression and recovery of the intestinal histopathology.

The Prevalence of Clonorchis sinensis and Its Related Factors at Goksung Area in the Basin of Sumjin River (섬진강유역(蟾津江流域) 곡성지역(谷城地域) 간흡충(肝吸蟲) 감염실태(感染實態) 및 관련요인(關聯要因))

  • A questionnaire survey including stool examination for the parasite ova, skin test, and enzyme-linked immunosorbent assay (ELISA) of the liver fluke to find the factors related with the prevalence of the parasite infestation was done toward 118 residents living in three villages of Song-jung, Yi-jung, and Chim-gok of Okok-myun, Goksung County, South Cholla Province. The results are as follows : 1. The prevalence of Clonorchis sinensis ova by the stool examination was 46.6%. The sensitivity and specificity of skin test to egg-positive cases were 63.5% and 68.9%. The sensitivity and specificity of ELISA to egg-positive clonorchiasis were 100% and 34.9%. 2. The prevalence of Clonchiasis was higher in male than in female. 45~64 year old group was the highest among age groups, and the longer the duration of living in the present residence was, the higher the prevalence was shown statistically significantly (p<0.01). 3. The present drinker had higher prevalence of Clonorchiasis significantly than the past drinker or non-drinker, and the more the frequency of drinking the higher the prevalence was shown significantly (p<0.05). 4. Those who took raw fish had significantly higher than persons who didn't take it or take cooked fish(p<0.05) and prevalence showed the tendency to be increased as the frequency to take raw fishes increased significantly(p<0.05). 5. Those who had taken a raw sweet fish(Plecoglossus altivelis) or snake-headed fish(Ophicephalus argus) showed significantly higher prevalence of infection with liver fluke than persons who didn't take the raw fish. 6. While 56.4% of persons infected with Clonorchiasis answered that they were healthy, only 13.0% of the subjects were careful for their health, and 41.8% experienced tests to diagnose liver flukes before they were examined by this survey. 7. While 56.4% of infected persons thought the prevalence was high in their community, 54.0% were afraid to be infected, 47.1% thought the infection might destroy their health, 25.5% took treatment drugs, and 18.2% took raw fish continuously after treatment. The above results confirmed that Goksung area was highly prevalent in Clonorchiasis, so it was necessary to push effective preventive program against liver fluke infection together with the program to diagnose and treat existing infected persons.

Proximate compositions of wild and cultured sweet smelt (Plecoglossus altivelis) muscles and eggs (천연 및 양식산 은어의 근육 및 난의 일반성분 조성)

  • Proximate compositions of muscles and eggs between wild sweet smelt which were caught in Seomjin river and Miryang river, and cultured sweet smelt which were purchased from certain cultured farms in Hadong and Miryang, were compared. Proximate compositions of wild sweet smelt muscles were $76.7\~77.5\%$ for moisture, $18.4\~19.9\%$ for protein, $2.41\~3.47\%$ for lipid, and $1.04\~1.33\%$ for ash content and there were almost no differences between the fishes from Seomjin and Miryang river. Protein and ash content in the cultured fish muscles were similar to those of the wild fish muscles, while the former contained much more lipid and carbohydrate, and less moisture than the latter, especially lipid content differed greatly according to their cultured farms. This might be the result of their different diet compositions. Lipid, protein and carbohydrate content in eggs of the wild and cultured fishes were much higher than those in their muscles. There was a negative correlation between moisture and lipid content in all samples; y= -0.2715x+24.903 (r= -0.7253, p<0.001).

Endemicity of Zoonotic Trematode Metacercariae in Fish from Deokcheon-gang (River) in Sancheong-gun, Gyeongsangnam-do, Republic of Korea

  • The endemicity of zoonotic trematode metacercariae (ZTM) was investigated with total 871 freshwater fishes (19 species) from Deokcheon-gang (a branch stream of Gyeongho-gang) in Sancheong-gun, Gyeongsangnam-do, Korea for 3 years (2018-2020). All fishes were examined with the artificial digestion method. The metacercariae of Clonorchis sinensis (CsMc) were detected in 233 (36.3%) out of 642 fish in 11 positive fish species (PFS), and their infection intensity was 27 per fish infected (PFI). Especially, in index fish, Puntungia herzi, of CsMc infection, prevalence was 64.2% and infection intensity was 37 PFI. Metagonimus spp. metacercariae (MsMc) were found in 760 (87.5%) out of 869 fish in 18 PFS and their infection intensity was 228 PFI. In sweet smelt, Plecoglossus altivelis, the prevalence of MsMc was 97.6% and their infection intensity was 3,570 PFI. Centrocestus armatus metacercariae were detected in 209 (29.4%) out of 710 fish in 8 PFS and their infection intensity was 1,361 PFI. Echinostoma spp. metacercariae were found in 293 (42.6%) out of 688 fish in 15 PFS and their infection intensity was 5 PFI. Metacercariae of Clinostomum complanatum and Metorchis orientalis were also detected in 2.7% and 21.2% fish in 4 PFS and their infection intensities were 3.1 and 3.4 PFI respectively. By the present study, it was confirmed that some species of ZTM including CsMc and MsMc are more or less prevalent in fishes from Deokcheon-gang in Sancheong-gun, Gyeongsangnam-do, Korea.

Antigenic localities in the tissued of Metagonimus yokogawai observed by immunogoldlabeling method (면역황금 표식법을 이용한 요꼬가와흡충의 조직내 항원성 부위에 관한 연구)

  • In order to determine the antigenic localization in the tissues of the adult Metagonimus yokegawai, immunogoldlabeling method was applied using serum immunoglobulins (IgG) of cats which were infected with isolated metacercariae from Plecoglossus altivelis. The sectioned worm tissue was embedded in Lowicryl HM 20 medium and stained with infected serum IgG and protein A gold complex (particle size: 12 nm) , It was observed by electron microscopy at each tissue of the worm. The gold particles were observed on the tegumental syncytium as well as cytoplasm of tegumental cells and epithelial lamella of the caecum. The gold particles were not observed on the basal lamina of the tegument, interstitial matrix of the parenchyma, the muscle tissue and mitochondria of the tegument. The gold particles were specifically labeled in the secretory granules in the vitelline cells. They were also labeled on the lumen of bladder and egg shell. The above findings showed that antigenic materials in the tissue of adult worms were specifically concentrated on the tegumental syncytium as well as cytoplasm of tegumental cells and epithelial lamella of the caecum.

Investigation of fish community structure and species diversity in two river estuary ecosystems, the Taehwa River and Changwon Stream, based on conventional survey and eDNA metabarcoding (어구조사 및 환경 DNA 메타바코딩을 이용한 태화강, 창원천 하구 생태계의 어류 군집 구조 및 종 다양성 평가)

  • River estuaries are dynamic and productive ecosystems with high regional biodiversity. Environmental DNA (eDNA) has become a useful approach to assessing biodiversity in aquatic ecosystems. This study was conducted to investigate fish community characteristics and species diversity in two river estuary ecosystems, the Taehwa River and Changwon Stream. We further compared conventional and eDNA metabarcoding analyses of the fish communities. The conventional survey was performed in May, July, and October 2022, while the eDNA analysis was conducted only in May. We observed various fish species with different life histories, including carp, goby, and marine fish. We also found that migratory fish, such as dace Tribolodon hakonensis, sweetfish Plecoglossus altivelis, and eel Auguilla japonica, occurred in the Taehwa River, suggesting high river connectivity. Marine fish species were predominant in the Changwon Stream, as this river is located close to the sea. The diversity indices showed that the Taehwa River generally had higher species richness, evenness, and diversity values than the Changwon Stream. A total of 9-19 species were detected in the conventional survey for the three sites, whereas 11-18 species were found from eDNA analysis. The findings indicate that the sensitivity of eDNA was similar to or higher than that of the conventional method. Our study findings suggest the efficiency and efficacy of eDNA-based fish community monitoring, although with some shortcomings in applying the genetic marker to Korean fish, including no clear-cut distinction for Korean endemic species and/or genetically closely related species groups.
