Evaluation of Antioxidant Activity of Platycodon grandiflorum (장생도라지 (Platycodon grandiflorum)의 항산화 활성 평가)

  • Platycodon grandiflorum, Doraji as Korean name, is one of the most widely used traditional oriental medicine for bronchial diseases and also used as a folk remedy for geriatric diseases and inflammatory diseases. In recent studies, it has been reported that some effect of P. grandiflorum is derived from its antioxidant activity, although there is still a lack of evidence to establish its oxy-radical scavenging activity. In this study, total oxy-radical scavenging capacity (TOSC) assay was used to evaluate antioxidant activity of total extracts (T-PG), polysaccharide fraction (Po-PG), and saponin fraction (Sa-PG) isolated from P. grandiflorum against peroxyl radicals and peroxynitrites. And MTT assay was taken to assess cyto-protective effects of T-PG, Po-PG and Sa-PG in H4IIE cells treated with hydrogen peroxide and tert-butylhydroperoxide. In the TOSC assay, Sa-PG showed strong oxy-radical scavenging capacity compared with T-PG and Po-PG. In cell-based assay, T-PG and Po-PG protected cells from oxidative stress, but Sa-PG did not protect cells because of cytotoxicity of Sa-PG. These results suggest that the saponin components of P. grandiflorum have relatively strong antioxidant capacity and cytotoxicity in rat hepatoma cells.

Apoptotic Cell Death of Human Lung Carcinoma A549 Cells by an Aqueous Extract from the Roots of Platycodon grandiflorum (길경 수용액 추출물에 의한 인체 폐암세포의 apoptosis 유발에 관한 연구)

  • Platycodi Radix, the root of Platycodon grandiflorum, commonly known as Doraji, is used as a traditional oriental medicine. Extracts from the roots of P grandiflorum have been reported to have wide ranging health benefits. In the present study, we investigated the effects of an aqueous extract from the roots of P. grandiflorum (AEPG) on the growth of human lung carcinoma A549 cells. Upon treatment with AEPG, a concentration-dependent inhibition of cell growth was observed and cells developed many of the hallmark features of apoptosis, including condensation of chromatin. Flow cytometry analysis confirmed that AEPG increased populations of apoptotic-sub Gl phase. Immunoblot and quantitative RT-PCR analyses indicated that the expressions of Bcl-2 was down-regulated but Bax was up-regulated in AEPG-treated A549 cells. The expression of active form of caspase-3 by AEPG treatment was markedly increased, and the levels of poly(ADP-ribose) polymerase (PARP) and $\beta$-catenin, its target proteins, were decreased in a concentration dependent manner. Taken together, these findings suggest that P. grandiflorum has strong potential for development as an agent for prevention against human lung cancer.

Platycodon grandiflorum Extracts Exhibits Anti-inflammatory Properties by Down-regulating MAPK Signaling Pathways Lipopolysaccharide-treated RAW264.7 Cells

  • Platycodon grandiflorum is a medicinal herb that is used to treat pulmonary and respiratory allergic disorders. The objective of this study was to investigate the protective effects of ethyl acetate extract of Platycodon grandiflorum (PGEA) against inflammation and to discern the molecular mechanism of PGEA in lipopolysaccharide (LPS)-induced signal pathways in RAW264.7 macrophage cells. PGEA suppressed the generation of nitric oxide (NO) and the expression of inducible NO synthase induced by LPS in RAW264.7 cells, and inhibited the release of pro-inflammatory cytokines induced by LPS in RAW264.7 cells. Western blot analysis showed that PGEA suppressed LPS-induced phosphorylation of p38 and c-Jun N-terminal kinase (JNK) but not extracellular signal-regulated kinase and $I{\kappa}-B{\alpha}$ degradation. Inactivation of JNK and p38 was effectively alleviated by PGEA, which subsequently affected the activation of c-Jun and c-Fos, which are the essential components of the activator protein-1 (AP-1) transcription complex. Taken together, the results indicate PGEA suppress the activation of p38, JNK, and AP-1, thereby inhibiting the generation of NO and pro-inflammatory cytokines, which affect the regulation of inflammation. PGEA may be useful for the treatment of various inflammatory diseases.

  • To investigate the antimutagenic effect of Platycodon grandiflorum DC, methanol extract of Platycodon grandiflorum DC was investigated. In Ames test, the methanol extract showed inhibitory effects of 80-90% on the mutagenicity induced by indirect mutagen of IQ(2-amino-3-methylimidazo[4,5-f]quinoline) and direct mutatgen of MNNG(N-methyl-N'-nitro-N-nitrosoguanidine) in Salmonella typhimurium TA 98 and TA 100. And then the methanol extract was further fractionated. Among the solvent extracted fractions from the methanol extract, the ethyl acetate fraction and butanol fraction exhibited the greatest antimutagenic effect suppressing the mutagenicity IQ and MNNG with inhibition rate of 99% and 98%.

Genetic Diversity and DNA Polymorphism in Platycodon grandiflorum DC. Collected from East-Asian Area

  • Broadening the genetic base of Platycodon grandiflorum DC. cultivar to sustain improvement requires assessment of genetic diversity available in P. grandiflorum DC.. The objective of this study was to analyze the genetic variation, genetic relationship among 48 samples collected from East-Asian Area by means of RAPD-PCR (random amplified polymorphic DNA-polymerase chain reaction) markers. From the 18 primers tested, produced total 211 bands with an average of 11.7 bands per primer and obtained 103 polymorphic band with an average of 5.7 bands per primer,s revealed relatively high percentage of polymorphic bands (48.8%). The genetic similarities calculated from RAPD data varied from 0.688 to 0.994 and were clustered to six major groups on a criterion of 0.78 similarity coefficient. The present study has revealed the significant genetic similarity among the samples tested. The analysis of genetic relationships in P. grandiflorum using RAPD-PCR banding data can be useful for the breed improvement.

Evaluation of Antibacterial, Antioxidant Fractionalities and Hair Loss Prevention Effect of Platycodon grandiflorum (도라지 추출성분의 항균·항산화 및 탈모예방 효과)

  • To investigate the effects of antioxidant activities and hair loss prevention of extracts from Platycodon grandiflorum, we've prepared chloroform (CF) and ethylacetate fractions (EA) extracted from P. grandiflorum. In the results of DPPH radical scavenging assay, the two fractions showed dose-dependent antioxidant activities. Furthermore, in the ABTS assay, the two fractions exhibited the inhibitory effect over 90% at 10, 50, 100, 200 mg/mL. To investigate the inflammation inhibitory effect, we used RAW264.7 cells, these extracts were inhibited inflammatory reaction by suppressing the production of nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) in dose-dependent manner. In the assay of HaCaT cells' proliferation, the 24 hr treatment of the extracts significantly accelerarted cell proliferation in the range of concentrations used. The two fractions inhibited the proliferation of Malssezia furfur, the bacteria induce the dandruff. Finally, the CF could significantly inhibit the DHT production at 1, 10, 50, $100{\mu}g/mL$, but EA showed the inhibitory effect at the concentration over $50{\mu}g/mL$. The overall results of this study suggest that the chloroform (CF) and ethylacetate fractions (EA) from P. grandiflorum could be a useful raw material for the hair loss prevention products.

The Effect of Platycodon grandiflorum Root Ethanol Extract on Blood Glucose, Lipid, Activities of Carbohydrate Metabolism Related Enzyme in Streptozotocin-Induced Diabetic Rats (도라지 뿌리 에탄올 추출물이 streptozotocin으로 유발된 흰쥐의 혈당지질, 당대사에 미치는 영향)

  • This study was done to investigate the antidiabetic effect of ethanol extract from Platycodon grandiflorum root in Streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose 45mg/kg.b.w. dissolved in citrate buffer(pH4.5). The ethanol extract of Platycodon grandiflorum root was orally administrated once a day for 7 days. The contents of serum glucose, triglyceride(TG) and total cholesterol were significantly decreased(p<0.05) in Platycodon grandiflorumt root treated group compared to the those of STZ-control group. Also the contents of hepatic glycogen and HDL-cholesterol, the activities of glucose-phosphate dehydrogenase(G-6-PDH) and glucokinase(GK) were significamtly increased (p<0.05). These results indicated that ethanol extract of Platycodon grandiflorum root would have antidiabetic effect in STZ-induced diabetic rats.

Effect of Saponin Content and Antioxidant Activities of Platycodon grandiflorum Radix by Cutting Length (도라지 절단 길이에 따른 사포닌 함량 및 항산화 활성)

  • Background : Platycodon grandiflorum radix used as a therapeutic agent for lung and respiratory diseases in traditional Korean medicine. The saponin and sugar content in P. grandiflorum root is dependent on the drying temperature and method. In this study, we investigated saponins and antioxidant contents in dried P. grandiflorum roots either not cut or cut to different lengths. Methods and Results : P. grandiflorum roots were either cut to 0.5, 1.0 or 2.0 cm in length or left uncut. They were dried at $45^{\circ}C$ and the effect of antioxidant substances, antioxidant ability, and saponin content were investigated. P. grandiflorum roots rapidly dried out when cut to short lengths. The saponin contents (platycodin D, polygalacin D, and deapioplatycodin D) was highest in the uncut P. grandiflorum roots. When the cut lengths were long, the levels of polyphenolic compound increased. However, 2,2'-azinobis-3-ethylbenzo-thiazoline-6-sulfonic acid (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities were higher in the samples cut to shorter lengths. Conclusions : These results suggested that the saponin and antioxidant content of P. grandiflorum roots could be maximized by controlling the drying rate of the roots via adjustments to the root cutting length.

  • Platycodi Radix, as one of the most important traditional oriental materia medica, is utilized frequently in clinical practice and dietary in China, Korea, and Japan. To evaluate definitely the quality of Platycodi Radix, and provide scientific data for commodity circulation, new product development and clinical application of Platycodi Radix, we summarized its medical history, application status, chemical compositions, bioactivities, cultivation, international circulation, quality evaluation, dosage and safety, and brought forward some constructive suggestions on further establishment of international criteria for quality control of Platycodon grandiflorum.
