Effects of polygalacin D extracted from Platycodon grandiflorum on myoblast differentiation and muscle atrophy (길경에서 추출한 polygalacin D가 근원세포 분화 및 근위축에 미치는 영향)

  • Purpose: The balance between synthesis and degradation of proteins plays a critical role in the maintenance of skeletal muscle mass. Mitochondrial dysfunction has been closely associated with skeletal muscle atrophy caused by aging, cancer, and chemotherapy. Polygalacin D is a saponin derivative isolated from Platycodon grandiflorum (Jacq.) A. DC. This study aimed to investigate the effects of polygalacin D on myoblast differentiation and muscle atrophy in association with mitochondrial function in in vitro and in zebrafish models in vivo. Methods: C2C12 myoblasts were cultured in differentiation media containing different concentrations of polygalacin D, followed by the immunostaining of the myotubes with myosin heavy chain (MHC). The mRNA expression of markers related to myogenesis, muscle atrophy, and mitochondrial function was determined by real-time quantitative reverse transcription polymerase chain reaction. Wild type AB* zebrafish (Danio rerio) embryos were treated with 5-fluorouracil, leucovorin, and irinotecan (FOLFIRI) with or without polygalacin D, and immunostained to detect slow and fast types of muscle fibers. The Tg(Xla.Eef1a1:mitoEGFP) zebrafish expressing mitochondria-targeted green fluorescent protein was used to monitor mitochondrial morphology. Results: The exposure of C2C12 myotubes to 0.1 ng/mL of polygalacin D increased the formation of MHC-positive multinucleated myotubes (≥ 8 nuclei) compared with the control. Polygalacin D significantly increased the expression of MHC isoforms (Myh1, Myh2, Myh4, and Myh7) involved in myoblast differentiation while it decreased the expression of atrophic markers including muscle RING-finger protein-1 (MuRF1), mothers against decapentaplegic homolog (Smad)2, and Smad3. In addition, polygalacin D promoted peroxisome proliferator-activated receptor-gamma coactivator (Pgc1α) expression and reduced the level of mitochondrial fission regulators such as dynamin-1-like protein (Drp1) and mitochondrial fission 1 (Fis1). In a zebrafish model of FOLFIRI-induced muscle atrophy, polygalacin D improved not only mitochondrial dysfunction but also slow and fast muscle fiber atrophy. Conclusion: These results demonstrated that polygalacin D promotes myogenesis and alleviates chemotherapy-induced muscle atrophy by improving mitochondrial function. Thus, polygalacin D could be useful as nutrition support to prevent and ameliorate muscle wasting and weakness.

Effect of $GA_3$ and Light Quality on Seed Germination in Three Campanulan Plants ($GA_3$ 및 파종(播種) 후(後) 광질(光質) 처리(處理)에 따른 도라지, 더덕, 만삼의 발아율(發芽率))

  • Gibberellin (GA) has been widely used to break seed dormancy for better stand establishment. The experiment was done to clarify the effect of $GA_3$ (concentration; period) and light quality (red; white; dark) after sowing on seed germination and radicle elongation of Campanulaceae (Platycodon grandiflorum; Codonopsis lanceolata; C. pilosula) to get an information on their field emergence. The germination test was carried out with 12 hours irradiation for 9 days after priming treatment. In the darkness, the mean germination rate of all the species was decreased in the order to P. grandiflorum, C. lanceolata, C. pilosula. Their mean germination rates and radicle lengths were increased with increased concentration to 0.1mM of $GA_3$. Earlier germination rate was higher but later one was less in 4-day $GA_3$ treatment than in 1- or 2-day $GA_3$ treatment. Light treatment. especially red light given after $GA_3$ treatment. eliminated the $GA_3$ treatment effect. Red light done after $GA_3$ treatment nearly blocked the germination of P. grandiflorum and C. pilosula but delayed that of C. pilosula compared to the other light quality treatments having the similar rate. In addition. the radicle elongation of all three species affected by light quality treatment showed the same result as the germination rate.

Detection Characteristics of Gamma-Irradiated Korean Medicinal Herbs by Using PSL, TL, and ESR (PSL, TL 및 ESR 분석에 의한 감마선 조사 한약재의 검지 특성)

  • The detection characteristics of gamma-irradiated ($0{\sim}10.0\;kGy$) medicinal herbs (Platycodon grandiflorum, Acanthopanax chiisanensis) were investigated by photostimulated luminescence (PSL), thermoluminescence (TL), and electron spin resonance (ESR). The results of the PSL, a first screening method in comparison with the TL, showed photon counts greater than 5,000 counts/60 s (positive) in the irradiated samples, while the non-irradiated samples yielded photon counts less than 700 counts/60 s (negative). The TL was also applied for the detection method of irradiated medicinal herbs and showed that the non-irradiated sample revealed a glow curve with a low intensity, while the irradiated samples showed a higher intensity. These results were normalized by re-irradiating the mineral grains with a irradiation dose of 1.0 kGy, and a second glow curve was recorded. The ratio of the intensity of the first glow curve ($TL_1$) to that after the normalization dose ($TL_2$) was determined and compared with the recommended threshold values. TL ratio ($TL_1/TL_2$) was below 0.007 for the non-irradiated sample and higher than 0.1 for all irradiated samples (above 1.0 kGy). ESR spectroscopy revealed specific signals (6.065 mT) derived from free radicals in cellulose containing irradiated medicinal herbs. The P. grandiflorum showed clearer signals than A. chiisanensis. From the results of our studies, the PSL, TL, and ESR determinations were found to be suitable for the detection of irradiated medicinal herbs such as P. grandiflorum and A. chiisanensis.

Germanium Contents of Soil and Crops in Gyeongnam Province (경남지역의 토양 및 농작물중 게르마늄 함량)

  • To investigate the germanium content in paddy soil in Gyeongnam province, 310 paddy soil samples were collected at 19 areas in Gyeongnam, Ulsan and Busan. Contents of germanium in paddy soils were analyzed in different topographies, soil types and soil textures. Average content of germanium in Gyeongnam was 0.24 mr/kg, those of Masan, Jinju and Hadong were above 0.30 mg/kg. Germanium content with different topographies were no difference. Germanium contents in different soil types were 0.27 mg/kg in well adapted soil and 0.23 mg/kg in poorly drained soil. Germanium contents in different soil textures were 0.27 mg/kg in silt loam and 0.23 mg/kg in sandy loam. To determine germanium content on agricultural product in the field, content of germanium in cereals, vegetables and fruits were analyzed. Germanium content of agricultural product was high in the order of cereals>vegetables>fruits. In case of vegetables, germanium contents were high in the order of leaf vegetables>root vegetables>fruit vegetables. Germanium contents were high with 62 and $65{\mu}g/kg$ in lettuce and young radish, respectively. To analyze the germanium content in medicinal plant, samples were collected from 19 medicinal plants at Hamyang areas. Germanium contents in Angelica keiskei, Ligusticum chuanxiong, Panax ginseng and Atractylodes macrocephala were relatively high with $100{\mu}g/kg$ above.

In vitro screening of the acetylcholinesterase inhibition, antioxidant activity, and neuronal cell protective effect of medicinal plant extracts (생약추출물의 acetylcholinesterase 저해, 항산화 및 신경세포보호 효과 in vitro 탐색)

  • This study investigated the effects (i.e., the acetylcholinesterase activity, lipid peroxidation, and neuronal survival) of 20 kinds of medicinal water extracts. The water extracts of three medicinal plants - Cornus officinalis, Glycyrrhiza glabra, and Angelica gigas - were found to be the most effective on acetylcholinesterase inhibitory activity. In the lipid peroxidation-generating system induced by $H_2O_2/FeSO_4$ in rat brain homogenates, Perilla frutescens, Polygonum multiflorum, Cinnamomun cassia, and G. glabra exhibited protective activity against lipid peroxidation. The neuronal cell death induced by L-glutamate in PC12 was suppressed by the water extracts of G. glabra, Cinnamomun cassia, Platycodon grandiflorum, and Mentha arvensis at the concentration of $100{\mu}g/mL$. Taken together, these results showed that the water extract of G. glabra has the potential anti-dementia activity, which suggests that it might provide an effective strategy for improving dementia.

Screening of Inhibitory activities on Angiotensin Converting Enzyme from Medicinal plants (약용식물의 Angiotensin Converting Enzyme 저해활성 탐색)

  • Angiotensin converting enzyme(ACE) belongs to the class of zinc protease and plays an important role in the regulation of blood pressure. In this experiment, we investigated the inhibitory activities of medicinal plant extracts on ACE. Fifty medicinal plants were selected and the extracts were prepared by refluxing with 70% methanol. Among the extracts, eleven medicinal plant extracts such as Sedum sarmentosum Bunge, Petasites japonicus(s.et z.) Max, Rubus coreanus, Morus bombycis Koidz, Acorus calamus var. angustatus, Glycyrhiza glabra, Equisetum hyemale, Portulaca oleracea L., Prunella vulgaris var. lilacina Nakai, Sorbus commixta Hedl, Allium thunbergii showed more than 50% inhibitory activities, and Paeonia suffruticosa Andr., lnula helenium, Acanthopanax senticosus Harms, Dendrobium moniliforme, Juglans mandshurica, Zizyphus jujuba, Leonurus artemisia, Aster scaber Thunb, Vitex rotundifolia, Platycodon grandiflorum, Prunus persica, Ligularia fischeri showed $40{\sim}49%$ inhibitory activities. Therefore these extracts which contain high ACE inhibitory activities may be useful as antihypertension agents and to the treatment of hypertension.

Quality Characteristics of Fermented Wild Grass Juice (산야초 발효액의 품질 특성)

  • The quality characteristics, enzyme activity, and antioxidative activity of fermented wild grass juices were investigated during fermentation and ripening for 6 months. Fermented wild grass juice was prepared from wild grasses (Oenothera biennis, Portulaca oleracea, and Rhus verniciflua) and sugar. Wild grasses and sugar mixed at the same ratio (1:1, w/w) and ripened at $20^{\circ}C$ for 3 months after fermentation for 3 months at $20^{\circ}C$. The $^{\circ}Bx$ of all fermented wild grass juices (FWGJ) decreased during fermentation. The pH of all FWGJ decreased gradually during fermentation and did not show any significant difference during ripening. Viscosity of FWGJ increased during fermentation but decreased during ripening periods in all tested samples. Total viable cell of FWGJ decreased after fermentation for 1 month, whereas lactic acid bacteria were not detected during fermentation. Enzyme activity was lower than 1 unit during fermentation and ripening period in all tested FWGJ samples. Enzyme activity of commercial FWGJ (Acanthopanax sessiliflorum, Oenanthe javanica, Plantago asiatica L., Platycodon grandiflorum, Orostachys japonicus A.) showed lower activity of 1 unit following invertase, cellulase, and amylase activity.

The Changes of Benzo[${\alpha}$]pyrene Content in Herbal Tea Containing Schizandra chinensis, Astragalus membranaceus, Platycodon grandiflorum and Liriope platyphylla Affected by Roasting Temperature (Roasting 온도에 따른 오미자, 황기, 길경 및 맥문동을 첨가한 한방차의 벤조피렌 함량 변화)

  • The following is the study result of herbal tea roasted at different temperatures between $80{\sim}140^{\circ}C$. Depending on treatment temperature the water content decreased, some carbonization occurred and crude ash content relatively increased. Also crude protein and crude fat changed little. Benzopyrene content (0.17~0.35ppb) showed a tendency to increase with higher treating temperature. From this result, the $B({\alpha})P$ content differed depending on the treatment temperature and raw materials. In case of roasting, the actual inside temperature is around $200^{\circ}C$ but since the surface temperature of the roaster reaches around $2,000^{\circ}C$ some portion of $B({\alpha})P$ content was presumed to be produced from the area that came in contact with this surface. Solid elution rate of herbal tea showed 0.18~0.35%(w/w) and the rate of solid elution decreased with higher roasting temperature. There was no big change in $80{\sim}110^{\circ}C$ treatment section but the solid elution decreased rapidly in $110{\sim}140^{\circ}C$ section. The reason for decreasing solid elution rate at higher treatment temperature is because the compact inner tissue makes elution difficult.

Monitoring and Risk Assessment of Heavy Metals in Perennial Root Vegetables (다년생 근채류 중 중금속 모니터링 및 위해성평가)

  • BACKGROUND: This study was carried out to survey the levels of heavy metals in perennial root vegetables and to assess dietary exposure and risk to the Korean population health.METHODS AND RESULTS: Perennial root vegetables (n=214) including Panax ginseng C.A mayer, Woodcultivated ginseng, Codonopsis lanceolata, and Platycodon granditloum were collected from markets or harvested from farmhouse in Korea. Lead(Pb), cadmium(Cd) and arsenic (As) analysis were performed with microwave device and inductively coupled plasma mass spectrometer. Limit of detection for heavy metals were 0.010~0.050 μg/kg, while limit of quantitation were 0.035~0.175 μg/kg. The recovery results were in the range of 76~102%. The average contents of heavy metals in perennial root vegetables were in the range of Pb 0.013(Panax ginseng C.A Mayer)~0.070 (Wood-cultivated ginseng) mg/kg, Cd 0.009(Panax ginseng C.A Mayer)~0.034(Codonopsis lanceolata) mg/kg, and As 0.002(Panax ginseng C.A Mayer)~0.004(Plafycodon grandiflorum) mg/kg, respectively. For risk assessment, daily intakes of heave metals were estimated and risk indices were calculated in comparison with reference dose. The dietary exposures of heavy metals through usual intake were Pb 0.070 μg/day, Cd 0.041 μg/day and As 0.008 μg/day, taking 0.03%, 0.08% and 0.0003% as risk indices, respectively.CONCLUSION: The risk level for Korean population exposed to heavy metals through intake of perennial root vegetables was far low, indicating of little possibility of concern.

Pesticide and Heavy Metal Residue Monitoring in 13 Types of Agroforestry Products in 2019 (2019년 유통 임산물 중 산나물류와 약초류, 과실류 13종의 농약 및 중금속 잔류 실태)

  • In accordance with the implementation of the Positive List System (PLS), the proper usage of pesticides is now being enforced. It is assumed that unregistered pesticides are being used on agroforestry products due to the low number of registered pesticides in the agricultural industry. In this study, pesticide and heavy metal residues were investigated in 13 types of products to determine the status of usage. The levels (%) of pesticides detected in Pimpinella beachscape, Platycodon grandiflorum, Codonopsis lancekolate, Artemisia dubia, Angelica gigas, Pyrus pyrifolia, and Punica granatum were 40.0, 20.0, 26.7, 13.3, 56.3, 57.1, 33.3, 26.7, 66.7, and 46.7%, respectively, while, those in Petridium aquilinum, Disoscorea batata and Senna tora were zero. Heavy metals (Pb, Cd) were detected only from P. grandisflorum and A. dubia. The pesticide usage and registration data by agroforestry product obtained in the study will be useful in the future for ensuring the safety of domestic agroforestry products.
