분심기음(分心氣飮)이 고혈압 백서와 인간유래 혈관내피세포주(ECV 304)에 미치는 영향에 대한 연구 (An Experimental Study of Effect on ECV 304 Cells, Platelet Rich Plasma and Rats treated with L-NAME by Boonsimgieum extract)

  • Object : This study was designed to research whether the protection and inhibitory effects of cardiovascular diseases in L-NAME induced rat or ECV 304 cell lines through the Cell morphological pattern, Tunel assay, LDH activity, heart rate, blood pressure and immunohistochemistric analysis by Boonsimgieum water extract Methods : Nitric oxide(NO) play an important role in normal and pathophysiological cells including as a messenger molecule, neurotransmitter, microbiocidal agent, or dilator of blood vessels and artheriosclerosis, hypertension, myocardial infarction, respectively. Endothelial cell products can modulate the magnitude of a response to a vasoconstrictor, as evinced by the greater constriction after endothelium removal or NO synthesis blockade. To investigate that Boonsimgieum in the potential contribution of the levels of nitric oxide generated by endothelial nitric oxide synthase (eNOS) and the mechanisms of protection against NG-nitro-L-arginine methyl ester (L-NAME), human ECV 304 cells, which normally do not express eNOS, were expressed by L-NAME. L-NAME stimulated rat or cells were found to be resistant to injury and delayed death following the Boonsimgieum. Inhibition of nitric oxide synthesis abolished the protective effect against L-NAME, thrombin and collagen exposure. Interestingly, such effects have been observed during stimulation with agents such as phenylephrine and KCl on L-NAME mediate rats, were damaged by the NOS inhibitor L-NAME. Result : As the result of this study, In group, the anti-apoptosis and necrosis in the cardiovascular system have a potential capacity for prevented, protected and treating the diseases of cardiovascular system, against the necrosis of rat and ECV 304 cells with Caspase 3 and calpain expression by L-NAME is promoted. Conclusion : these results demonstrate neuroprotective and memory enhancing effects of ZIBU, suggesting its beneficial actions for the treatment of AD.

Rubbery 고분가 막을 이용한 휘발성 유기화학물의 제거 및 회수 (Removal/Recovery of VOCs Using a Rubbery Polymeric Membrane)

  • 실리콘 코팅한 복합막을 이용하여 톨로엔이나 메탄올 같은 휘발성 유기화합물의 제거 및 회수에 관한 연구를 수행하였다. 사용된 막은 코팅층의 두께가 얇고($~1\mu$m), 지지층의 기고율이 커서 투과저항은 적었으며 코팅층과 지지층은 플라즈마 중합에 의해 결합되어 있어서 높은 압력에도 견딜 수 있었다. Feed의 유속은 60cc/min. 이하일 때 사용된 모듈에 위해 휘발성 유기화합물의 제거율은 96~99%이었으며, feed의 농도가 높을수록 제거율은 더 증가하였다. 이 공정은 휘발성 유기화합물의 농도가 높고 유속이 낮은 흐름을 처리하는데 아주 적합함을 알 수 있었다. 사용된 막을 통한 휘발성 유기 화합물들의 투과도는 $4~30 \times 10^{-9}gmol/sec \cdot cm^{2}\cdot cmHg$이었고, 질소의 투과도는 $3~9 \times 10^{-10}gmol/sec \cdot cm^{2} \cdot cmHg$의 범위에 있었다. 휘발성 유기화합물과 질소의 선택도는 유속과 휘발성 유기화합물의 농도에 따라 톨루엔/질소의 경우 10~55, 메탄올/질소의 경우 15~125의 값을 얻었다.

  • Radiation synovectomy is one of the most useful methods for treating patients with refractory synovitis because of its convenience, long-term effects, repeatability and the avoidance of surgery. In this study, we investigated the toxicity, stability and biodistribution of a rhenium-188 ($^{188}$Re)-tin colloid to evaluate its suitability as a synovectomy agent. Twenty four hours after injecting the $^{188}$Re-tin colloids (74 KBq/0.1 mL) into the tail vein of ICR mice, most of the $^{188}$Retin colloidal particles was found in the lungs. In addition, there were no particle size changes at either room temperature or at $37^{\circ}C$ after injecting the $^{188}$Re-tin colloids in human plasma and synovial fluid. In vitro stability tests showed that the $^{188}$Re-tin colloid remained in a colloidal form without a critical size variation over a 2-day period. We investigated the leakage of $^{188}$Retin colloids from the intraarticular injection site with gamma counting in New Zealand white rabbits. The $^{188}$Re-tin colloids (55.5 MBq/0.15 mL) were injected at the cavum articular and the mean retention percentage of the $^{188}$Re-tin colloid was 98.7% for 1 day at the injection site, which suggests that there was neither change in the particle size nor leakage at the injection sites. In the biodistribution study with the SD rats, the liver showed the highest radioactivity (0.0427% ID/organ) except for the injected knees (99.49%). In the SD rats, mild toxicities including the skin or a synovium inflammation were observed as a result of a radioactivity of 15 mCi/kg at the intraarticular injection site. However, there was no systemic toxicity. In the Ovalbumin (OVA)-induced arthritic rabbits, the $^{188}$Re-tin colloid improved the macroscopic, the histological score and reduced the knee joint diameter when compared to the arthritic control. In conclusion, a $^{188}$Re-tin-colloid is considered as a strong candidate for radiation synovectomy with a superior efficacy and safety.

기능성 소화불량증의 침치료 연구 동향 : Pubmed를 중심으로 (Trend of Acupuncture Treatment Study for Functional Dyspepsia)

  • Objectives: This study was conducted to help clinical studies for treating Functional Dyspepsia with acupuncture. We inspected tendency of studies of acupuncture treatment for functional dyspepsia and analyzed the assessing instrument for it by scrutinizing Pubmed recent publications. Materials and Methods: We inspected 8 theses and scrutinized their objectives, periods, participants, materials and methods, the instrument of assessment for the studies, and criteria and symptom score for questionaire, results and JADAD score. Results: 1. Most studies were published by oriental doctors except one which was published by an American doctor. More and more studies are being published in America and written in English recently. So it is considered that western doctors think that the acupuncture treatment is useful as unorthodox medicine for functional dyspepsia. 2. Among the 4 clinical studies, 2 studies were RCT and 1 study was not RCT and 1 study with vagueness. All 4 clinical studies were not double-blinded because they were about comparison between acupuncture treatment and medicine. 3. ST36, PC6, CV12, BL21 were chiefly used, and auricular point, ST40, SP6 were also used. Manual acupuncture were used in 2 studies, and low frequency pulse and electroacupuncture were used in other studies respectively. 4. 4 clinical studies used 0~3 points symptom score as the instrument of assessment. Additional assessments were on gastric emptying time, EGG and plasma level of motilin. All thesis were not verified of validity and reliability. 5. Among the 4 reviewed thesis, 2 theses were about acupuncture as one of the various treatment of functional dyspepsia, and the other 2 theses were about efficacy of acupuncture treatment. And the result generally showed effectivity of acupuncture treatment. Conclusion: Acupuncture treatment for functional dyspepsia is considered useful as in unorthodox medicine by western doctors, and it is necessary to provide objective instrument for assessing acupuncture treatment for functional dyspepsia.

Ginsenoside Rg3 ameliorates myocardial glucose metabolism and insulin resistance via activating the AMPK signaling pathway

  • Background: Ginsenoside Rg3 is one of the main active ingredients in ginseng. Here, we aimed to confirm its protective effect on the heart function in transverse aortic coarctation (TAC)-induced heart failure mice and explore the potential molecular mechanisms involved. Methods: The effects of ginsenoside Rg3 on heart and mitochondrial function were investigated by treating TAC-induced heart failure in mice. The mechanism of ginsenoside Rg3 for improving heart and mitochondrial function in mice with heart failure was predicted through integrative analysis of the proteome and plasma metabolome. Glucose uptake and myocardial insulin sensitivity were evaluated using micro-positron emission tomography. The effect of ginsenoside Rg3 on myocardial insulin sensitivity was clarified by combining in vivo animal experiments and in vitro cell experiments. Results: Treatment of TAC-induced mouse models with ginsenoside Rg3 significantly improved heart function and protected mitochondrial structure and function. Fusion of metabolomics, proteomics, and targeted metabolomics data showed that Rg3 regulated the glycolysis process, and Rg3 not only regulated glucose uptake but also improve myocardial insulin resistance. The molecular mechanism of ginsenoside Rg3 regulation of glucose metabolism was determined by exploring the interaction pathways of AMPK, insulin resistance, and glucose metabolism. The effect of ginsenoside Rg3 on the promotion of glucose uptake in IR-H9c2 cells by AMPK activation was dependent on the insulin signaling pathway. Conclusions: Ginsenoside Rg3 modulates glucose metabolism and significantly ameliorates insulin resistance through activation of the AMPK pathway.

사인으로부터 추출한 정유와 용매 추출물의 휘발성 물질 및 금속성분 분석 (Analysis of volatile compounds and metals in essential oil and solvent extracts of Amomi Fructus)

  • 한약재 중 항산화 작용이 있다고 알려진 사인(Amomi Fructus)을 SDE (simultaneous distillation extraction) 방법으로 정유(essential oil)를 추출하고 GC-MS (gas chromatography-mass spectrometry)를 이용하여 39개의 성분을 규명하였으며, 주요 성분은 camphor, borneol acetate, borneol, D-limonene 및 camphene이었다. 유기용매 3가지(헥세인, 다이에틸 에터 및 다이클로로메테인)를 이용하여 얻은 용매 추출물에서 정유성분 이외에 추가로 4개의 성분을 규명하였으며, 주요 5개 성분은 동일하였으나 상대적인 면적 비율은 차이가 있었다. 특정온도와 휘발 조건에서 정유와 용매 추출물에서 휘발하는 물질들의 종류와 상대적인 비율을 측정하기 위하여 dynamic headspace를 이용하여 특정 조건에서 휘발하는 물질들을 트랩에 포집한 후 GC-MS로 분석하였다. 사인으로부터 SDE 방법에 의해 정유를 추출하는 방법의 회수율을 장뇌(camphor)를 이용하여 측정하였으며, 장뇌의 회수율은 82.0% 이었다. 사인에서 Hg의 함량은 수은 분석기로 측정하였으며, Cd, Pb, Cr, Mn, Co, Ni, Cu 와 Zn은 ICP-MS (Inductively coupled plasma-mass spectrometer)로 측정하였다. 사인에서 Pb, Cd 및 Hg이 각각 0.72 mg/kg, <0.10 mg/kg 및 0.0023 mg/kg 검출되었으며, 이는 순도시험 허용기준치 이하의 값들이었다. 사인에서 Cr, Mn, Co, Ni, Cu와 Zn의 함량을 조사한 결과 Mn (213 mg/kg), Cu (8.29 mg/kg) 및 Zn (31.0 mg/kg)등이 비교적 높은 함량으로 검출되었으며, 정유와 용매 추출물에서도 Mn (0.65~9.08 mg/kg), Cu (1.16~4.40 mg/kg) 및 Zn (1.10~3.80 mg/kg)이 검출되었다. 이들 금속성분이 사인에서 기인하는 것인지 아니면 유통과정이나 취급과정에서 오염된 것인지의 여부 규명이 필요하며, 또한 이런 금속들이 생물학적 활성 연구에 어떤 영향 주는지도 규명할 필요가 있다.

구멍쇠 미역의 변비 완화 효과 및 혈액지방에 미치는 영향 (Effects of Agarum cribrosum on the Improvements in Treating Constipation and Plasma lipid Profiles)

  • 구멍쇠 미역의 변비 개선 효능을 검정한 결과, 흰쥐의 사료섭취량과 체중 변화는 정상군에 비해 구멍쇠 미역 투여군이 유의적으로 식욕억제와 비만예방에 효과가 있을 것으로 사료된다. 장통과시간은 대조구 853.1분이었고, 구멍쇠 미역 5% 급이군에서 789.7분으로 감소하였다. 변비를 유발시키면서 식이섬유를 급이한 흰쥐의 체중 대비 간의 무게는 모든 실험군에서 정상군과 차이가 없었으며, 신장의 무게도 각 실험군에서 차이가 나타나지 않았다. 또한, 소장 및 대장의 길이는 변비의 유무와 식이섬유 급원에 따른 군간 유의적인 차이가 없었다. 분변량을 측정한 결과 정상군의 1일 평균 분변량은 2.84 g, 대조군은 1.34 g, 구멍쇠 미역 5% 급이군은 1.57 g, 구멍쇠 미역 10% 급이군은 2.02 g으로 구멍쇠 미역의 섭취가 변비 증상 호전 효과가 있을 것으로 판단된다. 분변의 수분함량 측정 결과, loperamide를 단독 투여한 대조군 분변의 수분함량이 정상군에 비해 유의적으로 감소하여 loperamide 투여로 인해 변비가 유발된 것을 확인하였으며, 구멍쇠미역 5% 및 10%가 함유된 사료를 급이한 군은 정상군에 비해 분변의 수분함량이 각각 1.6배 및 2.1배 증가하였다. 구멍쇠 미역을 급이한 흰쥐의 혈청 중 cholesterol 및 triglyceride 함량 변화측정 결과, 혈청 중 total cholesterol 함량은 구멍쇠 미역 5% 급이군에서 loperamide로 변비를 유발한 대조군에 비해 3.5% 감소하였으나 유의적인 차이는 없었으며, 구멍쇠 미역 10% 급이군은 40.21 mg/dL로 33.5% 감소하여 구멍쇠 미역의 섭취량이 높아질수록 흰쥐의 혈중 cholesterol 함량이 감소하였다. GOT 및 GPT는 각 실험군에서 차이가 나타나지 않았으나, BUN 농도는 대조군에 비해 구멍쇠 미역 10% 급이군에서 낮게 나타났으며, 혈중 포도당 농도는 각 실험군 간에 유의적인 차이가 없었다. 간 조직 내의 총콜레스테롤 분석 결과, 구멍 쇠 미역 급이군 모두 정상군 및 대조군에 비해 총콜레스테롤 함량이 감소하였으며, loperamide 투여에 의해 감소되었던 HDL-콜레스테롤은 구멍쇠 미역 급이 시 대조군에 비해 증가하였으나 유의적인 차이는 없었다. 또한, 중성지방 함량은 총 콜레스테롤과 마찬가지로 구멍쇠 미역 급이군이 정상군과 대조군에 비해 감소하였다. 이와 같은 결과로 보아 구멍쇠 미역은 장 이동률을 증가시켜 장운동을 촉진하리라 생각되며, 변비 해소 작용도 나타내어 장 기능 개선에 효과가 있을 것으로 판단된다.

구강 편평세포암종 세포주에서 Cyclosporin A와 Taxol 투여시 PI-3 kinase/Akt1 Pathway에 의한 세포사멸 병용효과 (APOPTOTIC EFFECT IN COMBINATION OF CYCLOSPORIN A AND TAXOL ON ORAL SQUAMOUS CELL CARCINOMA CELL LINE THROUGH THE PI-3 KINASE/AKT1 PATHWAY)

  • Oral cancer take up 2-6% of all carcinomas and squamous cell carcinoma, which is the most common type in oral cancer, has a poor prognosis due to its high metastasis and recurrence rates. In treating oral cancer, chemotherapy to the primary, metastasized and recurrent lesion is a very important and useful treatment, even though its widespread usage is limited due to high general toxicity and local toxicity to other organs. Taxol, a microtubule stabilizing agent, is an anticancer drug that induces cell apoptosis by inhibiting depolymerization of microtubules in between the metaphase and anaphase of the cell mitosis. Recently, its effectiveness and mechanism on various tumor has been reported. However, not much research has been done on the application of Taxol to oral squamous cell carcinoma. Cyclosporin A, which is an immunosuppressant, is being used on cancers and when co-administered with Taxol, effectiveness of Taxol is enhanced by inhibition of Taxol induced multidrug resistance. In this study, Cyclosporin A with different concentration of Taxol was co-administered to HN22, the oral squamous cell carcinomacell line. To observe the cell apoptosis and the mechanisms that take part in this process, mortality evaluation of tumor cell using wortmannin, c-DNA microarray, RT-PCR analysis, cytometry analysis and western blotting were used, and based upon the observation on the effect and mechanism of the agent, the following results were obtained: 1. The HN22 cell line viability was lowest when $100{\mu}M$ of Wortmannin and $5{\mu}g/ml$ of Taxol were co-administered, showing that Taxol participates in P13K-AKT1 pathway. 2. In c-DNA microarray, where $1{\mu}g/ml$ of cyclosporine A and 3mg/ml of Taxol were co-administered, no up regulation of AKT1, PTEN and BAD c-DNA that participate in cell apoptosis was observed. 3. When $1{\mu}g/ml$ of Cyclosporin A was applied alone to HN22 cell line, no difference was found in AKT1, PTEN and BAD mRNA expression. 4. Increased AKT1, mRNA expression was observed when $3{\mu}g/ml$ of Taxol was applied alone to HN22 cell line. 5. When $1{\mu}g/ml$ of Cyclosporin A and Taxol($3{\mu}g/ml\;and\;5{\mu}g/ml$) were co-administered to HN22 cell line, PTEN mRNA expression increased, whereas AKT1 and BAD mRNA decreased. 6. As a result of cytometry analysis, in the group of Cyclosporin A($1{\mu}g/ml$) and Taxol($3{\mu}g/ml$) co-administration, increased Annxin V was observed, which shows that apoptosis occurred by deformation of plasma membrane. However, no significant difference was observed with vary ing concentration. 7. In western blot analysis, no caspase 3 was observed in the group of Cyclosporin A($1{\mu}g/ml$) and Taxol($3{\mu}g/ml$) co-administration. From the results of this study, it can be concluded that synergistic effect can be observed in combination therapy of Taxol and Cyclosporin A on oral squamous cell carcinoma cell line, where decreased activity of the cell line was observed. This resulted in decreased AKT1 and BAD mRNA and increased PTEN mRNA expression and when wortmannin and Taxol were co-administered, the viability decreased which confirms that Taxol decreases the viability of tumor cell line. Hence, when Taxol and cyclosporine A are co-administered, it can be assumed that cell apoptosis occurs through AKt1 pathway.
