Effects of Acute Change of Salinity on Physiological Response and Survival in Hybrid Striped Bass (Morone Chrysops male X Msaxatilis female) (빠른 염분변화가 Striped bass 잡종 (Morone Chrysops male X Msaxatilis female)의 생리적 반응과 생존에 미치는 영향)

  • Physiological responses and tolerance limit of hybrid striped bass $(Morone\;Chrysops\;male{\times}\;M.\;saxatilis\;female),$ were investigated on salinity changes from 33 psu to 0 psu and from 0 to psu 33 psu in two separate trials, respectively. In the acute salinity decrease from sea water (33 psu) to fresh water (0 psu), plasma level of cortisol, glucose, total protein, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were not affected, however, $Na^+,\;Cl^-$ and osmolality significantly decreased. In the acute salinity increase from fresh water to sea water (33 psu), plasma cortisol level did not change. Glucose and total protein concentrations in plasma and hematocrit (Hct) decreased, whereas AST, ALT, $Na^+,\;Cl^-$ and osmolality increased. The hybrid striped bass can overcome the acute changes of salinity, from 0 psu to 33 psu or from 33 psu to 0 psu, But salinity decrease stressed the fish much more than salinity increase.

  • Recently, the pop-up satellite archival tag (PSAT) is being used in studies as a method of using satellites for monitoring organisms. Because PSATs are attached directly on the organism being monitored, it is used mostly on larger species since the health of the organism is an important concern. For this reason, PSAT-based surveys are lacking in Korea with no studies on the physiological responses of organisms with PSAT attached. Accordingly, as a basic biomonitoring study using PSAT, the present study investigated the physiological changes in fish in response to the attachment of PSAT. The present study used red seabream (Pagrus major) and Korean rockfish (Sebastes schlegelii) as the experimental fish. The PSAT was attached to the muscle below the experimental fish's dorsal fin using a mono filament (n = 3). To investigate the changes in physiological responses according to PSAT attachment, blood samples were collected from all experimental fish, including the control (n = 3), at 1, 7, 14 and 21 days after the attachment. Upon blood sample collection, whole blood was used to measure hematocrit and hemoglobin levels. After separating the plasma, the separated plasma was used to measure the GOT, GPT, glucose, total protein, and cholesterol levels. Meanwhile, the plasma cortisol, superoxide dismutase, and catalase levels were measured using the ELISA method. The results showed that attaching the tag did not have any impact on the immunity and stress response of the experimental fish. The findings in this study also demonstrated the possibility of using PSAT for studying relatively smaller species living in the coastal waters of Korea.

Immune Response of Broiler Chickens Fed Diets Supplemented with Different Level of Chromium Methionine under Heat Stress Conditions

  • The objectives of this study were to investigate the immune responses of broiler chickens fed diets supplemented with different level of chromium methionine (CrMet) in heat stress (HS) condition. Two hundred and eighty eight male broiler chickens (Ross 308) were allocated to four treatment groups (supplementation with 0, 200, 400 or 800 ppb Cr in the form of CrMet) in a completely randomized design. The experiment was conducted at heat stressed condition and all birds were kept under temperature of $33{\pm}2^{\circ}C$. Antibody titers against Newcastle disease virus (NDV) and infectious bronchitis virus (IBV), heterophil to lymphocyte ratios (H/L), and concentration of plasma cortisol (CPC) were measured at 21 and 42 d. At 42 days of age two birds were chosen randomly from each replicate, slaughtered, spleen and bursa of Fabricius were collected, weighed and expressed as a percentage of live body weight. Antibody titers against NDV and IBV at 21 and 42 days of age in broiler fed supplemental CrMet were higher than in broiler chickens fed control diet (p<0.05). CPC level in broiler chickens fed CrMet were significantly (p<0.05) decreased. Increases in lymphocyte counts and consequently a decrease in heterophil to lymphocyte ratios in broiler chickens fed 800 ppb Cr were observed at 21 and 42 d. Supplementation with CrMet had no significant effect on lymphoid organs of broilers. The results suggest that dietary CrMet supplementation at a level of 800 ppb can improve some immune responses of broiler chickens under heat stress conditions.

The effects of supplementary diets on the water temperture stress in olive flounder, Paralichthys olivaceus (넙치, Paralichthys olivaceus의 수온 자극 스트레스에 대한 사료첨가제 투여 효과)

  • The effect of each dietary supplements (Undaria 5%, Undaria 10%, Obosan 0.5%, Wasabi leaf 2%, Wasabi stem 2%) on the water temperature fluctuation in juvenile olive flounder (Paralichthys olivaceus) was investigated. The response to stress was assessed in terms of effects on haematological and immunological, and resistance against Edwardsiella tarda infection. Plasma glucose and cortisol levels were significantly lower in 5% undaria and 2% wasabi leaf supplement groups than the controls after the first change of water temperature (P<0.05). The plasma lysozyme activities and the survival rates from E. tarda infection were significantly higher in the 5% undaria supplement groups than the control (P<0.05). These results suggest that the 5% undaria supplement seems to be contributable to the increased disease resistance on olive flounder.

Oxidative stress and non-specific immune responses in juvenile black sea bream, Acanthopagrus schlegelii, exposed to waterborne zinc

  • Juvenile black sea bream, Acanthopagrus schlegelii, were exposed to waterborne zinc (Zn) at concentrations of 0, 200, and $400{\mu}g/L$, at temperatures of 18 or $26^{\circ}C$ for 4 weeks. Superoxide dismutase (SOD) activities in the liver and gill of A. schlegelii significantly increased following exposure to waterborne Zn. Significant reduction in glutathione S-transferase (GST) activity in the liver and gill was observed following exposure to waterborne Zn. Glutathione (GSH) concentrations in the liver and gill also significantly decreased following exposure to waterborne Zn. Phagocytosis and lysozyme in the plasma and kidney were significantly increased following exposure to waterborne Zn. High water temperature increased alterations in the antioxidant and immune responses. The results of the present study suggest that waterborne Zn induced significant alterations in oxidative stress, increased immune responses and high temperature that trigger Zn toxicity.

Effect of Exercise Intensity on Unfolded Protein Response in Skeletal Muscle of Rat

  • Endoplasmic reticulum (ER) stress, unfolded protein response (UPR), and mitochondrial biogenesis were assessed following varying intensities of exercise training. The animals were randomly assigned to receive either low- (LIT, n=7) or high intensity training (HIT, n=7), or were assigned to a control group (n=7). Over 5 weeks, the animals in the LIT were exercised on a treadmill with a $10^{\circ}$ incline for 60 min at a speed of 20 m/min group, and in the HIT group at a speed of 34 m/min for 5 days a week. No statistically significant differences were found in the body weight, plasma triglyceride, and total cholesterol levels across the three groups, but fasting glucose and insulin levels were significantly lower in the exercise-trained groups. Additionally, no statistically significant differences were observed in the levels of PERK phosphorylation in skeletal muscles between the three groups. However, compared to the control and LIT groups, the level of BiP was lower in the HIT group. Compared to the control group, the levels of ATF4 in skeletal muscles and CHOP were significantly lower in the HIT group. The HIT group also showed increased PGC-$1{\alpha}$ mRNA expression in comparison with the control group. Furthermore, both of the trained groups showed higher levels of mitochondrial UCP3 than the control group. In summary, we found that a 5-week high-intensity exercise training routine resulted in increased mitochondrial biogenesis and decreased ER stress and apoptotic signaling in the skeletal muscle tissue of rats.

Anesthetic Effect and Physiological Response in Olive Flounder (Paralichthys olivaceus) to Clove Oil in a Simulated Transport Experiment

  • The optimum concentrations of clove oil as an anesthetic for olive flounder (Paralichthys olivaceus) and the stress response of the fish to clove oil anesthesia were determined over a range of water temperatures, and investigated in a simulated transport experiment using analysis of various water and physiological parameters. While the time for induction of anesthesia decreased significantly as both the concentration of clove oil and water temperature increased, the recovery time increased significantly (P<0.05). The plasma cortisol concentration in fish at each temperature increased significantly up to 12 h following exposure (P<0.05), then decreased to 48 h (P<0.05). The DO dissolved oxygen concentrations, pH values, and the fish respiratory frequencies decreased over 6 h following exposure to clove oil in all experimental groups (P<0.05), whereas the $NH_4{^+}$ and $CO_2$ concentrations in all experimental groups increased up to 6 h (P<0.05). The pH values and DO concentrations increased with increasing clove oil concentration (P<0.05) in the 6 h following exposure, and the $CO_2$ and $NH_4{^+}$ concentrations and the respiratory frequencies decreased with increasing clove oil concentration (P<0.05). The results of this experiment suggest that clove oil reduced the metabolic activity of olive flounder, thus reducing $NH_4{^+}$ excretion and $O_2$ consumption. In conclusion, clove oil appears to be a cost-effective and efficient anesthetic that is safe for use and non-toxic to the fish and users. Its use provides the potential for improved transportation of olive flounder.

Anesthetic and Physiological Effects of Clove oil and Lidocaine-HCl on the Grass Puffer, Takifugu niphobles

  • The aim of this study was to determine the physiological response and the applicable concentration ranges of anesthetic clove oil and anesthetic lidocaine-HCl, and to investigate the synergistic effect of a mixture of these two anesthetics on the in grass puffer (Takifugu niphobles). The anesthesia times decreased and the recovery times increased with increasing concentrations of clove oil and lidocaine-HCl. Applicable concentration ranges for long-term transportation requiring more than 1 hour were 2 ppm for clove oil and 50 ppm for lidocaine-HCl. With mixtures of the two anesthetics, the anesthesia time decreased as the admixture concentration of clove oil and lidocaine-HCl increased. Anesthesia times of experimental groups with the combined anesthetics were shorter than those with the same concentrations of clove oil or lidocaine-HCl alone. Plasma cortisol concentrations were highest at 6 hours in all experimental groups anesthetized with the mixture of clove oil and lidocaine-HCl, while all groups with clove oil or lidocaine-HCl alone had the highest plasma cortisol concentrations at 12 hours. Plasma glucose concentrations were highest at 12 hours in experimental groups anesthetized with the mixture of clove oil and lidocaine-HCl, while groups with clove oil or lidocaine-HCl alone had the highest plasma glucose at 24 hours. The results of this study provide basic information about anesthetics and the synergistic effect of mixtures of anesthetics in this fish species. This information should be useful for aquaculturists who require methods for safe and easy fish handling, and for transporters who require that minimal stress is imposed on fish during transport.

Combined effects of copper and temperature on Hematological constituents in the Rock fish, Sebastes schlegeli (조피볼락, Sebastes schlegeli의 혈액학적 성분변화에 미치는 구리 및 온도의 복합적 영향)

  • Copper ($CuSO_4$) has been widely used to control algae and pathogens in fish culture ponds. However, its toxic effects on fish depend not only on its concentration in water but also on water quality. The susceptibility of the rockfish, Sebastes schlegeli to copper was evaluated at three water-temperatures (WT; 18, 23 and $28^{\circ}C$) for 4 days. After the exposure of two copper concentrations (100 and $200{\mu}g/L$), a hematological effect was exerted on rockfish, by causing changes in red blood cell count and hematocrit value at $28^{\circ}C$. Total protein levels of the fish showed a tendency of co-increase with glucose depend on the WT, after copper exposure. However, the plasma calcium and magnesium levels were significantly increased at $200{\mu}g/L$ copper, regardless of the WT. Enzymes activities including ALT and LDH in serum were also significantly increased depend upon the copper treatment only. This indicates that inorganic components and enzymes activities were sensitive indexes to stress by toxicant such as copper. The cortisol levels were significantly elevated by both WT rising and copper treatment in serum of rock fish. In conclusion, these changes can be seen as an initial response to temperature stress and as a sustaining response to copper exposure. The present findings suggest that a simultaneous stress by temperature change and copper exposure could accelerate an alteration of hematological and plasma biological parameters in the rockfish.

Profiles of Glucocorticoid Receptor mRNA Expression and Physiological Changes in Response to Osmotic and Thermal Stress Conditions in Black Porgy (Acanthopagrus schlegeli) (염분과 수온 스트레스에 따른 감성돔의 glucocorticoid receptor mRNA 발현 특징과 생리적 변화에 관한 연구)

  • The present study investigated the expression of glucocorticoid receptor (GR) mRNA as a stress response during salinity changes (35, 10, and 0 psu) and water temperature changes (from $20^{\circ}C$ to $30^{\circ}C$, $1^{\circ}C$/day) in black porgy. We cloned the full-length GR cDNA from the kidney and examined its expression in the gill, kidney, and intestine by quantitative real-time PCR (QPCR) during salinity and water temperature changes. During salinity changes, the levels of GR mRNA in the gill, kidney, and intestine were highest at 0 psu, and the levels of plasma cortisol and glucose were elevated, but triiodothyronine ($T_3$) decreased. Also, during water temperature changes, the levels of GR mRNA in the gill, kidney, and intestine increased at $30^{\circ}C$. Plasma parameters also increased with an increase in water temperature. Therefore, this upregulation of GR mRNA was a good indicator of stress, such as those resulting from changes in salinity and water temperature.
