Objective: Heat stress poses an increasing threat for poultry production. Some amino acids have been found to play critical roles in affording thermotolerance. Recently, it was found that in ovo administration of L-leucine (L-Leu) altered amino acid metabolism and afforded thermotolerance in heat-exposed broiler chicks. Methods: In this study, two doses (35 and $70{\mu}mol/egg$) of L-Leu were administered in ovo on embryonic day 7 to determine their effect on rectal temperature (RT), body weight (BW) and thyroid hormones at hatching. Changes in RT, BW, and thermotolerance in post-hatched chicks were also analyzed. Results: It was found that in ovo administration of L-Leu dose-dependently reduced RT and plasma thyroxine ($T_4$) level just after hatching. In post-hatched neonatal broiler chicks, however, the higher dose of L-Leu administered in ovo significantly increased RT without affecting BW gain. In chicks that had been exposed to heat stress, the RT was significantly lowered by in ovo administration of L-Leu (high dose) in comparison with the control chicks under the same high ambient temperature (HT: $35^{\circ}C{\pm}1^{\circ}C$, 120 min). Conclusion: In ovo administration of L-Leu in a high dose contributed to an increased daily body temperature and afforded thermotolerance under HT in neonatal broiler chicks.
Background: It is known that large dogs who are fed lamb and rice diets are at increased risk to develop taurine-deficiency-induced dilated cardiomyopathy. Since dogs obligatorily conjugate bile acids (BA) with taurine, we determined whether rice bran (RB) or other fibers (cellulose; CL, beet pulp; BP) would affect BA excretion and/or the taurine status of dogs. Results: Eighteen medium/large mixed-breed dogs were given purified diets containing CL, BP, or RB for 12 weeks. Taurine concentrations in plasma and whole blood were significantly decreased at week 12. The BP group, compared to the CL or RB groups, showed significantly lower taurine concentrations in plasma ($6.5{\pm}0.5$ vs $20.4{\pm}3.9$ and $13.1{\pm}2.0{\mu}mol/L$, respectively, P < 0.01, $mean{\pm}SEM$) and in whole blood ($79{\pm}10$ vs $143{\pm}14$ and $127{\pm}14{\mu}mol/L$, respectively, P < 0.01), lower apparent protein digestibility ($81.9{\pm}0.6$ vs $88.8{\pm}0.6$ and $88.1{\pm}1.2%$, respectively, P < 0.01), and higher BA excretions ($5.6{\pm}0.1$ vs $3.4{\pm}0.5$ and $3.4{\pm}0.4{\mu}mol/g$ feces, respectively, P < 0.05) at week 12. Conclusions: These results do not support the hypothesis that RB is likely to be a primary cause of lamb meal and rice diets, increasing the risk of taurine deficiency in large dogs. However these indicate that BP may contribute to a decrease taurine status in dogs by increasing excretion of fecal BA and decreasing protein digestibility, thus decreasing the bioavailability of sulfur amino acids, the precursors of taurine.
Background: Heat-shock protein70 (HSP70) are intracellular protein chaperones, with emerging evidence of their association with various diseases. We have previously reported significantly elevated plasma-HSP70 (pHSP70) in pancreatic cancer. Current methods of pHSP70 isolation are ELISA-based which lack specificity due to cross-reactivity by similarities in the amino-acid sequence in regions of the protein backbone resulting in overestimated HSP70 value. Materials and Methods: This study was undertaken to develop a methodology to capture all isoforms of pHSP70, while further defining their tyrosine and serine phosphorylation status. Results: The methodology included gel electrophoresis on centrifuged supernatant obtained from plasma incubated with HSP70 antibody-coupled beads. After blocking non-specific binding sites, blots were immunostained with monoclonal-antibody specific for human-HSP70, phosphoserine and phosphotyrosine. Conclusions: Our novel immunocapture approach has distinct advantages over the commercially available methods of pHSP70 quantification by allowing isolation of molecular aggregates of HSP70 with additional ability to precisely distinguish phosphorylation state of HSP70 molecules at serine and tyrosine residues.
This study was designed to test the effects of dietary distillers dried grain (DDG) level on the growth performance, feed utilization, body composition and antioxidant activity of juvenile red seabream (Pagrus major). Six isonitrogenous and isocaloric diets were formulated to contain 0%, 5%, 10%, 15%, 20%, and 25% DDG from rice (designated as DDG0, DDG5, DDG10, DDG15, DDG20, and DDG25), respectively. Juvenile red seabream averaging $10.1{\pm}0.05g$ were randomly distributed into 400-L tanks in a flow through systems. Three replicate groups of fish were fed one of the experimental diets to visual satiation two times a day for 10 weeks. Survival, weight gain, feed efficiency, protein efficiency ratio and hepatosomatic index of fish were not affected by dietary DDG levels (p>0.05). Proximate and amino acid composition of whole body in juvenile red seabream were not affected by dietary DDG levels (p>0.05). Plasma content of total protein, glucose, cholesterol, glutamic-pyruvic transaminase, phospholipid and triglyceride were not affected by dietary DDG levels (p>0.05). 1, 1-Diphenyl-2-picryl-hydrazyl radical and alkyl radical scavenging activities in plasma and liver of fish were not affected by dietary DDG levels (p>0.05). The results of this experiment suggest that DDG has the potential to replace plant origin ingredients such as wheat flour and corn gluten meal and could be used up to 25% in diet without incurring negative effects on the growth performance of juvenile red seabream.
A 9-week feeding experiment was designed to test the effects of the dietary inclusion of distillers dried grain (DDG) on the growth performance, body composition, and antioxidant activity of juvenile rockfish Sebastes schlegeli. Five isonitrogenous and isocaloric diets were formulated to contain 0% DDG (DDG0), 7% and 14% DDG from rice (diets DDG-R7 and DDG-R14, respectively), as well as 7% and 14% DDG from rice and wheat flour (diets DDG-RW7 and DDG-RW14, respectively). Three replicate groups of juvenile rockfish averaging $68.0{\pm}0.4g$ were fed one of the diets to apparent satiation twice a day. Survival, weight gain, feed efficiency, and the protein efficiency ratio of fish were not affected by dietary DDG (P > 0.05). Proximate and amino acid compositions of the whole body were not affected by dietary DDG (P > 0.05). Plasma total protein, glucose, total cholesterol, glutamate oxaloacetate transaminase, glutamate pyruvate transaminase, phospholipid, and triglyceride were not affected by dietary DDG (P > 0.05). Radical scavenging activity on 1,1-diphenyl-2-picryl-hydrazyl (DPPH) in the plasma of rockfish fed the DDG-RW14 diet was higher than that of fish fed the DDG0 diet (P < 0.05). Hydroxyl radical scavenging activity in the liver of rockfish fed diets containing DDG was higher than that of fish fed the DDG0 diet (P < 0.05). Alkyl radical scavenging activity in the liver of rockfish fed the DDG-R7 diet was higher than that of fish fed the DDG0 diet (P < 0.05). These results suggest that DDG is a suitable ingredient for the partial replacement of wheat flour and can be used at a rate of up to 14% in the diet without incurring negative effects on the growth performance or body chemical composition of juvenile rockfish.

Purpose: MAT-I/III deficiency by MAT1A gene mutation causes isolated hypermethioninemia, which is considered to be a clinically benign disease. But in some patients, mental retardation, developmental delay, myelination disorder may be shown. This study was performed to find out the clinical manifestations and genetic characteristics of patients with isolated hypermethioninemia. Methods: Clinical, biochemical and genetic analysis were done to 10 patients with isolated hypermethioninemia who were referred to department of pediatrics, Soonchunhyang University Hospital from March 1999 to March 2012. Results: At first visit, all patients' mean plasma methionine level was 5.5 mg/dL (2.1-14.6) and there were no increase of amino acid levels including homocystine in all patients. Serum homocysteine level was evaluated in seven patients who visited after year 2003, and ranged from 4.96 to $11.15{\mu}mol/L$ (normal < $25{\mu}mol/L$). Methionine restricted diet was started to all patients. Nine patients who managed regularly showed normal development, but one patient whose initial plasma methionine level was 14.6 mg/dL showed language delay at 1 year of age and was diagnosed as mild mental retardation (IQ=66) at 6 years of age. Genetic analysis was done to eight patients, R264H mutation was identified in seven patients. Also, both R299C and R356Q mutation were identified in one patient. Conclusion: Clinical findings in patients with isolated hypermethioninemia were generally good, but one patient showed mental retardation and language difficulty. R264H mutation which usually inherits as an autosomal dominant trait was most frequently found in our patients, and R299C/R356Q mutation were also identified.
The present study was designed to investigate the hypoglycemic activity and lipid metabolism of ethanol (EtOH) extract from the aleurone layer of anthocyanin-pigmented (AP) rice in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing 210~240 g were divided into 4 groups, normal, diabetic control, and two experimental groups, and diabetes in rats was induced by injection of STZ (45 mg/kg, body weight) into tail vein. The EtOH extract of the powdered aleurone layer of AP rice was administered orally in diabetic rats for 14 days. In order to find the hypoglycemic effects in the animal model, the body weight, plasma glucose levels, cholesterol, HDL-cholesterol, triglyceride (TG), free fatty acid (FFA), aspartate aminotransferase (AST) and alanine amino- transferase activities (ALT) were determined. Oral administration of 1.0 81kg on the EtOH extract for 14 days resulted in a significant reduction in blood glucose, ALT, TG and FFA. However, in the case of 2.0 g/kg, the hypo-glycemic effects were not considerable. This results suggest that the EtOH extract might induce hypoglycemic effects in STZ-induced diabetic rats due to some photochemical components in the aleurone layer of AP rice.

Caseinomacropeptide (CMP) is a glycopeptide of 64 amino acid residues derived from the C-terminal of mammalian milk K-casein. Recently, human CMP (hCMP) was produced from the recombinant yeast Saccharomyces cerevisiae. In this study, the antiobesity activity of the recombinant hCMP (rhCMP) was investigated in vivo using Sprague-Dawley rats. The rhCMP did not affect the rats fed with a normal fat diet (fat content, $5.0\%$), but decreased the body weight gain of the rats fed with a high fat diet (fat content, $20\%$) by up to $19\%$. Autopsies revealed that the weights of the liver, kidney and adipose tissues decreased when the rats were given the rhCMP, which also reduced the lipid concentrations in the plasma and liver, but enhanced the fecal excretion of lipids. These results suggest that rhCMP prevent the accumulation of lipid by stimulating its fecal excretion, so could be used as a food supplement to alleviate the obesity problem caused by a high fat diet.

Growth hormone-releasing peptide-2 (GHRP-2, also named KP102) is a new hexapeptide of a series of synthetic growth hormone-releasing peptides (GHRPs) which stimulates the secretion of growth hormone (GH) in vitro and in vivo in several species including calf, sheep and pig. The GH-releasing activity of GHRP-2 is two to three times more effective than that of the original GHRP-6, and GHRP-1 in the rats and humans. To date, GHRP-2 seems to be the most potent member of the family of GHRPs. Since the GHRPs are short peptides (5-7 amino acid residues), they are synthesized easily and are not as readily degraded in plasma as GHreleasing hormone (GHRH). These features ameliorate their potential on domestic animals because of their chemical nature the GHRPs are efficacious when administered i.v. orally or orally. However, studies in cow, pig and sheep do not indicate such a close relationship between GHRH, somatostatin (SS) and GH, calling into question the general applicability of the human and rat models. Perhaps there is an important role for an endogenous GHRP in the regulation of GH secretion in domestic animals. This review provides an overview on the current knowledge of physiological role of GHRP-2 in domestic animals.
The purpose of this study was to establish the selection of indicators for estimating and factors affecting the requirement of vitamin B6. There has been a need to establish the human requirements of vitamin $B_6$ since vitamin $B_6$ is thought to be involved in more than one hundred biochemical reactions as a coenzyme in the metabolism of amino acids, glucose, and lipid, and the synthesis of neurotransmitters. For the review of the literature, this study included from early findings of the sixties to studies of 2009. This study suggests that plasma pyridoxal 5' phosphate (PLP) is the best single indicator of vitamin $B_6$ status for the healthy but not for the non-healthy. Erythrocyte aspartate aminotransferase and alanine aminotransferase activation by PLP as an indirect measure and urinary 4-pyridoxic acid excretion as a direct measure are useful as supporting indicators. Bioavailability, nutrient interaction, physiological need, and chronic diseases may increase the requirement for vitamin $B_6$. However, these effects can not be quantified due to insufficient evidences.
