Thirty Holstein calves were used to determine effects of age, environment and sex on blood metabolite concentrations during 1 to 90 d of age. Calves were weaned at 75 d of age. Environmental effects are grouped by the difference in month at birth and site of feeding. Blood samples were obtained every 2 or 3 d. The mean metabolite concentration every 3 d was used for the statistical analysis. Dairy bodyweight gain was not affected by environmental group and sex effect. Concentrations of plasma glucose, nonesterified fatty acids (NEFA), triglyceride, total cholesterol and total ketone changed with growth. These developmental changes in metabolite levels would be caused by ruminal maturation with increment of grain intake. Levels of plasma urea nitrogen, glucose, NEFA, triglyceride and total cholesterol drastically changed during a few weeks after birth, indicating that the physiological state in calves greatly changed during that time. Effects of the environmental group and sex were significant in almost all metabolites. Temperature influenced plasma metabolite concentrations. The plasma metabolite concentrations were affected more intensely by heat stress in the infant period than in the neonatal period.
A glucose clamp technique was used to investigate the effects of non-protein energy intake on tissue responsiveness and sensitivity to insulin for glucose metabolism in intact adults male goats. Three goats were fed diets at 1.0, 1.5 and 2.0 times of ME for maintenance, each for 21 d. Crude protein intake was 1.5 times of maintenance requirement in each treatment. Tissue responsiveness and sensitivity to insulin were evaluated using a hyperinsulinemic euglycemic clamp technique with four levels of insulin infusion, beginning at 13 h after feeding. Concentrations of plasma metabolites and insulin were also measured at 3, 6 and 13 h after feeding, for evaluating effects of non-protein energy intake on the metabolic status of the animals. Increasing non-protein energy intake prevented an increase in plasma NEFA concentration at 13 h after feeding (p = 0.03). Plasma urea-nitrogen and total amino-nitrogen concentrations decreased (p<0.01) and increased (p = 0.03), respectively, with increasing non-protein energy intake across time relating to feeding. Plasma insulin concentration was unaffected (p = 0.43) by non-protein energy intake regardless of time relating to feeding. In the glucose clamp experiment, increasing non-protein energy intake decreased numerically (p = 0.12) the plasma insulin concentration at half-maximal glucose infusion rate (insulin sensitivity), but did not affect (p = 0.60) maximal glucose infusion rate (tissue responsiveness to insulin). The present results suggest that an increase in non-protein energy intake may enhance insulin sensitivity for glucose metabolism, unlike responsiveness to insulin, in adult male goats. The possible enhancement in insulin sensitivity may play a role in establishing anabolic status in the body, when excess energy is supplied to the body.
The objective of this study was to compare the bioefficacy of L-lysine sulfate relative to L-lysine${\cdot}$HCl for 10 to 20 kg pigs. Two experiments were conducted to determine the bioefficacy of the two sources of lysine using daily gain, feed conversion, plasma urea nitrogen and nitrogen retention as the response criteria. In experiment 1, 168 crossbred barrows ($Landrace{\times}Large$ White), weaned at $28{\pm}3$ d ($9.07{\pm}0.78$kg body weight), were allotted to one of seven dietary treatments in a $2{\times}3$ (two lysine $sources{\times}three $ lysine levels) factorial arrangement of treatments with an added negative control treatment group. The basal diet was based on corn, peanut meal and soybean meal and provided 0.67% lysine. The basal diet was supplemented with 0.1, 0.2 or 0.3% lysine equivalents supplied from either L-lysine sulfate or L-lysine${\cdot}$HCl. Each treatment was fed to six pens of pigs with four pigs per pen. The trial lasted 21 days. The relative bioefficacy value of lysine in L-lysine sulfate using daily gain, feed conversion and plasma urea nitrogen as response criteria was 1.01, 1.05 and 1.04 of the lysine in L-lysine${\cdot}$HCl, respectively. In experiment 2, 42 crossbred ($Landrace{\times}Large$ White) pigs ($16.03{\pm}1.58$ kg body weight) were housed in stainless steel metabolism cages for 10 d and fed the seven diets used in the nitrogen-balance trial. The relative bioefficacy value of L-lysine sulfate was estimated to be 0.95 as effective as L-lysine${\cdot}$HCl for nitrogen retention on an equimolar basis. The t-test analysis revealed that bioefficacy of lysine in L-lysine sulfate was not significantly different from lysine in L-lysine${\cdot}$HCl, which was set at 1.00. In conclusion, L-lysine sulfate can be used instead of L-lysine${\cdot}$HCl to fortify lysine-deficient diets fed to 10 to 20 kg pigs.
The present experiment aimed to evaluate the effect of HMBi on the production performance and metabolism in dairy cows. Thirty multiparous Holstein dairy cows under similar conditions were randomly assigned to three dietary treatments; i) Control, a basal diet; ii) T1, a basal diet plus HMBi (0 g prepartum and 18 g postpartum); and iii) T2, a basal diet plus HMBi (10 g prepartum and 18 g postpartum). Treatments were initiated 21 d before expected calving and continued through 91 d postpartum. HMBi was top-dressed onto the total mixed ration of each cow. Treatments did not affect dry matter intake, plasma urea nitrogen, peak milk yield, days to peak milk yield, nonesterified fatty acid, glutamate pyruvate transaminase, glutamic oxalaetic transaminase, milk fat content, milk protein content, milk lactose content, and milk solid non-fat content. The milk composition yields were increased by the HMBi-supplemented treatment. The T1 and T2 treatments increased the yields of 4% fat-corrected milk yield, milk fat, milk protein, and milk lactose compared with the control. Although there was no difference in the milk composition of the control and T2-treated cows, the T2-treated cows exhibited higher milk fat yield (increased by 74 g/d), lower milk urea nitrogen (reduced by 3.41%), and plasma ${\beta}$-hydroxy butyrate than the control cows. The results indicate that HMBi supplementation to diet has beneficial effects, and that there is no difference between supplementation at prepartum and starting only at parturition.

The purpose of this study was to evaluate the effect of protected fat and heat-extruded soybean meal on the lactation performance of Holstein cows. Twenty-four cows, consisting of 20 lactating cows and 4 rumen-fistulated dry cows, were randomly allocated into four groups with 5 lactating cows and 1 fistulated cow in each group. A replicated 4${\times}$4 Latin square design with four 21 day periods, including 14 days of adaptation and 7 collection days within each period was employed. The experiment was a 2${\times}$2 arrangement, with or without heat-extruded soybean meal and protected fat inclusion. The dietary treatments consisted of supplements of (a) soybean meal (the control), (b) heat-extruded soybean meal, (c) protected fat, and (d) heat-extruded soybean meal and protected fat. The results showed that there were no significant differences in feed intake, milk yield, milk protein content, milk lactose content and body weight change between the dietary treatments. However, cows supplemented with protected fat showed a significantly increased (p<0.05) milk fat yield, 3.5% FCM and total solid yield. The increase in undegradable intake protein (UIP) via heat extruded soybean meal supplement significantly decreased the urea nitrogen concentration in the blood (p<0.05). Dietary fat inclusion significantly increased the blood cholesterol concentration (p<0.01) and decreased the ruminal pH value (p<0.01). Increased dietary UIP significantly decreased the ammonia nitrogen concentration in the rumen (p<0.01), but did not significantly influence the pH and VFA molar percentage in the rumen. It appears that dietary protected fat inclusion could improve milk fat and solid content. Increased dietary undegradable intake protein through heat extruded soybean meal did not improve milk yield. But it could alleviate the adverse effect of decreased milk protein due to dietary fat supplementation. Increased UIP could also decrease the ammonia nitrogen concentration in the rumen and plasma urea nitrogen concentration in the blood.
This work was conducted to investigate dietary supplementation of oyster mushroom fruiting bodies on biochemical and histological changes in hyper and normocholesterolemic rats. Six-week old female Sprague-Dawley albino rats were divided into three groups of 10 rats each. Feeding a diet containing a 5% powder of Pleurotus ostreatus fruiting bodies to hypercholesterolemic rats reduced plasma total cholesterol, triglyceride, low-density lipoprotein (LDL), total lipid, phospholipids, and LDL/high-density lipoprotein ratio by 30.18, 52.75, 59.62, 34.15, 23.89, and 50%, respectively. Feeding oyster mushrooms also significantly reduced body weight in hypercholesterolemic rats. However, it had no adverse effects on plasma albumin, total bilirubin, direct bilirubin, creatinin, blood urea nitrogen, uric acid, glucose, total protein, calcium, sodium, potassium, chloride, inorganic phosphate, magnesium, or enzyme profiles. Feeding mushroom increased total lipid and cholesterol excretion in feces. The plasma lipoprotein fraction, separated by agarose gel electrophoresis, indicated that P. ostreatus significantly reduced plasma ${\beta}$ and pre-${\beta}$-lipoprotein but increased ${\alpha}$-lipoprotein. A histological study of hepatic cells by conventional hematoxylin-eosin and oil red O staining revealed normal findings for mushroom-fed hypercholesterolemic rats. These results suggest that a 5% P. ostreatus diet supplement provided health benefits by acting on the atherogenic lipid profile in hypercholesterolemic rats.
This experiment was performed to examine the influences of high ambient temperature on milk production, nutrient digestibility, energy and protein sufficiency ratio, and plasma metabolites concentration in lactating cows. In a $2{\times}2$ crossover design, four multiparous lactating Holstein cows were maintained in a chamber under treatment of constant moderate ($18^{\circ}C$) ambient temperature (MT) or high ($28^{\circ}C$) ambient temperatures (HT). The DMI and milk protein yield were significantly lower in HT (p<0.05). The milk yield, milk lactose yield, and milk SNF yield tended to be lower in HT (p<0.10). No statistical differences for 4% fat-corrected milk and milk fat yield were observed. Rectal temperatures were significantly higher in HT than MT (p<0.05). The apparent DM, OM, ether extract, CF, and ash digestibility did not differ between treatments. On the other hand, the apparent CP digestibility was increased significantly (p<0.05) and nitrogen free extract tended to increase (p<0.10) in HT. The sufficiency ratio of ME and DCP intake for each requirement tended to be lower in HT than in MT (p<0.10). Concentrations of total protein (TP), albumin, and urea nitrogen in plasma did not differ between treatments. Plasma 3-methylhistidine (3MH) concentration as a marker of myofibrillar protein degradation tended to be higher in HT (p<0.15). In conclusion, high ambient temperature was associated with a lower energy and protein sufficiency ratio, and decreased milk protein production, even though the body protein mobilization tended to be higher.
This study compared the effects of two levels of crude protein (CP) supplementation during a close-up dry period on the productive performance, nitrogen balance, and blood profiles of Holstein cows. Eighteen cows (8 primigravid and 10 multiparous), 21 days prior to calving, were divided into four groups and fed a diet containing a low (11.5 to 12.7% CP) or high (14.1 to 15.3% CP) level of protein. Following parturition, all cows were offered the same diet. Nitrogen balance was measured 12 d to 10 d prior to the expected calving date. Whole feces and urine were collected. The higher close-up CP did not improve milk production during the early lactation period (14 w) in multiparous cows but tended to increase production by 2 kg of milk per day in primigravid cows. There were no differences in the weight of calves, the weight of the placenta, or the incidence of calving difficulty. Nitrogen intake, urine nitrogen and nitrogen retention were increased by the higher CP diet in primigravid and multiparous cows. All groups of cows showed a positive nitrogen balance. In particular, higher nitrogen retention was observed in primigravid cows fed the higher CP diet. With the exception of plasma urea nitrogen (PUN), non of the blood profiles were influenced by the CP level. The higher close-up CP increased the PUN during the period studied in primigravid and multiparous cows. These results indicate that a CP level of 14% during a close-up dry period is optimal for primigravid cows, and 12% CP level is sufficient for multiparous cows.
Comparative effects of oyster mushrooms on plasma and fecal lipid profiles and on liver and kidney function were evaluated in hyper and normocholesterolemic rats. Feeding of hypercholesterolemic rats a 5% powder of oyster mushrooms (Pleurotus ostreatus, P. sajor-caju and P. florida) reduced the plasma total cholesterol level by 37%, 21% and 16%, respectively and reduced the triglyceride level by 45%, 24% and 14%, respectively. LDL/HDL ratio decreased by 64%, 45% and 41% for P. sajor-caju, P. ostreatus and P. florida fed rats, respectively. Mushroom feeding also reduced body weight in hypercholesterolemic rats. However, it had no adverse effect on plasma bilirubin, creatinin and urea nitrogen level. Mushroom feeding also increased the total lipid and cholesterol excretion in the feces. The present study reveals that feeding of 5% oyster mushroom powder does not have detrimental effects on the liver and kidneys rather may provide health benefits for the cardiovascular-related complication by decreasing the atherogenic lipid profiles.
Objective: This study was conducted to investigate the effects of rumen-protected biotin (RPB) on growth performance, nutrient digestibility, nitrogen utilization and plasma biochemical parameters of Liaoning cashmere goats during the cashmere fiber growing period. Methods: Sixteen 6-month-old Liaoning cashmere twin-doelings (24.8±1.20 kg) were allocated to 2 diet groups that were individually ad libitum fed 30% concentrate and 70% forage diet (dry matter [DM]) by a paired experimental design. Goats of the control group were fed the basal diet, while goats belonging to the RPB group were fed the basal diet with 10 mg RPB/d per animal. The duration of the experiment was 16 weeks with two 8-week periods. Digestibility was determined at weeks 7 and 15, and other measures were taken every four weeks. Results: Compared with the control group, the average daily gain of the RPB group increased by 10.94% (p<0.05), and the intake of neutral detergent fiber was increased (p = 0.045). There were some increasing tendencies for the intake of DM, acid detergent fiber and ether extract (p = 0.070, 0.088, and 0.070, respectively). The intake and digestibility of N tended to increase (p = 0.062 and 0.093, respectively), while the N fecal excretion percentage of N intake was decreased (p = 0.093) in the RPB compared with the control group. N retention tended to increase (p = 0.084) with the addition of adding RPB to the diet. Plasma total protein was increased (p = 0.037), whereas the urea-N concentration was decreased (p = 0.049) in the RPB diet group compared with the control diet group. The levels of propionyl-CoA carboxylase (p<0.001) and methylmalonyl-CoA (p = 0.013) were increased in the RPB group. Conclusion: Supplementation of rumen-protected biotin in the diet of cashmere goats can enhance the utilization of N and improve daily weight gain during cashmere fiber growing period.
