Three experiments were conducted to determine the effects of dietary arginine concentrations on plasma free amino acid (PAA) concentrations in rainbow trout, Oncorhynchus mykiss (Walbaum). The first experiment was conducted to determine appropriate post-prandial and food deprivation sampling times in dorsal aorta cannulated rainbow trout averaging 519${\pm}$9.5 g (mean${\pm}$SD) at $16^{\circ}C$. Blood samples were taken at 0, 2, 3, 4, 5, 6 and 24 h after feeding (0 and 24 h blood samples were taken from the same group of fish). PAA concentrations increased by 2 h post-feeding and the concentration of all essential amino acids except histidine peaked at 5 h and returned to 0 time values by 24 h. In the second experiment dorsal aorta cannulated rainbow trout averaging 528${\pm}$11.3 g (mean${\pm}$SD) were divided into 6 groups of 4 fish to study the effect of dietary arginine levels on PAA. After 24 h food deprivation, each group of fish was fed one of six L-amino acid diets containing graded levels of arginine (0.48, 1.08, 1.38, 1.68, 1.98 or 2.58%) by intubation. Blood samples were taken at 0, 5 and 24 h after feeding. Post-prandial (5 h after feeding) plasma-free arginine concentrations (PParg) showed a breakpoint at 1.03% arginine in the diet and post-absorptive (24 h after feeding) plasma free-arginine concentrations (PAarg) showed a breakpoint at 1.38% arginine. PAarg increased linearly from fish fed diets containing arginine between 0.48% and 1.38%, and the concentrations remained constant from fish fed diets containing arginine at or above 1.38%, but were all below PParg at all time points. Results of the third experiment confirm the results that PParg concentrations from fish fed arginine deficient diets were higher than PAarg (0 or 24 h values). Thus, in contrast to mammals and birds, the PParg when arginine is present in the diet as the most limiting amino acid such that it severely limits growth, increases in plasma rather than decreases.
Taurine concentrations in blood and various tissues were measured in cats fed normal trurine diet(0.15% taurine) and trurine-free diet(0% taurine) for 5 weeks and 12 weeks, respectively. Cumulative body weight gain in young cats fed normal taurine diet and taurine-free diet for the initial 5 weeks were 463$\pm$43g and 383$\pm$53g respectively. Taurine concentration was one to two orders magnitude higher in most tissues than in plasma, among which the liver taurine-free diet for 12 weeks resulted in 98% reduction in plasma taurine concentration, 93% reduction in whole blood tarurine concentration, and 40~90% reduction in tissue taurine concentrations compared to the values for normal taurine diet group. Among the tissues tested, taurine depletion was most profound in heart and kidney, while liver and intestine were relatively resistant to taurine depletion. Whole blood taruined concentration appeared to be a better index for the body taurine status of cats as in more closely reflects the rate of taurine depletion in tissues. Taurine depletion in cats significantly increased the concentrations of essential and non-essential free amino acids in liver, while free amino acid concentrations in other tissues were not affected by taurine depletion.
The plasma levels of 23 free amino acids in both the umbilical circulation (umbilical vein and artery) and the maternal circulation (antecubital vein, uterine vein and iliac artery ) of 34 pregnant women were measured at delivery by the cesarean section. Each amino acid with the exception of glutamate was found to be in higher concentration in the fetal blood and the cord plasma characterized by hyperaminoacidemia with a marked increase of the lysine and ornithine levels. The linear relationships found between the amino acids concentrations of iliac artery and those of umbilical vein suggest three transport groups across the placental membrane One group consists of neutral amino acids whose slopes are equal to one and the other two groups are characterized by their slopes higher and lower than unity respectivly. This division into three groups is tentatively explained by the result of a dynamic equilibrium between active transport towards the fetus and diffusion back towards the maternal circulation.

Apo E polymorphism(e2, e3, e4) was among the first reported genetic polymorphism that explained part of the normal variation in plasma cholesterol concentrations. Among 62 normolipidemic healthy females, aged 19 up to 22 years, the relative frequencies of E3/3 was 0.806(n=50), E3/2 was 0.081(n=5), E3/4 allele was 0.113(n=7), and no E2/2, E2/4 and E4/4 were found. Based on the five samples of E2 allele, five subjects were randomly selected by E3 and E4 groups for the study of effects of apo E polymorphism on the distribution of serum lipid and amino acids profiles. No differences in the anthro pometric data among apo E isomers were found, otherwise the pulsation was higher in E4 than that in the others. There were no differences in plasma total HDL , HDL3 , HDL2 & LDL cholesterol, and apo A I concentrations. However, phenotype means significantly rank E2>E3>E4 allele in average TG levels(p=0.014), and rank E4>E3>E2 in total cholesterol levels(p=0.011). Atherogenic index(AI) such as lipoproteins was significantly increased in E2 & E4 than that in E3(p=0.045). Subjects with E3/2 allele had significantly higher concentrations of glutamine, phosphoserine and taurine, while subjects with E3/4 allele showed significantly lower concentrations of arginine and am inobutyrate and elevated level of phosphoserine in plasma com pared to those of E3/3 allele. Higher level of plasma taurine in subjects with E3/2 or E3/4 allele appears to be related to the elevated level of plasma total and LDL cholesterol concentrations compared to those of E3/3 allele.
The presence of replication-competent adenovirus (RCA) subpopulations in adenoviral vector products raises a variety of safety issues for development of therapies based on gene therapy. To analyze the differing effects of adenoviral vector and RCA in vivo, we examined alterations in amino acids (AAs) using rat plasma following injection of ${\beta}$-galactosidase expressing recombinant adenovirus (designated rAdLacZ) or RCA. Plasma AAs were examined by gas chromatography-mass spectrometry. A total of 16 AAs were positively measured. In the rAdLacZ group compared to the control group, the level of aspartic acid was significantly increased (Student's t-test), while the level of glutamic acid was significantly reduced. Additionally, in the RCA group compared to the control group, the level of four AAs, valine, leucine, and isoleucine as branched-chain amino acids, and proline were significantly increased, whereas the levels of three AAs, glycine, threonine, and glutamic acid were significantly reduced. Altered plasma free AA metabolic patterns in rAdLacZ and RCA groups, compared with the control group, may explain the disturbance of AA metabolism related to viral infection.
The effect of dietary components on cholecystokinin (CCK) release into plasma was investigated in chicks by feeding a meal through a stomach tube, followed by the CCK determination with specific CCK-8 antibody. In experimental 1, the results showed that both isolated soya protein and an amino acid mixture simulating the amino acid composition of the soya protein increased the release of CCK, though to a lesser extent with a delayed response in the former, when added to a protein-free diet. Among amino acids added singly to the protein-free diet, phenylalanine was more efficient than arginine and valine, exerting a response almost identical to the complete amino acid mixture. In experimental 2 and 3, by feeding the protein diets supplemented SBTI, piasma CCK level was promptly increased and this response was in a dose dependent fashion during the measurement time, being higher at 1000 than at 100 mg/kg diet. Since the SBTI supplementation did not affect crop emptying rates significantly, it was concluded that SBTI by itself enhanced CCK release into circulation.

Nine crossbred female pigs fitted with the bladder catheters were used to investigate the effects of dietary protein form on the efficiency of absorbed nitrogen for nitrogen retention in growing pigs. Combinations of the main protein sources were corn-soybean meal (CSM; slow + slow absorption rate form), corn-hydrolyzed casein (CAS; slow + rapid absorption rate form) and corn-porcine plasma (CPL; slow + intermediate absorption rate form). All experimental diets were formulated to be isonitrogenous (CP 11%) and isocaloric (3.5 Mcal/kg) and synthetic amino acids were added to the diet as required to maintain an equivalent amino acid profile among diets. Fecal digestibility of nitrogen was not different among treatments (p > 0.10). Ingested nitrogen was absorbed with an apparent efficiency of 82% to 84%. Mean nitrogen retention in pigs fed the CSM diet was as high as for pigs fed the CPL diet (0.74 g N/kg $BW^{0.75}$ per d), which was higher than the N retention rate in pigs fed CAS diet (0.68 g/kg $BW^{0.75}$ per d; P < 0.05). Apparent biological values (ABV = 100 ${\times}$ N retention/absorbed nitrogen) were 63.3%, 58.0% and 61.6% for CSM, CAS, and CPL groups, respectively (p < 0.05). There was no difference in mean energy digestibility among treatments. The efficiency of absorbed lysine utilization was significantly different among treatments (p < 0.05). Pigs fed the CAS diet were inferior to counterparts on the other diets in utilizing absorbed lysine. The ratios of free (and small peptide-bound) to protein-bound amino acids in CSM diet differed considerably from the CAS diet. This may affect the efficiency of amino acids utilization for nitrogen retention if hydrolyzed and intact amino acid pools reach the blood at different times.

The serum lipid-lowering effect of garlic was examined in hypercholesterolemic rats. The food intake was not significantly different by garlic powder supplementation. The inclusion of 1% garlic powder in a diets containing 1% cholesterol brought a reduction in the serum cholesterol and triglyceride. Liver lipids were not affected by the addition of l% garlic Powder in hypercholesterolemic rats. Earlier studies have suggested that the effect of garlic on serum cholesterol may be related to the sulfur-containing amino acids of products released during protein digestion. However, our results in rats fed garlic powder diet showed no specific effect on plasma free amino acid concentrations. In conclusion. this study clears shows that garlic intake is able to reduce the levels of serum cholesterol in hypercholesteroolemic rats. However, further studies have to be done in order to elucidate the mechanism underlying this phenomenon.

Effects of animo acids infusion into the abomasum on plasma growth hormone (GH) concentration were investigated using three wethers of 54 kg of average body weight. Wethers were infused with either 3.25 mmol/kg BW/day of sodium chloride solution (control), 3 mmol/kg BW/day of alanine (Ala), or 3 mmol/kg BW/day of aspartic acid (Asp) continuously for five days through an abomasum cathether in a $3{\times}3$ Latin square desing. On the day of starting infusion (day 0) and day 4 blood samples were collected from a jugular vein every fifteen minutes for six hours after feeding, and their GH concentrations were measured. Blood samples were also collected immediately before starting infusion (day 0), and before feeding of day 1, day 2 and day 4, and their plasma free amino acid concentrations were measured. In the animals infused with Ala, plasma free Ala concentration was increased by Ala infusion and it continued for four days. Plasma GH concentration of these animals increased on day 0, but this phenomenon disappeared on day 4. In the animals infused with Asp, the increase in plasma Asp concentration was observed only on day 1. Plasma GH concentration of these animals was not affected by Asp infusion. These results suggest that continuous Ala infusion stimulates GH secretion for a short period, but the effect would not last long, and that continuous Asp infusion does not affect plasma GH concentration.
A new technique combining forced-feeding and dorsal aorta cannulation was developed to monitor concentration of nutritions in the blood circulation and their metabolites in rainbow trout. To study the effect of dorsal aorta cannulation on stress, 30 rainbow trout (523$\pm$5.4 g; Mean$\pm$SD) were divided into 6 groups of 5 individuals each. A group was anesthetized and blood samples were taken at 0, 3, 6, 12, 24 or 48 h after dorsal aorta cannulation. Hematocrit peaked at 6 h and returned to 0 values by 12 h after dorsal aorta cannulation. Plasma cortisol and glucose concentrations also peaked at 6 h and returned to 0 values by 48 h after dorsal aorta cannulation. Based on the plasma cortisol and glucose concentrations, the rainbow trout recovered from the operation of dorsal aorta cannulation within 48 h. To compare the patterns of plasma free amino acid concentrations after force-feeding in the fish with dorsal aorta cannulation, 5 dorsal aorta cannulated individuals (511$\pm$6.2 g) were kept in a cage. After 48 h starvation, they were anesthetized and blood samples were taken at 0, 4, 8, 12, 24, 36 or 48 h after forced-feeding. The concentration of all plasma free amino acids, except isoleucine, leucine, phenylalanine, and tryptophan, also peaked at 4 h and returned to 0 values by 24 h after feeding. The combined technique allows forced-feeding and repeated sampling of blood in rainbow trout with minimum stress.
