Low Nourishment of Vitamin C Induces Glutathione Depletion and Oxidative Stress in Healthy Young Adults

  • The present study was conducted to assess the status of vitamin C among healthy young adults in relation to serum antioxidant parameters [glutathione (GSH), thiols, and total antioxidant capacity, (TAC)], and oxidative stress markers [malondialdehyde (MDA), and nitrites plus nitrates (NN)]. A prospective study included 200 young adults, and their dietary intake was assessed by using food diaries. Fasting plasma vitamin C, serum levels of GSH, thiols, TAC, MDA, and NN were measured using biochemical assays. It was observed that 38% of the enrolled subjects, n=76, had an adequate dietary intake of vitamin C (ADI group). Meanwhile, 62%, n=124, had a low dietary intake of vitamin C (LDI group) as compared to the recommended dietary allowances. The fasting plasma level of vitamin C was significantly higher in the ADI group as compared to the LDI group. Oxidative stress in the sera of the LDI group was evidenced by depletion of GSH, low thiols levels, impairment of TAC, an elevation of MDA, and increased NN. In the ADI group, positive correlations were found between plasma vitamin C and serum antioxidant parameters (GSH, thiols, and TAC). Meanwhile, the plasma vitamin C was negatively correlated with serum MDA and NN levels. This study reveals a significant increase of oxidative stress status and reduced antioxidant capacity in sera from healthy young adults with low intake of the dietary antioxidant, vitamin C.

Effect of Partial Substitution of Dietary Spray-dried Porcine Plasma or Fishmeal with Soybean and Shrimp Protein Hydrolysate on Growth Performance, Nutrient Digestibility and Serum Biochemical Parameters of Weanling Piglets

  • The present experiment was conducted to study the effects of partial replacement of spray-dried porcine protein (SDPP) or fish meal with soybean and shrimp protein hydrolysate (SSPH) on growth performance, nutrient digestibility and serum biochemical parameters in weaned pigs. Two hundred and forty 21${\pm}$2 d old pigs ((Pietrain${\times}$Duroc)${\times}$(Landrace${\times}$Large Yorkshire)) with initial weight of 6.9${\pm}$0.5 kg were randomly allocated to five dietary treatments with six replicates per treatment and eight piglets per replicate. The control diet (T1) contained 2% SDPP and 6% fishmeal, and SDPP for experimental diets T2 and T3 was replaced with 1% and 2% SSPH, respectively, on an iso-nitrogenous basis. The fishmeal for experimental diets T4 and T5 was replaced with 1% and 2% SSPH, respectively, also on an iso-nitrogenous basis. The experimental period was 21 days. The results showed that weaned piglets fed the diets containing 1% and 2% SSPH as a replacement for SDPP had similar average daily gain (ADG), average daily feed intake (ADFI), feed/gain (F/G), diarrhea rate and serum biochemical indices e.g. blood urea nitrogen (BUN), total serum protein (TP), albumin to globulin ratio (A/G), globulin (GLO), serum glucose (GLU), and immunoglobulin G (IgG) to those fed the control diet during 0-10 d and 0-21 d of the experiment. The substitution of 1% SSPH on an iso-nitrogenous basis for fish meal appeared to be beneficial for ADG (p = 0.59) and ADFI (p = 0.23) of piglets during the overall period. The digestibility of calcium was higher (p<0.01) in pigs fed diets containing SSPH than on the control diet. Addition of 1% SSPH on an iso-nitrogenous basis for fish meal could increase the digestibilities of dry matter and energy of the diet. Dietary replacement of fish meal with 1% and 2% SSPH had no effect on the concentrations of BUN, TP, A/G, GLO, GLU, and IgG. In conclusion, dietary SDPP or fish meal could partially replace SSPH without any adverse effect on growth performance, nutrient digestibility and serum biochemical parameters in weaned piglets.

Blood biochemical parameters and organ development of brown layers fed reduced dietary protein levels in two rearing systems

  • Objective: An experiment was conducted to evaluate the effect of different levels of crude protein (CP) and two rearing systems (cage and floor), on blood parameters and digestive and reproductive organ development of brown laying hens. Methods: A total of 400 Hisex Brown laying hens between 30 and 45 weeks of age were distributed in a completely randomized design and a 2×4 factorial arrangement, with main effects including two rearing systems (cage and floor) and levels of CP (140, 150, 160, and 180 g/kg), in a total of eight treatments and five replicates of 10 birds each with initial body weight of 1,877 g (laying hen in cage) and 1,866 g (laying hens in floor). The parameters evaluated were plasma total protein, albumin, uric acid, total cholesterol, relative weights of oviduct, abdominal fat, liver, gizzard, crest and dewlap, length of small intestine and oviduct. Results: The blood parameters were similar in birds reared in cage and floor systems. The birds reared on the floor showed greater small intestine and oviduct weight (%) and lower liver and pancreas weight (%). A significant interaction was observed between factors for the relative gizzard, crest and dewlap weight, serum protein, uric acid, and total cholesterol (p<0.05). The diets with 140 g/kg CP resulted in lower serum protein and lower cholesterol in birds reared in floor system, while birds reared in cage system showed no effect of CP on both parameters. Birds reared in cage and fed with 140 and 150 g/kg CP presented lower uric acid. The group of birds reared in floor system fed 180 g/kg had greater uric acid. Conclusion: The dietary protein level can be reduced up to 140 g/kg for Hisex Brown hens (30 to 45 weeks of age) without an important effect on metabolic profile and organ development in both rearing systems.

오리를 이용한 한방식품 추출액이 흰쥐의 지질함량에 미치는 영향 (Effect of Duck Extract on Lipids in Rats)

  • The aim of the present study was to investigate the effect of duck extract on lipids. Rats in the experimental group were orally administered with duck extract for four weeks. The half of the supplementation was composed of duck meat and the other of the supplementation was composed of oriental herbs. Weight gain, diet intake, and food efficiency ratio were compared between control and experimental groups. Organ weight, lengths of small/large intestines, and the following plasma biochemical parameters were also measured: hemoglobin, hemoglobin, GOT, GPT, blood glucose concentration, lipids of plasma, liver, feces. Spleen index (weight/100 g body weight) of the rats in the experimental group was significantly higher than those in the control group due to increase in fat intake from the supplementation of duck meat. The large intestines of the rats in the experimental group was significantly larger due to the supplementation with dietary fiber derived from herbs in the extract. There were significant higher levels of plasma hematocrit, GOT, total cholesterol, and HDL-cholesterol in the rats of the experimental group. From the results, although duck meat is a source of animal food, the fat profile of duck extract, such as the ratio of polyunsaturated fatty acids to saturated fatty acids appears to be beneficial to human health. These findings suggest a possible anti-hyperlipidemic effect of duck extract. (Korean J Nutrition 36(1): 3∼8, 2003)

Antidiabetic and Hypolipidemic Effects of Collybia confluens Mycelia Produced by Submerged Culture in Streptozotocin-Diabetic Rats

  • This investigation was undertaken to study the effects of oral administration (3 weeks) of Collybia confluens mycelial powder (CCMP) produced by a submerged culture on plasma glucose and other biochemical parameters in streptozotocin (STZ)-induced diabetic rats. Antidiabetic and hypolipidemic effects were proportionally increased with the increasing concentration of the CCMP for oral administration. The CCMP, at the dose of 400 mg/kg BW, substantially reduced the plasma glucose level by as much as 33.1 % as compared to the STZ-induced diabetic rats group. It also lowered the plasma total cholesterol, triglyceride, and low density lipoprotein (LDL) cholesterol by 22.9%, 19.9%, and 37.3%, respectively. The levels of total cholesterol and triglyceride in liver were reduced to the extent of by 13.5% and 18.8%, and the activity of alanine transaminase (ALT) and aspartate transaminase (AST) was decreased by 48.8% and 37.2%, respectively, under the influence of CCMP. The general components of CCMP were found to contain 26.18% carbohydrate, 3.67% crude ash, 4.02% crude fat, 22.55% crude protein, and 43.58% dietary fiber. The amino acid composition of the CCMP was also analyzed in detail.

Gender Differences in the Body Composition, Resting Energy Expenditure, and Leptin Levels of Obese Adults

  • The objective of this study was to examine how circulating leptin concentrations and resting energy expenditures (REE) are related to body composition in obese adults, and to examine differences in these parameters according to gender. Twenty-three subjects, 6 males and 17 females, were recruited from patients with a body mass index (BMI) of greater than 27 at the Obesity Clinic of the K University Hospital. Anthropometric assessments and biochemical analyses were performed, and REEs were measured. In spite of having similar BMI values the plasma leptin levels of females (20.0$\pm$6.5 ng/ml) were significantly higher (p<.05) than those of males (14.2$\pm$6.1) ng/ml). In females, plasma leptin concentrations were found to be positively related to body weight. BMI, waist-hip ratio (WHR), fat mass (FM), body fat, and to the circumferences of forearm, waist and hip (p<.0001). However, in males, plasma leptin concentrations were positively related only to suprailiac thickness (p<.05). The higher plasma leptin levels in females compared to males may, at least partially. be explained by the females' higher subcutaneous fat mass. Plasma leptin concentrations appeared to reflect not only total fat mass but also regional fat distribution, especially in females. REE values of males (2254.3$\pm$256.2 kcal/day) were significantly higher (p<.01) than those of females (1799.1$\pm$454.7 kcal/day). REE values for females were positively related to body weight, BMI, lean body mass (LBM), FM, body fat, and to the circumferences of waist and hip (p<.05); however, REE values for males were (positively) related only to LBM (p<.05). REE values were not related to plasma leptin concentrations for either males or females, indicating that the plasma level of leptin might not be a predictor for REE value.

Evaluation of Growth, Carcass, Immune Response and Stress Parameters in Naked Neck Chicken and Their Normal Siblings under Tropical Winter and Summer Temperatures

  • The performance of naked neck and normal chicken was evaluated with respect to growth, carcass, immune, biochemical and stress parameters under winter and summer seasons to assess the suitability of naked neck birds under high temperatures in the global scenario of climate change. The growth performance was significantly ($p{\leq}0.05$) higher in naked neck chicken in the summer season. The dressing percentage was significantly ($p{\leq}0.05$) higher in naked neck birds in both winter and summer season because of reduced plumage. The thigh, giblet and feather proportion significantly ($p{\leq}0.05$) varied between naked neck and normal chickens in summer season. The humeral immune response to sheep red blood cells (SRBC), Newcastle disease vaccine (NDV) and cutaneous basophil hypersensitivity (CBH) did not show any significant differences among the chicken groups. The protein and cholesterol concentration observed was within the normal ranges. The total cholesterol levels in plasma were significantly ($p{\leq}0.05$) lower in naked neck birds in both the seasons. H:L ratio was significantly ($p{\leq}0.05$) lower in summer season indicating less stress in naked neck chicken. Basophil and eosinophil concentration was significantly ($p{\leq}0.05$) higher in normal chicken in summer. The lipid peroxidation was higher in full feathered birds under summer stress. The enzyme glutathione reductase (GR) levels were significantly higher during the summer and varied significantly ($p{\leq}0.05$) between the normal and naked neck chicken in both seasons. The results indicated that the naked neck birds performed significantly better at high ambient temperatures with respect to growth, carcass and biochemical parameters. It was concluded that the ability of the naked neck chicken to adapt to high temperatures foresees a viable option for the biological mitigation of climate change.

Evaluation of the Oral Acute Toxicity of Black Ginseng in Rats

  • We studied the acute oral toxicity of black ginseng (BG) produced by heat process in rats. Single acute BG extract doses of 0, 5, 10, and 15 g/kg dissolved in saline were administered by oral gavage and the animals were kept under observation for 14 days. The single administration of BG extract up to 15 g/kg did not produce mortality, behavioral change or abnormal clinical signs in the rats. These results indicated that the oral $LD_{50}$ of the BG extract in the rats is higher than 15 g/kg. Compared to the control group, no treatment-related biologically significant effects of BG extract were noted in the measurements of the body weight or food intake. At the end of the period, the biochemical parameters and hematological parameters were analyzed in the plasma and blood. A histopathological examination of the liver and kidney was also conducted. Only the blood nitrogen urea and potassium levels in the biochemical indices showed significant differences at 10 and 15 g/kg doses of BG extract compared to the control group. These changes were not considered to be due to the toxicity. None of the other clinical chemistry parameters were affected. Therefore, these results indicate that the BG by heat processing is virtually nontoxic.

The Effects of Propolis on Biochemical Parameters and Activity of Antioxidant Enzymes in Broilers Exposed to Lead-Induced Oxidative Stress

  • This study aimed to determine the effects of vitamin C and propolis-supplemented feeds on some blood parameters, lipid peroxidation, and activities of some antioxidant enzymes in broilers exposed to oxidative stress. 360 three-day-old broiler chicks (Ross 308) were randomly divided into four treatment groups each containing 90 animals, including six replicate groups for each treatment. The experimental groups were designated for a 3-42 days period as follows: no supplement to basal ration (Control-Group I); supplement of 500 ppm vitamin C and 200 ppm lead (as lead acetate) to basal ration (Group II); supplement of 1 g/kg propolis and 200 ppm lead (as lead acetate) to basal ration (Group III); and supplement of 200 ppm lead (as lead acetate) to basal ration (Group IV). The highest TG level (86.83 mg/dl) was observed in the lead supplemented group; however, the lowest aspartate aminotransferase (SGOT) level (90.71 IU/L) was observed in the control group (p<0.05). The addition of lead increased the plasma malondialdehyde (MDA) level (p<0.01) compared to other treatments. However, the addition of vitamin C and propolis decreased the plasma MDA level close to control levels. The highest erythrocyte superoxide dismutase (SOD) activity was observed in the lead addition group (p<0.01) while no significant differences were observed for SOD activities of the control, vitamin C +lead, and propolis+lead groups. The plasma reduced glutathione (GSH) activity of the control ($2.30{\mu}mol$/ml) was significantly lower than the lead administered group ($6.20{\mu}mol$/ml) (p<0.01); while this parameter was determined to be similar to other groups. No significant differences were observed between groups for liver GSH activity, but heart GSH activity of the control was significantly higher in comparison to other treatments (p<0.05). To obtain similar antioxidant effects, it is recommend that using propolis (1 g/kg) and vitamin C (500 mg/kg) supplementation in broiler diets may overcome the adverse effects of oxidative stress originating from dietary lead.

울트라 마라톤(100 km)에서 삼투압 농도와 전해질 대사의 생화학적 요인 변화에 대한 안전성 평가 (Safety Assessment of Osmolality Concentration and Biochemical Factors Changes in Electrolyte Metabolism during an Ultra-marathon (100 km))

  • 본 연구는 22명의 남자 아마추어 선수를 대상으로 100 km 울트라 마라톤에서의 전해질 대사 반응의 안전성을 평가하기 위해 실시하였다. 대상자의 평균 연령은 $50.91{\pm}4.77$세, $VO_2max$$48.19{\pm}6.43mL/kg/min$, 그리고 평균 경기 완주시간은 $829.64{\pm}89.31$분이었다. 혈액검사에 포함된 전해질 지표로는 칼슘, 무기인, 마그네슘, 소디움, 포타슘, 염소, 총이산화탄소, 음이온 차이, 혈장량 변화와 삼투압 농도를 경기 전, 50 km 지점, 경기 종료 직후 측정하였다. 본 연구결과 소디움 농도는 경기 전보다 50 km 지점에서 감소하였고 50 km 지점보다 100 km 지점에서 감소하였으나, 고나트륨혈증 및 저나트륨혈증은 나타나지 않았다. 100 km 울트라 마라톤 경기에서 다른 모든 전해질 지표들의 거리별 변화는 참고범위 내에서 이루어졌으며, 혈장량 변화는 나타나지 않았다. 결론적으로 아마추어 지구력 선수들의 전해질 대사에 있어서 경기중 통계적인 차이는 있었으나 임상적으로 의미 있는 삼투압 농도와 전해질의 변화는 나타나지 않았으며 안전한 경기로 나타났다.
