It has been reported that Magnoliae cortex extract has antibacterial and antimicrobial activity against pathogenic microbes and Zea Mays L. extract is effective for improving gingival tissue health. The purpose of this study was to examine the anti-inflammatory and antimicrobial effects of Zea Mays L. and Magnoliac cortex extract mixtures through experimental peridontitis induced beagle dog model. Nine beagle dogs with experimentally induced periodontitis were selected. Baseline clinical indices which includes plaque index, gingival index, probing pocket depth, clinical attachment level, gingival fluid flow rate were recorded and microbial assays were done. Magnoliac cortex and Zea Mays L., mixed at 2:l ratio in 105mg capsular dosage, were taken by 3 capsule (Group I) or 6 capsule dosages (Group II) three times a day. After 4,8,12 weeks, clinical indices were recorded. All data of clinical indices were compared through one-way ANOVA with 95% confidence level. Clinical indices of group I and II showed significantly better results than those of control group. There were no significant differences between group I and II. In conclusion, it was confirmed that mixture of Magnoliae cortex and Zea May L. (mix ratio 2:1) possessed clinical improving effects to periodontitis.
To investigate the effects of carboxyethylgermanium sesquioxide (Ge-132) on the cyclophosphamide (CY)-induced immunotoxicity, hemagglutinin titer (HA-titer), splenic IgM plaque forming cells (PFC) and contact-delayed type hypersensitivity (CDTH) were assessed in mice. Ge-132 was orally administered alone (single dose of 300, 600, 900 mg/kg b.w.) or with CY (10 or 50 mg/kg, i.p.) to mice on the 2nd day before, simultaneously with, the 2nd day after immunization. Within Ge-132 alone-treated groups, HA-titer and PFC to SRBC were significantly and dose-dependently enhanced when compared with control group. HA-titer and PFC numbers suppressed by the treatment of CY alone were significantly restored by the concomitant treatment of CY and Ge-132. Also, Ge-132 significantly decreased DNFB-induced CDTH and inhibited the CY-enhanced CDTH. These results indicate that Ge-132 may be able to increase humoral immunity and inhibit the immunotoxicity by CY.
Objectives: This purpose of this study was to examine the trends in interventional research and analyze the contents and results of intervention through a systematic literature review of the domestic research literature, suggesting the effect of an oral care program on elementary school students. Methods: A total of 262 articles were retrieved initially, and the duplicate articles were then removed, excluded after screening titles, abstracts, and full-text articles. Finally, a total of 16 papers were used in the review. Results: The study included 9 articles with the nonequivalent control group pretest-posttest design and 7 with the one-group pretest-posttest design. All articles were of non-randomized studies. The most common index of the interventional effect was dental plaque. Further, oral health behaviors and knowledge, perception, self-efficacy, and DMFT index were common indices. In the quality assessment, in the "blinding of outcome assessment" among six items of ROBANS, 12 studies showed a high risk of bias. Conclusions: In order to develop an interventional oral health care program for elementary school students and evaluate the effectiveness, a strict research design and qualitative improvement of research reports are required, and continuous research should be conducted to develop a systematic protocol.

To find antitumor components in the hot water extract of the cultured mycelia of Ganoderma lucidum, protein-bound polysaccharides were purified and fractionated (Fr. I-V) by DEAE-cellulose ion exchange column chromatography and Sepbarose CL-4B gel filtration. When a dose of 20 mg/kg/day of each was, i.p., injected into ICR mice, the inhibition ratios against the solid form of sarcoma 180 were $64.2{\sim}75.8%$. The antitumor component was examined for immunological activity. It increased the amount of superoxide anion released by induced macrophages in peritoneal cavity to 1.8 times and the count of hemolytic plaque-forming cells (PFC) was increased to 4.4 times as compared with those of the control group. It contained 68.6% polysaccharide which consisted of mannose, glucose, galactose, fucose and xylose and 5.1% protein consisting of 17 amino acids. The contents of hexosamine were 0.78%. The molecular weight of Fr. V that showed the highest antitumor activity was $5.8{\times}10^4$ dalton by Sepharose CL-4B gel filtration. It was named lucidan.

A novel recombinant baculovirus Autographa californica nuclear polyhedrosis virus (ACNPV) producing the green fluorescent polyhedra was constructed and characterized. The recombinant virus was stably produced fluorescent polyhedra in the infected cells and the morphology of the polyhedra was nearly similar to that of wild-type AcNPV. For the production of the fluorescent polyhedral the green fluorescent protein (GFP) gene was introduced under the control of polyhedrin gene promoter of AcNPV by translational fusion in the front and back of intact polyhedrin gene. The recombinant baculovirus was named as CXEP, As expected, the 93 kDa fusion protein was expressed in the CXEP-infected cells. Interestingly, however, the cells infected with CXEP also showed a 33 kDa protein band as cells infected with wild-type AcNPV. The results of Southern blot analysis and plaque assay suggested that two types of baculoviruses expressing the GFP fusion protein or only native polyhedrin were formed through homologous recombination between two polyhedrin genes in the same orientation. Thus, this system can be applied for the production of recombinant polyhedra with foreign gene product of diverse interest.
Magnetic resonance-guided focused ultrasound (MRgFUS) is an emerging new technology with considerable potential to treat various neurological diseases. With refinement of ultrasound transducer technology and integration with magnetic resonance imaging guidance, transcranial sonication of precise cerebral targets has become a therapeutic option. Intensity is a key determinant of ultrasound effects. High-intensity focused ultrasound can produce targeted lesions via thermal ablation of tissue. MRgFUS-mediated stereotactic ablation is non-invasive, incision-free, and confers immediate therapeutic effects. Since the US Food and Drug Administration approval of MRgFUS in 2016 for unilateral thalamotomy in medication-refractory essential tremor, studies on novel indications such as Parkinson's disease, psychiatric disease, and brain tumors are underway. MRgFUS is also used in the context of blood-brain barrier (BBB) opening at low intensities, in combination with intravenously-administered microbubbles. Preclinical studies show that MRgFUS-mediated BBB opening safely enhances the delivery of targeted chemotherapeutic agents to the brain and improves tumor control as well as survival. In addition, BBB opening has been shown to activate the innate immune system in animal models of Alzheimer's disease. Amyloid plaque clearance and promotion of neurogenesis in these studies suggest that MRgFUS-mediated BBB opening may be a new paradigm for neurodegenerative disease treatment in the future. Here, we review the current status of preclinical and clinical trials of MRgFUS-mediated thermal ablation and BBB opening, described their mechanisms of action, and discuss future prospects.
본 연구는 대전 양치교실사업의 2년 6개월경과 시점에서 구강병 예방효과를 추구조사연구로 보고자 하였다. 조사대상은 2008년부터 2010년 5월 현재 양치교실이 설치되어 있는 대전 동구 D초등학교 5학년 학생 70명을 실험군으로 선정하였으며 대조군으로는 지리적, 경제적 상태, 학교 규모가 비슷할 것으로 예측되며 학교장의 협조를 얻을 수 있었던 대전 동구 S초등학교 5학년 학생 70명을 선정하였다. 조사방법은 구강검사로 치면세균막검사, 치아우식증검사를 양치교실 경과 6개월 시점, 18개월 시점, 30개월 시점에 실시하였으며, 30개월 시점에서는 치은지수검사, 치은열구액 수집을 추가하여 치은열구액 내 MMP-9 을 분석하였으며, 구강보건지식도 및 행위도를 분석하였다. 1. 치아우식증 관련지수에 있어서 2년 6개월 경과함에 따른 양치교실군과 대조군의 유의한 차이가 없었다. 2. 치면세균막지수는 양치교실군이 대조군에 비해 유의하게 낮았다. 3. 치주질환 관련지수인 치은지수, 치석지수는 양치교실군과 대조군의 유의한 차이가 없었다. 4. 치은열구액 내 MMP-9의 농도에 있어서 양치교실군이 대조군에 비해 낮았으나 유의한 차이가 없었다. 5. 구강보건지식에 있어서 양치교실군이 대조군에 비해 지식상태가 높은 것으로 나타났다. 6. 구강보건행위도, 1일 잇솔질 횟수, 교내 양치도구 준비에 있어서 양치교실군과 대조군의 유의한 차이가 없었다. 결론적으로 양치교실사업에 있어 치면세균막제거의 효과는 검증할 수 있었으나 치아우식증예방, 치주질환예방 효과는 불분명하며 행위도나 양치도구 구비 여부를 보았을 때 현재 대전 D초의 양치교실사업이 효율적으로 운용되고 있지 않은 것으로 판단되기 때문에 양치교실사업의효과를 보기위해서는 양치교실 설치 뿐 아니라 적절한 운용프로그램과 운용에 대한 책임 소재, 학교측과 학부모의 관심이 있어야 할 것으로 사료되었다.
Dental plaque is a film of microorganisms on the tooth surface that plays an important part in the development of caries and periodontal diseases. Streptococcus mutans (S. mutans) is present in almost all types of dental plaque. Teeth and their supporting structure, the gums (gingiva) are subjected to infection by S. mutans that causes cavities and pyorrhea which, if left untreated, can eventually lead to gingivitis. Various chemical agents have been evaluated over the years with respect to their antimicrobial effects in the oral cavity; however, all are associated with side effects that prohibit regular long-term use. The present study was designed to investigate the effect of Aralia continentalis (Araliaceae) extracts on the growth, acid production, adhesion, and insoluble glucan synthesis of S. mutans. The methanol extract of A. continentalis showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.25, 0.5, 1, 2 and 4 mg/ml compared to the control group. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 60% at the concentration of 0.25 mg/ml and complete inhibition was observed at the concentration of 4 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan from sucrose, methanol extract of A. continentalis showed more than 10% inhibition over the concentration of 0.25 mg/ml. The synthesis of insoluble glucan was decreased in the presence of 0.25 - 4 mg/ml of the methanol extract of A. continentalis. Hence, we conclude that A. continentalis might be a candidate of anticaries agent.
생체내의 유해산소를 제거하는 superoxide dismutase (superoxide : superoxide oxidoreductase E.C.1.15.1.1) 중 세포질내에서 그 활성을 지니는 인체의 세포질 superoxide dismuta~ie (SODl) 유전자를 사람의 간 cDNA library로부터 동위원소로 표지된 oligonucleotide probe를 이용, in situ plaque hybridization 방법으로 선별 분리하여 내장균 벡터로 클로닝하였다. 이 클론은 SOD1 유전자의 5"L"TR과 3’UTR을 포함한 1.6 kb 정도의 cDNA였다 SOD1 구조유전자만을 선택적으로 분리하기 위해서 ATG를 포함하는 sense strand primer와 3’UTR 부위의 antisense strand primer를 이용하여 중합효소연쇄반응(Polymerase Chain Reaction) 방법을 써서 SOD1 구조유전자 부위만을 선택적으로 증폭시켰다. Taq DNA polymerase에 의해 증폭된 DNA를 벡터 pUCl9의 multiple cloning site (MCS) 내의 Hinc II 위치에 넣였으며 이 insert DNA를 M13 mp19으로 옮겨 dideoxy chain termination 방법으로 sequenase를 사용하여 염기서열을 결정하였다. 클론닝된 cDNA는 153개의 아미노산을 포함하고 있는 하나의 open reading frame (ORF)을 가셨다. 중합효소연쇄반응에 의해 이때 증폭된 SOD1 구조유전자를 $\lambda P_{L}$ 프로모터를 포함하고 있는 발현 벡터 pUPL에 옮긴 후 대장균에서 대량으로 발현시켰다. 이때 발현된 단백질 SOD1은 고유의 효소활성을 가지고 있었다.
본 실험에서는 S. mutans KCTC 3065을 이용하여 lactic acid를 첨가하였을 때 일어나는 스트레스 반응을 살펴보고자 하였으며 다음과 같은 결론을 얻었다. 1. 배지에 lactic acid를 농도별로 첨가하여 농도에 따른 생육저해현상을 조사한 결과, 배 지에 첨가 된 lactic acid의 함량에 비례하여 S. mutans의 성장이 완만해지며, 지수증식기일 때 lactic acid를 첨가 하였을 경우 유도기일 때 lactic acid를 첨가한 경우보다 균의 성장이 활발한 것을 알 수 있었다. 2. 대수증식기의 균주를 취하여 0, 60, 70, 80 mM 농도의 lactic acid를 처리한 후 colony의 생성으로 균주의 생존유무를 관찰한 결과, 70 mM의 농도로 lactic acid를 처 리하고 90분후부터는 colony의 밀도가 급격히 낮아지며생존에 영향을 받는 것을 관찰할 수 있었으며, lactic acid의 농도가 80 mM이상 에서는 생존이 거의 불가능한 것으로 나타났다. 3. Acid stress 동안에 지방산 조성의 변화를 관찰한 결과, $C_{18:1}$은 30.92%에서 33.89%로 증가하였으며 $C_{14:0}$은 5.02%에서 2.62%로 $C_{16:0}$은 39.16%에서 33.69%로 감소하였다. 4. Acid stress 동안에 단백질 패턴의 변화를 관찰한 결과, 약 70 kDa, 60 kDa, 45 kDa, 40 kDa 그리고 23 kDa의 단 백질 발현이 증가되는 것을 볼 수 있었다.
