Aloe in vitro culture was attempted to induce callus and regeneration ability from different explant sources onto MS medium with 0.5mg/l NAA plus 1.0mg/l BA. Anthers that no developed any callus and plant regeneration, while only four out of 274 filament explants induced calli at cut edge without regenerated plants. Twenty ovary explants regenerated four direct plantlets without via callus from the base of epidermal tissues. Regenerated plants on the root tip gave 2n=14 of chromosome numbers.
Explants of Lactuce sativa cultivar, chungchima, were co-cultivated with Agrobacterium tumefaciences LBA4404, EHA101 strains containing nptll gene and ferritin gene encoding iron storage protein from soybean for transformation. Through initial selection of regenerated explants by culturing on a kanamycin and carbenicillin containing MS medium, multiple shoots were obtained after 2 months of culture. For a complementary step of selection, putative transgenic shoots were transferred to 1/2 MS basal medium supplemented with 100 mg/L kanamycin and 500 mg/L carbenicillin. The selected shoots were tested with PCR analysis using nptll, ferritin specific primers whether ferritin gene was introduced to genome of the plants. These results confirmed that produced the specific PCR bands in the putative transgenic lines. Additionally the Northern blot showed that transcripts of ferritin gene were detected in mature leaf of the transgenic lines. These results suggest that ferritin gene be successfully integrated and transcribed in the putative transgenic lettuce plants.
This study was conducted to screen the antagonistic bacteria which inhibit the growth of plant pathogen, fusarium wilt(Fusarium oxysporum) occurred in tomato plants in greenhouse. We isolated an effective bacterial strains and investigated into the antifungal activity of the antagonistic microorganism and it’s identification. Ten bacterial strains which strongly inhibited Fusarium oxysporum were isolated from the nature, and the best antagonistic bacterial strain designated as KC175, was selected. The antagonistic strain KC175 was identified to be the genus Bacillus sp. based on the morphological and biochemical characterization. The Bacillus sp. KC175 showed 58.2% of antifungal activity against the growth of Fusarium oxysporum. By the bacterialization of the culture broth and the heat bacterialization culture filtrate of it, Bacillus sp. KC175 showed 91% and 18% of antifungal activity, respectively.

This study aims at detecting the three dimensional gripping points for the transplanting robot system to grip in the process of developing transplanting robot system, which is one of the automation systems for transplanting tissue culture. The stereo vision system equipped with two cameras has been used to detect the gripping points of the plant stem. The method for matching the plants of the image information which came from two cameras was to measure the total numbers of pixels, leaves, and the heights of the plants. The gripping points were detected near the roots after extracting the stem parts by the standard deviation of the X axis according to the Y axis. The performance test of the developed program showed that the detecting errors of the gripping points were 0∼1mm for X axis and 1∼2mm for Y & Z axis. The mean running time of the program was about 3 seconds.
Antioxidant activity of Astragalus membranaceus, Polygonatum stenophyllum, Angelica gigas, Acanthopanax sessiliflorus and Angelica pubescens extracts cultivated in Jecheon prepared with different solvents were determined by 1,1-diphenyl-2-picrylhydrzyl(DPPH) radical scarvenging ability and thiocyanate method . The highest hydroxy radical scarvenging activity was shown in Acanthopanax sessiliflorus, while the lowest was in Polygonatum stenophyllum. Angelica gigas showed strong antioxidant activity by thiocyanate method. Methanol extracts and water extracts showed higher antioxidant activities than ethanol extracts. Angelica pubescens showed the highest polyphenol contents and Acanthopanax sessiliflorus showed the highest flavonoid contents. Direct correlation between the hydroxy radical scavenging activity and polyphenol contents or flavonoid contents was established by simple regression(r>0.8) in each solvent extracts. Among medicinal plants cultivated in Jecheon, Acanthopanax sessiliflorus and Angelica pubescens showed strong antioxidant activities. These results suggest that methanol or water extracts of several medicinal plant cultivated in Jecheon could be used as natural antioxidants.
Mesophyll protoplasts derived from young leaves of Nicotiana rustica and N. tabacum cv Burley 21 were fused with the aid of polyethylene glycol(PEG). Cytological examination of protoplasts after PEG treatment revealed 12.8 % heterokaryocytes. After 7 weeks culture, the hybrid calli showing greenish white with a compact appearance were selected in contrast to parental type calli tinged with white or green color. The somatic hybrid plants were verified by morphological, biochemical and cyclological analysis. A heterosis effect for plant vigor and height was observed but the shape of leaves and flower characteristics were intermediate between N. tabacum and N. rutstica. The isozyme banding patterns for peroxidase of somatic hybrid lines were compared with the parent species. A number of isozyme bands derived from both parental species were found in the hybrids. Somatic hybrid plants have been successfully backcrossed to the parental N. tabacum particularly with somatic hybrid plants as female parents. These hybrid plants yielded small seeds, only few which were germinable.

In the present study, we have tested the effects of 21 salt marsh plants on cell proliferation of mouse immune cells (spleen and thymus) using MTT assay in culture. The methanolic extracts of six salt marsh plants (Rosa rugosa, Ixeris tamagawaensis, Artemisia capillaris, Tetragonia tetragonoides, Erigeron annus, and Glehnia littoralis) showed very powerful suppressive effects of mouse immune cell death and significant activities of cell proliferation in vitro. Especially, the methanolic extract of Rosa rugosa was found to have fifteen times compared to the control treatment, demonstrating that Rosa rugosa may have a potent stimulation effect on immune cell proliferation. These results suggest that several salt marsh plants including Rosa rugosa could be useful for further study as an immunomodulating agent.

During the past two decades, China has seen her great progress in plant biotechnology. Since the Chinese market of herb medicine is huge, while the plant resources are shrinking, particular emphasis has been placed in plant tissue and cell cultures of medicinal plants, this includes fast propagation, protoplast isolation and regeneration, cell suspension cultures and large scale fermentation. To optimize culture conditions for producing secondary compounds in vitro, various media, additives and elicitors have been tested. Successful examples of large scale culture for the secondary metabolite biosynthesis are quite limited : Lithospermum ery throrhizon and Arnebia euchroma for shikonin derivatives, Panax ginseng, P. notoginseng, P. quinquefolium for saponins, and a few other medicinal plants. Recent development of genetic transformation systems of plant cells offered a new approach to in vitro production of secondary compounds. Hairy root induction and cultures, by using Ri-plasmid, have been reported from a number of medicinal plant species, such as Artemisia annua that produces little artemisinin in normal cultured cells, and from Glycyrrhiza uralensis. In the coming five years, Chinese scientists will continue their work on large scale cell cultures of a few of selected plant species, including Taxus spp. and A. annua, for the production of secondary metabolites with medicinal interests, one or two groups of scientists will be engaged in molecular cloning of the key enzymes in plant secondary metabolism.
Eighteen endophytic fungi with different colony morphologies were isolated from the roots of Nymphoides peltata growing in the Dalsung wetland. The fungal culture filtrates of the endophytic fungi were treated to Waito-c rice seedling to evaluate their plant growth-promoting activities. Culture filtrate of Y2H0002 fungal strain promoted the growth of the Waito-c rice seedlings. This strain was identified on the basis of sequences of the partial internal transcribed spacer region and the partial beta-tubulin gene. Upon chromatographic analysis of the culture filtrate of Y2H0002 strain, the gibberellins (GAs: $GA_1$, $GA_3$, and $GA_4$) were detected and quantified. Molecular and morphological studies identified the Y2H0002 strain as belonging to Aspergillus clavatus. These results indicated that A. clavatus improves the growth of plants and produces various GAs, and may participate in the growth of plants under diverse environmental conditions.

The mechanism of the mulch film cutter assembly designed as an integral part of a rice transplanting device was developed for mulching cultivation of early season culture rice. This mulch film cutter assembly was directly attached to the transplanting device of the rotary type Japanese transplanter. The principle is that the knife cut a planting slit on the polyethylene film while the planting finger immediately plants seedling into the soil through the planting slit. Computer results implied that the knife of the mulch film cutter assembly can make appropriate planting slit on the polyethylene film as the planting finger effectively plants the seedlings into the paddy soil through the same planting slits.
