Effects of Light Emitting Diodes on Growth and Morphogenesis of in vitro Seedlings in Platycodon grandiflorum (도라지 배양묘의 생장 및 형태형성에 미치는 발광다이오우드의 효과)

  • To clarify the possibility of plant production under red, green. blue, and red+blue using light emitting diodes (LEDs) and fluorescent lamps (control), the effects of light quality on the growth and morphogenesis of in vitro seedlings in Piatycodon grandiflorum were examined. The plantlets grown under the LEDs resulted in taller plants with greater stem than fluorescent lamps. The shortest shoot length, 3.8 cm, was observed in the control and the longest one, 13.4 cm, in the red light. But the shoot length was 5.6 cm under red LED with supplemental blue(red+blue light). This results indicate that red LED may be suitable, in proper combination with other wavelengths of light. The root length under red light was significantly smaller among the treatments. The plantlets grown under red+blue light had lower shoot dry weight, higher dry matter than other lights-grown plantlets. Among the various growth parameters measered, there was an indication that leaf area was controlled by the LEDs. Leaf area of a plantlets developing under green light was about 2.4 times wider than that of plantlets grown under red LED (10.1 $\textrm{cm}^2$ in area). The dry matter rate per plantlet among the treatments was greater in plantlets grown under the red/blue LEDs in comparison with that grown under other LEDs. Chlorophyll contents in plantlets grown under the red, green, blue and red/blue LEDs were 2%, 7% 20% and 10% lower, respectively, than those in plant grown under fluorescent lamps.

Effects of Cultural Conditions on the In Vitro Propagation of Plantlets derived from Apical Meristem in Ipomoea batatas L. (고구마 정단분열조직 유래 식물체의 기내 증식에 미치는 배양조건의 영향)

  • Sweet potato is a crop vegetatively propagated by vine cuttings, an ineffective method for maintaining pathogene-free stock plants. As an alternative method, single-node cultures of virus-free plantlets derived from apical meristem in sweet potato (cv. Yulmi) was examined. Effective pH range, sugar concentration and nodal order were investigated to establish an in vitro mass propagation system with high quality virus-free stock plantlets to farmhouse. Although the plantlets grew at wide range of pH, the most effective pH of the medium was 4.8 in single-node cultures. High sugar concentration of 60∼80 g/L resulted in increased growth response in shoot length, root length, number of node, leaf area and fresh and dry weight of shoot and root, whereas reducing sugar contents below 6% was showed reduced growth response. The first node including meristem tip was the best for the rapid growth of plantlets and the other nodes also showed a very similar growth response. Uniform plantlet can be obtained massively at the same time by culture of single node except for the first node including meristem tip. In conclusion, the most effective pH range and sugar concentration of medium for the growth of plantlets via single-node cultures was 4.8, 60∼80 g/L respectively. The first node was the best for the rapid propagation of plantlets in nodal order.

Effects of Excising In Vitro-Formed Roots on Acclimatization of Micropropagated Cassava Plantlets (카사바의 미세증식에서 기내 발생 부정근의 절단이 순화에 미치는 영향)

  • The in vitro plantlets of cassava (Manihot esculenta Crantz cv. MColl 22) could be regenerated from nodal explant cultures in a liquid MS basal medium containing 0.01 mg/L zeatin for 2 weeks. The plantlets of 1.5∼2.5 cm in shoot length were transplanted to a glass bottle containing fine sand and acclimated under non-sterile conditions after excising their intact roots by: 1) prune leaving roots base of 1∼1.5 cm; 2) complete removal of roots; and 3) cutting off the rooting zone. The majority of in vitro-formed intact roots continued growth after transferred to soil, and all of the damaged roots stopped further growth. The plantlets with excised roots began to develop new roots within 7∼10 days after being transferred to a glass bottle, and a few of the pruned roots developed lateral roots from the remaining portion. Pruning and removal of in vitro roots resulted in a high survival rate (>87%), and did not significantly affect ex vitro root regeneration and acclimation, but the plantlets in which the rooting zone had been cut-off showed 73% survival rate. Pruning or removal of in vitro roots before transfer of plantlets is recommended for useful method of commercial micropropagation because of easier handling and high survival rate of plantlets.

The Photoautotrophic Culture System Promotes Photosynthesis and Growth of Somatic Embryo-derived Plantlets of Kalopanax septemlobus (독립영양방식 액체대량배양 시스템하에서 배양한 체세포배 유래 음나무 기내묘의 생장과 광합성)

  • A photoautotrophic micropropagation methodology in liquid culture medium has a number of advantages for large-scale propagation of plants. This paper describes an improved system for the mass propagation via somatic embryogenesis of the medicinal plant Kalopanax septemlobus Nakai. Somatic embryo-derived young plantlets of K. septemlobus were cultured either under heterotrophic conditions with sucrose on half-strength MS medium with $30gL^{-1}$ sucrose, under heterotrophic conditions without sucrose, or under photoautotrophic conditions (MS liquid medium without sucrose under forced aeration) for four weeks before transferring the plantlets for acclimatization. Plantlets grown under photoautotrophic conditions had more leaves, higher chlorophyll content, a higher net photosynthetic rate (NPR), and a higher survival rate. The results indicate that the photoautotrophic conditions with a forced ventilation system are effective in enhancing the growth of plantlets and the rate of net photosynthesis. The plantlets grown under photoautotrophic conditions had a high survival rate (92%) upon ex vitro transplantation. Our study shows that autotrophically produced plantlets acclimatize better and sooner upon ex vitro transplantation than conventionally cultured plants.

Effects of Cutting Time and Scions Section in Root Cuttings of Lacquer Tree (Rhus verniciflua Stokes) (옻나무 근삽시 삽목시기와 삽수의 절단 효과)

  • Effects of cutting time and scions section on shooting and plantlets growth characteristics were investigated to increase plantlets products by root cuttings in lacquer tree. Shoot induction from scion section was developed lately as it planted in earlier season. However, shooting rates were over 84% or more in cut planting on 15, 25 March and 5 April than sinee 15 April. Shoots were developed evenly as late cut planting. Generally, plant height, branch number, leaf number, stem diameter, and leaf area per plantlets was excellent in cut planting on 25 March and 5 April. Shooting rate from the root scion varied greatly by root sectionings. The rate of the 10 cm-long-scion was reached 84%, whereas only 54% in no trimmed root scion after 10 weeks of cutting. Regenerated plantlets from root scion without cut were slightly good in shape, but their height, number of branch per plantlets, and stem diameter were not significant.

Biological Management of Virulent Fusarium Species on Asparagus with Avirulent Fusarium Species In Vitro (비병원성(非病原性) Fusarium균(菌)을 이용(利用)한 아스파라거스의 병원성(病原性) Fusarium균(菌)의 생물적(生物的) 방제(防除))

  • Fusarium oxysporum was isolated most frequently, followed by F. moniliforme, and F. solani from infected asparagus plants grown in the field. In pathogenicity tests both with seedlings and plantlets, F. moniliforme showed higher virulence than Fusarium oxysporum did in general. Fusarium moniliforme showed more consistent virulence on both seedlings and plantlets than F. oxysporum did. Fusarium oxysporum showed higher virulence on plantlets than on seedlings. Fusarium solani showed very weak or no sign of virulence on seedlings and plantlets, respectively, in both tests. In protection tests with plantlets, most protection of asparagus against virulent fusarial infections occurred when challenge isolates were inoculated five or seven days after inoculation of protective fusarial species. Avirulent F. oxysporum was a more effective protective agent against infection of F. moniliforme than it was against F. oxysporum. Fusarium solani was more effective against infection of F. oxysporum than it was against F. moniliforme.

Characteristics of Plantlets Redifferentiated from F1 Hybrid between Panax ginseng and Panax quinquefolius (고려인삼과 미국삼의 종간잡종으로부터 재분화된 식물체의 특성)

  • The characteristics of plantlets redifferentiated from calli of F1 hybrid between Panax ginseng and Panax quinquefolius were investigated. Growth of plantlets redifferentiated from F1 hybrid was superior to the plants redifferentiated from Korean ginseng. Stem color of plantlets redifferentiated from F1 hybrid was more purple than that from Korean ginseng and leaf color of the former was also greener than that of the latter. Chunpoong, Yunpoong and Seonweon which are belonged to Korean ginseng showed same PCR band(A), while American ginseng showed different PCR band (B) in Internal Transcribed Spacer (ITS) region. F1 hybrid exhibited both A and B PCR band which belonged to Korean ginseng and American ginseng, respectively. F1 hybrid calli and plantlets redifferentiated from F1 hybrid calli showed same PCR band with that of F1 hybrid plant in ITS region. Therefore it was confirmed that piantlets redifferentiated from F1 hybrid exhibited genetic stability in ITS region.

Mass-production of Eleutherococcus seoulensis Seedlings Through Somatic Embryogenesis (체세포배 형성을 통한 서울오갈피(Eleutherococcus seoulensis) 묘목의 대량생산)

  • This study was conducted to establish the optimal condition for acclimatization from somatic embryos of Eleutherococcus seoulensis. Torpedo-shaped embryos of Eleutherococcus seoulensis were cultured on 1/3 MS and WPM media supplemented with $GA_3$ (3.0, 5.0 mg/L) for 4 weeks. Plentlets were transferred to 1/2 SH solid medium with 1.0 mg/L $GA_3$ and 0.2% activated charcoal for shoot and root elongation and them elongated plantlets further developed on 1/2 SH medium for 4 weeks. Developed plantlets further elongated into well-shaped leaf and root system on 1/3 SH medium under ventilation condition for 4 weeks. Plantlets grew normally on 1/3 SH basal medium, were acclimated on various soil. Survival frequency of plantlets was influenced by soil type(peatmoss+perlite, perlite, soil on Nam mountain). The highest survival rate to soil was more than 70% when plantlets were 1/3 SH medium under ventilation condition in Nam mountain soil. These results indicate that the systematic procedure of plant production in Eleutherococcus seoulensis could be practically applicable for mass propagation.

Effect of Kelpak® on the promotion of in vitro rooting in transgenic rose plantlets (Kelpak® 침지 처리에 의한 형질전환 장미 기내 식물체 발근 촉진)

  • In order to promote in vitro rooting in SOD2-transgenic rose plantlets, which were not well rooted in a rooting medium (MS medium with NAA $0.03mg{\cdot}L^{-1}$), we dipped the plantlets into liquid $Kelpak^{(R)}$ before placing them in the rooting medium. After 4 weeks, $Kelpak^{(R)}$ significantly promoted in vitro rooting in the plantlets. Therefore, $Kelpak^{(R)}$ can be used successfully to aid in the in vitro rooting of rose plantlets with roots that are not well-generated.

Efficient Propagation by Bioreactor System of Korean Native Seosanjong in Ginger

  • For the purpose of establishing an efficient propagation through airlift bioreactor system of Zingiber of officinale Rosc. Korean native Seosanjong, the effect of different factors and bioreactor on cultured plantlets were investigated. The highest number of plantlets, fresh weight per plant was obtained from explants when cultured in MS liquid medium including 0.3 mg/L NAA and 2.0 mg/L kinetin for 40 days. A 10 L bottle type bubble bioreactor, compared with 250 mL Erlenmeyer flask, was more efficient, producing 4.7 plantlets or from 1.5 to 1.6 times more than did the Erlenmeyer flask. The results demonstrate the rapid mass propagation of airlift bioreactor to produce normal ginger.
