Validation of Korean Meat Products and Processed Cheese for the Detection of GMO using p35S and tNOS Primers

  • In this study, 543 samples of press hams, sausages, processed ground meat and processed cheese acquired from retail markets in Seoul and Gyeonggi province in Korea from 2005 to 2010 were monitored using a one-step multiplex polymerase chain reaction (PCR) method that involves the amplification of specific soya or maize endogenous genes and the amplification of 35S promoter (p35S) and nopaline synthase terminator (tNOS) for GMO detection. Among the 543 samples, 477 samples were amplified for maize and/or soybean endogenous genes. Although one sausage sample collected in 2008 showed amplification of tNOS, the result was assumed to be false positive based on the results from further tests of other sausage samples of the same brand. Our results demonstrate the absence of GM soya and/or maze of livestock products in the Korean market during 2005-2010. In addition, the one-step multiplex PCR using previously constructed primer sets appears to be useful as a screening method for the detection of GMOs in processed livestock products. However, more specific methods should be established and employed to detect the event-specific GM gene for positive reaction samples by screening tests in processed livestock products.

  • Tofu has been consumed as source of protein in Asia for hundreds of years and it was first known in US and Europe by Asian immigrants during 1900s. Lately it is being spotlighted for excellent plant-based protein that has nutritional value. Tofu has long been the most widely used ingredients in Asia and it has been developed into various forms such as tofu, yuba, fried tofu, tofu sheet, fermented tofu and more according to food culture. With development of equipment, coagulant, packaging and pasteurization, now we can have advanced flavor, productivity and distribution of tofu. Tofu has been brought to customer's attention, people who prefer more health oriented, sustainable and eco-friendly food during COVID-19 pandemic season. Furthermore, this global trend is expected to be continued. In response to the trend we need more study on new texture of tofu, substitution of meat, dairy, and various commercialization of HMR in future.

소규모 식육포장처리업 HACCP 평가항목 개선 연구 (A Study on the Improvement of HACCP Evaluation Items in Small Scale Meat Packaging Plant)

  • 소규모 식육포장처리장은 일반규모 작업장에 비해 종업원의 숫자가 적기 때문에 전반적인 위생상태 개선 및 HACCP System의 적용을 위해서 일반 규모 작업장에 비해 간단히 적용할 수 있도록 해야 할 필요가 있는 것은 사실이다. 하지만 여전히 축산식품 안전사고가 많이 나타나고 있는 시점에 국민건강 안전을 위해서라도 식육포장처리장에서 많이 발생하는 문제점, 그리고 그 위해의 심각도가 커서 사회·경제적으로 파급 여파가 큰 중요 관리항목에 대해서는 규모가 작은 업소에서도 철저히 준수해야 할 필요가 있다고 판단되며, 이는 HACCP 심사에도 연계하여 그에 대한 평가를 진행할 필요가 있다고 사료된다. 본 연구결과 도출을 위해 지난 3년간의 HACCP 심사 시 지적사항 발생빈도를 분석하고, 기존 논문자료 등을 참고하여 중요도를 결정하여 새로운 소규모 HACCP 평가항목을 도출하였다. 다만, 본 연구에서 개발된 소규모 식육포장처리업 HACCP 평가항목은 통계적, 문헌적 자료를 검토하여 도출한 것으로 실제 소규모 식육포장처리장 작업 현장에서 꼭 평가가 필요한 항목인지에 대해서는 추후 현장적용을 통해 추가적인 검토가 필요할 것으로 사료되고, 현행 『식품 및 축산물 안전관리인증기준』 고시에 따른 일반규모 축산물 HACCP 평가는 업종별 특성에 맞추어 평가항목이 다르게 적용되어 있으나, 소규모의 경우에는 식육판매업 및 식용란수집 판매업을 제외한 다른 업종은 전부 동일한 평가표를 이용하여 평가를 하도록 되어 있어 해당 업종의 특성에 맞도록 평가항목을 분리하는 등의 연구도 필요할 것으로 판단된다. 아울러 인력 부족 등으로 인하여 소규모 식육포장처리장에서 쉽게 적용하기 어려운 위해분석 등 HACCP 관리 평가항목에 대해서는 정부에서 쉽게 적용할 수 있는 모델 개발 등을 통해 소규모 식육포장처리장에서 무리 없이 HACCP을 적용할 수 있도록 유도하는 것이 중요한 것으로 판단된다.

HACCP 적용을 위한 도계처리 공정내 미생물 오염의 분석 (Analysis of Microbial Contamination in Poultry Slaughtering Operations for the Application of HACCP)

  • The application of HACCP system, which was adopted by Codex Alimentarius Committee for the safe meat and poultry production, is one of the urgent task for competing in the world trade markets. But there have been no useful analytical studies to identify the causes of contamination in the poultry meat processing plants in Korea. This study was conducted to investigate the potential hazards during the operations by the microbiological examination for the poultry meat processing plant (20,000 birds capacity a day) located in Kangwon province. In spite of air contamination of work places, it may not directly affect the surface contamination of poultry meats. But the risk of Campylobacter jejuni/coli contamination was high. The number of total count was decreased about ten times, but remarkable changes of microbial contamination could not be recognized in each procedure during the operations. The washing water was already contaminated as much as $10^{3-6}CFU/ml$ in SPC before the operations. It means that to keep water tanks hygienic is a primary step to prevent the occurrences of microbial contamination. The overflow and recirculation of water in scalding, washing, and chilling was aslo an important factor for a hygienic control. Based on this study, the followings could be regarded as an important factors for hygenic control in the poultry slaughtering plants on a small scale. The temperature of water used for scalding should be constantly maintained on a required temperature, and the overflow rate of 1~1.5 liter per bird. The carcass surface and the body cavity should be washed thoroughly and the cross-contamination due to facilities, workers, and tools should be prevented. The chilling water sholud be maintained under 5$\circ$C of temperature with ice and overflow, and residual chlorine level of 50 ppm.

식육포장처리업의 HACCP 운용실태 분석 (Evaluation of HACCP system implementation in meat packaging industry)

  • The purpose of this study was to determine food safety practices and procedures based on Hazard Analysis Critical Control Point (HACCP) system and to suggest more effective method of HACCP implementation in meat packaging industry in Korea. We used the non-compliance rate of each evaluation item to compare the weak points of prerequisite requirements and HACCP. The prerequisite items related to facility, equipment, and tools showed inadequate level of requirements or unsanitary conditions for proper HACCP operation. A lack of understanding of sanitation standard operation procedures was identified as a fundamental barrier to HACCP implementation. High rate of non-compliance in HACCP items compared to prerequisite requirements signify that small businesses have potential difficulties of applying HACCP due to lack of technical expertise, financial resources for prerequisite requirements, and available personnel to prepare and operate HACCP plan. Also we suggest to revise and minimize current critical control points (CCPs). Time-temperature control of cold-storage rooms for carcasses and final products could be performed by control points of prerequisite requirements. As the occurrence frequency getting lower, metal detector should be replaced by intensified training of sanitary handling and safety procedure. This will be more effective and preventive measures against physical contaminants including metal particles. In conclusion, control point of prerequisite requirement may replace CCP in the plant with simple processing line and no heating process such as meat packaging industry.

Study on the current research trends and future agenda in animal products: an Asian perspective

  • This study aimed to analyze the leading research materials and research trends related to livestock food in Asia in recent years and propose future research agendas to ultimately contribute to the development of related livestock species. On analyzing more than 200 relevant articles, a high frequency of studies on livestock species and products with large breeding scales and vast markets was observed. Asia possesses the largest pig population and most extensive pork market, followed by that of beef, chicken, and milk; moreover, blood and egg markets have also been studied. Regarding research keywords, "meat quality" and "probiotics" were the most common, followed by "antioxidants", which have been extensively studied in the past, and "cultured meat", which has recently gained traction. The future research agenda for meat products is expected to be dominated by alternative livestock products, such as cultured and plant-derived meats; improved meat product functionality and safety; the environmental impacts of livestock farming; and animal welfare research. The future research agenda for dairy products is anticipated to include animal welfare, dairy production, probiotic-based development of high-quality functional dairy products, the development of alternative dairy products, and the advancement of lactose-free or personalized dairy products. However, determining the extent to which the various research articles' findings have been applied in real-world industry proved challenging, and research related to animal food laws and policies and consumer surveys was lacking. In addition, studies on alternatives for sustainable livestock development could not be identified. Therefore, future research may augment industrial application, and multidisciplinary research related to animal food laws and policies as well as eco-friendly livestock production should be strengthened.

Inhibitory effect of natural extract mixtures on microbial growth and lipid oxidation of sausages during storage

  • Large amounts of additives are used during meat product processing to maintain product quality and shelf life. With the growing interest in healthy foods, natural plant-based additives are being used as alternatives to synthetic additives. In this study, six types of natural extracts with excellent antibacterial activity were selected, and their antibacterial and antioxidant activities against four types of pathogens were evaluated in various combinations. In addition, the pH, color, amount of thiobarbituric acid reactive substances (TBARS), and growth of pathogenic microorganisms were analyzed during the storage of sausages treated with various combinations of these extracts. The natural extract mixtures exhibited different antibacterial activities, depending on the combination. Compared to grapefruit seed extract, a mixture of natural extracts extracted with ethanol (M4) reduced the Escherichia coli content by more than 99.9% after 8 days of storage and slowed the growth of Listeria monocytogenes and Salmonella spp. by more than 80% after 14 days. Compared to untreated (NC) and grapefruit extract (PC)-treated sausages, sausages treated with the natural extract mixtures showed a significant decrease in CIE L* and an increase in CIE a* and CIE b* (p < 0.05). The pH value was significantly lower in sausages containing natural extract mixtures than in the NC and PC sausages (p < 0.05). The natural plant extract mixtures significantly prevented lipid oxidation (p < 0.05). In summary, different types of natural extract mixtures have a synergistic effect when used together, suggesting that natural preservatives can generally inhibit the growth of microorganisms and oxidation of processed meat.

Effect of feeding a diet comprised of various corn silages inclusion with peanut vine or wheat straw on performance, digestion, serum parameters and meat nutrients in finishing beef cattle

  • Objective: The objective of this study was to compare the feeding value, meat nutrients and associative effects of a diet comprised of various corn silages inclusion with peanut vine or wheat straw in finishing beef cattle. Methods: One hundred and eighty Simmental crossbred beef steers were blocked and assigned to the follow treatments: i) whole plant corn silage-based diet (control, WPCS), ii) mixed forages-based diet (replacing a portion of corn silage with wheat straw, WPCSW), iii) corn stalklage-based diet (CS), and iv) sweet corn stalklage-based diet (SCS). Each group consisted of 5 repeated pens with 9 steers/pen. The diets were formulated to be isonitrogenous and isoenergetic with same forage to concentrate ratio. Experimental diets were fed for 90 d. Results: The effective ruminal degradability of dry matter and crude protein were highest for WPCS diet (p<0.05), for neutral detergent fiber was highest in SCS diet (p<0.05). The average daily gain was greater for cattle offered the WPCS diet, intermediate with WPCSW and SCS and lowest with CS (p<0.001). The concentration of non-esterified fatty acid in serum was higher for steers fed with CS and SCS diets than those offered WPCS and WPCSW steers (p<0.001). The treatments did not affect the general nutritional contents and amino acids composition of Longissimus dorsi of steers (p>0.05). Conclusion: The corn silage-based diet exhibited the highest feeding value. The sweet corn stalklage and wheat straw as an alternative to corn silage offered to beef cattle had limited influence on feeding value and meat nutrients. However, the value of a corn stalklage-based diet was relatively poor. To sum up, when the high quality forage resources, such as corn silage, are in short supply, or the growth rate of beef cattle decreases in the later finishing period, the sweet stalklage and wheat straw could be used as a cheaper alternative in feedlot cattle diet without sharp reducing economic benefits.

도축장에서 HACCP 시행주체의 HACCP 운용수준 및 성공적 시행에 대한 인식실태 (A Awareness Survey of HACCP Implementation in the Korean Slaughterhouses)

  • 위해요소중점관리제도(Hazard Analysis Critical Control Point : HACCP)는 세계적으로 식품의 안전성을 보증하는 가장 과학적이고 체계적인 위생관리기법으로 널리 인정되고 있고, 우리나라도 법적으로 도축장에 연차적으로 의무적으로 적용토록 하고 있다. 본 연구는 이러한 HACCP를 국내 도축장에 시행하는데 있어 기본토대를 파악함으로서 향후 효과적이고 체계적인 HACCP이 시행되는데 기여할 수 있도록 하기 위한 것이다. 이를 위하여 HACCP 시행주체인 영업자(124명),종업원(363명) 및 HACCP 담당공무원(170명)의 HACCP 시행능력 및 인식실태를 설문조사를 통하여 분석하였다. 본 연구결과, HACCP의 이해정도는 도축장 영업자가 34.7%, 종업원 21.1%, 관계공무원이 57.1% 으로 공무원이 비교적 HACCP에 대한 이해정도가 높은 것으로 나타났으며, HACCP수행의지는 영업자 93.5%, 영업자 88.9%, 공무원 93.6%으로 모두 시행의지가 있는 것으로 나타났다. 설문조사에서 나타난 도축장에서의 HACCP 수행의 가장 큰 잇점은 도축장 위생수준의 향상(54.1%), 식육판매증가(17.6%), 위생식육 생산에 대한 자긍심(9.6%), 식육수출증가(7.5%) 등으로 나타났으며, 시행시 가장 큰 장애요인으로는 도축장 경영난(33.2%), 도축장의 낮은 시설수준(27.3%), 종업원의 낮은 위생의식(19.4%), 및 HACCP의 이해부족(12.3%)으로 나타났다. 현 시점에서 도축장에서 효과적인 HACCP를 시행하기 위한 가장 시급한 조치로는 도축장의 시설수준향상(36.6%), 교육과 훈련을 통한 HACCP 기반 확대(17%), HACCP시행 도축장을 위한 특혜부여(11.8%), 그리고 정부와 산업체간의 긴밀한 협조체제 (10.3%)로 나타났다. 이번 연구결과는 HACCP를 시행하기 위한 효과적인 방법을 제시하며, 도축장 영업자에게는 그들 자신만의 효율적인 HACCP 계획을 수립할 수 있도록 도와줄 뿐만 아니라, 국내 도축장의 HACCP 시행여건을 체계적으로 분석·제공함으로서 정부등 관련업계에서 HACCP시행 정책 등을 수립하는데 있어 기초자료로 활용될 수 있다.

Radiological Safety Assessment for a Near-Surface Disposal Facility Using RESRAD-ONSITE Code

  • Radiological impact analyses were carried out for a near-surface radioactive waste repository at Gyeongju in South Korea. The RESRAD-ONSITE code was applied for the estimation of maximum exposure doses by considering various exposure pathways based on a land area of 2,500 ㎡ with a 0.15 m thick contamination zone. Typical influencing input parameters such as shield depth, shield materials' density, and shield erosion rate were examined for a sensitivity analysis. Then both residential farmer and industrial worker scenarios were used for the estimation of maximum exposure doses depending on exposure duration. The radiation dose evaluation results showed that 60Co, 137Cs, and 63Ni were major contributors to the total exposure dose compared with other radionuclides. Furthermore, the total exposure dose from ingestion (plant, meat, and milk) of the contaminated plants was more significant than those assessed for inhalation, with maximum values of 5.5×10-4 mSv·yr-1 for the plant ingestion. Thus the results of this study can be applied for determining near-surface radioactive waste repository conditions and providing quantitative analysis methods using RESRAD-ONSITE code for the safety assessment of disposing radioactive materials including decommissioning wastes to protect human health and the environment.
