In Vitro Propagation of Cindium officinale Makino Through Shoot Tip Culture (천궁의 경정배양을 통한 기내번식)

  • This experiment was conducted to identify the optimal in vitro propagation condition of Cnidii rhizoma (Cnidium officinale Makino). It was effective to reduce contamination and improve regeneration of shoot when shoot tips taken in July were cultured in 1/2 strength Murashige and Skoog medium supplemented with 500 mg/L carbenicillin disodium 1.0 mg/L BA and 1.0mg/L $GA_3$followed by surface sterilization of explant source in solution of 1% sodium hypochlorite for 20 minutes. When shoot tips were 쳐cultured in 1/2 strength MS medium with 0.5 mg/L BA and 60 g/L sucrose, shoot elogation and subsequent multiplication of the formed shoot were favorable than in other media. Regenerants were well rooted in 1/2 strength MS medium containing 3.0 mg/L NAA.

Efficient Transmission and Propagation of Tomato Chlorosis Virus by Simple Single-Leaflet Grafting

  • Tomato chlorosis virus (ToCV), a member of the genus Crinivirus, has caused an epidemic disease in tomato worldwide. ToCV is phloem-limited and transmitted by whiteflies in a semi-persistent manner, but not by mechanical inoculation. Experimental propagation of ToCV has been performed primarily by using whitefly-mediated inoculation. To develop a simple and convenient method for transmission of ToCV, we investigated grafting single-leaflets from tomato plants infected with ToCV to recipient tomato seedlings. Forty-one of 46 tomato seedlings tested were grafted successfully with single-leaflets infected with ToCV. Among them, 36 seedlings (87.8%) were systemically infected with ToCV and developed typical symptoms. Our results demonstrated that single-leaflet grafting could provide a sufficient amount of inoculum for the transmission of ToCV to the grafted seedlings.

  • To develop restoration technologies of natural environment, which is destroyed through the urbanization, industrialization, and the increase of leisure industry, the development of environmental restoration material and those application is greatly needed. Concerning this current issue, an experiment has been conducted to test and an experiment has been conducted to test and analyze the conditions of nutritonal propagation of willow cutting, which is known as a plant with rapid propagation and good growth capability under bad soil conditions and its wide ecological amplitude. 24 various kinds of willow cuttings -2~6 cuttings for each species -were placed into the earth for 57 days and measured the lengths and the diameters of willow cuttings, the possibility of root formation, the total number, and the total length of new shoots and roots. 23 among 24 willow cuttings tested were showing a great growing capacity and a number of new shoots and roots were observed in a relatively short time. This proved that salix is an adequate plant material for quick environmental restoraton . Besides, those willow cuttings could be classified for different site conditions through comparing the ratio of the underground part to the aboveground in total length. The results are as follows; S. dependens, S. matsudana, S. babylonica are the most suitable species for quick vegetaton and S. gracilistyla,S. myricoides,S. alba 'sercia' for erosioni control.

  • In this paper, The methodology of Type-2 fuzzy set-based Radial Basis Function Neural Network(T2RBFNN) is proposed for Sewage Treatment Process and the simulator is developed for application to the real-world sewage treatment plant by using the proposed model. The proposed model has robust characteristic than conventional RBFNN. architecture of network consist of three layers such as input layer, hidden layer and output layer of RBFNN, and Type-2 fuzzy set is applied to receptive field in contrast with conventional radial basis function. In addition, the connection weights of the proposed model are defined as linear polynomial function, and then are learned through Back-Propagation(BP). Type reduction is carried out by using Karnik and Mendel(KM) algorithm between hidden layer and output layer. Sewage treatment data obtained from real-world sewage treatment plant is employed to evaluate performance of the proposed model, and their results are analyzed as well as compared with those of conventional RBFNN.

IBA Treatment of Poplar Cuttings and Soil Composition Amendment for Improved Adaptability and Survival

  • Poplar trees from the Salicaceae family over the years have been utilized for various reasons which include prevention of deforestation as well as phytoremediation. This study aims to determine the optimal pre-treatment and soil conditions required for propagation of poplar cuttings for increased initial adaptability and survival rate. Five poplar clones (Hanan, 110, 107, DN-34, 52-225) were selected for IBA, soil composition treatments on propagation. IBA pre-treatment of cuttings were utilized 0, 10, and 100 mg l-1 concentrations. Soil compositions were amended with TKS-2+perlite 2:1 (v:v) and sandy clay loam mixed with artificial soil. According to the greenhouse results 10 mg l-1 of IBA showed a significant increase in plant height whereas 100 mg l-1 inhibited plant growth except in clone 110. Soil composition severely affected root growth and hence overall growth of the clones. Sandy clay loam soil had poor to stunted growth compared to TKS-2+perlite.

  • Apple stem grooving virus (ASGV) is a destructive viral pathogen of pome fruit trees that causes significant losses to fruit production worldwide. Obtaining ASGV-free propagation materials is essential to reduce economic losses, and accurate and sensitive detection methods to screen ASGV-free plantlets during in vitro propagation are urgently necessary. In this study, ASGV was sensitively and accurately quantified from in vitro propagated apple plantlets using a reverse transcription droplet digital polymerase chain reaction (RT-ddPCR) assay. The optimized RT-ddPCR assay was specific to other apple viruses, and was at least 10-times more sensitive than RT-real-time quantitative PCR assay. Furthermore, the optimized RT-ddPCR assay was validated for the detection and quantification of ASGV using micropropagated apple plantlet samples. This RT-ddPCR assay can be utilized for the accurate quantitative detection of ASGV infection in ASGV-free certification programs, and can thus contribute to the production of ASGV-free apple trees.

In Vitro Mass Propagation and Soil Adjastment of Zanthoxylum piperitum var. inerme Makino through Apical Meristem Culture (生長點 培養에 依한 민초피나무(Zanthoxylum piperitum var. inerme Makino)의 器內 大量 增殖 및 土壤 活着)

  • This study was conducted to investigate the effect of growth regulators and medium composition on the growth of each stage in apical meristem culture for mass propagation of Zanthoxylum piperitum var. inerme Makino. The source material, shoot tip segments were taken from three-years old graft trees. Apical meristems were cultured in vitro on basal MS, GD, WS, half strength MS(1/2MS) and half strength GD(1/2GD) media supplemented with various concentrations for growth regulators(BA, IBA) and inorganic nutrients. The results summarized are as follows: 1. In culture establishment stage, ratio of culture establishment was 96.7% and the best resuit was obtained using MS medium supplemented with 1.0mg/l BA and 0.2mg/l IBA. 2. In shoot multitication stage, both shoot multiplication and growth were achieved in average 5.6cm. These results were obtained on in MS medium supplemented with 1.0mg/l BA and 0.2mg/l IBA. 3. In roothing stage, phloroglucinol(PG) acted as IBA synergist in root initiation. The most faverable combinations for root development was half-strength MS medium supplemented with 162mg/l PG and 0.2mg/l IBA, and ratio of rooting was 58.0%. 4. In Vitro formed plantlets were transplanted to paper pots in greenhouse with 85% of relative humidity. 96% of survival rate was obtained from artificial soil mix having same volume of sand, vermiculite, peat, and soil.

Effect of Growth Regulator and Sterilization Method on Multiple Shoot Induction through Sucker and Stem Node Culture in Ramie(Boehmeria nivea Hoooker et Arnot) (모시풀 조직배양에서 소독방법 및 생장조절제의 Multiple Shoot 유기 효과)

  • This experiment was carried out to establish the system of mass propagation through tissue culture using sucker and stem in Ramie. The sterilization for tissue culture of Ramie was the better treatment of 2% NaClO for 20 minute into ultrasonic cleaner than the others, and so rate of contamination was 3.3%, and it was able to produce 96% healthy plant. The effect of growth regulator was superior to mixed treatment of 0.02mg/$\ell$ NAA, 1.5mg/$\ell$ BA, 0.lmgmg/$\ell$ GA$_3$, which it was not formed callus and but produced 96% healthy plant. The effect of propagation was higher in culturing of the stem node than the sucker in cultural part, local variety than improved ones. The effect of acclimatization was superior to pretreatment of 30 minute after soaking in 100ppm NAA, transplanting on bed soil which mixed to ratio of vermiculite : soil : sand =1 : 2 : 1, the transplanted plants were grown all normal.

Conditions of In Vitro Spore Germination and Prothallium Culture for Sporophyte propagation of Polystichum braunii (Spenn.) Fée (좀나도히초미(Polystichum braunii (Spenn.) Fée) 포자체 증식을 위한 기내 포자 발아와 전엽체 배양 조건)

  • This study was conducted to investigate the optimal conditions for spore germination, prothallus propagation, sporophyte formation, and seedling growth in Polystichum braunii (Spenn.) $F{\acute{e}}e$. The rate of spore germination and early prothalium development was high in Knop (41.2%), which had low mineral content. The optimal medium for prothallus propagation and sexual organ formation was 2MS medium (2% sucrose). Among the various mixtures of cultivation soil (bedding soil, peat moss, perlite, and decomposed granite), a mixture of bedding soil and decomposed granite at a ratio of 2:1 (v:v) had a positive effect on sporophyte formation (276.3 ea/$7.5m^2$). The most efficient conditions for promoting the growth of sporophytes were pots filled with only bedding soil.

Development of Refining Methods in Phragmites Communis and Imperata Cylindrica seed (갈대와 띠 종자의 정선기술 개발)

  • The efficient refinement of seed is required to reduce the cost and labor input in artificial propagation of wild plant. This study was carried out to develop methods for collecting and refining tiny seeds from wild plants. For obtaining Phragmites communis seeds, the inflorescence was cut into small fragments using a Straw Cutter and subsequently detached pappus hairs from seed coat by Hammer Mill. The primary refined seeds were passed 1.0 mm sieve. The screened seeds were subjected to Seed Blower with wind speed of 0.25 mㆍsec-1 to collected intact and well-ripen seeds. The seeds of Imperata cylindrica were refined as follows. Inflorescences were cut using a Straw Cutter first. The pappus was removed from cut fragments using a Hammer Mill and subsequently subjected to Seed Scarifier at 500rpm for 60 sec. for further separation. The separated seeds were passed 1.0 mm screen and collected after blowing with Seed Blower of wind speed of 0.15 mㆍsec-1. When the amount of seed was too little to refine with Seed Scarifier and Blower, the procedure was slightly modified from the procedure described above. The crude seed mixture obtained from Hammer Mill step was hand-refined roughly and then immersed into cone. (95%) sulfuric acid for 2 min. and collected floating portion after dilution of sulfuric acid solution 100 times with tap water. The collected seeds were dried and passed 0.149 mm sieve. During seed refining process using mechanical or sulfuric acid treatments, a small portion of damaged seed were evolved, however, the amount was not noticeable as compared to the total amount of collected seeds. Because the germination percentages between hand-refined seeds and seeds refined by above methods were not statistically different, the developed procedures for refining tiny seed of wild plants are helpful to reduce the cost and labor input in artificial propagation of two species.
