Response of Turnip to Botrytis cinerea Infection and Their Relationship with Glucosinolate Profiles (잿빛곰팡이병에 대한 순무잎의 반응과 Glucosinolate 함량변화)

  • In order to determine the response of turnip leaves to various races of Botrytis cinerea, fourteen different races were infected to the leaf surface and measured the lesion size. Based on lesion size of the pathogen on the leaves, turnip showed susceptible response to four races, moderate to seven races and resistant to three races. Four glucosinolate (GLS) compounds, such as butenyl GLS, indole-3ylmethyl GLS (I3M), 4-methoxy-indole-3-ylmethyl GLS (4MOI3M) and normal methoxy-indole-3ylmethyl GLS (NMOI3M) were identified in turnip leaves infected with Botrytis cinerea. Leaves infected with resistant races showed higher GLS contents as compared with the leaves infected with susceptible races. Contents of I3M in the leaves with resistant races were 2.5 times as high as those in uninfected leaves, whereas I3M in the leaves with susceptible races showed lower contents than those in untreated leaves. Leaves infected with resistant races showed 4MOI3M and NMOI3M contents 2.3 and 2.7 times as high as those in uninfected leaves, respectively. GLSs in the infected leaves were most abundant at 5~10 mm area from center of the lesion. However, the GLSs in 5~10 mm area were rapidly degradated at leaves with susceptible races which resulted in continuous expansion of the lesion on the leaves, whereas no degradation was obseveved at leaves with resistant races. These results suggest that accumulation and degradation of GLS compounds in turnip leaves closely related to the susceptibility and resistance of turnip leaves to Botrytis races.

Fusarium Wilt of Winter Daphne (Daphne odora Thunb.) Caused by Fusarium oxysporum

  • Severe wilt disease epidemic was found on winter daphnes (Daphne odora Thunb.) cultivated in farmers, nurseries in Suncheon, Jeonnam in 2003. Typical symptoms appeared on the leaves of winter daphne as yellowish wilts and turned brown from the lower leaves on the same plant. Severely infected leaves were defoliated, resulting in blight of stems and eventual death of the entire plant. Black decayed vascular tissues were distinctly observed in a wilted plant. Fusarium sp. was isolated from the diseased plants repeatedly and its pathogenicity was confirmed by artificial inoculation on healthy plants. The fungus was identified as Fusarium oxysporum on the basis of the morphological and cultural characteristics on potato dextrose agar and carnation leaf agar. The optimum temperature for fungal growth was around $25{\circ}C$ and the fungal growth was inhibited by metconazole, triflumizole and trifloxystrobin on potato dextrose agar. This is the first report on the wilt disease of winter daphnes caused by F.oxysporum in Korea.

Studies on Salinity and Growth of Rice at Seosan Reclaimed Land (서산 간척지의 염도와 벼의 생육에 관한 연구)

  • The effects of salinity on the growth and production of rice were studied at Seosan reclaimed land from July to October, 1995. The plant height, the number of living aleaves, dead leaves and total leaves, the number of the grains and the dry weight of the grains per individual, and the dry weight of above the ground in $25cm{\times}25cm$ quadrat were investigated on 5 plots whers were different salinity. The plant height, the number of living leaves and total leaves and the number of grains and the dry weight of grains per individual, and the dry weight of above the ground decreased as the salinity of water increase and the number of dead leaves of rice increased as the salinity of water decrease. The effect of salinity on the reproductive production is severer than the vegetative production. Because of the salinity, the growth and the production of the rice at Seosan reclaimed land are worse than the normal rice field.

  • This experiment was carrie out to investigate the amount of essential of leaves and fruits of Magnolia sieboldii, Magnolia hypoleuca, Evodia danillii male and female, Phellodendron amurense male and female and Zanthoxylum schinifolium male and emale by Karlsrube set on August, August, September and October in 1994. Essential oil yield was highest in the fruits of Zanthoxylum schinifollium leaves on Augus the amount of essential oil of Magnolia sieboldii and Magnolia hypoleuca leaves was more than the one of Evodia danillii, Phellodendron amurense and Zanthoxylum schnifolium leaves. The amount of essential oil were not diferent between male and female leaves.

Morphological characteristics of leaves and trichomes in reciprocal hybrids of Quercus dentata and Q. serrata

  • The morphological characteristics of leaves, trichomes, and acorns in 8- to 11-year-old reciprocal hybrids of Quercus dentata and Q. serrata were investigated. The reciprocal hybrids had obovate to obovateelliptic leaves that were smaller than those of Q. dentata but similar to Q. serrata in their leaf size, obtuse leaf apex, narrow-tipped leaf base, and shape of small to intermediate-sized serrations. The ray length of stellate hairs was intermediate between those of the parents, and the hairs showed dense growth. Long single hairs are present among the stellate hairs on the abaxial side of the leaves. Therefore, the hybrids of Q. dentata and Q. serrata could be identified by the size and shape of leaves, shape of serrations, and type of leaf trichomes.

[ $^{14}CO_2$ ] Assimilation and Metabolism of $^{14}C-$Assimilates in Whole Plants of Spring Barley In Relation to Adult-Plant Resistance to Powdery Mildew (흰가루병에 대해 성체식물 저항성을 지닌 봄보리에서 $^{14}CO_2$ 동화와 $^{14}C-$동화산물의 대사)

  • The effect of powdery mildew infection on the $^{14}CO_2$ assimilation and metabolism of $^{14}C-$assimilates was studied with spring barley cultivars, susceptible Peruvian and adult-plant resistant Asse at the four-leaf stage. No consistent differences between Peruvian and Asse were revealed in $^{14}CO_2$ assimilation and metabolism of $^{14}C-$assimilates in healthy whole plants. In the two cultivars, $^{14}CO_2$ assimilation and translocation of assimilates decreased as the number of infected leaves increased. Despite the same infection intensity, $^{14}CO_2$ assimilation was less inhibited in Asse than Peruvian. Infection reduced the fixation of $^{14}CO_2$ in noninfected fourth leaves of Peruvian more severely than that of Asse. Infection of the lower 3 leaves also inhibited the incorporation of 14 C into carbohydrates such as fructose and glucose in noninfected fourth leaves and their translocation into leaf sheathes, the inhibitions being greater in Peruvian than Asse. In the infected third leaves, there was a reduction of 14 C-activity in carbohydrates, more $^{14}C-$labeled fructose and glucose being retained in Peruvian. The stimulation of $^{14}C-$organic acid synthesis in all plant organs was more pronounced in Peruvian than Asse. Powdery mildew markedly increased the incorporation of $^{14}C$ into amino acids in infected third and noninfected fourth leaves, but reduced their translocation to the leaf sheathes. A greater rise of $^{14}C-$ activity in some amino acids in the two leaves was found in Peruvian than Asse.

Scopoletin Production Related to Induced Resistance of Tobacco Plants Against Tobacco mosaic virus

  • A fluorescent material was accumulated in inoculated leaves showing necrotic local lesions of tobacco plants with N gene, Nicotiana tabacum cvs. Xanthi-nc NN, Samsun NN, Burley 21 and KF 114, and N. glutinosa, and Datura stramonium at the early growth stages by the inoculation of Tobacco mosaic virus (TMV). It was identified as a coumarin phytoalexin, scopoletin. Although the material was most prominently produced in TMV-inoculated tobacco leaves with local necrotic lesions, its accumulation was also noted in uninoculated leaves of TMV-inoculated plants. Its accumulation was somewhat greater in high resistance-induced leaves than low resistance-induced and intact leaves. Scopoletin treatment induced the expression of a pathogenesis-related protein, PR-1, prominently at the concentration of 500 or 1000 ${\mu}$g/ml. This suggests that scopoletin is a phytoalexin abundantly accumulating in N gene-containing resistant plants in response to TMV infection, and may be related to hypersensitive responses (HR) and systemic acquired resistance (SAR) in the resistant tobacco plants.

Effects of DTPA on Microelements in Soybean and Bush Bean (대두 및 강낭콩내 미량원소의 농도 및 분포에 미치는 DTPA의 영향)

  • Hawkeye(Fe-chlorosis resistant) and PI 54619-5-1(Fe-chlorosis sensitive) soybeans were grown with and without DPTA(diethylene triamine pentaacetic acid) in Yolo loam soil. The major purpose of the study was to compare leaf-stem distribution of microelements for different treatments which increase concentrations of microelements in plants to evaluate the role of the chelating agent in increasing translocation of the microelements. Plant responses and yields were recorded and Fe, Mn, Zn, Cu, Al, Co, N, Sn, Pb and Mo contents of leaves and stems were determined by emission spectrography. Sulfur(soil pH4) increased leaf concentrations of Mn, Zn, Cu, CO, Ni, Sn and Pb. DTPA, particularly at 50ppm in soil, increased leaf concentrations of Fe, Mn, Zn, Cu, Co, Ni and Mo. It increased Ti in leaves for the PI 54619-5-1 soybeans only. DTPA increased the ratios of the concentration in leaves to that in stems for Fe, Zn, Cu, Al, Ti, CO, Ni and Mo. Sulfur which increased the microelement concentration in both leaves and stems did not have this effect. DTPA increased the ratio at soil pH 6 and 8.5 in leaves to that in stems of the bush bean plants for Fe, Zn, Cu, Ni, but to a lesser extent in bush beans than in soybeans. PI 54619-5-1 soybeans tended to contain less of most of the metals than did Hawkeye soybeans.

Effect of a Sudden Increase in Light Intensity on Normalized Difference Vegetation Index (NDVI) Reflected from Leaves of Tobacco (급격한 광도 변화가 담배 잎에서 반사되는 Normalized Difference Vegetation Index에 미치는 영향)

  • Normalized Difference Vegetation Index (NDVI) has played an important role in assessing green plant biomass through remote sensing on global scale since the early 1970s. The concept of NDVI is based on the fact that green plants show higher reflection in near-infrared region than in visible region of the electromagnetic spectrum. However, it is well known that the relocation of chloroplasts in plant leaf cells may dramatically change the optical properties of plant leaves. In this study I traced the changes in the reflectance and transmittance properties of Tobacco leaves at the wavelengths of 660 and 800 nm after a sudden increase in light intensity. The results showed that NDVI of leaves gradually decreased from 72.7% to 69.9% when exposed to a sudden increase in light intensity from 30 to $1,200{\mu}mol/m^2{\cdot}s$. This means that the error resulting from the physiological status of the plant should be accounted for a more precise understanding of ground truth corresponding to the data from the remotely acquired images.

GUS Expression by CaMV 35S and Rice Act1 Promoters in Transgenic Rice

  • To determine the patterns and the levels of expression of the cauliflower mosaic virus (CaMV 35S) promoter and the rice actin 1 (Act1) promoter in rice, transgenic rice plants containing CaMV 35S-$\beta$-glucuronidase (GUS) and Act1-GUS constructs were generated and examined by fluorometric and histochemical analyses. The fluorometric analysis of stably transformed calluses showed that the activity of the rice Act1 promoter was stronger than that of the CaMV 35S promoter in rice cells. In a histochemcial study of the transgenic rices, it was shown that the GUS activity directed by the CaMV 35S promoter was localized mainly in parenchymal cells of vascular tissues of leaves and roots and mesophyll cells of leaves. These results are similar to those of potato, a dicot plant. In contrast, rice plant transformed with Act1-GUS fusion construct revealed strong GUS activity in parenchymal cells of vascular tissue, mesophyll cells, epidermal cells, bulliform cells, guard subsidiary cells of leaves and most cells of the root, suggesting that the rice Act1 promoter is more constitutive than the CaMV 35S promoter. It was also confirmed that in both types of transgenic rice little or no staining was localized in metaxylen tracheary elements of vascular tissue from leaves or roots. These results indicate that the rice Act1 promoter can be utilized more successfully for expression of a variety of foreign gene in rice than the CaMV 35S promoter.
